This study was conducted to investigate the suitability of frozen oysters as a raw material for the preparation of seafood products by measuring the concentrations of harmful microorganisms and chemicals in thawed flesh. The microbial concentrations in thawed oysters were 2.3-5.0 log CFU/g for viable cell counts, not detected (ND)-1.0 log CFU/g for coliform bacteria, and ND for Escherichia coli and pathogenic bacteria such as Staphylococcus aureus, Salmonella spp., Listeria monocytogenes, Vibrio parahaemolyticus, Enterohemorrhagic Escherichia coli (EHEC), and Clostridium perfringens. In frozen oysters, the heavy metal concentration for viable cell counts was ND-0.030 mg/kg, for lead was ND-0.393 mg/kg, and for cadmium was 0.021-0.597 mg/kg. Benzo(a)pyrene, shellfish poison (paralytic shellfish and diarrhetic shellfish poisons), and radioactivity were not detected in the thawed oysters. These results suggest that frozen oysters can be safely used as a raw material for the preparation of seafood products.

This study assessed microbial contamination in seven fried fish pastes sold in Southeast Asian traditional markets during summer. It measured viable cell count, coliforms, Escherichia coli, fungi, and Staphylococcus spp. It also qualitatively analyzed Vibrio parahaemolyticus, Salmonella spp., Bacillus cereus, Listeria monocytogenes, and Clostridium perfringens. The average viable cell count, coliforms and fungi were detected as 6.34 (3.84-8.13), 2.16 (1.00-3.55), and 3.92 (1.85-7.74) log10 CFU/g, respectively. Staphylococcus spp. was detected at 4.59 (2.10-7.63) log10 CFU/g. Some samples had high contamination levels: viable cell count (8.13 log10 CFU/g), fungi (7.74 log10 CFU/g) and S. aureus (7.63 log10 CFU/g). However, E. coli was not detected in any samples (ND, <1 log10 CFU/g). V. parahaemolyticus, Salmonella spp., B. cereus, L. monocytogenes, and Cl. perfringens were also not detected in the samples. The microbial contamination data provide insight into managing microbial contamination and ensuring the safety of fried fish pastes in traditional summer markets.

This study was conducted to achieve develop organic sludge recycling technology as sludge make a prey of earthworm. Therefore sludge treatment and recycling technology is an important field by which this research project to solve landfill site and reduction treatment expense using vermicomposting treatment process on the waste sludge from the biological wastewater treatment plant. In experimental results on the optimum conditions of vermicomposting of nightsoil treatment sludge, survival rates were observed 98.3% in temperature of $10-15^{\circ}C$, 75% in pH 5.8-7.5 and 100% in density of $1/79.8cm^3$, respectively. Liveweight changes of earthworm were increased 266% in temperature of $10-15^{\circ}C$, 227% in pH 5.8-7.5 and 325 % in density $1\;cap./79.8cm^3$, respectively. Casting production rate were generated 0.06 g/cap./day in temperature $20-25^{\circ}C$, 0.065 g/cap./day in pH 5.8-7.5 and 0.1 g/cap./day in density $1\;cap./79.8cm^3$, respectively. Cocoon production numbers were observed 3.8 ea. /cap.in $10-15^{\circ}C$, 2.95 ea./cap.in pH 5.8-7.5 and 3.16 ea./cap. in $1\;cap./79.8cm^3$ during 6 weeks, respectively. pH was droped by 6.2 to 5.7, volatile solids was decreased by 2.9%, $NH_3-N$ were also reduced by $6.984{\mu}g/g$ to $0.991{\mu}g/g$. $NO_3-N$, however, were increased by $3.213{\mu}g/g$ to $7.706{\mu}g/g$. Fecal coliforms and pathogenic bacteria are analyzed by microbiological method to assess public health safety of casting. Number of fecal coliform groups were reduced 88.6-99.1% (Avg. 95.7%) approximately. And pathogenic bacteria such as Salmonella, Shiegella and Vibrio, were not isolated from the earthworm cast.
The bacteriological and physiochemical analysis of seawater in Tongyeong harbor was conducted to evaluate sanitary conditions, The samples were collected at 6 stations established once a month from January to December, 2000. During the study period, the ranges of temperature, transparency, chemical oxygen demand, dissolved oxygen, dissolved nitrogen, phosphate and chlorophyll-a were $6.8\sim25.2^{\circ}C,\;1.0\sim2.5\;m,\;1.79\sim2.41\;mg/L,\;5.7\sim10.1\;mg/L,\;6.59\sim10.53{\mu}g-at/L,\;0.56\sim1.01{\mu}g-at/L\;and\;1.21\sim9.54\;mg/m^3$, respectively, The viable cell counts of seawater in Tongyeong harbor ranged from $3.0\times10^4CFU/mL\;to\;6.9\times10^6CFU/mL$. The coliform group and fecal coliform MPN's of the samples were ranged $23\~4,600\;MPN/100\;mL$ (means 540 MPN/100 mL) and $11\~1,600\;MPN/100\;mL$ (means 210 MPN/100 mL), respectively, The coliform group was classified with IMViC reactions and pathogenic vibrios were analyzed. Two hundred eighteen strains that were obtained from seawater samples in Tongyeong harbor represented Escherichia coli group, $66.1\%$; Citrobacter freundii group, $11.0\%$; Enterobacter aerogenes, $9.6\%$; and unknown, $13.3\%$, respectively. During the study period, infectious bacteria such as Vibrio cholerae O1, Salmonella sp. and Shigella sp. were not detected from the samples, but detection ratios of V. parahaemolyticus, V cholerae non-O1 and V. vulnificus were $10.0\sim30.1\%$.
