  • Korean Panax ginseng C. A. Meyer (P. ginseng) is a well-known and one of the most important tonic herbs used in traditional Korean medicine. The pharmacological effects of P. ginseng have been reported by many researchers. Nevertheless, little is known between the mechanism of action and the active compounds. In this study, we performed a comprehensive proteomic analysis and protein categorization in order to understand the physiological characteristics of the major components in the adventitious roots of P. ginseng. Whole proteins extracted from the cultured adventitious roots of P. ginseng were separated by two-dimensional polyacrylamide gel electrophoresis (2-DE). Among the 1000 spots which were detected by silver staining, 113 spots were labeled and identified by matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF MS). Our results showed that 40 proteins were identified among the 113 spots, with a hit ratio of 35.3%. A number of proteins identified on the 2-DE gels (30%; 16 spots) were involved in energy metabolism. These proteomic data will be helpful to better understand the physiological and pharmacological effects of P. ginseng.

The Assessment of Carbendazim, Cyazofamid, Diethofencarb and Pyrimethanil Residue Levels in P. ginseng (C. A. Meyer) by HPLC

  • A fast and simple high-performance liquid chromatography (HPLC) method for the simultaneous determination of four pesticides having fungicide properties has been proposed for Panax ginseng, C. A. Meyer grown for 4, 5, or 6 years. Analytical separation was performed on C18 columns using ultraviolet detector under gradient conditions. Spiked blank samples were used as standards to counteract the matrix effect observed in the chromatographic determination. The HPLC response for all pesticides was linear, with determination coefficients > 0.9986. The average rate of recovery for pesticides spiked with 2 fortification levels was > 72% with relative standard deviations < 9%. The limits of quantification (LOQ) ranged from 0.03 to 0.16 ppm. These LOQs were lower than the respective maximum residue limits (MRL) established by the Korean Food and Drug Administration (KFDA), except for cyazofamid. The proposed method was used to determine pesticide residue levels in samples of ginseng obtained from Jeonnam Province (Republic of Korea). None of the pesticides were found in ginseng samples grown for 4, 5, or 6 years.

홍삼제품의 산업현황 (The Industry Status of Red Ginseng Products)

  • 인삼(Panax ginseng C.A. Meyer)은 두릅나무과(Araliaceae)에 속하는 식물로 동아시아에서 2,000여년 전부터 보기(補氣)제로 사용된 중요한 약재이다. 홍삼은 인삼의 뿌리를 찐 것으로, 고려시대 이전부터 쪄서 말리는 가공법이 있었던 것으로 알려져 있다. 건강기능식품에서 말하는 홍삼제품은 홍삼을 원재료로 하여 이를 분말화하거나 물이나 주정으로 추출하여 농축 또는 발효하여 식용에 적합하며, 진세노사이드Rg1과 Rb1의 합이 0.8~34 mg/g이 되어야 한다. 2011년 한국의 건강기능식품 시장은 약 3조 6,000억원에 이르며, 홍삼시장은 1조 2,000억원에 이른다. 2007년 6,420억원이었던 홍삼시장이 5년만에 두배가 된 것이다. 홍삼시장이 성장하면서 한국인삼공사(정관장)와 농협중앙회(NH한삼인), 동원F&B(천지인), 웅진식품(다채움수) 등의 회사가 기초연구와 다양한 제품개발을 통하여 경쟁하고 있다. 한국인삼공사의 경우, 최근 식품의약품안전청으로부터 홍삼의 '항산화' 기능에 대하여 건강기능식품기능성원료 인정을 받았고, '홍삼정타브렛'을 출시하였으며 '홍이장군' 브랜드를 리뉴얼하였다. 농협중앙회에서는 이미지 향상을 위한 '지성이면 한삼인' 시리즈와 짜 먹는 '뽀로로 홍삼 젤리'를 출시하였고, 동원F&B은 '홍송조화건조'를 제조공정에 도입하였고, 캡슐형태의 제품을 출시하였으며, 최근에는 꼬마버스 타요를 활용하여 '천지인 꼬마버스 타요' 시리즈를 개발하였다. 웅진식품, 대상웰라이프 등에서도 사포닌이 흡수되기 좋게 만든 '발효홍삼' 제품을 출시하고 있다. 향후 홍삼시장의 지속적인 성장을 위해 기초연구와 소비자의 니즈에 맞는 국내시장만이 아닌 해외시장까지 나아갈 수 있는 특화된 제품 개발이 절실히 요구되는 시점이다.

Studies on the mechanism of cytotoxicities of polyacetylenes against L1210 cell

  • This study was performed to investigate the mechanism of in vitro cytotosic actions of polyacetylenes which are panaxydol, panaxynol and panaxytriol isolated from Panax ginseng C. A. Meyer. DNA synthesis of L1210 cells was significantly inhibited with dose dependent pattern when L1210 cells were treated for 1 hour with over 5 .mu.g/ml of polyacetylenes. Panaxydol which had the most potent cytotoxicity among three polyacetylenes showed also the strongest inhibitory effect on DNA synthesis. Intracellular cyclic AMP levels of L1210 cells treated with 2.5 $\mu$g/ml of panaxydol or panaxytriol were significantly elevated on the incubation duration. The elevation of cyclic AMP levels by panaxytriol was higher than that by panaxydol, but no significant increase in cyclic AMP by panaxynol was observed. All three polyacetylenes had no effect on glycolysis of L1210 cells. Electron microscopic observations revealed that polyacetylenes caused damage to plasma membranes of L1210 cells in proportion to their cytotoxicities at each $ED_{50}$ value (panaxydol > panaxynol> panaxytriol). These results suggest that cytotoxicities of polyacetylenes against L1210 cells might be mediated by elevated cyclic AMP level, even though the relationship among their cytotoxicities, inhibitory effect on DNA synthesis and ability to elevation of cyclic AMP level are not fully agreed, and might be also related to membrane damage.

