Wilt symptoms were observed in basil (Ocimum basilicum) plants grown in a vinyl greenhouse located in Gokseong, Korea, during crop disease surveys conducted in August 2022. The symptoms appeared as wilting of the plants and brown to dark brown longitudinal streaks on the stems at or above the soil line. The disease incidence among the plants in the vinyl greenhouse was 5-20%. Six isolates of Fusarium sp. were obtained from stem lesions and identified as Fusarium oxysporum species complex based on their morphological characteristics. Among the isolates, two were used for phylogenetic analysis and pathogenicity test. Phylogenetic analysis revealed that these isolates belonged to F. oxysporum. Pathogenicity of the isolates was confirmed through artificial inoculation test. The symptoms induced by the isolates were similar to those observed in basil plants in the investigated vinyl greenhouse. This is the first report of F. oxysporum causing Fusarium wilt in basil in Korea.
The objective of these experiments was to develop an attenuated thermostable Newcastle disease virus(NDV), CBP-1 strain isolated from infected pheasants. Safety, pathogenicity and protective effects against velogenic NDV were also investigated to evaluate if the attenuated NDV, CBP-1 strain could be a candidate for a new NDV vaccine strain. CBP-1 strain was passaged up to the 173 times by nine days old embryonated eggs and chicken embryo fibroblast(CEF) cell cultures. Its sensitivitly to lipid solvents and low pH, thermostability, mean death time(MDT), intracerebral pathogenicity index(ICPI) of one day old chicks and intravenous pathogenicity index(IVPI) of four weeks old chicks were examined. Safety, boosting and protective effects were tested by chicks mortality. CBP-1 NDV strain had significant thermostability at 56$\^{C}$ for 30 minutes. by hemagglutinin activity and egg infectivity test, but was not resistant to lipid solvent. It showed possibility to use as a feed or water vaccine because of the resistance to low pH. MDT, ICPI and IVPI of CBP-1 were attenuated from 51.5, 1.96, 2.60 to 112.4, 1.12, 1.45. These results implied that the 173rd passages in embryonated egg and CEF cell cultures induced a substantial attenuation of the pathogenicity of the parent virus, changing the virulence from velogenic to intermediate between mesogenic and lentogenic. After vaccination with CBP-1 at one day old by drinking water mortality was 17.5%. However, spray vaccination with B1 at one day old, CBP-1 at two weeks ild and challenge with velogenic Kyojeongwon strain at four weeks old showed 93.5% survival rate. Mortality of chicks, vaccination with 173rd passaged CBP-1 strain at one day old, two weeks old and challenge with Kyokeongwon strain at four weeks old, was 20.0%. The results of these studies indicated that partial attenuated CBP-1 strain tended to be a low safety for ND of broiler chicks and would need to be more successive attenuation.
Fusarium oxysporum, F. moniliforme, F. solani, F. equiseti, F. semitectum, and F. sporotrichioides were detected from seeds, roots and cultivated soil of Phaseolous vidissimus collected from Kyung-gi Provincial Rural Development Administration. The rate of seedling desease incidence was $60\%$ by testing of seed germination using a large petri-dish. According to the blotter method, F. moniliforme showed $7\%$ infection at seed-coat and $2\%$ at cotyledon and embryo. Their pathogenicities of F. moniliforme, F. semitectum, F. equiseti, and F. sporotrichioides isolated from seeds were recognized on seedlings by water-agar test tube methods. F. oxysporum and F. solani isolated from infected-roots had their pathogenicity by water-agar test tube method but were weakly pathogenic by soil treatment method. Their pathogenicities of F. oxysporum. F. solani and F. uiseti isolated from cultivated-soil were recognized by water-agar test tube method. These F. oxysporum and equiseti isolates had their pathogenicities but F. solani was weakly pathogenic by soil treatment method.

