In this review, the terminology that is used to describe ileal amino acid (AA) digestibilities in piglet feed ingredients is defined. If one accepts that the determination of AA digestibilities should be based on the ileal analysis method, one should consider that ileal digesta contains variable amounts of endogenous crude protein (CP), which originates mainly from digestive secretions, sloughedoff epithelial cells and mucins. The ileal endogenous CP and AA losses are separated into basal ileal endogenous CP and AA losses ($IAAL_{B}$), which are not influenced by the feed ingredient composition, and specific ileal endogenous CP and AA losses ($IAAL_{S}$), which are induced by feed ingredient characteristics such as level and type of fiber and anti-nutritional factors (ANF). Depending how ileal endogenous CP and AA losses are considered in the measurement of CP and AA digestibilities, digestibility values are expressed as apparent (AID), standardized (SID), or true (TID) ileal digestibilities of CP and AA. The main concern associated with the use of AID values in diet formulation for pigs is that they are not additive in mixtures of feed ingredients. Consequently, the concept of standardized ileal CP and AA digestibilities was introduced by correcting AID values for basal ileal endogenous CP and AA losses ($IAAL_{B}$). The correction for both $IAAL_{B}$ and $IAAL_{S}$ yields TID values, however, routine procedures to measure $IAAL_{S}$ are not yet available. In principle, SID values should be preferred, because they represent the fundamental properties of the feed ingredient. There exist only few reports on SID of CP and AA in feedstuffs frequently used in piglet nutrition. These include soybeans (SB), soybean meal (SBM), soy proteins (SP), soy protein concentrate (SPC), soy protein isolate (SPI), corn gluten (CG), wheat gluten (WG), pea protein (PeaP), potato protein (PotP), fish meal (FM) and whey proteins (WP), but the results obtained are inconsistent. Differences in SID values within feed ingredients may, at least in part, be attributed to different processing conditions or inherent differences of the assay feed ingredients. Moreover, there is some evidence that the determination of SID values and $IAAL_{B}$ in piglets may be confounded by the dietary CP level of the assay diet, age and (or) body weight (BW), the level of feed intake or the methodological approach used to determine $IAAL_{B}$.
This study focused on the usefulness of plant extracts for a city organic farm. Insecticidal activity of 8 plant species, plants crude extracts by ethanol and water, against third instar larvae (susceptible strain) of Plutella xylostella was evaluated in this study. Ethanol(25%) extracts of plant materials had better insecticidal activity than water extracts at the room temperature. Insecticidal activity were investigated by leaf disc spray method. Five plant extracts (Capsicum annuum, Chelidoniun majus, Leonurus sibiricus, Coptis japonica, Mentha piperascens) showed over 50% insecticidal activity to P. xylostella at the concentration of $1500mg{\cdot}L^{-1}$. Extract of C. annuum showed the highest insecticidal activity (83.3%) against P. xylostella. Phytotoxicity was not observed on Raphanus sativus and Brassica campestris with spray application. Seed germination and growth of two plants were not affected. Extract of C. japonica revealed 73.3% control efficacy against P. xylostella of R. sativus and showed 70% control efficacy against those of B. campestris, respectively, in plastic house. Insecticidal activity against various insects was observed in C. japonica extract as 87.4% for R. sativus and 74.3% for B. campestris in the field. These results suggested that extracts of C. annuum, C. japonica showed the highest insecticidal activity, and could be used as city farm insecticides for organic farming.
To investigate phytotoxic substances in Artemisia argyi, the donor plant, and their biological activities, seed germination and seedling growth of receptor plants such as Arundinella hirta, Echinochloa crus-galli, Rumex crispus and Lactuca sativa were examined at different concentrations of aqueous extracts of the donor plant. Germination of four receptor species was inhibited by the extracts, while seedling growth was decreased to a lesser degree than in the germintion test. Germination, seedling growth and dry weight growth of Achyranthes japonica grown in pot were proportionally inhibited by the extracts. Volatile substances emitted from A, argi plant caused slight inhibition in the germination and seedling growth of the receptor species. Essential oil of the plant extracted by Karlsruker's apparatus inhibited growth of microorganisms and callus growth of Pinellia ternata and Oryza sativa. The GC /MS method was employed for analysis and identification of allelochemicals from A. argyi leaves. Sixty-one chemical substances such as a-pinene, camphene, 1. 8-cineol, etc. were identified from essential oil of A. argyi. The results of this experiment on seed germination, seedling growth, microorganism culture and tissue culture indicated that naturally occurring chemical substances from A. argyi would be responsible for the growth inhibition of plants studied.
