Community Analysis and Pathogen Monitoring in Wild Cyprinid Fish and Crustaceans in the Geum River Estuary (금강 하구 자연수계 생물체의 군집 분석 및 질병 원인체 검사)

  • Freshwater farms are primarily located adjacent to rivers and lakes, facilitating the introduction and spread of pathogens into natural systems. Therefore, it is necessary to continuously monitor natural aquatic organisms, the breeding environment, and infection rates by pathogenic organisms. Fish and crustaceans were sampled 4 times in the Geum River estuary in 2016. The samples were analyzed for the presence of pathogens for reportable communicable diseases, including KHVD (koi herpesvirus disease), SVC (spring viraemia of carp), EUS (epizootic ulcerative syndrome) and WSD (white spot disease); parasite abundance was also examined. The dominant fish species were deep body bitterling Acanthorhodes macropterus (21.4%), followed by skygager Erythroculter erythropterus (12.7%). For crustaceans, Palaemon paucidens and Chinese mitten crab Eriocheir sinensis were dominant. Sixty fish and 36 crustacean species were examined for reportable communicable diseases. When using a specific primer set for each disease, PCR analysis did not detect any reportable communicable diseases in the samples. Some instances of Dactylogyrus, copepods, nematodes and metacercaria were detected. However, the PCR results indicated that the metacercaria were not Clonorchis sinensis.

Characterization of the Myxosporean Parasite Isolated from Emaciated Olive Flounders Paralichthys olivaceus on Jeju Island (제주도 여윔증상 넙치(Paralichthys olivaceus)로부터 분리한 점액포자충의 특성 분석)

  • To investigate the causes of emaciation in cultured olive flounder Paralichthys olivaceus in Korea. We performed histological examinations and polymerase chain reaction (PCR) with a new primer set. In most cases, the most severe emaciation was observed in the abdominal area Using PCR on extracted livers, kidneys, spleens, gills, brains, and intestines, we found that areas around the kidneys and intestines were as almost always positive. In significantly emaciated fish, PCR was positive in all internal organs except the gills. In addition, the homology of 812-bp nucleotide sequences of the 28S rRNA gene was more than 99% in emaciated fish. Partial homology with Myxobolus spp. and Cystodiscus axonis, whose data were obtained from GenBank was 86% and 88%, respectively. Histological examinations detected spores in kidneys and intestines but not in other organs. We also performed cohabitation experiments to determine whether infections could be exchanged among species or only within species. Uninfected olive flounder and red sea bream, Pagrus major, cohabitating with emaciated olive flounder showed 100% and 0% cumulative mortality, respectively. Thus the cause of emaciation in cultured olive flounder of Korea is likely due to a new parasite.

First report and characteristics study of Cymothoids isolated from cultured flathead grey mullet (Chelon haematocheilus) (양식 가숭어(Chelon haematocheilus)에서 최초로 분리된 갈고리벌레과 Cymothoids의 특성 연구)

  • Mullet is an important marine aquaculture fish species in Korea, with a total of 7,237 tons produced as of 2022, making it the 5th most produced marine aquaculture fish species. In this study, ectoparasites presumed to be isopods were discovered in the fins of farmed flathead grey mullet (average weight 550 g), and the characteristics of the parasites were confirmed. The length of the parasite was 5 to 18 mm, and 3 to 7 parasites were infected per fish. To analyze the characteristics of the parasites, molecular biological identification and phylogenetic analysis were performed using the cytochrome c oxidase subunit I (COI) gene, and it was confirmed to be most closely related to Nerocila japonica in the Cymothoidae family. To confirm the parasite control effect, a direct exposure drug sensitivity test was conducted on five types of aquatic drugs and fresh water, trichlorfon was confirmed to be effective.

Failure of Toxoplasma - Vaccination in Mice Born to Immune Mothers (Toxoplasma 면역모체로 부터 출산된 신생마우스에 있어서 Vaccination 효능 저하)

  • Female ICR mice were vaccinated against Toxoplasma gondii(RH strain), infected 2 weeks later and after recovery mated to normal ICR males. Control matings were with normal ICR females. The progeny of the above matings were weaned at 1 week, vaccinated at 1, 2, 3, 4 or 5 weeks of age and infected 2 weeks later with lethal Toxoplasma tachyzoites. As assessed by survival, the effectiveness of vaccination among offspring of vaccinated-recovered mothers was greatly impaired than that of control mice, with respective of age : where mice did survive, recovery was greatly delayed relative to the controls. The protective effect of vaccination among infants born to control mothers was also blocked by maternal specific antibodies, by administration of high-titered specific antibodies or by transfer of nylon wool adherent immune-spleen cells, but was augmented by transfer of nylon wool passed immune-spleen cells. These results indicate that this impairment of vaccination may be due to the transmission of maternal specific antibodies to the offspring which acts to suppress both priming by the vaccine and the generation of parasite-specific helper T cells.

