Effects of Rubus coreanus and Artemisia princeps Extracts on the Ultraviolet B-Induced DNA Damage Responses in HaCaT Cells (HaCaT 세포에서 자외선 B에 의해 유도된 DNA 상해반응에 대한 복분자와 쑥 추출물의 효과)

  • We investigated the effects of extracts from Rubus coreanus (RC) and Artemisia princeps var. orientalis (AP) on DNA damage response in ultraviolet B (UVB)-exposed HaCaT cells. Cell activity upon treatment for 24 h with RC or AP alone was similar to or greater than that of the nontreated control. When UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, cell activity increased in a concentration-dependent manner. Nuclear fragmentation analysis showed that postincubation with RC or AP decreased UVB-induced apoptosis by about 20% and 15%, respectively, of that in cells postincubated with growth medium. When UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, the level of cyclobutane pyrimidine dimer decreased in a concentration- dependent manner. Western blot analysis showed that treatment of cells not exposed to UVB with RC or AP alone did not significantly change the levels of phospho-p53 and GADD45 protein. Interestingly, when UVB-exposed cells were postincubated for 24 h in medium containing RC or AP, phospho-p53 and GADD45 levels decreased in a concentration dependent manner. Our results suggest that RC and AP extract assist the survival of UVB-exposed cells in parallel with a decrease in levels of UVB-induced DNA damage and damage-response proteins, such as p53 and GADD45.

Increase in the Th1-Cell-Based Immune Response in Healthy Workers Exposed to Low-Dose Radiation - Immune System Status of Radiology Staff

  • Objectives: Radiation is one of the most important sources of free radical (such as reactive oxygen species) production, which plays an essential role in the etiology of over hundred diseases. The aim of the study was to investigate some immune parameters and hematological indices in healthy workers of the Radiology Department, University Hospital of Mashhad, Iran. Methods: The study was performed on 50 healthy workers: 30 radiology staff as the case group and 20 laboratory workers as the control group. The radiation dose received by the radiology staff participating in the study was less than the annual maximum permissible level, 50 millisievert. Hematological parameters, lymphocyte proliferation and cytokine production were studied in both groups. Results: Among healthy radiology workers, the hematological indices did not differ statistically; however, their proliferation indices and $IFN-{\gamma}$ levels showed significant increases in parallel with decreases in the IL-4 levels as compared to controls. The immune system of workers exposed to low-dose ionizing radiation was found to be shifted from a Type 2 to a Type 1 response to promote cellular immunity. Conclusion: Based on our data, exposure to low-dose ionizing radiation may decrease the prevalence, frequency, and recurrence of various cancers and infectious diseases because of an increase in Th1-cell-based response, thus leading to more protection of the human body against tumor cells and foreign agents and possibly increased longevity. However, due to high rate of fluoroscopy use for interventional radiology, we suggest continuing research projects on radiation protection and hazards to prevent irreversible damage. As a recommendation, in future studies, radiology staff with a weakened immunity due to high radiation exposure should be considered as good choices to be treated using acupuncture techniques because acupuncture has been demonstrated to enhance the function and the number of immune cells.

Effect of Organic Materials and the Removal of Apical Shoot on Controlling Cucumber Downy Mildew (유기농업자재와 순지르기를 이용한 오이 노균병 방제)

  • This study investigated the effect of organic materials (Bordeaux, Loess-sulfur) and the removal of apical shoot against downy mildew disease on cucumber cultivated in greenhouse. Five kinds of Bordeaux were made by adjusting mixing ratio of lime and copper sulfate in order to elucidate the optimal combination. The 4-6type Bordeaux was selected as the most effective combination for controlling cucumber downy mildew. Loess-sulfur showed inhibitory activity against cucumber downey mildew, but it was less effective than Bordeaux. It was confirmed that apical shoot cutting could reduce the incidence of cucumber downy mildew disease by 56.3%. When apical shoots of susceptible cucumber variety were cut at different leaf stages, disease incidence by early apical shoot cutting treatment was lower than that of late apical shoot cutting treatment. However in a resistant variety, 'Heukryungsamcheok', disease incidences of all cucumber apical shoot cutting treatments were lower than that of non-cutting treatment, but there was no differences between apical shoot cutting treatments due to low disease incidences. In addition, when organic materials and apical shoot cutting treatment were carried out in parallel, the combined treatments of organic materials and apical shoot cutting showed low disease incidence of cucumber downy mildew compared to untreated control. The lowest disease incidence of cucumber downy mildew was recorded in the combined treatment of 4-6type Bordeaux and apical shoot cutting. This study confirmed that apical shoot cutting can reduce the disease incidence of cucumber downy mildew and the combined treatment of apical shoot cutting and organic materials showed higher suppressive effect against cucumber downy mildew.

