Outbreak and Spread of Bacterial Canker of Kiwifruit Caused by Pseudomonas syringae pv. actinidiae Biovar 3 in Korea

  • A bacterial pathogen, Pseudomonas syringae pv. actinidiae (Psa), is a causal agent of kiwifruit bacterial canker worldwide. Psa biovar 3 (Psa3) was first detected in 2011 at an orchard in Dodeok-myeon, Goheung-gun, Jeonnam Province in Korea. In this study, we present the results of an epidemiological study regarding Psa3 occurrence on kiwifruit orchards in Korea for the period of 2013 to 2015. Since the first detection of Psa3 in 2011, there was no further case reported by 2013. However, Psa3 was rapidly spreading to 33 orchards in 2014; except for three orchards in Sacheon-si, Gyeongnam Province, most cases were reported in Jeju Island. Entering 2015, bacterial canker by Psa3 became a pandemic in Korea, spreading to 72 orchards in Jeju Island, Jeonnam, and Gyeongnam Provinces. Our epidemiological study indicated that the first Psa3 incidence in 2011 might result from an introduction of Psa3 through imported seedlings from China in 2006. Apart from this, it was estimated that most Psa3 outbreaks from 2014 to 2015 were caused by pollens imported from New Zealand and China for artificial pollination. Most kiwifruit cultivars growing in Korea were infected with Psa3; yellow-fleshed cultivars (Yellow-king, Hort16A, Enza-gold, Zecy-gold, and Haegeum), red-fleshed cultivars (Hongyang and Enza-Red), green-fleshed cultivars (Hayward and Daeheung), and even a kiwiberry (Skinny-green). However, susceptibility to canker differed among cultivars; yellow- and red-fleshed cultivars showed much more severe symptoms compared to the green-fleshed cultivars of kiwifruit and a kiwiberry.

Studies on the Strength of Cement Mortars with Surface Crosslinked cPSA Absorbent (표면이 가교된 Crosslinked Poly(sodium acrylate) 흡수제가 첨가된 시멘트 모르타르의 강도 특성 연구)

  • To study the effect of surface crosslinked layer on the crosslinked poly(sodium acrylate) (cPSA) absorbent, we synthesized several surface crosslinked cPSAs with 5, 10 and 20 g of ethylene glycol dimethacrylate (EGDMA) by an inverse emulsion polymerization method to delay the absorption of excess water in concrete. We measured the compressive and flexural strength of mortars having 0.5, 1.0 and 1.5 wt% cPSA-EGDMA. We observed the increase of compressive and flexural strength of the cPSA-EGDMA added cement mortars except for the 0.5 wt% cPSA-EGDMA (20 g) added cement mortar. 1.0 wt% cPSA-EGDMA (5 g) added cement mortar showed about 16% and 10% increased compressive and flexural strength than those of plain cement mortar. To study the effect of porosity on compressive and flexural strength, we used FE-SEM and porosimeter. FE-SEM analysis showed swollen cPSMAEGDMA (5 g) filled between calcium silicate hydrate (C-S-H) crystals. We observed the decreased porosity of the cPSA-EGDMA added cement mortars than that of plain cement mortar. 1.0 wt% cPSA-EGDMA (5 g) cement mortar showed the lowest porosity of 16.5%.

Elution Behavior of Copper(II) and Iron(II) Ions by Phenol Sulfonic Acid on Chelating Resin (킬레이트 수지에서 Phenol Sulfonic Acid에 의한 구리(Ⅱ)와 철(Ⅱ) 이온의 용리현상)

  • The elution behavior of copper(Ⅱ) and iron(Ⅱ) ions by phenol sulfonic acid(PSA) as an eluent on chelating resin, Amberlite IRC-718 have been investigated. When copper(Ⅱ) and iron(Ⅱ) solutions were adsorbed on the resin and eluted with various concentration of PSA, two peaks of each ion were appeared in the elution curve. These two peak areas were changed according to the PSA concentration. Using these phenomena, the stability constants of complex formation between the two ions and PSA were calculated. The values are 7.0 for copper(II) and $4.5{\times}10^4$ for iron(II), respectively.

