Biological Control of Phytophthora Blight and Anthracnose Disease in Red-pepper Using Bacillus subtilis S54 (Bacillus subtilis S54 균주를 이용한 고추 역병과 탄저병의 생물학적 방제)

  • Phytophthora blight and anthracnose disease caused by Phytophthora capsici and Collectotrichum gloeosporioides are the most important devastating diseases of red pepper plants, worldwide. Five different bacterial isolates were isolated from the red pepper rhizosphere and non-rhizosphere soil and subsequently tested for antagonistic activity against P. capsisi and C. gloeosporioides. The area of the inhibition zone was taken as a measure for antagonistic activity. Among the 5 isolates tested, S54 exhibited a maximum antagonistic activity under in vitro and in vivo conditions. In greenhouse studies the isolate has successfully reduced the disease symptom. Protect value was 80.8% (Phytophthora blight) and 81.9% (Anthrancnose disease), whereas the infection rate of control plants was 21.3% and 23.2%. Based on the 16S rDNA sequence and API 50CHB Kit analysis the most effective isolate was identified as Bacillus subtilis. The results of the study indicate that the stratin S54 could be used as an potential biological control of Phytophthora blight and anthracnose disease of red pepper.

  • SCAI(spent CANDU fuel bundle serial number identifier) was developed to read serial numbers of spent fuel bundles in the spent fuel storage. For the purpose of effectively identifying the serial number of fuel bundle. SCAI was composed of underwater camera & light part. guiding & supporting part and control & monitor part. So it is easy to assemble and disassemble, and operate. It was tested to read serial numbers of spent fuel bundles loaded in basket during the recent spent fuel transfer campaign at Wolsong Unit 1. And it was also applied to read serial numbers of spent fuel bundles discharging from the initial core at Wolsong Unit 3 by slight change of camera and light. Inspectors could easily operate SCAI after several practices in the storage pond, which was a user friendly. And SCAI provided clear and immediate picture for identification of serial numbers of spent fuel bundles. It was interally evaluated that SCAI greatly contributed to cut inspection efforts for national and international safeguards at Wolsong power plant.

Effects of Browning Products and Charcoal on the Degradation of Aflatoxin B, in Korean Soy Sauce (Kanjang) and its Model System (간장 및 모델시스템에서 간장 갈색물질과 숯이 Aflatoxin $B_1$의 파괴에 미치는 영향)

  • The effects of browning Products (BP) from Kanjang(soy sauce) and charcoal on the degradation of aflatoxin $B_1(AFB_1)$ in Kanjang and its model system were studied. Approximately 60% of $AFB_1$ was degraded in the presence of 0.05% BP at pH 7 of phosphate buffer after 2 days of incubation at $30^{\circ}C$. The mutagenicity of the $AFB_1$ which reacted with the BP was decreased to about 50% and 70% in Salmonella typhimurium TA98 and TA100 strains, respectively (p<0.05). When a few pieces of charcoal were added to home made Kanjang, $AFB_1$ was quite stable for 5days at $30^{\circ}C$, however, about 80% of $AFB_1$ was removed when the charcoal was either in distilled water or in 20% of NaCl solution after 2 days of incubation. Activated carbon instead of the charcoal removed $AFB_1$ completely in the all samples under the same conditions.

Physicochemical Properties and Physiological Activities of Ulsan Sweet Persimmon Peel.Flesh According to Cultivars (울산지역 생산 단감 과피.과육의 품종별 이화학적 특성 및 생리활성)

  • This study was performed to confirm the physicochemical properties and physiological activities according to cultivar and parts of Ulsan sweet persimmons. The proximate composition of the sweet persimmons was moisture $77.68{\sim}84.65%$, crude ash $0.3{\sim}0.6$, crude fat $0.29{\sim}0.39$ and crude protein $0.52{\sim}0.61%$. Confirmed organic acids and free sugars in sweet persimmons were malic, citric, succinic, and fumaric acids, along with fructose, glucose and sucrose. The major fatty acids were linolenic, vaccenic and palmitic acids. Also, K is the highest mineral in sweet persimmons that accounts for the majority of mineral, more than 60% as $84.9{\sim}103.7mg%$. Contents of vitamin C, total phenolic compounds and flavonoids were the highest in Fuyu peel as 139.91, 169.36 and 60.79mg%, respectively, Electron donating ability and xanthine oxidase inhibition were the highest in Fuyu peel as $30.6{\sim}91.2$ and $26.3{\sim}91.9mg%$, respectively. Nitrite scavenging activity was higher in peel than flesh as $51.0{\sim}76.4mg%$ with slight difference depending on cultivar.

