Profiling of endogenous metabolites and changes in intestinal microbiota distribution after GEN-001 (Lactococcus lactis) administration

  • This study aimed to identify metabolic biomarkers and investigate changes in intestinal microbiota in the feces of healthy participants following administration of Lactococcus lactis GEN-001. GEN-001 is a single-strain L. lactis strain isolated from the gut of a healthy human volunteer. The study was conducted as a parallel, randomized, phase 1, open design trial. Twenty healthy Korean males were divided into five groups according to the GEN-001 dosage and dietary control. Groups A, B, C, and D1 received 1, 3, 6, and 9 GEN-001 capsules (1 × 1011 colony forming units), respectively, without dietary adjustment, whereas group D2 received 9 GEN-001 capsules with dietary adjustment. All groups received a single dose. Fecal samples were collected 2 days before GEN-001 administration to 7 days after for untargeted metabolomics and gut microbial metagenomic analyses; blood samples were collected simultaneously for immunogenicity analysis. Levels of phenylalanine, tyrosine, cholic acid, deoxycholic acid, and tryptophan were significantly increased at 5-6 days after GEN-001 administration when compared with predose levels. Compared with predose, the relative abundance (%) of Parabacteroides and Alistipes significantly decreased, whereas that of Lactobacillus and Lactococcus increased; Lactobacillus and tryptophan levels were negatively correlated. A single administration of GEN-001 shifted the gut microbiota in healthy volunteers to a more balanced state as evidenced by an increased abundance of beneficial bacteria, including Lactobacillus, and higher levels of the metabolites that have immunogenic properties.

  • This paper presents design and operational characteristics of 150 MW pulse power system for high current pulse forming network to control trigger time. The system is composed of two capacitor bank modules. Each capacitor bank module consist of a trigger vacuum switch, 9k 33kJ capacitor, an energy dump circuit, a crowbar circuit and a pulse shaping inductor and is connected in parallel. It is controlled by trigger controller to select operational module and determine triggering time. Pspice simulation was conducted about determining parameters of components such as crowbar circuit, capacitor, pulse forming inductor, trigger vacuum switch and predicting results of experiment circuit. The result of the experiment was in good agreement with the result of the simulation. The various current shapes with 300~650 us pulse width is formed by sequential firing time control of capacitor bank module. The maximum current is about 40 kA during simultaneous triggering of two capacitor bank modules. The developed 150 MW pulse power system can be applied to high current pulse power system such as rock fragmentation power sources, Rail gun, Coil gun, nano-powers, high power microwave.

Effect of Cetylpyridinium Chloride on Gingival Inflammation and Plaque Accumulation (염화 세틸피리디늄 분말의 치태제거와 치은염 완화 효과에 관한 임상시험)

  • To evaluate the effect of cetylpyridinium chloride in a powder form when used concomitantly with a commercial flouride containing tooth paste on inhibition of plaque formation, on gingivitis, and on irritation of oral mucosa, a double--blind, randomized parallel study was set up. Cetylpyridinium chloride was incorporated into a ligh brown colored powder with menthol added for scent. There were no diffemces between the active agent and the placebo which did not contain cetylpyridinium chloride in appearance, color, smell, taste, or dispenser. 98 healthy volunteers were assigned to one of two groups: brushing 3 times a day with flouride toothpaste and cetylpyridinium chloride powder, or brushing 3 times a day with flouride toothpaste and placebo. Before the test period, the subjects received through tooth cleaning and polishing. At basteline, GI, PI, BOP, and GCF of the Ramfjord teeth were measured in the experimental and placebo groups including 58 and 42 subjects repectively, After 4 weeks, GI, PI, BOP, GCF, compliance, irritation of the oral mucosa(redness, pus drainage, edema) and adverse reactions were measured. The PI, GI, and BOP of the experimental group recorded at baseline are 0.19${\pm}$0.19, 0.42${\pm}$0.31, and 0.08${\pm}$0.15. These scores showed significant decrease after 4 weeks of test period(0.11${\pm}$0.15, 0.22${\pm}$0.24, 0.02${\pm}$0.09 repectively at the end of the study) and inhibition of plaque accumulation and resolution of gingival inflammation could be observed. GCF shoed slight increase but this was not statistically different. Comparison of changes in measured scores of control and experimental groupshow GI, PI of the test indices have decreased. Test group showed significantly greater decrease in gingivitis and plaque accumulation after 4 weeks. GCF and BOP also showed greater decrease thant the control group but this difference was not statistically significant. At no time of the study period did any of the subjects show signs of irritation of the oral mucosa or adverse reactions. Following conclusions could be obtained from this study. The combined use of cetl powder and flouride toothpaste showed greater inhibition of plaque accumulation and greater decrease of gingivitis than use of flouride toothpaste with placebo agent.

