  • Oxygen isotope ratios (${\delta}^{18}O$) of three Pacific oyster (Crassostrea gigas) specimens from the Neolithic Yeondae-do shell midden site, Tongyeong, Korea, were analyzed to determine the seasonality of shellfish gathering and site occupation. Oxygen isotope samples were taken from the left valve hinge sections of the specimens. Oxygen isotope values ranged between -0.1 ‰ and -2.4 ‰, between -0.2 ‰ and -2.9 ‰, and between 0.3 ‰ and -2.8 ‰ in oyster specimen #one, #two and #three, respectively. The isotope profiles showed seasonal temperature cycles, providing information related to the seasonality of shellfish gathering and site occupation. Hinge-edge oxygen isotope values of the specimens showed decreasing trends after passing through maximum values (winter), indicating that they formed during spring. Thus it can be assumed that during spring season, oysters were gathered and the site was occupied.

The Effects of PAHs (Polynuclear Aromatic Hydrocarbons) Exposure on Fertilization and Larval development of the Pacific Oyster, Crassostrea gigas

  • To evaluate the effect of PAHs on fertilization and larval development of the Pacific oyster, Crassostrea gigas via in vivo exposure for 30 days. Conditioned C. gigas adult brood stock was taken from oyster farm in May 2003 and raised in flow through tank at 20-22$^{\circ}C$ with feeding equivalently mixed marine microalgae: Chaetoceros simplex, C. gracilis, Isochrysis galbana and Tetrasemis tetrathele. The oysters were exposed to PAHs cocktail at 200ppb (10 species) until they are fully riped with an untreated group. Motility of sperm, Fertilization and The percent of D-shaped larvae was measured under microscope (Olympus BX-50). Fertilization capability using dry sperm and eggs from both non- and PAHs treated brood stocks was significantly reduced at the treated group (P<0.01). In addition to the origin effect, it was obvious that fertilization of eggs from both group were adversely suppressed in consistent with PAHs level (P<0.01). The percent of D-shaped larvae in treated group was significantly reduced comparing to non-treated. The percent was adversely correlated with PAHs level, which was more significantly reduced in treated group. Therefore, it was obvious that PAHs exposure on the Pacific oyster has adverse affects on the sperm as well as larvae in both maternal and acute source.

IQF 굴(Crassostrea gigas) 복합엑스분을 이용한 굴 소스의 가열향기 성분 (Volatile Flavor Constituents of Cooked Oyster Sauce Prepared from Individually Quick-frozen Oyster Crassostrea gigas Extract)

  • The pacific oyster Crassostrea gigas has a desirable taste and flavor that differs from those of other fish and shellfish. In order to develop a high value-added product from individually quick-frozen oyster extract (IQFOE), we prepared an oyster sauce from IQFOE and characterized its volatile compounds using vacuum simultaneous steam distillationsolvent extraction / gas chromatography / mass spectrometry. The moisture, crude protein, crude ash, salinity, pH and volatile basic nitrogen contents of the oyster sauce were 60.6%, 8.2%, 9.2%, 9.3%, 5.7 and 21.0 mg/100 g, respectively. Seventy-six volatile compounds were detected in the cooked odor of the oyster sauce. These volatile compounds included 14 esters, including ethyl acetate, 13 nitrogen- containing compounds, including 2,4,6-trimethyl pyridine, 13 acids, including hexadecanoic acid, 12 alcohols, including ethyl alcohol and 6-methyl heptanol, 6 alkanes, 5 aldehydes, including benzaldehyde, 5 ketones, including 1-(2-furanyl)-ethanone, 4 furans, including 2-furancarboxaldehyde and 2-furanmethanol, 3 aromatic compounds, including d-limonene, and 1 miscellaneous compound. Esters, acids and nitrogen-containing compounds, and alcohols were the most abundant compounds in the odor of the cooked oyster sauce, with some aldehydes, ketones, and furans.

저염수에서 이매패류 참굴(Crassostrea gigas)의 패각운동 (Shell Valve Movement of Pacific Oysters, Crassostrea gigas, in Response to Low Salinity Water)

  • 본 연구는 참굴(Crassostrea gigas)의 패각운동을 이용하여, 연안역에서 발생하는 저염수에 대한 조기경보가능성을 살펴보았다. 30 psu와 20 psu에서 패각운동은 각각 $7.32{\pm}3.21$회/hr와 $7.11{\pm}3.90$회/hr였으며, 파형과 횟수는 차이가 없었다(t-test, p>0.001). 하지만 10 psu와 5 psu에서는 모든 개체가 폐각상태를 지속하였다. 수온과 염분의 복합실험결과, Group 1(수온 $15^{\circ}C$ ${\times}$ 염분 15 psu)은 20~30 psu에서 보인 패각운동 후(약 2~3시간), 장시간 폐각을 하였다. Group 2(수온 $30^{\circ}C$ ${\times}$ 염분 15 psu)에서는 Group 1의 패각 개폐운동보다 더 빠르고 자주 나타나, 참굴의 생리적인 위기상황에 대한 신호를 나타내었다. 따라서 이러한 파형은 하계 저염수 출현 시 나타낼 수 있는 조기경보 신호로 충분히 활용될 수 있을 것으로 보인다.

