Seed-borne Pathogenic Bacterium Interact with Air-borne Plant Pathogenic Fungus in Rice Fields

  • Air-borne plant pathogenic fungus Fusarium graminearum and seed-borne plant pathogenic bacterium Burkholderia glumae are cause similar disease symptoms in rice heads. Here we showed that two pathogens frequently co-isolated in rice heads and F. graminearum is resistant to toxoflavin produced by B. glumae while other fungal genera are sensitive to the toxin. We have tried to clarify the resistant mechanism of F. graminearum against toxoflavin and the ecological reason of co-existence of the two pathogens in rice. We found that F. graminearum carries resistance to toxoflavin as accumulating lipid in fungal cells. Co-cultivation of two pathogens resulted in increased conidia and enhanced chemical attraction and attachment of the bacterial cells to the fungal conidia. Bacteria physically attached to fungal conidia, which protected bacterium cells from UV light and allowed disease dispersal. Chemotaxis analysis showed that bacterial cells moved toward the fungal exudation compared to a control. Even enhanced the production of phytotoxic trichothecene by the fungal under presence of toxoflavin and disease severity on rice heads was significantly increased by co-inoculation rather than single inoculation. This study suggested that the undisclosed potentiality of air-born infection of bacteria using the fungal spores for survival and dispersal.

고추냉이 추출물의 항균효과에 관한 연구 (Studies on the Antimicrobial Effect of Wasabi Extracts)

  • The volatile components of Eutrema wasabi were prepared by distillation and ether extraction. The extracts were similar in the color, the odor, and the antimicrobial activity to allylisothiocyanate, the main ingredient of wasabi essential oil. The antimicdrobial activity of the extracts was evaluated and it was found that these had the antibacterial effect against a broad spectrum of pathogenic bacterial such as Staphylococcus, Escherichia, Pseudomonas and Salmonella.

콩 꼬투리에서 서식하는 세균 및 콩나물 부패균의 밀도 변화 (Population Density Changes of Bacteria Causing Soybean Sprout Rot on Soybean Pods)

  • Bacterial population densities on soybean pods from Chungnam province ranges 105~106 CFU/$\textrm{cm}^2$, whereas those of bacteria causing sprout rot ranged 0~103 CFU/$\textrm{cm}^2$. Erwinia chrysanthemi, Xanthomonas campestris pv. glycines, Staphylococcus sp., and Micrococcus sp. were identified as pathogenic bacteria causing soybean sprout rot. The population density of X. campestris pv. glycines was higher than those of other bacteria.

C. elegans Behavior of Preference Choice on Bacterial Food

  • Caenorhabditis elegans is a free living soil nematode and thus in its natural habitat, C. elegans encounters many different species of soil bacteria. Although some soil bacteria may be excellent sources of nutrition for the worm, others may be pathogenic. Thus, we undertook a study to understand how C. elegans can identify their preferred food using a simple behavioral assay. We found that there are various species of soil bacteria that C. elegans prefers in comparison to the standard laboratory E. coli strain OP50. In particular, two bacterial strains, Bacillus mycoides and Bacillus soli, were preferred strains. Interestingly, the sole feeding of these bacteria to wild type animals results in extended lifespan through the activation of the autophagic process. Further studies will be required to understand the precise mechanism controlling the behavior of identification and selection of food in C. elegans.

파이로시퀀싱을 이용한 한강상수원보호구역 수계 중의 세균 다양성 (Analysis of Bacterial Diversity in Water from the Han River Water Source Protection Area via a Pyrosequencing Assay)

  • Objectives: We investigated bacterial diversity in the Han River water resource protection area in order to provide basic microbiological information on the drinking water safety of the Seoul metropolitan region. Methods: Samples were collected in the spring and winter, but not during the rainy season. Pyrosequencing, gene amplification, and extraction of nucleic acids were employed in this study. Results: In total, 57 and 48 operational taxonomic units were respectively analyzed in samples collected during spring and winter. Proteobacteria were predominant in all samples. The samples contained phylogenetically diverse bacterial communities, with eleven major phyla and 36 genera. Cyanobacteria were predominant in the spring samples, but not in the winter samples. The predominant species in the samples collected during both seasons belonged to the genus Aquamicrobium and Bradyrhizobium. Moreover, no pathogenic bacteria were detected in the samples. Conclusion: Proteobacteria were predominant in the samples from the Han River water source protection area. Cyanobacteria were more predominant in the spring samples than in the winter samples, but Aquamicrobium and Bradyrhizobium were predominant in both sampling seasons.

  • Potato scab, an important disease that affects developing tubers, causes a major problem in potato cultivation. The major potato cultivation areas in Korea are located in two Northern provinces, Gangwon and Gyeonggi, and two Southern provinces, Jeju island, and South Jeolla. In these areas, potato scab is widely distributed and has caused severe problem in potato cultivation. Therefore, potato-growing areas were surveyed for identification and distribution of potato scab pathogens from 1996 to 1999. Pathogenic Streptomyces strains were isolated from potato scab lesions and six representative Streptomyces species were characterized based on their phenotypic and molecular characteristics including, pathogenicity, physiological and morphological properties, analyses of 16SrRNA genes and 16S-23S ITS region, DNA relatedness, production of thaxtomin A, and the presence of nec1 and ORFtnp gene homologs. Three species were identified as previously described Streptomyces scabies, S. turgidiscabies, and S. acidiscabies, while other three species having distinct phenotypics properties were identified as novel S. luridiscabiei, S. puniciscabiei, and S. niveiscabiei.

