Preparation and Characterization of Microparticles of $Poly(\gamma-glutamic\;acid)$ Containing Lysozyme by means of Supercritical Anti-Solvent (SAS) Precipitation Process (초임계 반용매 침전법에 의한 라이소자임이 내포된 폴리감마글루탐산의 미세입자 제조 및 특성)

  • The sub-micron particles of poly ($\gamma-glutamic$ acid) (PGA) containing lysozyme have been prepared using supercritical antisolvent (SAS) precipitation process at various experimental conditions such as pressure, temperature, inner diameter of nozzle, and concentration. As overall results of the application of SAS process to this system, the smaller size powder has been produced as (i) increasing pressure, (ii) decreasing temperature, (iii) decreasing the inner diameter of nozzle, and (iv) decreasing the concentration of PGA and lysozyme. It is found by means of FT-IR analysis that during SAS process, the composition has changed from the original composition of PGA : lysozyme=50 : 50 into PGA : lysozyme=33 : 67 at final product powder. It means that PGA has higher solubility for the mixed solvent of carbon dioxide and dimethyl sulfoxide (DMSO). Due to such difference of solubility, this particle forms the core-shell structure of which the core consists mainly of lysozyme. It is also found that the residual DMSO amount of $7.8\times10^{-3}wt%$ exists inside the powder.

Numerical simulation of turbidity currents intruding into a reservoir (저수지로 유입되는 부유사 밀도류의 수치모의)

  • This study proposes a numerical model which is able to simulate turbidity currents intruding into a reservoir and resulting sediment depositions. The proposed model is applied to laboratory experiments by Toniolo and Schultz (2005), and propagation of turbidity currents, morphological change, and trap of suspended sediment are simulated. It is simulated that the turbidity current after plunging at the foreset of the model delta, propagates along the bottom. The thickness of the turbidity current increases significantly after being blocked by the dam, and this effect is propagated in the upstream direction. In addition, it is simulated that the foreset moves in the downstream direction due to both the bedload and suspended load and the thickness of the bottom set increases due to the suspended load. It is found that the height of the intake affects the thickness of the turbidity current and the location of the internal hydraulic jump. The impact of the height of the intake on the trap efficiency is not clear in the experimental results, however, overall trap efficiency is predicted quite successfully by the model. Also, sensitivity analysis is carried out, and the results indicates that the particle size affects the trap efficiency most.

  • Silkworm fed on the mulberry leaf mixed with silver nanoparticle to produce silver-nanoparticle embedded cocoon. Comparative analysis of silver content of cocoon shell, percentage of pupation and percentage of cocoon-shell weight showed that the optimum concentration and the feeding period of mulberry leaf mixed with silver nanoparticle were 500 ppm and the period from 3 day 5 instar to mounting of silkworm. The silver content of cocoon was observed variously by silkworm breedings. C212 variety makes pale yellow cocoon with the highest silver content(69%). Using the scanning electron microscope, we showed that the size of silver nanoparticles in silk was observed from 26.98 to 99.81nm. Silver-nanoparticle embedded silk is expected to use as high valuable application owing to the different functional properties including antibiotic characteristics and mechanical and electronic properties. The applicable fields expected is antistatic and/or electronic products with biological degradable natural materials.

Mass Mortalities of Cultured Striped Beakperch, Oplegnathus fasciatus by Iridoviral Infection (이리도 바이러스 감염에 의한 양식 돌돔, Oplegnathus fasciatus의 대량폐사)

  • From August to October 1998, over 60% mortality of cultured striped beakperch Oplegnathus fasciatus was occurred in net cages along the southern coast of Korea. Moribund fish showed some clinical signs of lethargic behavior, dark coloration or decoloration, severe gill anemia and enlargement of spleen. Also enlarged basophilic cells showing Feulgen -positive reaction were observed in the tissue section of spleen, kidney, liver and heart of the diseased fish. GF cells inoculated with spleen homogenate of diseased fish produced cytopathic effect of enlarged and rounded cells, therefore the causative virus was isolated from diseased fish. Striped beakperch fingerlings intraperitoneally inoculated with the causative virus ($10^4TCID_{50}$/0.1 ml) revealed symptoms similar to those of naturally infected fish and died from 7 to 14 days post injection. Transmission electron microscopy revealed that the causative virus was enveloped icosahedral particle with 120~130 nm in diameter. PCR products of the expected size (500 bp) were amplified with a primer set based on the ATPase gene of RSIV(red sea bream iridovirus) using template DNAs which were extracted from the spleen of diseased fish and GF cells inoculated with the causative virus. According to the analysis of nucleotide sequence of these PCR products, the sequence from ATPase cDNA gene of the causative virus showed 95% homology with that of RSIV. These results indicate that the mass mortality in the cultured striped beakperch was caused by the infection of iridovirus similar to RSIV.