Evaluation of Physical Properties of Korean Pine (Pinus koraiensis Siebold & Zucc.) Lumber Heat-Treated by Superheated Steam (과열증기 열처리 잣나무재의 물성 평가)

  • In this study, the method for heat treating wood using superheated steam (SHS) was designed and applied. The physical and mechanical properties of Korean Pine (Pinus koraiensis Siebold & Zucc.) lumber heat-treated by SHS at $170^{\circ}C$ and 0.4 MPa for 10 hours were compared with those of non-treated and normal heat-treated wood. The amount of adsorbed water and equilibrium moisture content of the SHS treated wood were lower than non-treated wood. On the other hand the compressive strength parallel to grain and the bending strength of SHS treated wood were higher than those of non-treated wood. The hygroscopicity of SHS treated wood was similar to normal heat treated wood at $220^{\circ}C$. Internal checks that often occur during normal heat treatment were not developed at SHS treatment. Also, SHS treatment are effective in control of internal checks occurrence and resin exudation.

Immunochemical Studies for the Characterization of Purified $(Na^+,\;K^+)-ATPase$ and Its Subunits with a Special Reference of Their Effect on Monovalent Cation Transport in Reconstituted $(Na^+,\;K^+)-ATPase$ Vesicles

  • A highly purified $(Na^+,\;K^+)-ATPase$ from the rectal gland of Squalus acanthias and from the electric organ of Electrophorus electricus has been used to raise antibodies in rabbits. The 97,000 dalton catalytic subunit and glycoprotein derived from the rectal gland of spiny shark were also used as antigens. The two $(Na^+,\;K^+)-ATPase$ holoenzymes and the two shark subunits were antigenic. In Ouchterlony double diffusion experiments, these antibodies formed precipitation bands with their antigens. Antibodies prepared against the two subunits of shark holoenzyme also formed precipitation bands with their antigens and shark holoenzyme, but not with eel holoenzyme. These observations are in good agreement with inhibitory effect of these antibodies on the catalytic activity of $(Na^+,\;K^+)-ATPase$ both from the shark and the eel, since there is very little cross-reaction between the shark anticatalytic subunit antibodies and the eel holoenzyme. The maximum antibodies titer of the anticatalytic subunit antibodies is found to be 6 weeks after the initial single exposure to this antigen. Multiple injections of the antigen increased the antibody titer. However, the time required to produce the maximum antibody titer was approximately the same. These antibodies also inhibit catalytic activity of $(Na^+,\;K^+)-ATPase$ vesicles reconstituted by a slow dialysis of cholate after solubilization of the enzyme in a presonicated mixture of cholate and phospholipid. In these reconstituted $(Na^+,\;K^+)-ATPase$ vesicles, effects of these antibodies on the fluxes of $Na^+$, $Rb^+$, and $K^+$ were investigated. Control or preimmune serum had no effect on the influx of $^{22}Na^+$ or the efflux of $^{86}Rb^+$. Immunized sera against the shark $(Na^+,\;K^+)-ATPase$ holoenzyme, its glycoprotein or catalytic subunit did inhibit the influx of $^{22}Na^+$ and the efflux of $^{86}Rb^+$. It was also demonstrated that these antibodies inhibit the coupled counter-transport of $Na^+$ and $K^+$ as studied by means of dual labeling experiments. However, this inhibitory effect of the antibodies on transport of ions in the $(Na^+,\;K^+)-ATPase$ vesicles is manifested only on the portion of energy and temperature dependent alkali metal fluxes, not on the portion of ATP and ouabain insensitive ion movement. Simultaneous determination of effects of the antibodies on ion fluxes and vesicular catalytic activity indicates that an inhibition of active ion transport in reconstituted $(Na^+,\;K^+)-ATPase$ vesicles appears to be due to the inhibitory action of the antibodies on the enzymatic activity of $(Na^+,\;K^+)-ATPase$ molecules incorporated in the vesicles. These findings that the inhibitory effects of the antibodies specific to $(Na^+,\;K^+)-ATPase$ or to its subunits on ATP and temperature sensitive monovalent cation transport in parallel with the inhibitory effect of vesicular catalytic activity by these antibodies provide direct evidence that $(Na^+,\;K^+)-ATPase$ is the molecular machinery of active cation transport in this reconstituted $(Na^+,\;K^+)-ATPase$ vesicular system.