  • 인삼(Panax ginseng C.A. Meyer) 엽소병에서 chorophyll의 bleaching 현상에 대한 생리적 반응기작을 조사, 분석한 결과 chorophyll의 bleaching 현상은 singlet oxygen ($^1O_2$)에 의한 photooxidation 과 catalase와 peroxidase의 inactivation에 의한 $H_2O_2$의 autooxidation이었다. Chorophyll의 bleaching 현상에 원인으로 추리된 $H_2O_2$를 농도별로 인삼잎 절편에 처리하였던 바, chorophyll의 함량과 chorophyll의 흡수스펙트럼은 자연광 처리시 유발된 bleaching 현상과 일치하였으며, saponin 첨가로 인하여 bleaching 현상은 현저히 촉진되었다.

인삼 모상근의 성장 및 Ginsenosides 생성에 미치는 광의 효과 (Growth and Ginsenosides Production of Hairy Root (Panax ginseng C.A. Meyer) via Light Energy)

  • The effects of light on the growth and ginsenosides production were examined in the hairy roots of Panax ginsen C.A. Meyer induced by Agrobacterium rhizogines A4. The 9rowth of ginseng hairy roots in 1/2MS liquid medium was significantly decreased with an increment of light intensity (1,000~7,000 lux). The growth of hairy roots under 7,000 lux condition was decreased at 17% compared to the dark condition. The production of 7 ginsenosides in hairy root was very high in 3,500 lux condition. The production of ginsenoside-Rg, and Rf increased 3.3 and, 2.4 times respectively as compared to dark condition. The growth of hairy roots was inhibited by blue light, while ginsenosides production was increased. The sucrose demands of hairy roots was examined in light condition(3,500 lux). The growth of hairy roots in 1/2MS liquid medium with various sucrose concentrations(1~4%) was high in IVp sucrose, while ginsenosides production was high in 3% sucrose condition. The growth and ginsenosides production were high when hairy roots were cultured in dark condition for 1 week and then transferred to light condition(3,500 lux) for 4 weeks. It is suggested that ginsenosides production could be accelerated by light intensity of specific wavelength in cultures of ginseng hairy roots.

  • A major polyacetylene compound from Panax ginseng roots, heptadeca-1-en-4,6-diyn-3,9,10-triol, was irradiated with 300 nm UV light to obtain a photooxidized acetylenic compound having the conjugated en-on-diyne chromophore, heptadeca-1-en-4,6-diyn-9,10-diol-3-one. The same oxidation product was obtained exclusively by 4-(dimethylamino) pyridinium chlorochromate at room temperature.

Age-dependent Distribution of Fungal Endophytes in Panax ginseng Roots Cultivated in Korea

  • Fungal endophytes were isolated from 1-, 2-, 3-, and 4-year-old ginseng roots (Panax ginseng Meyer) cultivated in Korea. The isolated fungal endophytes were identified based on sequence analysis of the internal transcribed spacer and morphological characterization by microscopic observations. A total of 81 fungal endophytes were isolated from 24 ginseng roots. Fungal endophytes were classified into 9 different fungal species and 2 unknown species. Ginseng roots that were 1-, 2-, 3-, and 4-years old were colonized by 2, 6, 8, and 5 species of fungal endophytes, respectively. While Phoma radicina was the most frequent fungal endophyte in 2-, 3-, and 4-year-old ginseng roots, Fusarium solani was the dominant endophyte in 1-year-old ginseng roots. The colonization frequencies (CF) varied with the host age. The CF were 12%, 40%, 31%, and 40% for 1-, 2-, 3-, and 4-year-old ginseng roots, respectively. We found a variety of fungal endophytes that were distributed depending on the age of ginseng plants.

인삼 캘러스 원형질체의 배양에 따른 세포벽 재생의 전자현미경적 연구 (An Electron Microscopic Study on the Cell Wall Regeneration of Culture Panax ginseng Callus Protoplast)

  • 인삼(Panax ginseng C. A. Meyer) 캘러스로부터 분리한 원형질체를 배양하면서 배양시간에 따른 원형질체의 미세구조 변화와 원형질막 표면을 전자현미경으로 조사하였다. 3일 동안 배양된 원형질체에서는 조면소포체, 리보좀, 골기체, 미토콘드리아, 전색소체 등의 세포기관들의 수가 증가하였고 미소관도 관찰되었다. 골기체 주변에는 많은 골기소낭들이 형성되어 세포질 전반에 존재하였고, 이들 소낭들은 원형질막 바깥으로 돌출되어 돌기를 형성하기도 하였다. 소포체에서 유래된 액포속에는 액포의 함입에 의하여 골기소낭들이 들어 있는데, 이들 액포들은 융합에 의한 exocytosis로 원형질막 근처로 이동하여 원형질막과 융합한 다음, 세포벽 물질을 원형질막 바깥으로 방출하여 원형질막에 침적하였다. 전색소체는 많은 전분립을 함유하고 있었고, 미소관들은 원형질막 근처에서 막과 평형으로 배열하고 있었다. 배양된 원형질체의 표면에는 섬유소로 구성되어 있는 원섬유들이 서로 연결되어 있는 것이 관찰되었다.
