Behavioral Pharmacological Studies of Nootropic Candidates on the Central Dopaminergic Activity Rats (중추도파민신경활성에 미치는 뇌기능개선 후보약물의 행동약리학적 연구)

  • The present study was undertaken to elucidate the behavioral characteristics of nootropic candidates, entrophenoxine, N-methyl-D-glucamine, piracetam and red ginseng saponin components on stereotyped sniffing behavior induced by apomorphine in rats. Apomorphine, a direct dopaminergic receptor agonist, induced stereotyped behaviors including sniffing licking growing and biting in a dosedependent manner, and that behaviors were completely inhibited when measured at 1 week after 6-ydroxydopamine(6-HDA) treatment. Centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin(TS), panaxatriol (PT), and Rg1 enhanced but panaxadiol (PD) inhibited, whereas piracetam and Rb1 were not effective of the sterotyped sniffing behavior induced by apomorphine(1mg/kg). The enhanced stereotyped behavior by centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin, panaxatriol(PT), and Rg1 seems to have a similarity to entrophenoxine, N-methyl-D-glucamate in modulating of dopaminergic neuroal activity and also my be useful for the nootropic candidates.

  • The studies on the saponins of Korean ginseng, Panax ginseng C.A. Meyer, were performed according to the cultivating locations, sampling seasons, plant parts, and growing stages. The changes in saponin content in the course of manufacturing Red ginseng and Ginseng extract were observed. In this paper, a new method for the determination of the total and the individual saponin glucosides was proposed and applied to the samples under study. The method employing Digital Densitorol DMU-33C (Toyo electric Co., Japan) followed the separation of the saponins by means of a preparative thin layer chromatography. The saponin contents and their fractional distribution were summarized as follows: 1. The average concentrations(% plant dry weight) of semi-purified saponins in the roots of Korean ginseng planted in the various locations were 5.0%(Keumsan), 6.0% (Kimpo), and 5.4% (Pocheon), respectively. 2. There were 3.3% saponins in White ginseng(Rhizome) and 12.7% saponins in Ginseng tail (Fibrous root). 3. Regarding the year of growth, the contents of saponins were 90.3mg (2-year-old ginseng), 254.4mg (3-year-old ginseng), 404.2mg (4-year-old ginseng). 999.6mg (5-year-old ginseng), and 1377.1mg (6-year-old ginseng) respectively, and the saponin factions containing panaxatriol as an aglycone increased. 4. Thin layer chromatography revealed that Red ginseng yielded many saponins which Shibata et al. designated as $ginsenoside-Rb_1$ (22.1%), $-Rb_2(15.4%)$, -Rc(12.6%), -Re (15.7%), and $-Rg_1$, (9.3%). 5. 29.9% of crude saponins were isolated from ethanolic extract of Panax ginseng fibrous root and their extraction yield was 94.2% of fibrous root saponin.

Influences of Fusurium sozani and Phytophthoya cactorum on the Changes in Saponin Components of Korean Ginseng (Panax ginseng C.A. Meyer) (Fusarium solani와 Phytophlhora cactorum이 고려인삼의 사포닌 성분변화에 미치는 영향)

  • Influnces of Fuiarium solani and Phytophthora cactorum infection on the changes in saponin components of Korean ginseng (Panax ginseng C.A. Meyer)roots and some of the biology of those fungi in relation to ginseng root were investigated. Mycelial growth of F. solani was decreased as increasing concentration of the water extracts of fresh ginseng roots, while that of P. cactorum was increased as increasing the concentration of the water extracts and crude saponin. Mycelial growth of F. solani, however, was increased as increasing concentration of crude ginseng saponin upto 20 ppm, while it was tended to be decreased when the concentration was higher than 50 ppm. Nystatin also suppresed the growth of F. solani as increasing its concentration, but it did not affected on the growth of p. cactorum. Ginsenoside Ra and Ro components were not detected in ginseng roots inoculated with F. solani or P. cactorum. Panaxadiol gisenosides were increased by 3.0%, whereas panaxatriol ginsenosides were decreased by 34.9% in ginseng roots inoculated with F. iolani. In ginseng roots inoculated with P. cactorum panaxadiol ginsenosides were increased by 21.1%, but panaxatriol ginsenosides were decreased by 23.5%. PD/PT ratio in ginseng roots inoculated with F. solani or P. cactorum were equally increased by 58.4% in spite of differences in the change of panaxadiol and panaxatriol ginsenosides. The total saponin components of ginseng roots inoculated with F. solani or P. cactorum were decreased by 17.8% and 2.5%, respectively.