  • Construction of focused library of 2-benzylimino-1,3-thiazolines 7 through molecular modification of 3-alkyl-2-phenylimino-1,3-thiazolines 1 which showed selective fungicidal activity against rice blast and their fungitoxic activity against 6 kinds of typical plant diseases was described. Fifty four compounds of focused library of 2-benzylimino-1,3-thiazolines 7 were synthesized from the reaction of the corresponding $\gamma$-chloroacetoacetanilides 6 with N-benzyl thioureas 5 by parallel synthetic methodology. As results of fungicidal screening against the typical plant diseases, twenty eight kinds of 7 at 100 ${\mu}g$ $mL^{-1}$ showed the control value over 50% against tomato late blight.

  • Most of the recent researches for ATM switches have been based on multistage interconnection network known as regularity and self-routing property. These networks can switch packets simultaneously and in parallel. However, they are blocking networks in the sense that packet is capable of collision with each other Mainly Banyan network have been used for structure. There are several ways to reduce the blocking or to increase the throughput of banyan-type switches: increasing the internal link speeds, placing buffers in each switching node, using multiple path, distributing the load evenly in front of the banyan network and so on. Therefore, this paper proposes the use of recirculating shuffle-exchange network to reduce the blocking and to improve hardware complexity. This structures are recirculating shuffle-exchange network as simplified in hardware complexity and Rank network with tree structure which send only a packet with highest priority to the next network, and recirculate the others to the previous network. after it decides priority number on the Packets transferred to the same destination, The transferred Packets into banyan network use the function of self routing through decomposition and composition algorithm and all they arrive at final destinations. To analyze throughput, waiting time and packet loss ratio according to the size of buffer, the probabilities are modeled by a binomial distribution of packet arrival. If it is 50 percentage of load, the size of buffer is more than 15. It means the acceptable packet loss ratio. Therefore, this paper simplify the hardware complexity as use of recirculating shuffle-exchange network instead of bitonic sorter.

Biochemical Analysis of Interaction between Kringle Domains of Plasminogen and Prion Proteins with Q167R Mutation

  • The conformational change of cellular prion protein ($PrP^C$) to its misfolded counterpart, termed $PrP^{Sc}$, is mediated by a hypothesized cellular cofactor. This cofactor is believed to interact directly with certain amino acid residues of $PrP^C$. When these are mutated into cationic amino acid residues, $PrP^{Sc}$ formation and prion replication halt in a dominant negative (DN) manner, presumably due to strong binding of the cofactor to mutated $PrP^C$, designated as DN PrP mutants. Previous studies demonstrated that plasminogen and its kringle domains bind to PrP and accelerate $PrP^{Sc}$ generation. In this study, in vitro binding analysis of kringle domains of plasminogen to Q167R DN mutant PrP (PrPQ167R) was performed in parallel with the wild type (WT) and Q218K DN mutant PrP (PrPQ218K). The binding affinity of PrPQ167R was higher than that of WT PrP, but lower than that of PrPQ218K. Scatchard analysis further indicated that, like PrPQ218K and WT PrP, PrPQ167R interaction with plasminogen occurred at multiple sites, suggesting cooperativity in this interaction. Competitive binding analysis using $\small{L}$-lysine or $\small{L}$-arginine confirmed the increase of the specificity and binding affinity of the interaction as PrP acquired DN mutations. Circular dichroism spectroscopy demonstrated that the recombinant PrPs used in this study retained the ${\alpha}$-helix-rich structure. The ${\alpha}$-helix unfolding study revealed similar conformational stability for WT and DN-mutated PrPs. This study provides an additional piece of biochemical evidence concerning the interaction of plasminogen with DN mutant PrPs.