On the Control Effects of Some Chemicals to Rice White-tip Nematode (Aphelenchoides os besseyi) after Transplanting of Rice (모낸 후의 벼이삭 선충(Aphelenchoides besseyi)에 대한 약제방제에 관하여)

  • Field experiments were carried out to find out effective chemicals and number and time of a chemical application for the control of the rice white-tip nematode, Aphelechoides besseyi, parasite on rice plant. Ethoprop and carbofuran were significantly effective to make increase as much as $19\%,\; or\; 260g/3.3m^2$ of unhulled grain yield. Application of carbofuran as soon as the white-tip symptom of the disease appears in field is recommended for the effective control of the disease.

Malondialdehyde, Glutathione, and Nitric Oxide Levels in Toxoplasma gondii Seropositive Patients

  • The aim of this study was to investigate the difference in the serum malondialdehyde (MDA), glutathione (GSH), and nitric oxide (NO) levels between normal and T. gondii-infected patients. To this end, MDA, GSH, and NO levels in the sera of 37 seropositive patients and 40 participants in the control group were evaluated. In Toxoplasma ELISA, IgG results of the patient group were $1,013.0{\pm}543.8$ in optical density (mean ${\pm}$ SD). A statistically significant difference was found between patients and the control group in terms of MDA, GSH, and NO levels. A decrease in GSH activity was detected, while MDA and NO levels increased significantly. Consequently, it is suggested that the use of antioxidant vitamins in addition to a parasite treatment shall prove useful. The high infection vs control ratio of MDA and NO levels probably suggests the occurrence as a mechanism of tissue damage in cases of chronic toxoplasmosis. Moreover, it is recommended that the patient levels of MDA, GSH, and NO should be evaluated in toxoplasmosis.

  • Phytophagous insects were surveyed for biological control of weeds in the southern part of korea from April 20 to July 31, 1987. Phytophagous coleopteran insects representing 12 families, 52 genera and 61 species, hemipteran insects 7 families, 18 genera and 20 species, homopteran insects 4 families, 16 genera and 21 species, hymenopteran insects 2 families, 2 genera and 3 species, and orthopteran 2 familes, 6 genera and 6 species were recorded from 58 weeds. Some chrysomelids and orthopteran insects were potential agents for biological control of weeds.

  • Plasmodium vivax is more challenging to control and eliminate than P. falciparum due to its more asymptomatic infections with low parasite densities making diagnosis more difficult, in addition to its unique biological characteristics. The potential re-introduction of incidence cases, either through borders or via human migrations, is another major hurdle to sustained control and elimination. The Republic of Korea has experienced re-emergence of vivax malaria in 1993 but is one of the 32 malaria-eliminating countries to-date. Despite achieving successful nationwide control and elimination of vivax malaria, the evolutionary characteristics of vivax malaria isolates in the Republic of Korea have not been fully understood. In this review, we present an overview of the genetic variability of such isolates to increase understanding of the epidemiology, diversity, and dynamics of vivax populations in the Republic of Korea.

Anti-Parasitic Activity of Lespedeza cuneata Extract on Causative Agent of Nosemosis Type C, Nosema ceranae

  • Although honeybees (Apis mellifera) are crucial for maintenance of the ecosystem, population of honeybee has been steadily decreasing due to diseases including nosemosis. Nosemosis is a disease caused by Nosema ceranae and is now considered as a major threat to honeybees. N. ceranae is a microsporidian that stays in form of spore even before the infection, which makes it harder to control than other pathogens. People are now aware of this parasite, however, cure and preventive candidates for nosemosis are hardly found until today. In this study, in vitro experiment of Lespedeza cuneata treatment to prevent nosemosis were done using Trichoplusia ni cell line, BTI-TN5B1-4. Normal T. ni cells exhibited round shape without abnormal size. On the other hand, when N. ceranae were treated, cells deteriorated and some cells abnormally enlarged due to N. ceranae infection. Interestingly, treatment of T. ni cells with L. cuneate extract protected abnormal cell shape induced by N. ceranae infection to normal shape. Some N. ceranae spores were observed outside of the cells. Effective concentration range for N. ceranae control were experimented. Lowest concentration which can control nosemosis were 50 ㎍/mL. When the concentration of L. cuneata extract was exceeded 200 ㎍/mL, cytotoxicity started to show up.

Possibility of non-invasive diagnostic method for Kudoa septempunctata using a hyperspectral camera

  • Kudoa septempunctata, a myxozoan parasite, usually presents without any signs and primarily infects adult fish. The invasive diagnostic methods, such as tissue biopsy, can identify pathogens, but cause economic losses because they require killing the fish. In this study, we conducted a monitoring of four fish farms located on Jeju Island, to investigate the potential for non-invasive diagnosis of K. septempunctata using hyperspectral cameras. It provides spectral information from R000_B000_G000 to R255_B255_G255 for a total of 3,282 olive flounder (Paralichthys olivaceus). Each object is imaged with 2,000 data points, allowing comprehensive spectral analysis by comparing images obtained from negative control objects to positive control objects. Noticeable differences were observed in the brightness or pallor of the positive control images. This suggests the potential utility of hyperspectral imaging as a non-invasive diagnostic tool for detecting K. septempunctata infections in fish populations.