Effect of Ginseng Saponins on the Biosynthesis of Prostaglandins (인삼 사포닌 성분이 프로스타글란딘류 생성에 미치는 영향)

  • The effects of Ginseng saponins on the in vitro biosynthesis of prostaglandins were examined in order to identify the role of some Ginseng components on the regulation of arachidonic arid metabolism. The productions of prostaglandin $E_2$ (PG$E_2$), $F_2$ (PGF2), thromboxane $B_2$(TX$B_2$) and 6-ketoprostaglandin Fl (6-Keto-PGF1) from [3Hl-arachidonic acid were evaluatpf by radiochromatographic analysis with rabbit kidney microtome, human platelet homogenate and bovine aortic microsome. The amounts of the total prostaglandins produced by cyclooxygenase activity and malondialdehyde from arachidonic acid didn't show significant changes in the presence of Ginseng saponins. Both of panaxadiol and panaxatriol didn't affect the production of PG$E_2$ while the formations of PG$F_2$( and TX$B_2$( were nearkedly reduced and the production of prostacyclin was increased. The formation of TXBE was reduced by ginsenoside $Rb_2$, Rc, and Re, however the production of 6-Keto-PGF1 was increased dose dependently up to 1 mg/ml. Moreover, platelet aggregations induced by arachidonic acid and U46619 (9.11-methanepoxy PG$H_2$), TX$A_2$ mimetics, were also inhibited by three ginsenosides. The effect of G-Re on prostacyclin synthetase was inhibited by tranylcypromine, prostacyclin synthetase inhibitor. These results suggest that Ginseng saponins may not directly act on cyclooxygenase but affect on the divergent pathway from endoperoxide.

Effect of the Contents Ratio of Panaxadiol Ginsenosides Extracted from Various Compartment of Ginseng on the Transcription of Cu/Zn Superoxide Dismutase Gene (홍삼의 각 부위에서 추출된 Panaxadiol분획의 함량비에 따른 유해산소제거효소(Cu/Zn Superoxide Dismutase) 유도효과)

  • Cu/Zn superoxide dismutase (SOD1) is a protective enzyme responsible for the dismutat ion of superoxide radicals within the cell by converting superoxide radicals to oxygen and hydrogen peroxide, which is in turn changed to oxygen and water by catalase. Previously, we reported that the panaxadiol (PD) and its ginsenoside $Rb_2$ induced the expression of SOD1 gene through AP2 binding site and its induction. Here, we examined the effect of subfractions of panaxadiol ginsenosides, which were extracted from different parts of ginseng root that possess various ratios of panaxadiol to panaxatriol, on the induction of SOD1 gene expression. To explore this possibility, the upstream regulatory region of SOD1 was linked to the chloramphenicol acetyl transferase (CAT) structural gene and introduced into human hepatoma HepG2 cells. We observed that the transcriptional activation of SOD1 was proportional to the contents ratio of panaxadiol ginsensides. Consistent with this results, the total extract portion prepared from the finely-hairy root, which contains the higher ratio of panaxadiol to panaxatriol about 2.6, increased the SODl transcription about 3 fold. This results suggest that the panaxadiol fraction could induce the SOD1 and total extract of the ginseng finely-hairy root would be a useful material as a functional food for the SOD1 inducer.

Effects of Red Ginseng Component on the Antioxidative Enzymes Activities and Lipid Peroxidation in the Liver of Mice (홍삼 추출물 투여가 생쥐간에서 항산화 효소 활성과 지질과산화에 미치는 효과)

  • The effects of each component (water extracts, alcohol extracts, lipophillic extracts, total saponin, panaxadiol, panaxatriol) of red ginseng on the antioxidative enzyme activities were investigated in the liver in order to screen antioxidative components of red ginseng. 20∼25g ICR mouse which were pretreated with 50 mg/kg body weight of red ginseng component for 15 days. The ability of red ginseng component to protect against oxidative damage to the mouse liver was examined by determining the level of lipid peroxidation (MDA), hydroperoxide (H$_2$O$_2$) and the activities of superoxide dismutase (SOD), catalase. The hepatic total-SOD activity was highest in lipophilic extracts group and panaxadiol group next (p<0.01). The content of hepatic hydroperoxide was lowest in the order of panaxatriol group and alcohol extracts group (p < 0.01). The hepatic catalase activity in the liver was highest in order of lipophillic extracts group (p <0.01) and total saponin group (p<0.05). Finally the lipid peroxidation (MDA) level was lowest in lipophillic extracts group, alcohol extracts group and panaxadiol next (p <0.01). In conclusion, the order of effectiveness of antioxidants was to be lipophillic extracts>panaxadiol >total saponins.