  • 수소에너지는 다양한 원료로부터의 수소생산을 위한 반응기술 및 생산물로부터 수소 정제를 위한 분리기술의 확립과 더불어, 대형화 시스템부터 소형 시스템에 이르는 공정기술을 확보하는 것이 다가오는 청정 대체에너지 체제에 대비하기 위하여 필요하다. 이를 위해서는 생산된 수소 혼합물에서부터 수소를 분리 정제하는pressure swing adsorption (PSA) 의 개발이 필수적이다. 이 기술은 이미 다양한 분야에 성공적으로 상용화 적용되어 기술의 타당성을 제시하고 있으나, 국내의 경우 수입에 의존하고 있어 이를 설계 할 수 있는 공정모사기 (simulator)의 개발이 우선되어야 한다. 따라서 효율적으로 PSA 공정 및 scale-up기술을 확보하기 위해서는 전산모사기 개발의 선행이 필수적이다. PSA 공정의 전산모사기는 물질수지, 에너지수지, 모멘텀수지와 더불어 흡착평형과 속도식이 결합되어 개발되어야 한다. 특히 공정에 다양한 단계가 적용되기 때문에 복잡한 boundary condition이 적용되며, 연속순환공정이라 하더라도 각 단계가 discrete 하게 해석되어야 한다. 따라서 공정모사는dynamic simulator로 개발되어야 정확도를 확보할 수 있다. 본 연구에서는 제철소에서 발생하는 수소혼합물이 WGSR 반응기를 거쳐 수소의 농도를 향상 시키고, 이를 유입가스로 사용하는 $H_2$ PSA 공정 모사기를 개발하고자 한다. 수소 생산을 위한 PSA 공정 모사기 개발을 통하여 95% $H_2$ 순도와 90% 회수율 규모의 수소를 생산할 수 있는 PSA 공정의 설계 기술기반을 확보하고자 한다.

Cell Cycle Regulation and Antioxidant Activity of Psammaplin A, A Natural Phenolic Compound from Marine Sponge

  • Psammaplin A (PSA), a naturally occurring biophenolic compound has been demonstrated to deliver significant cytotoxicity to many cancer cell lines. In this article, we investigated the effect of PSA on cell cycle progression of lung cancer cells (A549). It was found that PSA could slightly perturb the cell cycle progression of A549 cells and lead to the cell cycle arrest at G2/M phase, indicating PSA might disturb the mitosis process of A549 cells. In addition, inspired by the two phenolic groups in the structure of PSA, the antioxidant activity of it has been evaluated. Although PSA was weak in scavenging the stable free radical 1,1-diphenyl-2-picrylhyrazyl (DPPH), it showed stronger ABTS radical scavenging activity than ascorbic acid in TEAC assay. Furthermore, it was found that PSA could effectively prevent DNA strand scission induced by oxidative stress. These results suggest that PSA have both cell cycle regulation and antioxidant activities. Herein, we suggest that PSA would be a very interesting and promising candidate to be developed as a multi-function drug.

Influence of Respiration on Photosynthetic Electron Transport in psaB Mutants from Cyanobacterium Synechocystis sp. PCC6803 (Cyanobacterium Synechocystis sp.PCC6803 psaB 돌연변이주의 광합성 전자전달에 미치는 호흡의 영향)

  • The influence of respiration on photosythetic electron transport were investigated in the Wid type and psaB mutants from Syneehocystis sp. PCC6803. The amount of glucose uptake in the wild type was proportional to the glucose concentration added in wild type and less than that of psaB mutants in the dark. It was suggested that psaB mutants more depend on the glucose than the wild type. It was investigated how the activities of isocitrate dehydrogenase(IDH) and glucose-6-phos-phate dehydrogenase(G6PDH) were changed. The activities of IDH were very low. While, the ac-tivities of G6PDH were much higher than that of IDH. These results agree to the reports that ex-ogenous glucose was dismilated aerobically via Oxidative Pentose Phosphate Pathway in heterotrophic cyanobacteria. PsaB mutants showed high G6PDH activity in the presence of glucose as well as in the dark and high respiratory activities especially in the dark. It was also investigated how photosynthetic electron transport activities were changed. PsaB mutants showed higher photosynthetic electron tranasport activities than wild type in the presence of glucose as well as in the dark. In the results, it was proposed that photosynthetic electron transport between PS I and PS U was complemented by respiratory electron transport through the NADPH generated by Dxidative Pentose Phophate Pathway in psaB mutant from Synechocystis sp. Application of Dynamic Probabilistic Safety Assessment Approach for Accident Sequence Precursor Analysis: Case Study for Steam Generator Tube Rupture

  • The purpose of this research is to introduce the technical standard of accident sequence precursor (ASP) analysis, and to propose a case study using the dynamic-probabilistic safety assessment (D-PSA) approach. The D-PSA approach can aid in the determination of high-risk/low-frequency accident scenarios from all potential scenarios. It can also be used to investigate the dynamic interaction between the physical state and the actions of the operator in an accident situation for risk quantification. This approach lends significant potential for safety analysis. Furthermore, the D-PSA approach provides a more realistic risk assessment by minimizing assumptions used in the conventional PSA model so-called the static-PSA model, which are relatively static in comparison. We performed risk quantification of a steam generator tube rupture (SGTR) accident using the dynamic event tree (DET) methodology, which is the most widely used methodology in D-PSA. The risk quantification results of D-PSA and S-PSA are compared and evaluated. Suggestions and recommendations for using D-PSA are described in order to provide a technical perspective.

