Glycosaminoglycans(PT -Gag) were isolated from the porcine testis. From the PT -Gag, we obtained two different types of Gag fractions using Dowex macro porous Resin MSA-1 column, PT -Gag-1.5% NaCl and PT -Gag-16% NaCl. Various biological activities of the GAGs were examined in aspect of anticoagulant and immunomodulating activity. The anticoagulant activity of the GAGs was evaluated by activated partial thromboplastin time (aPTT ) assay and thrombin time (TT) assay. The GAGs of porcine testis markedly incresed the clotting times of both of aPTT and TT. showing that PT-Gag-16% NaCl was more effective than PT-Gag-1.5% NaCl. The immunomodulating activityof the GAGs was examined in relation to regulation of xytoxine prodution of murine peritoeal maerophages. Taken together. GAGs isolated from porcine testis possess bilolgical functions such as anticoagulant and immunomodulating activity.

The monitoring of heparin therapy is using almost aPTT assay. This study is compare to estimating aPTT therapeutic range using in vitro heparin-spiked sample and aPTT therapeutic range using in vivo heparin-treated sample. Normal pooled plasma was collected from 20 healthy representative individuals. 11 concentration of heparinized plasmas from 0 U/mL to 1.0 U/mL at intervals of 0.1 U/mL made by addition of heparin to normal pooled plasma were measured aPTT. The aPTT therapeutic range was performed through correlation analysis between heparin level 0.2 to 0.4 U/mL and aPTT. 30 plasmas from patients on heparin therapy were measured aPTT and anti-Xa activity. The aPTT therapeutic range was performed through correlation analysis between anti-Xa activity 0.3 to 0.7 U/mL and aPTT. The aPTT therapeutic range corresponded by heparin level-vs-aPTT value regression analysis was 60.7 to 102.4 seconds. The aPTT therapeutic range corresponded by anti-Xa activity-vs-aPTT value regression analysis was 85.3 to 147.5 seconds. The validation of heparin sensitivity using in-vitro heparin sample was not considered. The establishing aPTT therapeutic range is recommended anti-Xa activity using in-vivo sample.
Background: Bleeding and thrombosis are major complications associated with high mortality in extracorporeal membrane oxygenation (ECMO) management. Anticoagulant therapy should be adequate to reduce thrombosis. However, related studies are limited. Methods: We retrospectively reviewed all patients supported with ECMO at a single institution between January 2014 and July 2022 and included those on all types of ECMO using the Permanent Life Support System. Patients were classified into two groups according to their measured mean activated partial thromboplastin time (aPTT) during ECMO management: a high-anticoagulation (AC) group (aPTT, ≥55 seconds; n=52) and a low-AC group (aPTT, <55 seconds; n=79). The primary outcome was thrombotic or bleeding events during ECMO. Results: We identified 10 patients with bleeding; significantly more of these patients were in the high-AC group (n=8) than in the low-AC group (15.4% vs. 2.5%, p=0.01). However, thrombus events and oxygenator change-free times were not significantly different between the two groups. Four patients in the high-AC group died of bleeding complications (brain hemorrhage, two; hemopericardium, one; and gastrointestinal bleeding, one). One patient in the low-AC group developed a thrombus and died of ECMO dysfunction due to circuit thrombosis. Conclusion: Heparin did not significantly improve thrombotic outcomes. However, maintaining an aPTT of ≥55 seconds was a significant risk factor for bleeding events, especially those associated with mortality.
This study was undertaken to know the effect of fat diet (for eight weeks) on changes of inflammatory markers [tumor necrosis factor (TNF-${\alpha}$) and prostaglandin $E_2$ ($PGE_2$)] and blood coagulation system [platelet aggregation function (PAF), prothrombin time (PT), activated partial thromboplastin time (aPTT)] in rats. Serum TNF-${\alpha}$, $PGE_2$, biochemical markers, PAF, PT, aPTT, and body weight were measured and compared between the control (normal diet-rats) and the fat group (fat diet-rats). The weights in the fat group were higher than those of the control group. TNF-${\alpha}$, $PGE_2$, glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT), and creatinine levels were greater in the fat group compared with the control group. The degree of platelet aggregation was lower, whereas PT and aPTT levels were longer in the fat group than in the control group. These findings have shown that fat diet may cause inflammatory response, diabetes, liver and renal dysfunction, and disturbances of fibrinolysis and coagulation system.
Oleanolic acid (OA), a triterpenoid known for its anti-inflammatory and anti-cancer properties, is commonly present in several medicinal plants but its anticoagulant activities have not been studied. Here, the anticoagulant properties of OA were determined by monitoring activated partial thromboplastin time (aPTT), prothrombin time (PT), fibrin polymerization as well as cell-based thrombin and activated factor X (FXa) generation activities. Data showed OA prolonged aPTT and PT significantly and inhibited thrombin catalyzed fibrin polymerization. In addition, OA inhibited the activities of thrombin and FXa and inhibited the generation of thrombin or FXa in human endothelial cells. OA also inhibited TNF-${\alpha}$-induced tissue factor expression on human endothelial cells. In accordance with these anticoagulant activities, OA showed an anticoagulant effect in vivo. These results indicate that OA possesses antithrombotic activities and suggest that daily consumption of a herb containing OA may be preventing thrombosis in pathological states.