Ginseng root rot caused by Cylindrocarpon destructans is one of the most destructive diseases of ginseng(Panax ginseng). We analyzed the features of the species through pathogenicity test and genetic diversity analysis of C. destructans in Korea, for its application as basic data to attempt for effective control. C. destructans isolated from rotted ginseng roots exhibited a variety of colonial colors on media. It was assumed that there may exist genetic diversity in the population by the diversity of pathogenicity among isolates observed when artificially inoculated into ginseng roots. Pathogenicity tests using ex vivo wound inoculation with agar mixture inoculation on ginseng roots were performed similar results as were observed appear to be useful for rapid pathogen inspection. According to RAPD analysis results, Korean C. destructans isolates formed a single genetic group which can be distinguished readily from closely related other fungi. C. destructans group was divided into two small groups. Therefore, we were able to confirm pathogenicity and genetic difference between the isolates in each of the groups of the pathogen.
Anthracnose of pepper casued by Colletotrichum spphas been a great problems for pepper production in Korea and China. Especially Colletotrichum gloeosporioides was found predominantly over cultivation areas during infection periods and caused severe rots on bath unripe and ripe fruits that resulted in major yield losses. In this study, comparison of Colletrichum spp.isolated from Korea and China in morphology and pathgenicity, and RAPD-PCR analysis were conducted. Based on morphological characteristics, the pathogen isolates, K1 and C1, K2 and C2, and K3 and C3 were identified as Colletotrichum gloeosporioides (G) type, C. gloeosporioides (R) type and C. coccodes, respectively. in pathogenicity test, K1 and C1, and K2 and C2 were found to attack mainly fruits and to be the most virulent among isolates. K3 and C3 were strongly virulent to leaves and seedling. Pathogenicity between Korean and Chinese isolates. K3 and C3 were strongly virulent to leaves and seedling. Pathogenicity between Korean and Chinese isolates did not show any difference. Results of the RAPD-PCR analyses indicate the varying levels of molecular diversity within and between Colletotrichum spp.of Korea and China. The similarities between K1 and C1, K2 and C2, and K3 and C3 were 85.71%, 71.43% and 50.0% respectively.
This study were performed to distribution of dermatophytes isolated from pet dogs in Pusan area and pathogenicity of the isolates. During the period from March 1993. to April 1994, 201 pet dogs of house breeding were examined. The result of obtained were summarized as followes; 1. Isolation rate of dermatophytes were 4.7%(7/149) in no lesion and 23.1%(12/52) in lesion of dog's skin. 2. According to breeding conditions, isolation rate of dermatophytes were 1) 3.8% in no lesion and indoor breeding, 2) 16.7% in lesion and indoor breeding, 3) 13.5% no lesion and outdoor breeding, and 4) 37.5% in lesion and outdoor bleeding. 3. Among the 201 Pet dogs, 16 strains of M. cams(84.2%), 1 of M. gypseum(5.3%), and 2 of T. mentagrophytes (10.5%) were isolated. 4. For Pathogenicity test, one strain of each M. canis, M, gypseum, T. mentagrophytes isolated from skin lesions were inoculated to the skin of the posterior back of guinea pigs. All of dermatophytes tested were induced skin lesions. Among them, M. cams and T. mentagrophytes were more virulent than M. gypseum.
A partial characteristics and pathogenicity of Aeromonas salmonicida strain isolated from diseased cultured eel were studied. On the biochemicial characteristics of the isolate strain was classified into atypical Aeromonas salmonicida : the characteristics of the isolate strain differed from those of the subspecies of Achromogenes, masoucida and salmonicida. It strain was named atypical Aeromonas salmonicida EL-1 and it grew at optimal pH 7.5 and l% Nacl. In the pathogenicity test, at one to two days post infection, the motality of artificially infected eels with $1{\times}10^8$ and $1.0{\times}10^7$ cfu/fish group was 100%, and at three days pos-infection, in the $1.0{\times}10^6$ /cfu /fish infected group, the epizootic haemorrhagic ulcer was developed, and haemorragic red sports were formed around abdomen and fins and its died (40%). Total cell protein peptide bands of the A. salmonicida EL-1 were presented betweeon 11.8~102 Kd in molecular weight by the electrophoretic analysis and appeared 28 bands in all. Drug sensitivity of atypical A. salmonicida EL-1 strain was sensitivity to chlorampenicol, gentamicin, kanamycin, streptomycin and sulfamethoxazole /trimethoprim and intermidiate to ampiclin and neomycin resistant to cephalotin, erythromycin, lincomycin and tetracycline.

Biotyping of Vibrio vulnificus strains isolated from marine environments along the south coast of Korea showed that the majority of the isolates (94.7%) belonged to biotype 1 and the remaining isolates (5.3%) belonged to biotype 2. Analysis of 16S rRNA V. vulnificus strains isolated from marine environments using a multiplex polymerase chain reaction (PCR) revealed that 78.7% were type A and 21.3% were type B. Random amplified polymorphic DNA (RAPD) was used to analyze the genomic differences in V. vulnificus among the biotype 2 strains isolated from marine environments (newly isolated strains group) and reference strains obtained from infected eels (reference strains group). The two groups had distinctly different profiles of the amplicons produced from RAPD. Additionally, biochemical comparison of these strains revealed that all four strains isolated from marine environments differed from the strains isolated from eels in their ability to promote D-mannitol fermentation. Two (NH 1 and NH 2) out of four isolates of biotype 2 from marine environments showed pathogenicity in eels Anguilla japonica in a challenge test. These isolates did not agglutinate with antisera against V. vulnificus NCIMB 2137 (serovar E), ATCC 27562 (non-serovar E), and ATCC 33816 (atypical serovar E).
Pear skin stains on 'Niitaka' pears, which occur from the growing stage to the cold storage stage, reportedly negatively influence the marketing of pears. These stains on fruit skin are likely due to a pathogenic fungus that resides on the skin and is characterized by dark stains; however, the mycelium of this fungus does not penetrate into the sarcocarp and is only present on the cuticle layer of fruit skin. A pathogenic fungus was isolated from the skin lesions of infected fruits, and its pathogenicity was subsequently tested. According to the pathogenicity test, Mycosphaerella sp. was strongly pathogenic, while Penicillium spp. and Alternaria spp. showed modest pathogenicity. In this present study, we isolated the pathogenic fungus responsible for the symptoms of pears (i.e., dark brown-colored specks) and identified it as Mycosphaerella graminicola based on its morphological characteristics and the nucleotide sequence of the beta-tubulin gene. M. graminicola was pathogenic to the skin of 'Niitaka' pears, which are one of the most widely growing varieties of pears in South Korea.

To obtain useful data on root rot in Korean ginseng, we performed phylogenetic analysis and pathogenicity test for Cylindrocarpon destructans isolated from peony. Cylindrocarpon destructans isolates from peony were proven to cause ginseng root rot. The isolate KACC44663 was identified as Ilyonectria robusta under the new classification system, which belongs to the I. radicicola species complex. This is the first report of the pathogenic isolate, which was isolated from another host plant, but not ginseng, that can cause root rot disease on ginseng in Korea.