Effect of increasing levels of rice distillers' by-product on growth performance, nutrient digestibility, blood profile and colonic microbiota of weaned piglets

  • Objective: This study was conducted to evaluate the effects of diets containing different wet rice distillers' by-product (RDP) levels on growth performance, nutrient digestibility, blood profiles and gut microbiome of weaned piglets. Methods: A total of 48 weaned castrated male crossbred pigs, initial body weight 7.54±0.97 kg, and age about 4 wks, were used in this experiment. The piglets were randomly allocated into three iso-nitrogenous diet groups that were fed either a control diet, a diet with 15% RDP, or a diet with 30% RDP for a total of 35 days. Chromium oxide was used for apparent digestibility measurements. On d 14 and d 35, half of the piglets were randomly selected for hemato-biochemical and gut microbiota evaluations. Results: Increasing inclusion levels of RDP tended to linearly increase (p≤0.07) average daily gain on d 14 and d 35, and decreased (p = 0.08) feed conversion ratio on d 35. Empty stomach weight increased (p = 0.03) on d 35 while digestibility of diet components decreased. Serum globulin concentration decreased on d 14 (p = 0.003) and red blood cell count tended to decrease (p = 0.06) on d 35, parallel to increase RDP levels. Gene amplicon profiling of 16S rRNA revealed that the colonic microbiota composition of weaned pigs changed by inclusion of RDP over the period. On d 14, decreased proportions of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge, and increased proportions of Prevotellaceae_ge, Prevotella_2, and Prevotella_9 were found with inclusion of RDP, whereas opposite effect was found on d 35. Additionally, the proportion of Lachnospiraceae_ge, Ruminococcaceae_ge, Ruminococcaceae_UCG-005, and Bacteroidales_ge in RDP diets decreased over periods in control diet but increased largely in diet with 30% RDP. Conclusion: These results indicate that RDP in a favorable way modulate gastrointestinal microbiota composition and improve piglet performance despite a negative impact on digestibility of lipids and gross energy.

  • In this paper, a decoupled dual servo (DDS) stage for ultra-precision scanning system with large working range is introduced. In general, dual servo systems consist of a fine stage for short range and a coarse stage for long range. The proposed DDS also consists of a $XY\theta$ fine stage for handling and carrying workpieces and one axis coarse stage. Its coarse stage consists of air bearing guide system and a coreless linear motor with force ripple. The fine has four voice coil motors(VCM) as its actuator. According to a VCM's nature, there are no mechanical connections between coils and magnetic circuits. Moreover, VCM doesn't have force ripples due to imperfections of commutation components of linear motor systems - currents and flux densities. However, due to the VCM's mechanical constraints the working range of the fine is about $25mm^2$. To break that hurdle, the coarse stage with linear motors is used to move the fine about 500mm. Because of the above reasons, the proposed DDS can achieve higher precision scanning than other stages with only one servo. With MATLAB's Sequential Quadratic Programming (SQP), the VCMs are optimally designed for the highest force under conditions and constraints such as thermal dissipations due to its coil, its size, and so on. And for their movements without any frictions, guide systems of the DDS are composed of air bearings. To get precisely their positions, a linear scale with 5nm resolution are used for the coarse stage's motion and three plane mirror laser interferometers with 5nm for the fine's $XY\theta$ motions. With them, on scanning the two stages have same trajectories. The control algorithm is named Parallel method. The embodied ultra-precision scanning system has sub 100nm following error and in-positioning stability.