  • The repeated silica gel colum chromatographies of EtOAc fraction, showing anticonvulsant activity, obtained from MeOH extracts of Schizandra chinensis B. fruits led to isolation of a sesquiterpenoid, four lignans and a sterol glycoside. Their chemical structures were determined to be chamigrenal, gomisin A, gomisin H, gomisin N. schizandrin and daucosterol. Among them, schizandrin and daucosterol inhibited GABA degrative enzymes, succinic semialdehyde dehydrogenase and succinic semialdehyde reductase, respectively. It is postulated that the schizandrin and daucosterol are able to elevate the neurotransmitter GABA levels in central nervous system by inhibitory action on GABA degrative enzymes and act as anticonvulsant drugs.

Vascular Relaxation Induced by the Water Soluble Fraction of the Seeds from Oenothera Odorata (월견자 물 분획층을 이용한 혈관이완 기전에 관한 연구)

  • In the present study, vasorelaxant effect of the extract of seeds of Oenothera odorata (SOO) and its possible mechanism responsible for this effect were examined in vascular tissues isolated from rats. Changes in vascular tension, 3',5'-cyclic monophosphate (cGMP) levels were measured in thoracic aorta rings from rats. Methanol extract of seeds of Oenothera odorata relaxed endothelium-intact thoracic aorta in a dose-dependent manner. A dose-dependent vascular relaxation was also revealed by treatment of ethylacetate, n-butanol, and H2O (aqua extract of seeds of Oenothera odorata , ASOO) extracts partitioned from methanol, but not by hexane extract. However, the vascular relaxation induced by ASOO were abolished by removal of endothelium of aortic tissues. Pretreatment of the endothelium-intact vascular tissues with NG-nitro-L-arginine methyl ester (L-NAME) or 1H-[1,2,4]-oxadiazole-[4,3-α]-quinoxalin-1- one (ODQ) significantly inhibited vascular relaxation induced by ASOO. Moreover, incubation of endothelium-intact aortic rings with ASOO increased the production of cGMP. However, ASOO-induced increases in cGMP production were blocked by pretreatment with L-NAME or ODQ. The vasorelaxant effect of ASOO was attenuated by tetraethylammonium (TEA), 4-aminopyridine, and glibenclamide attenuated. On the other hand, the ASOO-induced vasorelaxation was not blocked by verapamil, and diltiazem. Taken together, the present study demonstrates that ASOO dilate vascular smooth muscle via endothelium-dependent NO-cGMP signaling pathway, which may be closely related with the function of K+ channels.

Evaluation of flexible criteria for river flow management with consideration of spatio-temporal flow variation (시·공간적 유량 변화를 고려한 탄력적 하천관리 기준유량 산정 및 평가)

  • An Idea to estimate flexible criteria for river water use permits was proposed that takes the spatio-temporal flow variation along the river into account, which was applied to the Keumho River, one of the tributary of the Nakdong River in Korea. This idea implies the temporal division of four periods with different criteria, combining flood/non-flood seasons and irrigation/non-irrigation periods, while a single one has been applied throughout the year in the current practice. Through flow regime analysis of daily natural flow simulations at Dongchon and Seongseo, the control points of the study area, Q355 and 1Q10 for non-flood and non-irrigation period, Q275 for non-flood and irrigation period, Q185 for flood and irrigation period were suggested respectively. So, those values that subtract instream flow were determined as the flexible criteria in each season. From the comparison of current practice and the proposed method, it was estimated that $10.6\;million\;m^3/year$ is available for more water use permits without additional development of water storage. Therefore, it is conceived that flexible criteria for river water use permission suggested in this study can contribute to improve the national policies for more efficient water resources management in the future.