부안산 참굴(Crassostrea gigas) 추출물로부터 피부 상재균에 대한 새로운 항균 펩타이드, cgCAFLP의 탐색 및 정제 (Screening and Purification of a Novel Antibacterial Peptide, cgCAFLP, Against Skin Pathogens from the Extract of the Pacific Oyster Crassostrea gigas from Buan in Korea)

  • This study was performed to screen the antimicrobial activities of the extract from the Pacific oyster Crassostrea gigas against skin pathogens and to purify the relevant antibacterial peptide. The acidified extract showed potent antibacterial activities against gram-positive and gram-negative bacteria but showed no activity against Candida albicans and no significant cell toxicity. Among acne-causing pathogens, the acidified extract showed potent antibacterial activity only against Staphylococcus aureus, and its antibacterial activity was completely abolished by treatment with trypsin or chymotrypsin, and was inhibited by salt treatment. The acidified extract showed strong DNA-binding ability but did not show bacterial membrane permeabilizing ability. Based on antimicrobial activity screening and cytotoxic effects, a novel antibacterial peptide was purified from the acidified gill extract using solid-phase extraction, cation-exchange, and reversed-phase HPLC. The resulting peptide had a molecular weight of 4800.8 Da and showed partial sequence homology with the carbonic anhydrase 4 (CA4) protein in the hard-shelled mussel. Overall, we purified a novel antibacterial peptide, named cgCAFLP, which is related to carbonic anhydrase 4 (CA4) protein, against skin pathogens. Our results suggest that the Pacific oyster extract could be used as an additive to control some acne-related skin pathogens (S. aureus).

  • We explored the efficiency of natural antibacterial agents used to enhance the self-life and sensory properties of shucked and packed Pacific oyster Crassostrea gigas, which are in high demanded. First, we screened natural resources exhibiting antibacterial activity against food spoilage and pathogenic bacteria. Of theses, ignited oyster shell powder (IOS) and the natural food preservative, lactic acid bacteria fermented powder (LBF), were selected for further study considering the efficacy, mass production, and cost. The addition of 0.1% IOS (W/V) and 0.5% LBF (W/V) to shucked and packed oyster optimally extended the shelf-life without affecting the sensory evaluation. The results obtained in this study will provide a clue to enhance self-life in raw oyster products.

Regulation of adductor muscle growth by the IGF-1/AKT pathway in the triploid Pacific oyster, Crassostrea gigas

  • We investigated the insulin-like growth factor 1 (IGF-1)/AKT signaling pathway involved in muscle formation, growth, and movement in the adductor muscle of triploid Pacific oyster, Crassostrea gigas. Large and small triploid oysters (LTs and STs) cultured under identical conditions were screened, and the signaling pathways of individuals with superior growth were compared and analyzed. mRNA and protein expression levels of actin, troponin, tropomyosin, and myosin, proteins important in muscle formation, were higher in LTs compared with STs. Expression levels of IGF-1, IGF binding protein (IGFBP), and IGFBP complex acid-labile subunit were also higher in LTs compared with STs. Phosphorylation of the IGF receptor as well as that of AKT was high in LTs. In addition, the expression of phosphomammalian target of rapamycin and phospho-glycogen synthase kinase $3{\beta}$ was increased and the expression of Forkhead box O3 was decreased in LTs. Therefore, we suggested that the IGF-1/AKT signaling pathway affects the formation, growth, and movement of the adductor muscle in triploid oysters.

갯벌참굴 (Crassostrea gigas) 양식을 위한 부착기질별 하나굴 종묘 생산과 수평망식 시설을 이용한 배수체별 치패 성장 비교 (The Production Efficiency of Cupped Oyster Crassostrea gigas Sprat According to Clutch and Growth Comparing Diploid and Triploid Oysters in Off-bottom Culture for Tidal Flat Utilization)

  • To produce cupped oyster sprat consistently, we investigated the efficiency of nine settlement substrates using selective breeding of Pacific oyster Crassostrea gigas, with the goal of popularizing off-bottom culture on the west coast of Korea. We also compared the growth and survival of selectively bred sprat (diploid) with triploid sprat from an off-bottom culture system. Considering, the attachment rate and detachment efficiency, producing cupped oyster in a polypropylene gunny bag proved to be the most effective method. There were no differences in shell growth or total weight between the diploids from selective breeding and triploids from off-bottom culture for 5 months. However, the survival rate was 8 times higher in the diploids than the triploids. Transplanting sprat from selective breeding is one way to restore oyster farms and nurture off-bottom culture along the west coast.