Security Problem of National Major Facility's Parking Lot and its Improvement Method -Focused on Doonchi(Waterside) Parking Lot of National (국가중요시설의 주차장 보안의 문제점과 개선방안: 국회둔치주차장을 중심으로)

  • National Assembly is a constitutional institution that is required to first consult the will of the people and it should do its effort continuously so that security of citizens using parking lot would be enhanced at the same time while improving parking service in order to increase customer satisfaction of the people. Under this recognition, in this study, Doonchi parking lot of National Assembly under consigned management was first reviewed in a perspective of criminal prevention through environmental design(CPTED) and particularly, fence installation and reinforcement work for securing 'territoriality' and operation of all round shooting camera and installation of No-trespassing warning board at entrance were suggested. Second, it was recommended to change independent control system in which CCTV security system of National Assembly Doonchi parking lot is operated separately from National Assembly safety situation room and integrate it with National Assembly safety situation room(revised to double safety system) and performance of CCTV camera was made to be increased to over 2m. In addition, video recording mode was converted to NVR mode for application to IP camera in the future and in order to avoid dead zone of security monitoring area and based on site inspection result, addition 3 places of newly installing CCTV were indicated. Third, it was recommended to introduce parking fare billing and management system through unmanned equipment in parking lot management and operation.(specialized management of professional parking service provider was reviewed). By doing so, risk of cash handling by charging personnel was removed by reducing current 7 working personnel to 3 and particularly, by converting parking lot management mode being operated temporarily from 9 A.M. to 9 P.M. at present to 24 hours operation mode and providing more specialized parking service, citizens visiting National Assembly were provided with convenience and image of National Assembly was also enhanced. This study was carried out in parallel with various literature and case studies, including data from the Office of the Defense Protection in the National Assembly.

Intensity-modulated radiotherapy for stage I glottic cancer: a short-term outcomes compared with three-dimensional conformal radiotherapy

  • Purpose: To investigate the differences in treatment outcomes between two radiation techniques, intensity-modulated radiotherapy (IMRT) and three-dimensional conformal radiotherapy (3DCRT). Materials and Methods: We retrospectively analyzed 160 (IMRT = 23, 3DCRT = 137) patients with stage I glottic cancer treated from January 2005 through December 2016. The IMRT was performed with TomoTherapy (16 patients), volumetric-modulated arc therapy (6 patients), and step-and-shoot technique (1 patient), respectively. The 3DCRT was performed with bilateral parallel opposing fields. The median follow-up duration was 30 months (range, 31 to 42 months) in the IMRT group and 65 months (range, 20 to 143 months) in the 3DCRT group. Results: The 5-year overall survival and 3-year local control rates of the 160 patients were 95.7% and 91.4%, respectively. There was no significant difference in 3-year local control rates between the IMRT and 3DCRT groups (94.4% vs. 91.0%; p = 0.587). Thirteen of 137 patients in the 3DCRT group had recurrences. In the IMRT group, one patient had a recurrence at the true vocal cord. Patients treated with IMRT had less grade 2 skin reaction than the 3DCRT group, but this had no statistical significance (4.3% vs. 21.2%; p = 0.080). Conclusion: IMRT had comparable outcomes with 3DCRT, and a trend of less acute skin reaction in stage I glottic cancer patients.

Performance Characteristics of Water-Chilling Heat Pump Using CO2 on Control of Inverter Frequency (인버터 주파수 제어에 따른 CO2용 수냉식 열펌프의 성능 특성)

  • The performance characteristics of water-chilling heat pump using CO2 for the control of inverter frequency was investigated experimentally. An experimental apparatus is consisted of a compressor, a gas cooler, an expansion valve, an evaporator and a liquid receiver. All heat exchangers used in the test rig are counter flow type heat exchangers with concentric dual tubes, which are made of copper. The gas cooler and the evaporator consist of 6 and 4 straight sections respectively arranged in parallel, each has 2.4m length. The experimental results summarize as the following: for constant inlet temperature of evaporator and gas cooler, as mass flow rate, compression ratio and discharge pressure increases with the inverter frequency. And heating capacity and compressor work increases, but coefficient of performance(COP) decreases with the inverter frequency of compressor. As inlet temperature of secondary fluid in the evaporator increases from $15^{\circ}C$ to $25^{\circ}C$, compression ratio and compressor work decreases, but mass flow rate, heating capacity and COP increases with the inverter frequency of compressor. The above tendency is similar with performance variation with respect to the variation of inverter frequency in the conventional vapor compression refrigeration cycle.