Tenebrio molitor, rich in protein, is used as an alternative protein source. Many studies have evaluated Tenebrio molitor larvae (TML) for anti-obesity effects, fatty liver relief, antithrombosis, and antioxidant effects. In this study, we prepared an experimental diet by adding Tenebrio molitor larvae oil (TMLO) to feed and administered it to adult male ICR mice for six weeks. The study assessed the weight gain of mice, prothrombin time (PT), activated partial thromboplastin time (aPTT), blood coagulation time, and the activities of coagulation factor VII and coagulation factor XII. The weight gain of mice was suppressed in the groups fed with TMLO, indicating a potential anti-obesity effect. The blood coagulation time was delayed in the TMLO-fed groups, as evidenced by amplified PT and aPTT values. Furthermore, the activities of coagulation factor VII and coagulation factor XII were diminished in the TMLO-treated groups, indicating a potential thrombosis relieving effect. The concentrations of thromboxane B2 and serotonin declined in the TMLO-fed groups, signifying potential blood circulation effects. Finally, blood triglyceride and LDL-cholesterol concentrations were reduced in the TMLO-fed groups. In summary, TMLO supplementation may have an inhibitory effect on liver damage and blood clot formation caused by obesity.
We evaluated the newly developed plastic sodium citrate tubes for routine coagulation test by direct comparison with glass citrate theophylline adenosine dipyridamole (CTAD) tubes. Blood was drawn from 100 patients into glass CTAD tubes and plastic sodium citrate tubes. After collection, samples were centrifuged at 1500 ${\times}$g for 15 min at $22^{\circ}C$. Prothrombin time (PT), activated partial thromboplastin time (aPTT), fibrinogen were measured by using the Coagrex-800 (IRC, Japan). We used comparison plot by linear regression model and difference plot graphs to compare the results of the independent measurements of PT, aPTT, fibrinogen between glass CTAD tubes and plastic sodium citrate tubes. On the comparison study between glass CTAD tubes and plastic sodium citrate tubes, the correlation coefficients (R) were 0.99 for PT, 0.97 for aPTT and 0.97 for fibrinogen. This results implicated good correlation of each parameter between two tubes. Although the difference plot graph analysis showed statistically significant differences between glass and plastic tubes for PT, aPTT and fibrinogen, the range of difference was acceptable according to the CLSI/NCCLS guideline. The plastic sodium citrate tubes showed good correlation with the glass CTAD tubes, so it can substitute glass citrate tube for routine coagulation tests.
Piperlonguminine (PL), an important component of Piper longum fruits, is known to exhibit anti-hyperlipidemic, antiplatelet and anti-melanogenic activities. Here, the anticoagulant activities of PL were examined by monitoring activated-partial-thromboplastin-time (aPTT), prothrombin-time (PT), and the activities of thrombin and activated factor X (FXa). The effects of PL on the expressions of plasminogen activator inhibitor type 1 (PAI-1) and tissue-type plasminogen activator (t-PA) were also tested in tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) activated HUVECs. The results showed that PL prolonged aPTT and PT significantly and inhibited the activities of thrombin and FXa. PL inhibited the generation of thrombin and FXa in HUVECs. In accordance with these anticoagulant activities, PL prolonged in vivo bleeding time and inhibited TNF-${\alpha}$ induced PAI-1 production. Furthermore, PAI-1/t-PA ratio was significantly decreased by PL. Collectively, our results suggest that PL possesses antithrombotic activities and that the current study could provide bases for the development of new anticoagulant agents.
This study was performed to prove the clinical effects of Kyejibokryoung hwan(KBH), Jeodangtang(JDT), Kyejibokryounghwan & Jeo-dangtang(KJT) by way of experimental methods. The intravascular coagulation was induced by injection of endotoxin into the caudal vein of rats. And liquid extracts of Kyejibokryounghwan, Jeodangtang, Kyejibokryounghwan & Jeodang- tang were administerd orally to the rats. Then the number of platelets, concentration of fibrinogen, FDP(fibrin-fibrinogen degradation products), prothrombin time and PTT(partial thromboplastin time) were measured. The results were obtained as follows ; 1. The number of platelets was significantly increased in KBH and KJT-treated groups in comparison with the control group. 2. Fibrinogen was significantly increased in all sample groups as compared with the control group. 3. FDP was insignificantly decreased in all sample groups but have not significant. 4. Prothrombin time was significantly shortened in JDT and KJT-treated groups as compared with the control group. 5. PTT was significantly shortened in only KJT -treated groups as compared with the control group. From the above results, it was concluded that Kyejibokryounghwan, Jeodang tang, Kyejibokryounghwan & Jeodangtang can be applied effectively in the disease of thrombosis.
