  • This study was aimed to investigate the effects in different immersion doses of garlic, Allium sativum, juice to modulate on the nonspecific immune responses of the olive flounder, Paralichthys olivaceus, artificially prechallenged with Streptococcus iniae BS10 and Edwardsiella tarda KE-1, respectively. The nonspecific immune responses of the tested fish were assessed in term of skin mucus lysozyme activity, the change of bacterial cell counts in organs, the number of lymphocytes and neutrophils in blood, and SOD activity. Almost groups of the prechallenged with either S. iniae BS10 or E. tarda KE-1 fish which had been immersed in garlic juice showed the enhanced skin mucus lysozyme activity, the number of lymphocytes and neutrophils in blood, and SOD activity in the kidney but the decreased the number of bacterial cell in surveyed organs. RPS in the group immersed in 0.25 g/L of garlic juice was much higher than in other immersed test groups. These results suggested that the garlic juice immersion can be effective on enhancement of the nonspecific immune responses and the protective ability of olive flounder to the artificial challenge with S. iniae BS10 and E. tarda KE-1.

  • This experiment was carried out to measure the content of lactoferrin, IgA, $IgG_1,\;IgG_2$, in Holstein colostrum, and to test the effect of it's colostrum on the antibacterial activity to pathogenic bacteria and the growth stimulation of lactic acid bacteria. Colostrum was collected at the first, second, and third day after parturition in summer and winter season. The levels of lactoferrin, IgA, $IgG_1,\;and\;IgG_2$ in Holstein cow colostrum were 0.30 mg/mL, 0.37 mg/mL, 4.00 mg/mL, 0.37 mg/mL, respectively, on the first day of the summer season whereas they were 1.16 mg/mL, 2.60 mg/mL, 13.35 mg/mL, 1.30 mg/mL on the first day of the winter season, postpartum. Heat treated ($65^{\circ}C$ for 30 min) or non-treated colostrum showed antibacterial activity toward Escherichia coli. The growth of commercial mixed strains (Bifidobacterium longum, Lactobacillus acidophilus, Streptococcus themophilus), L. acidophilus, L. casei, L. bulgaricus, and L. lactis subsp. cremoris were improved in first, second and third day colostrum compared to normal milk. Commercial miked strains (B. longum, L. acidophilus S. themophilus) lowered the pH to 4.97-5.22 and 4.89 while increasing the titratable acidity to 0.75-0.88% and 0.70% in colostrum and normal milk, respectively. However, L. casei, L. bulgaricus, L. lactis subsp. cremoris lowered the pH to 5.96-6.47 and 6.5-6.8 while increasing the titratable acidity to 0.29-0.48% and 0.20-0.25% in colostrum and normal milk, respectively.

  • Physical, chemical and biological hazards of strawberry farms at the cultivation stage were analyzed to establish the GAP(Good Agricultural Practice) system. Samples were collected from the plants, cultivation environments(water, soil and air), and personal hygiene (hand, glove, and clothes) of three strawberry farms(A, B, and C) and were tested to analyze physical, chemical (heavy metals and pesticide residues), and biological(sanitary indications and foodborne pathogens) hazards. Physical hazards such as insects and pieces of metal and glass were found in the strawberry farms and can be potential bow for strawberry products. Heavy metal and pesticide residue as chemical hazards were detected at levels lower than the regulation limit. In case of biological hazards, total bacteria and coliform were detected at the levels of 1.6~7.3 and 1.3~5.6 log CFU/g, leaf, mL, hand or $100cm^2$. However, Escherichia coli was not detected in all samples. Bacillus cereus and Staphylococuus aureus were detected at levels of ${\leq}$ 1.1~6.1 log CFU and 4.7~5.4 log CFU/g, mL, hand or $100cm^2$, whereas Listeria monocytogenes, E. coli O157 and Salmonella spp. were not detected in all samples. This study demonstrates that various harzards were in strawberry farms at the growing stage. Therefore proper management such as GAP is needed to prevent the occurrence of food poisoning associated with the hazards revealed in this study.