Comparison of anti-inflammatory, antioxidant and anti-allergic effects of Ganoderma species mycelial extracts (영지버섯 균주별 균사체 추출물의 항염, 항산화 및 항알러지 효능 비교 분석)

  • This study wascarried out to compare the medicinal effects of various Ganoderma species mycelial extracts. Among 6 Ganoderma species mycelial extracts by using 100% MeOH, G. species ASI-7150 showed the highest antioxidant effect. In nitric oxide (NO) production and ${\beta}$-hexosaminidase release inhibition assay, the treatment of G. lucidum ATCC64251 (Taiwan) mycelia extracts most effectively inhibited NO production in LPS-stimulated RAW264.7 cells and ${\beta}$-hexosaminidase release. In addition, the treatment of all 6 Ganoderma species mycelial extracts were not affect on RAW264.7 cell viability. Although this preliminary research has thrown up many questions in need of further investigation, it will serve as a base for further studies of medicinal effects of various Ganoderma species.

Genetic Toxicity Test of o-Nitrotoluene by Ames, Micronucleus, Comet Assays and Microarray Analysis

  • o-Nitrotoluene is used to synthesize artificial dyes and raw materials of urethane resin. In this study, we have carried out in vitro genetic toxicity tests and microarray analysis to understand the underlying mechanisms and the mode of action of toxicity of onitrotoluene. TA1535 and TA98 cells were treated with o-nitrotoluene to test its toxicity by basic genetic toxicity test. Ames and two new in vitro micronucleus and COMET assays were applied using CHO cells and L5178Y cells, respectively. In addition, microarray analysis of differentially expressed genes in L5178Y cells in response to o-nitrotoluene was analyzed using Affymatrix genechip. The result of Ames test was that o-nitrotoluene treatment did not increase the mutations both in base substitution strain TA1535 and in frame shift TA98. o-Nitrotoluene has not increased micronuclei in CHO cells. But onitrotoluene increased DNA damage in L5178Y cell. Two-hundred two genes were initially selected as differentially expressed genes in response to o-nitrotoluene by microarray analysis and forty four genes among them were over 2 times of log fold changed. These forty four genes could be candidate biomarkers of genetic toxic action of o-nitrotoluene related to induction of mutation and/or induction of micronuclei and DNA damage. Further confirmation of these candidate markers related to the DNA damage will be useful to understand the detailed mechanism of action of o-nitrotoluene.

Three-Dimensional Analysis of the Mesophyll Plastids Using Ultra High Voltage Electron Microscopy (초고압전자현미경에 의한 엽육세포 색소체 미세구조의 3차원적 분석)

  • Image processing by ultra high voltage electron microscopy (UHVEM) and tomography has offered major contributions to research in the field of cellular ultrastructure. Furthermore, such advancements also have enabled the improved analysis of three-dimensional cellular structures in botany. In the present study. using UHVEM and tomography, we attempted to reconstruct the three-dimensional images of plastid inclusions that probably differentiate during photosynthesis. The foliar tissues were studied Primarily with the TEM and further examined with UHVEM. The spatial relationship between tubular elements and the thylakoidal membrane and/or starch grains within plastids mainly have been investigated in CAM-performing Sedum as well as in $C_4$ Salsola species. The inclusion bodies were found to occur only in early development in the former, while they were found only in mesophyll cells in the latter. The specimens were tilted every two degrees to obtain two-dimensional images with UHVEM and subsequently comparison has been made between the two types. Digital image processing was performed on the elements of the inclusion body using tilting, tomography, and IMOD program to generate and reconstruct three-dimensional images on the cellular level. In Sedum plastids, the inclusion bodies consisted of tubular elements exhibiting about 20 nm distance between elements. However, in Salsola, plastid inclusion bodies demonstrated quite different element structure, displaying pattern, and origin relative to those of the Sedum. The inclusion bodies had an integrative relationship with the starch grains in both species.