Analysis on Vortex Streets Behind a Square Cylinder at High Reynolds Number Using a Large-Eddy Simulation Model: Effects of Wind Direction, Speed, and Cylinder Width (큰에디모의 모형을 이용한 높은 레이놀즈 수에서의 사각 기둥 후면의 와열 분석: 풍향과 풍속, 기둥 너비의 영향)

  • This study investigates turbulent flow around a square cylinder mounted on a flat surface at high Reynolds number using a large-eddy simulation (LES) model, particularly focusing on vortex streets behind the square cylinder. Total 9 simulation cases with different inflow wind directions, inflow wind speeds, and cylinder widths in the x- and y-directions are considered to examine the effects of inflow wind direction, speed, and cylinder widths on turbulent flow and vortex streets. In the control case, the inflow wind parallel to the x-direction has a maximum speed of $5m\;s^{-1}$ and the width and height of the cylinder are 50 m and 200 m, respectively. In all cases, down-drafts in front of the cylinder and updrafts, wakes, and vortex streets behind the cylinder appear. Low-speed flow below the cylinder height and high-speed flow above it are mixed behind the cylinder, resulting in strong negative vertical turbulent momentum flux at the boundary. Accordingly, the magnitude of the vertical turbulent momentum flux is the largest near the cylinder top. In the case of an inflow wind direction of $45^{\circ}$, the height of the boundary is lower than in other cases. As the inflow wind speed increases, the magnitude of the peak in the vertical profile of mean turbulent momentum flux increases due to the increase in speed difference between the low-speed and high-speed flows. As the cylinder width in the y-direction increases, the height of the boundary increases due to the enhanced updrafts near the top of the cylinder. In addition, the magnitude of the peak of the mean turbulent momentum flux increases because the low-speed flow region expands. Spectral analysis shows that the non-dimensional vortex generation frequency in the control case is 0.2 and that the cylinder width in the y-direction and the inflow wind direction affect the non-dimensional vortex generation frequency. The non-dimensional vortex generation frequency increases as the projected width of the cylinder normal to the inflow direction increases.

An Antiviral Mechanism Investigated with Ribavirin as an RNA Virus Mutagen for Foot-and-mouth Disease Virus

  • To prove whether error catastrophe /lethal mutagenesis is the primary antiviral mechanism of action of ribavirin against foot-and-mouth disease virus (FMDV). Ribavirin passage experiments were performed and supernatants of $Rp_1$ to $Rp_5$ were harvested. Morphological alterations as well as the levels of viral RNAs, proteins, and infectious particles in the BHK-21 cells infected using the supernatants of $Rp_1$ to $Rp_5$ and control were measured by microscope, real-time RT-PCR, western-blotting and plaque assays, respectively. The mutation frequency was measured by sequencing the complete P1- and 3D-encoding region of FMDV after a single round of virus infection from ribavirin-treated or untreated FMDV-infected cells. Ribavirin treatment for FMDV caused dramatically inhibition of multiplication in cell cultures. The levels of viral RNAs, proteins, and infectious particles in the BHK-21 cells infected were more greatly reduced along with the passage from $Rp_1$ to $Rp_5$, moreover, nucleocapsid protein could not be detected and no recovery of infectious virus in the supernatant or detection of intracellular viral RNA was observed at the $Rp_5$-infected cells. A high mutation rate, giving rise to an 8-and 11-fold increase in mutagenesis and resulting in some amino acid substitutions, was found in viral RNA synthesized at a single round of virus infection in the presence of ribavirin of $1000\;{\mu}M$ and caused a 99.7% loss in viral infectivity in contrast with parallel untreated control virus. These results suggest that the antiviral molecular mechanism of ribavirin is based on the lethal mutagenesis/error catastrophe, that is, the ribavirin is not merely an antiviral reagent but also an effective mutagen.