An appropriateness review on the road tunnel ventilation standards by pollutants site measurement and case study (오염물질 현장측정 및 사례조사를 통한 도로터널 환기기준의 적정성에 관한 연구)

  • In this study, a series of site measurement of particulate and gases pollutants at five tunnels were carried out along with case studies to review the suitability of the current road tunnel ventilation design standards. Previous studies by other researchers have shown that the ratios of the level of measurement to the standard were 27.9%, 1.6% and 3.4% for TSP, CO and NOx, respectively. Those measured in this site study shows even lower ratios; the ratios were 2.6%, 0.8% and 0.3%, for TSP, CO and NOx, respectively. The particle size analysis of TSP for the five tunnels shows that PM10 including tire wear and re-suspended road dust exceeded 20.4%. This implies that non-exhaust particulate matter must be taken into account, since the current design standards for the particulate matter (visibility) include only the engine emission. Based on the recent research results, for vehicle emission rate and slope-speed correction factors, revision of ventilation design standards for pollutants is required. WRA (PIARC) also emphasizes the necessity of the ventilation design standards for pollutants. In addition, enactment of a new road tunnel ventilation system operation standard or guideline is strongly recommended when considering the low operating rate of the ventilation system with jet-fans.

The Effective Recovery of Gold from the Invisible Gold Concentrate Using Microwave-nitric Acid Leaching Method (마이크로웨이브-질산침출방법에 의한 비가시성 금의 회수율 향상)

  • This study aimed to liberate gold from invisible gold concentrate (Au = 1,840.00 g/t) through microwave nitric acid leaching experiments. For the purpose, this study conducted microwave-nitric acid leaching experiments and examined nitric acid concentration effect, microwave leaching time effect and sample addition effect. The results of the experiments were as follows: Au (gold) contents were not detected in all of the microwave leaching conditions. In the insoluble-residue, weight loss rate tended to decrease as the nitric acid concentration, microwave leaching time and sample addition increased. In an XRD analysis with solid-residue, it was suggested that gypsum and anglesite were formed due to dissolution of calcite and galena by nitric acid solution. When a fire assay was carried out with insoluble-residue, it was discovered that gold contents of the solid-residue were 1.3 (Au = 2,464.70 g/t) and 28.8 (52,952.80 g/t) times more than those of concentrate. But in the gold contents recovered, a severe gold nugget effect appeared. It is expected that the gold nugget effect will decrease if a sampling method of concentrate is improved in the microwave-nitric acid leaching experiments and filtering paper with smaller pore size is used for leaching solution and burned filter paper is used for sampling in lead-fire assay.

Hydraulic Characteristics of Mountainous Forest Soils in Korea and Applicability of Pedotransfer Functions

  • Pedotransfer functions (PTFs) were developed for each soil horizon to estimate hydraulic characteristics of mountainous forest soils in South Korea. Twenty one dominant soil series from 8 soil catenae such as granite-originated catena and volcanic ash-originated catena were selected for the study; gravel contents of selected soils were 10% or lower. Saturated conductivity (Ks) was measured for each horizon in situ. Particle size distribution and organic matter content of each horizon were also determined. Based on correlation analysis with total data set, sand separate showed positive relationship with Ks ($r=0.24^*$) while clay separate had negative relationship with Ks ($r=-0.29^{**}$). The correlation coefficients of sand, clay, and organic matter content with Ks increased to $0.41^{**}$, $-0.67^{***}$, and $0.58^{***}$, respectively, using data from granite- or gneiss-originated catena with exception of volcanic ash-originated catena and sedimentary rock-originated catena. Determination coefficients of PTFs were 0.31 for A horizon, 0.25 for B, and 0.35 for C with all data set while those were 0.74 for A, 0.48 for B, and 0.54 for C. Organic matter was a dominant factor affecting Ks in A horizon but clay content was selected as the only factor influencing Ks in C horizon. It implies that PTFs should be developed with understanding characteristics of parent materials and horizons. Developed PTFs for granite- or gneiss-originated catena were following: A horizon: Log ($K_s{\times}10^7$) = -0.031C + 0.398OM + 3.49 B horizon: Log ($K_s{\times}10^7$) = -0.028C + 0.141OM + 4.05 C horizon: Log ($K_s{\times}10^7$) = -0.072C + 4.66 where C is clay separate (%) and OM is organic matter content ($g\;kg^{-1}$). The unit of Ks is cm $sec^{-1}$.