Forensic Evaluation of Prostate-Specific-Antigen (PSA) Rapid Test Kit for Identification of Human Semen (전립선특이항원검사 Kit에 의한 정액의 신속 검출법)

  • It would be one of the most important tests that determination of semen in crime scene as a matter of significant evidences. Recently, it has been developed for the identification of semen in forensic specimens which was used simply, easily and reproductively. In this study, Prostate-Specific-Antigen (PSA) Rapid Test kit was evaluated for the forensic identification of semen and compared with one step semen inspection forensic rapid test kit. The sensitivity and specificity of the rapid PSA kit were examined in addition to the stability of PSA. The positive band of rapid PSA kit shown even with 1,000,000-fold diluted semen, which was at least 100 timed higher than qualitative one step semen inspection forensic rapid test kit. PSA was detected in urine from normal male adult, however, it was not detected in urine from young boys and female body fluids. It was shown that PSA was very stable to resist boiling for 20 minutes and the effect of bacteria. In crime scene investigation, rapid PSA kit is expected to help to identify semen easily in the evidences.

Comparison of the Formula of PSA, Age, Prostate Volume and Race Versus PSA Density and the Detection of Primary Malignant Circulating Prostate Cells in Predicting a Positive Initial Prostate Biopsy in Chilean Men with Suspicion of Prostate Cancer

  • Background: Combining risk factors for prostate cancer into a predictive tool may improve the detection of prostate cancer while decreasing the number of benign biopsies. We compare one such tool, age multiplied by prostate volume divided by total serum PSA (PSA-AV) with PSA density and detection of primary malignant circulating prostate cells (CPCs) in a Chilean prostate cancer screening program. The objectives were not only to determine the predictive values of each, but to determine the number of clinically significant cancers that would have been detected or missed. Materials and Methods: A prospective study was conducted of all men undergoing 12 core ultrasound guided prostate biopsy for suspicion of cancer attending the Hospital DIPRECA and Hospital de Carabineros de Chile. Total serum PSA was registered, prostate volumecalculated at the moment of biopsy, and an 8ml blood simple taken immediately before the biopsy procedure. Mononuclear cells were obtained from the blood simple using differential gel centrifugation and CPCs identified using immunocytchemistry with anti-PSA and anti-P504S. Biopsy results were classed as positive or negative for cancer and if positive the Gleason score, number of positive cores and percent infiltration recorded. Results: A total of 664 men participated, of whom 234 (35.2%) had cancer detected. They were older, had higher mean PSA, PSA density and lower PSA-AV. Detection of CPCs had high predictive score, sensitivity, sensibility and positive and negative predictive values, PSA-AV was not significantly different from PSA density in this population. The use of CPC detection avoided more biopsies and missed fewer significant cancers.Conclusions: In this screening population the use of CPC detection predicted the presence of clinically significant prostate cancer better than the other parameters. The high negative predictive value would allow men CPC negative to avoid biopsy but remain in follow up. The formula PSA-AV did not add to the predictive performance using PSA density.

A New Quantification Method for Multi-Unit Probabilistic Safety Assessment (다수기 PSA 수행을 위한 새로운 정량화 방법)

  • The objective of this paper is to suggest a new quantification method for multi-unit probabilistic safety assessment (PSA) that removes the overestimation error caused by the existing delete-term approximation (DTA) based quantification method. So far, for the actual plant PSA model quantification, a fault tree with negates have been solved by the DTA method. It is well known that the DTA method induces overestimated core damage frequency (CDF) of nuclear power plant (NPP). If a PSA fault tree has negates and non-rare events, the overestimation in CDF drastically increases. Since multi-unit seismic PSA model has plant level negates and many non-rare events in the fault tree, it should be very carefully quantified in order to avoid CDF overestimation. Multi-unit PSA fault tree has normal gates and negates that represent each NPP status. The NPP status means core damage or non-core damage state of individual NPPs. The non-core damage state of a NPP is modeled in the fault tree by using a negate (a NOT gate). Authors reviewed and compared (1) quantification methods that generate exact or approximate Boolean solutions from a fault tree, (2) DTA method generating approximate Boolean solution by solving negates in a fault tree, and (3) probability calculation methods from the Boolean solutions generated by exact quantification methods or DTA method. Based on the review and comparison, a new intersection removal by probability (IRBP) method is suggested in this study for the multi-unit PSA. If the IRBP method is adopted, multi-unit PSA fault tree can be quantified without the overestimation error that is caused by the direct application of DTA method. That is, the extremely overestimated CDF can be avoided and accurate CDF can be calculated by using the IRBP method. The accuracy of the IRBP method was validated by simple multi-unit PSA models. The necessity of the IRBP method was demonstrated by the actual plant multi-unit seismic PSA models.