The alone and combined effects of bacteriocin produced from Enterococcus faecalis MJ-213 and potassium sorbate against the food-borne pathogenic bacteria were studied. Bacteriocin minimal inhibitory concentration (MIC) values for Staphylococcus aureus ATCC 6538 and Salmonella enteritidis ATCC 13076 were 50 and 100 ${\mu}g$/ml, respectively. Bacteriocin (100 ${\mu}g$/ml) alone was active against S. aureus and S. enteritidis, but it was lower in antimicrobial effectiveness than the combination of bacteriocin (100 ${\mu}g$/ml) with potassium sorbate (100 ${\mu}g$/ml), which reduced initial counts (6 log cycle) of S. aureus and S. enteritidis by 1 and 3 log cycle, respectively. The bactericidal activity of bacteriocin of E. faecalis MJ-213 heated at $100^{\circ}C$ for 30 min or $121^{\circ}C$ for 15 min was markedly decreased as compared with the control. Moreover, the activity of bacteriocin was completely abolished by pepsin or protease II, but not affected by ${\alpha}$-amylase or lipase. The activity of bacteriocin adjusted to pH 6.0-8.0 showed almost the same inhibition ratio compared with the bacteriocin unadjusted pH, and though the inhibition ratio against pathogenic bacteria was reduced than the control, the bacteriocin was stable at pH 4.0 or 10.0, relatively. Furthermore, the combined treatment of bacteriocin and potassium sorbate than the alone treatment of bacteriocin significantly decreased (p<0.05) the viable cell counts of S. aureus or S. enteritidis inoculated on grind beef during storage at $4^{\circ}C$.
In this study, antibacterial activity against pathogenic strains and antioxidant activity were measured using the red pine leaf distilled concentrate. The results of the antibacterial activity measured using an emulsion of the red pine leaf distilled concentrate by the paper disc method showed the antibacterial activities against three Gram negative pathogenic strains, E. coli, S. typhi and Vibrio parahaemolyticus and exhibited growth inhibitions of 12 mm, 10 mm and 9 mm at a 5.0% (v/v) concentration, respectively. In addition, all three strains also showed growth inhibitions even at 0.5% (v/v) concentration. However, no antibacterial activity was exhibited against gram positive bacteria. The results of the antibacterial activity using the red pine leaf distilled concentrate measured by the turbidity method, the same antibacterial activities against three gram negative pathogenic strains, E. coli, S. typhi and V. parahaemolyticus as results of the paper disc method. V. parahaemolyticus showed more than 50% growth inhibition compared to the negative control at a concentration of 5% (v/v), E. coli exhibited 33.5% growth inhibition at 4 hr incubation, and S. typhi showed 65.1% and 44.6% growth inhibitions at 4 and 5 hr incubations, respectively. Antioxidant activities of an emulsion of the red pine leaf distilled concentrate were measured by DPPH and ABTS methods. DPPH method showed the highest activity of 55.81% at a 1.0% (v/v) concentration. ABTS method exhibited the highest activity of 18.44% at a 1.0% (v/v) concentration. Through this study, it is expected that the developments of the food and the cosmetics with enhanced functionality by utilizing the antioxidant and antibacterial activities of the red pine leaf distilled concentrate.
In the late summer of 1990 and 1991, mass mortality occured among cage-cultured grouper, Epinephelus septemfasciatus in south cost of Korea. The moribund fish didn't feed and became pale or dark chestnut colour and irregularly swimmed due to the loss of equilibrium, finally the diseased fish fell down side away on the bottom or the surface of cage showing the bent of body and died. The diseased fish showed the extensive hemorrahge in brain, the swelling of spleen and bile duct as the specific syptoms of internal organs. So the gill, skin and other organs of the diseased fish were examined for the presence of pathogenic parasites and bacteria. The parasitic Trichodina sp. were detected only from the gill lamella of the diseased fish, but these parasites seemed to be not a direct causative agents that induced the gross mortality of the cultured grouper. because these parasites were also observed in normal grouper, yellowtail, red seabream and rock bream co-cultured with the diseased grouper in same or near cages. In the viral examination, although isolation of the causative agent by the use of estabilshed cell Lines, RTG-2 and CHSE-214, was not succeed, the normal grouper inoculated intramuscularly with the filtered homogenate of the organs of the diseased fish showed the same external and internal signs with the naturally infected grouper. They died within a week. By using the naturally and the artificially infected fishes, electron microscopic observation revealed numerous hexagonal or polygonal particles in the cytoplasm of liver cells. Based on the these results, we suggest that the mass mortality of the cultured grouper would be occurred by the infection of a viral agent.