A pot experiment was conducted to find the nitrogen fixing activities associated with ice rhizoplane in nine varieties at various growth stage by acetylene reduction technique. Results obtained ware Summarized as follows. 1. Nitrog en fixing activites associated with rice rhizoplane differ by the rice variety and the growth stage. The activities increased with progressing of growing time, and maximum activities were obtained from the ear forming stage to the heading stage. 2. The application of rice straw did not greatly enhanced the nitrogen fixing activities associated with rice rhizoplane. However, somewhat increasing tendency was obtained in the Kataktara, TKM-6, Centrypartna. Daegujo, and Jodongji varieties. 3. The nitrogen fixed during the one rice cropping period was estimated as 0.55kg of N per 10a by Milyang-23 variety to 2.6kg of N per 10a by Jodongji variety. 4. The amount of rice root showed no significant relationship with the amount of nitrogen fixed during the growing period of rice plant.
In order to isolate novel oligotrophic bacteria exhibiting antifungal activities, soils were collected from pepper-cultivated fields of Yeongyang, Jecheon, Nonsan, Eumsong and Goesan area in Korea. From soils in pepper cultivated area, a total of 9,354 strains were isolated as oligotrophic bacteria by the R2A dilution method. Among 9,354 oligotrohic bacteria candidates, 1A strain was selected by screening against Phytophthora capsici causing phytophthora blight of hot pepper in the greenhouse and field. The strain was identified as Bacillus vallismortis based on its 16S rDNA sequence and key characteristics as compared with those of authentic cultures of B. vallismortis(KACC 12149) and B. mojavensis(KACC 12096). The strain showed broad spectrum of antibiotic activity in vitro test, as revealed in its strong inhibitory activity to the genera Phytophthora, Collectotrichum, Botrytis and Fusarium, but not to Rhizoctonia and Magnaporthe. In pot experiments, infection rate of hot pepper in the non-treated pots was about 89%, while it was only 29% in the pots treated with 1A strain. The result indicated B. vallismortis 1A is a potential biocontrol agent for phytophthora blight of hot pepper
Mutation tachniques inducing more useful mutations and reducing somatic effects need to be improved for crop breeding. Seeds of barley varieties ; Dema, Grosso were treated with two types of mutagens ; 1) chemical treatment: single treatment or double treatment of two mutagens (N-nitroso-N-methylurea ; MNH, Sodium Azide; NaN$_3$) 2) gamma ray irradiation treatment. After treatment, half of seeds were used for germination test and half of seeds were sown to the field. With the higher dose of mutagen both chemical and gamma ray were plants treated, the higher rate of growth reduction rate was in M$_1$ seedling. In chemical treatment, germination rate of seeds, growth rate of coleoptile and root in double treatment of chemical mutagens were better than single treatments, especially in same dose. Growth inhibition rate of plant in double treatment of 1.0mM MNH(0.5mM MNH + 0.5mM MNH), for example, were less than one of plants of single treatment of 1.0mM MNH in pot and petri dish test. Growth reduction rate of culm and fertility rate in M$_1$ plants double treated in same dose of single treatment were also less than single one. With the higher dose of mutagen both chemical and gamma ray were plants treated, the higher frequency of chlorophyll mutants was in M$_2$ seedling. The rate of chlorophyll mutants in double treatment of chemical mutagens were higher than single treatment. Double treatment methods can be a improved method for induction of new good mutants, which were induced more useful mutations and reduced harmful somatic effects.
An efficient method of plant regeneration from Acanthopanax chiisanensis somatic embryos was developed. Cotyledonary somatic embryos were obtained in liquid Murashige and Skoog (MS) medium from embryogenic cell suspension cultures. They were desiccated for 0 to 72 hr and then cultured on MS medium containing NAA, BA, GA$_3$, (0-0.5mg/L). The highest multiple shoots formation (100%) was obtained from 72 hr desiccated somatic embryos on ifs medium with 0.5mg/L NAA+0.5mg/L BA or 0.5 mg/L NAA+0.5mg/L BA+0.5mg/L GA$_3$ after 6 weeks culture. Plant conversion from multiple shoots was not high. The highest plant conversion from multiple shoots was obtained on 1/3MS medium with 1.0mg/L GA$_3$. Converted plantlets were transferred to ex vitro condition and the highest survival rate (70%) of the plantlets was obtained on plastic pots containing vermiculite and sand. These results indicate that micropropagation procedure can be applied for an efficient mass propagation of Acanthopanax chiisanensis.