A Study on the Effects to the Change of Social Capital from Participation in Elderly Employment Programs (노인일자리사업 참여가 사회적 자본 변화에 미치는 영향 연구)

  • This study has been conducted to assess the effectiveness of the elderly employment programs through change of social capital by understanding how the elderly employment programs in the elderly employment policy of Korea has effected the change of social capital. The method of research was to design the research in the nonequivalent control groups design from the quasi-experimental design for the 500 elderly who participated and 250 who have not participated in the elderly employment programs through distributing structured questionnaire by classifying into before and after the project during 7 months from March 1, 2010 to September 30, 2010 to those above 65 living in Changwon. The social capital which is an important variable among the measurement index of the study has been set as trust, network, and social participation fields based on the aforementioned theoretic background. When the fact that there is a partial difference in the change of social capital of the elderly according to the participation in the elderly employment programs and the timing (before/after) is looked at in terms of change of social capital after controlling the general character of the elderly and the pre-score, there are differences per timing according to the participation in the elderly employment programs. Thus, the participation in the elderly employment programs had a significant difference in the change of social capital. It is possible to see that the trust, network, and social participation of the social capital all increased in the elderly with difficulties in their living status. It can be seen that the participation in the elderly employment programs brings forth increase of social capital for the elderly with difficulties in their living status. Among the social capitals, the elderly who participated in the elderly employment programs showed that the trust increased the most. Consequently, in order to increase the comprehensive social capital, the programs must be carried out focused on the vulnerable class in parallel to the elderly employment programs for the general elderly of other classes.

Report of the 3rd Japan-Korea Workshop on Acupuncture and EBM;Protocol development for the acupuncture trial on the osteoarthritis of the knee

  • The 3rd Japan-Korea Workshop on Acupuncture and EBM was held at Kanazawa on June $16^{th}$. From Korea team, 4 papers were presented. Dr. Hahn introduced a new approach of data analysis on series of n-of-1 trials using the Bayesian statistics. It offered important information for the future n-of-1 trials. Dr. Park clearly demonstrated the significance of various sham devices proposed and stressed the importance of research questions when we choose the control intervention in RCT. Dr. Lee reported the results of survey in Korean Medical Doctors (KMD) for their point selection and techniques to the distal and local points. Dr. Kim presented the results of face to face survey on the KMD with 28 items for acupuncture treatment on the knee OA. Finally, a draft of protocol was introduced by Dr. Kim. The title was "multi-center, a randomized, single blinded, two arms, parallel-group study to compare the effectiveness and safety of 'individualized acupuncture' and 'standardized minimal acupuncture' in Korean and Japanese patients with knee osteoarthritis (Phase IV)". From Japan team, 7 speakers presented their comments and proposals on the protocol. Dr. Takahashi introduced several issues regarding n-of-1 trials and pointed out the importance of obtaining generalizability from n-of-1 trials. Dr. Shichidou pointed the importance of research design, selection of outcome measures and reduction of biases. Dr. Itoh presented the results of point selection for the knee OA based on the literature survey. Dr. Sumiya introduced several differences between KMD and Japanese acupuncturists based on the questionnaire used in KMD survey. Dr. Furuya demonstrated a result of press tack needle and its sham device on shoulder stiffness. Dr. Yamashita introduced the results of literature survey regarding adverse events occurred by acupuncture on knee OA. Dr.Tsukayama stressed the importance of responsibility of Institutional Review Board (IRB) for the conduction of clinical trials. After several issues were discussed, the need of continued meeting for final protocol development was agreed, then the workshop was closed.