Objectives: This study was performed to analyze cases of food poisoning outbreaks reported in Chungcheongnamdo Province in 2019 and report it as effective data for preventing food poisoning in the future. Methods: Food poisoning outbreaks were analyzed to detect virus, bacteria, and protozoa according to the Manual for Detection of Foodborne Pathogens in Outbreaks to clarify the causes of food poisoning that occurred in Chungcheongnam-do Province in 2019. Results: Among the 79 cases of food poisoning outbreaks, 59 cases (74.7%) were in general restaurants, 15 cases (19.0%) in food service institutions, three cases at banquets, and two cases in take-out food. The 42 cases at general seafood restaurants made up the majority of food poisoning in Chungcheongnam-do. Food poisoning pathogens were shown in 13 cases (86.7%) out of the 15 cases at food service institutions, and 10 cases were related to Norovirus. Among the 79 cases, food-borne pathogens were identified in 35 cases of outbreaks, accounting for 44.3%. The confirmed pathogens were as follows: bacteria (24 cases), Norovirus (12 cases) and Kudoa septempunctata. (five cases). The food-borne bacteria were pathogenic E. coli (12 cases), Staphylococcus aureus (six cases), Salmonella spp. (two cases), Campylobacter jejuni, Bacillus cereus, Clostridium perfringens, and Vibrio parahaemolyticus (one case). Conclusions: To prevent food poisoning, it is necessary to analyze regional characteristics and environments and to hold a campaign for the prevention of food poisoning based on that analysis. In addition, when food poisoning occurs, the results of analyzing its cause and spread based on accurate epidemiological survey need to be shared.
Aquaculture continues to be an ever-growing sector. However, high-density farming increases disease outbreaks due to deteriorating water quality and internal stress. To prevent disease, the most common method chemotherapy is using antibiotic administration. In this study, probiotic bacteria were isolated from Korean traditional foods, such a Gochu pickle and cutlassfish salted seafood. Various bacteria were isolated, and their 16S rDNA sequences were analyzed. The antimicrobial activities of four isolates from Gochu pickle and seven isolates from cutlassfish salted seafood were assayed, in addition to the antibacterial activity of culture pellet and supernatant. The antibacterial activity of the pellet was higher than that of the supernatant. Isolate JKM-2 showed the highest antibacterial activity against Streptococcus iniae (43 mm), S. parauberis (40 mm), S. mutans (35 mm), and Vibrio vuinificus (26.5 mm). The sequences of the isolated strains were compared with those of Bacillus subtilis (97.71%), B. tequilensis (97.71%), Brevibacterium halotolerans (97.71%), B. subtilis (97.63%), B. subtilis (97.63%), B. mojavensis (97.54%), B. vallismortis (97.46%), B. nanillea (97.45%), B. methylotrophicus (97.37%), and B. ssiamensis (97.37%). Future through analysis and new strains confirmed the bacterial cell material investigation of JKM-3, and to ensure sufficient stability, it is desired to verify the utility value as a substitute material for antibiotics by application to the form of the industry.

In this study, we isolate and identify bacteria from seawater collected from Jeju coast, to evaluate the antimicrobial activity against the fish pathogenic bacteria. 14 bacterial strains were isolated and identified using physiological, biochemical and molecular tools. Antibacterial activity of all the 14 isolates were screened against four major fish pathogens namely, two Gram-positive: Streptococcus iniae, Streptococcus parauberis and two Gram-negative: Vibrio anguillarum, Edwardsiella tarda. Results revealed that among the 14 isolates, MK-11 was found to have antibacterial activity against S. iniae, S. parauberis, V. anguillarum Particularly, S. iniae was susceptibility with the MIC value of $250{\mu}g/ml$. The biochemical and physio-chemical results reveal that MK-11 had the sugar-alcohol disassemble ability of the D-sorbitol and D-mannitol. Also the utilization of the yeast extract, sorbitol and di-potassium phosphate were noted to be high. The optimum culture condition such as pH and temperature was recorded as pH 6.0, $25^{\circ}C$ and along with 1% NaCl which differs from the previous reports particularly in nutrient resolutions. As results of the analysis of 16S rDNA sequences, MK-11 show the high similarity with Paenibacillus polymyxa, P. jamilae, P. brasilensis 99.78%, 99.43%, 99.39%, repectively. Hence, in the present study, the isolated Paemibacillus sp. MK-11 from Jeju seawater possesses the antibacterial activity against fish pathogens and it could be used as a new antibiotic agents against the gram positive fish pathogens.