Identification of mono- or poly-specific monoclonal antibody to Porphyromonas gingivalis heat-shock protein 60

  • Purpose: The aim of this study was to define the immunoreactive specificity of Porphyromonas gingivalis (P. gingivalis) heat shock protein (HSP) 60 in periodontitis and atherosclerosis. Methods: In an attempt to define the cross-reactive bacterial heat-shock protein with human self-antigen at molecular level, we have introduced a novel strategy for cloning hybridoma producing anti-P. gingivalis HSP 60 which is polyreactive to bacterial HSPs or to the human homolog. Results: Five cross-reactive clones were obtained which recognized the #19 peptide (TLVVNRLRGSLKICAVKAPG) among 37 synthetic peptides (20-mer, 5 amino acids overlapping) spanning the whole molecule of P. gingivalis HSP 60. We have also established three anti-P. gingivalis HSP 60 monoclonal antibodies demonstrating mono-specificity. These clones recognized the #29 peptide (TVPGGGTTYIRAIAALEGLK). Conclusions: Peptide #19 and #29 of P. gingivalis HSP 60 might be important immunoreactive epitopes in the immuno-pathogenic mechanism of bacterial antigen-triggered autoimmune diseases.

Xanthomonascampestris pv. pruni와 Erwinia nigrifluens에 의한 복숭아 및 자두의 세균성구멍병 (Shot Hole of Peach and Japanese Plum Caused by Xanthomonas campestris pv. Pruni and Erwinia nigriflens in Korea)

  • 1998년 나주와 밀양에서 재배중인 복숭아와 자두에서 세균성 구멍병이 발생하였다. 복숭아와 자두의 병반으루부터 분리한 균주의 세균학적 특성을 조사한 결과, 병원세균 중 2균주는 Xanthomonas campestris pv. pruni로, 3균주는 Erwinia nigrifluens로 동정되었다. E. nigrifluens는 국내에서는 복숭아와 자두의 세균성 구멍병으루 처음 분리되었다. 이 균주는 X. campestris pv. pruni에 의한 병징과 구분하기가 곤란하고, 2종류의 병워세균이 동시에 침입하여 발병하는 것으루 관찰되었다. 이와 같은 이유로, E. nigrifluens와 X. compestris pv. pruni에 의한 복숭아와 자두의 세균병을 각각 \"복숭아, 자두의 세균성구멍병\"로 명명할 것을 제안한다.할 것을 제안한다.

입상활성탄 부착세균과 염소소독 연구 (Bacterial attachment on granular activated carbon and effect of chlorine disinfection)

  • The authors investigated the bacterial community attached to granular activated carbon(GAC) particles and the susceptibility of the community to chlorine disinfection. The study was carried out at the G Water Treatment Plant in Seoul, which was in full-scale operation. Bacteria attached to the surface of GAC increased gradually with treatment from $0.4{\times}106{\sim}8.5{\times}106 CFU/g$. TOC removal was under 1.0 mg/L due to increased bacterial community on the surface of GAC. It was found that TOC removal was closely related with physical and biological parameters such as pore volume and the number of attached bacteria. When the washed and the attached cells were disinfected with 1.0mg/L of chlorine for 1 hour, the washed cells with chlorination could be controlled, but the number of the attached cells increased gradually. The results suggest a possibility that the treatment and disinfection barriers can be penetrated and pathogenic bacteria may break into the drinking water supplying system.

Antimicrobial Properties of Cold-Tolerant Eucalyptus Species against Phytopathogenic Fungi and Food-Borne Bacterial Pathogens

  • Mechanol extracts of three cold-tolerant eucalyptus trees-Eucalyptus darlympleana, E. gunnii and E. unigera were screened for antimicrobial activity against twenty two phyto-pathogenic fungi and six food-borne bacterial pathogens. E. unigera showed the antagonistic activity against all the tested pathogens. Among the tested fungal pathogens, Pythium species were highly sensitive to the leaf extracts. Especially, P. vanterpoolii, a causal agent of leaf blight in creeping bentgrass (Agrostis palustris), was completely inhibited by the extracts. The eucalyptus extracts were also effective in inhibiting the fungal growth of Botrytis cinerea and Phomopsis sp. isolated from the lesions of kiwifruit soft rot during post-harvest storage. Escherichia coli O-157 was less sensitive to the inhibition than the other bacterial pathogens tested. It was likely that Gram positive bacteria-Bacillus subtilis and Streptococcus mutans were more sensitive to the eucalyptus extracts than Gram negative bacteria-Escherichia coli, Salmonella enteritidis and Pseudomonas aeruginosa. Our findings suggest that the cold-tolerant eucalyptus species have antimicrobial properties that can serve the development of novel fungitoxic agents or food preservatives.