During the study period, the means of temperature was range from 5.3 to $24.9^{\circ}C$ (mean $17.7{\pm}0.4^{\circ}C$), transparency range from 1.4 to 2.5 m (mean $1.8{\pm}0.5m$), suspended solid ranged from 16.2 to 35.8 mg/L (mean $24.2{\pm}2.2mg/L$), chemical oxygen demand ranged from 1.42 to $3.29mgO_2/L$ (mean $2.06{\pm}0.55mgO_2/L$), dissolved oxygen ranged from 6.7 to 9.5mg/L (mean $7.9{\pm}0.6mg/L$), respectively. Seafood, if eaten raw, carries the risk of food poisoning. Seafood poisoning is often cause by pathogenic microorganism originating from fecal contamination, such as Salmonella sp., Shigella sp. and norovirus. Fecal coliforms are an important indicator of fecal contamination. Therefor, data on fecal coliform are very important for evaluating the safety of fisheries in coastal areas. So, we investigated the sanitary indicate bacteria. In this study, 56 sea water samples were collected from the Sacheon Bay, and total and fecal coliforms were compared and analyzed. The coliform group and fecal coliform MPN's of sea water in Sacehon Bay were ranged from <1.8~7,900 MPN/100mL (GM 214.7 MPN/100mL) and <1.8~330 MPN/100mL (GM 9.7 MPN/ 100mL), respectively. Total coliforms were detected in 75.0% of the samples and 76.2% of the total coliforms were fecal coliforms. During the study period, the means of water content, ignition loss, COD, and acid volatile sulfates in sediments in Sacheon Bay were $53.28{\pm}2.58%$, $9.38{\pm}0.42%$, $14.23{\pm}3.36mgO_2/g$, $0.09{\pm}0.07mgS/g$, respectively.

  • This study was conducted to find out the effect of herbicides on soil micro-organism and soil enzyme in loam soil, and on pathogenic microorganism in continuous pepper cropping soil. The result was summarized as follows: When herbicides were treated, the number of soil microorganism generally decreased at the early stage of incubation, and gradually increased at 30 days incubation. The number of fungi was significantly decreased seven times comparison with control plot. The number of actinomycetes was increased at 20 days incubation and that of pytium was increased for all days incubation in Linuron treatment plot. Although the activity of soil enzyme tended to decrease when herbicides were treated, the activities of urease and phosphatase were higher than that of control plot at the early stage of incubation in MO, Linuron and Simetryne treatment plot. In Simetryne treatment plot, the activity of protease was increased at the early stage, and suddenly decreased after 30 days incubation. There was no significant effect of herbicides on the activities of ${\beta}$-glucosidase and polygalacturonase. The activity of cellulase was inhibited at the early stage of incubation, but that of cellulase was higher than that of control plot after 20 days incubation. High significance was showed the correlation coefficient between soil microorganisms except fungi and soil respiration as herbicides were treated, and the total microorganism and soil respiration in Linuron plot. when Dicamba and Simetryne were treated, the correlation coefficient between the total microorganism and the activities of protease and urease were considerably significant.

  • In recent years, there has been an increasing public concern about fecal contamination of water, air and agricultural produce by pathogens residing in organic fertilizers such as manure, compost and agricultural by-products. Efforts are now being made to control or eliminate the pathogen populations at on-farm level. Development of efficient on-farm strategies to mitigate the potential risk posed by the pathogens requires data about how the pathogens prevail in livestock manure composts and organic fertilizers. Microbiological analysis of livestock manure composts and organic fertilizers obtained from 32 and 28 companies, respectively, were conducted to determine the total aerobic bacteria count, coliforms, Escherichia coli count and the prevalence of Staphylococcus aureus, Bacillus cereus, Salmonella spp., Escherichia coli O157:H7, Listeria monocytogenes, and Cronobacter sakazakii. The total aerobic bacteria counts in the livestock manure composts and organic fertilizers were in the range of 7 to $9log\;CFU\;g^{-1}$ and 4 to $6log\;CFU\;g^{-1}$, respectively. In the livestock manure composts, coliforms and E. coli were detected in samples obtained from 4 and 2 companies, respectively, in the range of 2 to $5log\;CFU\;g^{-1}$ and $2log\;CFU\;g^{-1}$. In the organic fertilizers, coliforms and E. coli were detected in samples obtained from 4 and 1 companies, respectively, in the range of 1 to $3log\;CFU\;g^{-1}$ and $2log\;CFU\;g^{-1}$. In 3 out 32 compost samples, B. cereus was detected, while other pathogens were not detected. In 28 organic fertilizers, no pathogens were detected. The complete composting process can result in the elimination of pathogens in livestock manure compost and organic fertilizer. The results of this study could help to formulate microbiological guidelines for the use of compost in environmental-friendly agriculture. This research provides information regarding microbiological quality of livestock manure compost and organic fertilizer.