Preparation and Characterization of Chitosan-coated PLGA Nanoparticle (키토산이 코팅된 PLGA 나노입자의 제조 및 특성)

  • In this study, poly lactic-co-glycolic acid (PLGA) nanoparticles (PNP) were prepared through double (w/o/w) emlusion and emulsifying solvent-evaporation technique using PLGA, which has biocompatibility and biodegradability. To maximize stability and bioavailability of the particles, chitosan-coated PLGA nanoparticles (CPNP) were prepared by charge interaction between PNP and chitosan. We demonstrated that CPNP can be utilized as a drug carrier of oral administration. The chemical structure of CPNP was analyzed by 1H-NMR and FT-IR, and all characteristic peaks appeared, confirming that it was successfully prepared. In addition, particle size and zeta potential of CPNP were analyzed using dynamic light scattering (DLS) while morphological images were obtained using transmission electron microscope (TEM). Thermal decomposition behavior of CPNP was observed through thermogravimetric analysis (TGA). In addition, the cytotoxicity of CPNP was confirmed by MTT assay at HEK293 and L929 cell lines, and it was proved that there is no toxicity confirmed by the cell viability of above 70% at all concentrations. These results suggest that the CPNP developed in this study may be used as an oral drug delivery carrier.

A Study on the Characteristics of CdS Thin Film by Annealing Time Change (열처리시간 변화에 의한 CdS 박막 특성에 관한연구)

  • This paper uses a multiplex deposition sputter system and aims to improve transmittance and reduce production costs by depositing a CdS thin film on an ITO glass substrate. When manufacturing CdS thin films, we wanted to find excellent conditions when manufacturing solar cells by changing heat treatment time. It was observed that thickness and sheet resistance were not significantly different depending on heat treatment time changes. The specific resistance was measured from a minimum of 6.68 to a maximum of 6.98. When the heat treatment time was more than 20 minutes, the transmittance was measured to be more than 75%. When the heat treatment time was 10 minutes, the bandgap was 3.665 eV and more than 20 minutes was 3.713 eV, which was measured as the same result. The XRD analysis showed that the structure of CdS was hexagonal and only CdS thin films were deposited without any other impurities. The result of calculating the FWHM was a maximum of 0.142 when the heat treatment time was 20 minutes, and a minimum of 0.133 when the heat treatment time was 40 minutes, so there was no significant difference in the FWHM when the heat treatment time was changed. The particle size was measured at 11.65 Å when the heat treatment time was 40 minutes, and at 10.93 Å when the heat treatment time was 20 minutes.

Anthracite Oxygen Combustion Simulation in 0.1MWth Circulating Fluidized Bed (0.1 MWth 급 순환유동층에서의 무연탄 연소 전산유체역학 모사)

  • The combustion characteristics of anthracite, which follow a complex process with low reactivity, must be considered through the dynamic behavior of circulating fluidized bed (CFB) boilers. In this study, computational fluid dynamics (CFD) simulation was performed to analyze the combustion characteristics of anthracite in a pilot scale 0.1 MWth Oxy-fuel circulating fluidized bed (Oxy-CFB) boiler. The 0.1MWth Oxy-CFB boiler is composed of combustor (0.15 m l.D., 10 m High), cyclone, return leg, and so on. To perform CFD analysis, a 3D simulation model reactor was designed and used. The anthracite used in the experiment has an average particle size of 1,070 ㎛ and a density of 2,326 kg/m3. The flow pattern of gas-solids inside the reactor according to the change of combustion environment from air combustion to oxygen combustion was investigated. At this time, it was found that the temperature distribution in air combustion and oxygen combustion showed a similar pattern, but the pressure distribution was lower in oxygen combustion. addition, since it has a higher CO2 concentration in oxygen combustion than in air combustion, it can be expected that carbon dioxide capture will take place actively. As a result, it was confirmed that this study can contribute to the optimized design and operation of a circulating fluidized bed reactor using anthracite.