  • The possibility of utilizing greet amount of by-product of ginseng (Panax ginseng C.A. meyer) plant-that is, production of ter from ginseng leaf, was studied and the results are summarized as follows: 1. Ginseng leaf contains more soluble matter than tea leaf (Thea sinensis) and the soluble matter is easily extracted by hot water. 2. Ginseng leaf has less tannin (2.2%) than yea leaf (7.89%). Therefore, it has less astringency than tea. 3. Vitamin C content of ginseng leaf is not compared with that of tea leaf. In fact, ginseng leaf contains Vitamin C $50{\sim}110$ times of tea leaf. 4. Ginseng leaf contains $5.7{\sim}8.5%$ glycoside (dammaranes) and the ratio of panaxadiol to panaxatriol is $0.54{\sim}0.75$ that is, panaxatriol contents is high. 5. For the acceptability of the product related with the soluble matter contents and color the method of extracting 2g of ginseng leaf product in 200ml of water for 3 minutes is recommended. 6. As a result of evaluating the flavor characteristics and effective components of the products, product D which is produced by the process of steaming, drying and roasting is considered to have the best quality.

Inhibitory Effects of Panaxatriol from Panax ginseng C. A. Meyer on Phosphoinositide Breakdown Induced by Thrombin in Platelets

  • In this study, we have investigated the effect of panaxatriol (PT) on phosphoinositides (PIS) breakdown and $Ca^{2+}$-elevation in thrombin-induced platelet aggregation. Thrombin (5U/ml), a potent platelet agonist which activates phospholipase $C_{\beta}$ via protease activated receptor (PAR), hydrolyzed PIS in platelet membrane. The phosphatidylinositol 4, 5-bisphosphate $(PIP_2)$ was hydrolyzed after 10 sec of the thrombin-stimulation, and both the phosphatidylinositol 4-monophosphate (PIP) and phosphatidylinositol (PI) were brokendown after 30 sec of the thrombin-stimulation. However, PT inhibited the thrombin-stimulated hydrolysis of $PIP_2$, PIP, and PI. On the other hand, thrombin increased the level of phosphatidic acid (PA) which is phosphorylated from diacylglycerol (DG) generated by PIS-hydrolysis. However, Pr inhibited the thrombin-increased PA level non-significantly. Thrombin increased cytosolic free $Ca^{2+}([Ca^{2+}])_i$) up to 72% as compared with control $(30.8{\pm}0.9 nM)$ in intact platelet. However, PT (100 ${\mu}g/ml$) inhibited the thrombin-elevated $[Ca^{2+}]_i$ to 100%. These results suggest that PT may have a beneficial effect on platelet aggregation-mediated thrombotic disease by inhibiting thrombin-induced platelet aggregation via suppression of the $[Ca^{2+}]_i$ level and PIS breakdown.

뇌중 dopamine 활성에 작용하는 뇌기능개선 약물의 효능검색

  • 뇌기능개선 약물들(nootropic drugs)은 항치매약물로 기대되나 현재까지 약리학적으로 공통점이 보고되어 있지는 않다. 최근 수종의 뇌기능 개선약물의 작용이 중추도파민신경 활성을 매개로 함이 보고된바 있다. 따라서 apomorphine 투여에 의해 유발되는 상동행동을 지표로 하여 현재 사용중인 뇌기능개선약물과 국내 자원생약 성분의 작용을 비교 검토하였다. 1. apomorphine 투여에 의하여 유발된 상동행동에 대하여 centrophenoxine 과 N-methyl -D-glucamine은 현저한 상승효과를 나타내었으나 Piracetam은 영향을 미치지 않았다. 2. 인삼사포닌 성분 중 panaxatriol 및 Rgl 성분은 현저한 상승효과를 나타내었으나 total saponine, panaxadiol 및 Rbl 성분은 거의 영향을 미치지 않았다.

  • The saponins isolated form the herb of Panax ginseng C.A. Meyer were investigated as compared with ginseng root saponins. By adopting DEAE cellulose ion exchange chromatography the pure saponins were isolated from Korean ginseng roots and leaves. The ginseng root and leaf saponins showed some differences in the pattern of the two-dimensional thin layer chromatogram. The ratio of panaxadiol to panaxatriol in the saponins was 1.7 in the roots and 3.5 in the leaves. Infra-red spectrum of ginseng leaf saponins isolated by liquid chromatography was identical with that of root saponins.