A total of 35 phosphate solubilizing bacterial strains were isolated from waste mushroom bed of Agaricus bisporus in Buyeo-Gun, Chungnam and screened for the production of indole acetic acid (IAA). The best IAA producing strain was identified as Pantoea rodasii using 16S rRNA analysis. In addition to the IAA production, this strain could act as an efficient phosphate solubilizer (1100 ${\mu}g$$ml^{-1}$ after 5 days of incubation) also. The selected strain was cultured under different conditions in order to assess the optimum conditions for maximum IAA production. The nutrient broth (NB) medium was recorded as the best medium, where the maximum IAA production (229 ${\mu}g$$ml^{-1}$) was recorded at the start of stationary phase (12 hours after inoculation) of the bacteria growth. The performance of the strain was found to be maximum at the temperature of $30^{\circ}C$ followed by $25^{\circ}C$. IAA production was found to be increased with increasing tryptophan concentration (from 0.1 to 0.6%), however beyond this limit, a slight reduction in IAA production was observed. The strains' ability to produce IAA was further confirmed by extraction of crude IAA and subsequent TLC analysis. A specific spot from the extracted IAA preparation was found corresponding with the standard spot of IAA with same $R_f$ value. The results of HPLC analysis conducted in identifying and quantifying the IAA production more precisely, are in agreement with the results of the assessment done with colorimetric method. As revealed by the results of the pot experiment, the isolated strain could significantly enhance the growth (as measured by shoot and root growth) of mung bean plants compared to that of non-inoculated plants. Therefore it can be concluded that the present strain, Pantoea rodasii has great potential to be used as bio-inoculants.
This research was carried out as a basic survey to provide more effective and improved food service for the soliders of the Armed Forces who are serving to defend the Republic of Korea subjects were selected randomly who were serving in the army for a short-term. Following is the result of a survey regarding the situation of their meals, dietary life, and their food preference of 190 kinds of foodstuffs. As a result of surveying the dietary life of the soldiers, they indicated that there was dissatisfaction in the current Food Service because it lacked tasted and the same kind of meals were being served too frequently with regard to this, many subjects expressed that they were eating out because of the quality of they get from the regular meal service the meals, that is they were eating in restaurants to resolve their dissatisfaction over the quality of the meals. It was also surveyed that in the current meals there is a need to diversify ways in preparing food, increase the number of dishes, and improve the tasts of the food by using improved cooking method. It seemed that the soldiers were satisfied in the quantitative level of the current food service but that there is a need to improve the meals in quality. Following is the result of a survey on the preference of foodstuffs. In case of rice as the stationary food, mixture of five-grain plain white cooked rice showed a higher propensity compared to plain whte boiled rice. This shows that soliders prefer various cereals in their rice. Soliders liked soup made from meat better than vegetable soup or soup made from fish. Especially there was a high performance for beef soup. Soliders liked One-dish meals such as fried rice or boiled rice with asserted mixtures, noodled, and dumplings on the whole-soliders generally liked vegetables and they liked uncooked vegetables better than cooked vegetables. Generally everyone liked beverages and there was a high preference for dessert. Among various rice cakes soliders liked songpyon(rice cake steamed on a layer of pine needles) and injolmi (cake made from glutinous rice). On the other hand, soliders did not like food prepared with fish, biscuits and candy very much. In the way of preparing food, soliders liked food that were fried in deep far rather than food cooked in water. Also they liked pot stem better than soup. As examined in the survey, those surveyed wanted improvement in the quality and diversity of the food rather than increase the number of food and quality. Therefore there is a need for diversified supply and demand of quality of meal service.

Oxidative stress is the main limiting factor in crop productivity. To solve global environmental problems using the plant biotechnology, we have developed on the oxidative stress-tolerant transgenic tall fescue plants via Agrobacterium-mediated genetic transformation method. In order to develop transgenic tall fescue (Festuca arundinacea Schreb.) plants with enhanced tolerance to multiple environmental stresses, nucleotide diphosphate kinase gene under the control of CaMV35S promoter were introduced into genome of tall fescue plants. Proteomic analysis revealed that transgenic tall fescue not only accumulated NDP kinase 2 protein in their cells, but also induced several other antioxindative enzyme-related proteins. When leaf discs of transgenic plants were subjected to cold stress, they showed approximately 30% less damage than wild-type plants. In addition, transgenic tall fescue plants showed normal growth when transgenic plants were subjected to $4^{\circ}C$ for 3 days treatments. These results suggest that transgene is important in ROS scavenging by induction of antioxidative proteins, and could improve abiotic stress tolerance in transgenic tall fescue plants.