  • This study was performed to investigate the extent of microbial contamination, the presence of enterotoxin genes, and the antibiotic susceptibility of Bacillus cereus in 58 red pepper plants and 43 environmental samples (soil, irrigation water, and gloves) associated with the plant cultivation. The detected counts of total aerobic bacteria, coliform bacteria, Escherichia coli, Bacillus cereus, and Staphylococcus aureus were lower in these samples, as compared to the regulations of standards for foods; moreover, pathogens, such as E. coli, E. coli O157:H7, Listeria monocytogenes, and Salmonella spp., were not detected. Genes encoding hemolysin BL enterotoxins (hblA, hblC, and hblD) as well as non-hemolytic enterotoxins (nheA, nheB, and nheC) were detected in 23 B. cereus specimens that were isolated from the test samples and had β-hemolytic activity. Interestingly, B. cereus is resistant to β-lactam and susceptible to non-β-lactam antibiotics. However, in this case, the isolated B. cereus specimens exhibited a shift from resistant to intermediate in response to cefotaxime and from susceptible to intermediate in case of rifampin, trimethoprim-sulfamethoxazole, vancomycin, clindamycin, and erythromycin. Therefore, the levels of B. cereus should be monitored to detect changes in antibiotic susceptibility and guarantee their safety.

  • Pintail (Anas acuta) is the major wintering bird in South Korea and known as a major mediator of the highly pathogenic avian influenza (HPAI). Pintail migrates long distances between Russian Siberia and Korea. This species prefers a rice paddy area as their winter habitat. The purpose of this study is to provide the data necessary for the conservation and management of bird habitats in Korea by understanding the wintering home-range and habitat of pintail in Korea. We captured six pintails using a cannon-net in the winter of 2015 and attached the GPS-mobile phone based telemetry (WT-300) on them to study the wintering home-range and wintering habitat. We analyzed the tracking location data using ArcGIS 9.0 Animal Movement Extension and calculated Kernel Density Estimation (KDE) and Minimum Convex Polygon (MCP). The average home-range in the wintering ground analyzed by MCP was $677.3km^2$ (SD=130.2, n=6) while the maximum and minimum were $847.7km^2$ and $467.5km^2$, respectively. Extents of home-range analyzed by KDE were $194.7km^2$ (KDE 90%), $77.4km^2$ (KDE 70%), and $35.3km^2$ (KDE 50%). The pintails mostly used both sea and paddy field as habitat in the winter season and utilized paddy fields more during the nighttime and than the daytime. We concluded that the home-range and habitat of pintails in the winter could be used as the reference data for the preservation of species, management of habitats, and coping with a breakout of HPAI.

  • In order to verify whether Zophobas atratus is an edible insect, the nutrients and harmful substances of Z. atratus larvae reared with an artificial diet (AD) and wheat bran (WB) were compared and analyzed. Based on dry weight, the crude protein content of Z. atratus larvae reared with an AD was 62.4%, 1.4 times higher than that of those reared with WB (45.2%). The crude fat content was 20.5% in the AD group, 2.3 times less than in the WB group (46.3%). The leucine content was 1.4 times higher in the AD group (4.2%) than in the WB group (3.0%). The glutamic acid content of nonessential amino acids was 1.3 times higher in the AD group (7.0%) than in the WB group (5.3%). The oleic acid content was 1.4 times higher in the WB group (37.0%) than in the AD group (26.7%). The potassium content was 1.1 times higher in the AD group (975.9 mg/100 g) than in the WB group (872.9 mg/100 g). According to the results of the toxic substances analysis, the lead and cadmium levels of the WB and AD groups were standard for edible insects. Pathogenic microorganisms, such as E. coli and salmonella, were not detected in either group. According to the results of the present analysis of nutrition and harmful substances, Z. atratus larvae raised on an AD are safe and contain various nutrients. Therefore, such larvae could be useful sources of food and feed.

  • Hearing loss is a group of clinically and genetically heterogeneous disorders characterized by congenital- to adult-onset deafness with frequent additional symptoms such as myopathy, nephropathy, and optic disorders. It is commonly divided into two types: syndromic, with no other symptoms, and nonsyndromic, with other symptoms. Autosomal recessive hearing loss is relatively frequent in Pakistan, which may be due in part to frequent consanguineous marriages. This study was performed by whole exome sequencing to determine the genetic causes in two Pakistani consanguineous families with autosomal recessive hearing loss. We identified a pathogenic homozygous variant (p.Leu326Gln in MYO7A) in a family with prelingual-onset hearing loss and two variants of uncertain significance (p.Val3094Ile in GPR98 and p.Asp56Gly in PLA2G6) in a family with early-onset hearing loss concurrent with muscular atrophy. The missense mutations in MYO7A and PLA2G6 were located in the highly conserved sites, and in silico analyses predicted pathogenicity, while the GPR98 mutation was located in the less conserved site, and most in silico analysis programs predicted its nonpathogenic effect. Homozygosity mapping showed that both alleles of the homozygous mutations identified in each family originated from a single founder; spread from this single source might be due to consanguineous marriages. This study will help provide exact molecular diagnosis and treatment for autosomal recessive hearing loss patients in Pakistan.
