The C. militaris belongs to entomopathogenic fungi, which have their specific sequences in internal transcribed spacer regions (ITS1 and ITS2) depending on species. In this study, to identify the phylogenetic relationship of the strain hybrided by mating of C. militaris, we compared DNA sequences of ITS regions and 5.8S ribosomal DNA (rDNA) repeat unit of hybrid strain and its parental strains. The result revealed that hybrid strains are C. militaris species. In addition, cordycepins produced by hybrid strains and other strains of C. militaris were analyzed by HPLC with 20mM $KH_2PO_4$ of mobile phase and C-18 columns. The result indicated that the strain hybrided by mating produce higher concentration of phytochemical cordycepin than other C. militaris strains.
An antioxidant- producing bacterium was isolated from sea water in Jeju island. The isolated strain, SC2-1 was gram-positive, catalase positive, facultatively anaerobic, oxidase negative, motile and small rods. The strain utilized sucrose, dextrose, fructose, mannitol and maltose as a sole carbon and energy source and sodium chloride was required for the bacteria growth. The radical scavenging activity of the culture supernatants was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) method. This bacterium was identified based on cellular fatty acids analysis and 16S rDNA sequencing, and then named Exiguobacterium sp. SC2-1. The modified optimal medium compositions required the addition of maltose $2.5\%(w/v)$, yeast extract $1.5\%(w/v)$ and $KH_{2} PO_{4} 0.05\%(w/v)$ in marine broth (Difco. Co. USA). Antioxidant activity of under optimal culture conditions were $93\%$.
Four bacterial strains capable of catabolizing 4-chlorobiphen!;l (4CB) were isolated from the industrial waste waters. The bacterial isolates designated as PO$. P20, P27, and P1242. respectively, were examined for their catabolic activities. And in order to examine molecular homology of the 4CB catabolizing genes of these bacterial isolates. Southern hybridization was conducted with bphABC genes of P. p.srudoalculigrnrs KF707 as a DNA probe. The metabolites of 2-hydroxy-6-0x0-6-(4'-chlorophenyl)hexa-2 .4-dienoic acid and Cchlorobenzoate were detected to be produced by the isolatc:~ in the MM2 liquid cultures. But Cchlorobenzoate was further catabolized to produce 4.-hydroxybenzoate by DJ-12, P08. and P27. but not by P20 and P1242. As results of hybridization, homologous regions were commonly observed in Xhol fragments of 2.2 and 1.8 kb and in EcoRl fragment of 11 kb in the DJ- 12. P08, and P27 isolates. But in any restriction enzyme digests ot the P20 and PI242 isolates. homologous region was not detected. A study aimed at identifying putative drought responsive genes that confer tolerance to water stress deficit in tea plants was conducted in a 'rain-out shelter' using potted plants. Eighteen months old drought tolerant and susceptible tea cultivars were each separately exposed to water stress or control conditions of 18 or 34% soil moisture content, respectively, for three months. After the treatment period, leaves were harvested from each treatment for isolation of RNA and cDNA synthesis. The cDNA libraries were sequenced on Roche 454 high-throughput pyrosequencing platform to produce 232,853 reads. After quality control, the reads were assembled into 460 long transcripts (contigs). The annotated contigs showed similarity with proteins in the Arabidopsis thaliana proteome. Heat shock proteins (HSP70), superoxide dismutase (SOD), catalase (cat), peroxidase (PoX), calmodulinelike protein (Cam7) and galactinol synthase (Gols4) droughtrelated genes were shown to be regulated differently in tea plants exposed to water stress. HSP70 and SOD were highly expressed in the drought tolerant cultivar relative to the susceptible cultivar under drought conditions. The genes and pathways identified suggest efficient regulation leading to active adaptation as a basal defense response against water stress deficit by tea. The knowledge generated can be further utilized to better understand molecular mechanisms underlying stress tolerance in tea.
Bacterial strain No. P08 isolated from wastewater at the Cheongju industrial complex was found to be capable of degrading 4-chlorobenzoate under aerobic condition. P08 was identified as Comamonas sp. from its cellular fatty acid composition and 16S rDNA sequence. The fcb genes, responsible for the hydrolytic dechlorination of 4-chlorobenzoate, were cloned from the plasmid pJJl of Comamonas sp. P08. The fcb gene cluster of comamonas sp. PO8 was organized in the order fcbB-fcbA-fcbTl-fcbT2-fcbT3-fcbC. This organization of the fcb genes was very similar to that of the fcb genes carried on the chromosomal DNA of pseudomonas sp. DJ-12. However, it differed from the fcbA-fcbB -fcbC ordering of Arthrobacter sp. SU. The nucleotide sequences of the fcbABC genes of strain P08 showed 98% and 53% identities to those of Pseudomonas sp. DJ-12 and Arthrobacter sp. SU, respectively. This suggests that the fcb genes might have been derived from Pseudomonas sp. DJ-12 to form plasmid pJSl in Comamonas sp. P08, or that the fcb genes in strain DJ-12 were transposed from Comamonas sp. P08 plasmid.
Recently the production of transgenic animals to express foreign proteins in mammary glands has been a routine procedure. However, it still takes a considerable time and effort, and is faced with various technical challenges until the protein of interest is successfully made. Thus, a development of an a vitro model for mamm a ary-specific gene expression for recombinant genes was carried out in this study. To this end, bovine $\alpha$$_S1$ casein cDNA was inserted at the multiple cloning site of pMSG vector under the control of MMTV promoter. MCF$_7$ cells were tran sfected with pMSG $\alpha$$_S1$ CN by CaP0$_4$ precipitation. Transfectants were selected in HAT medium and induced with dexamethasone. The cells were analyzed with chicken anti-casein and FITC-labeled rabbit anti-chicken antibodies. The results showed that dexamethasone induced 30-40 fold increase in the MMTV- $\alpha$$_S1$ casein e expression. Therefore MCF$_7$ cells, which have multiple steroid receptors, along with pMSG vector can be used as an in vitro model for the study of mammary-specific gene expression.
Human brain EST data provide important clues for our understanding of the molecular biology associated with the function of the normal brain and the molecular pathophysiology with brain disorders. To systematically and efficiently study the function and disorders of the human brain, 45,773 human brain ESTs were collected from 27 human brain cDNA libraries, which were constructed from normal brains and brain disorders such as brain tumors, Parkinson's disease (PO) and epilepsy. An analysis of 45,773 human brain ESTs using our EST analysis pipeline resulted in 38,396 high-quality ESTs and 35,906 ESTs, which were coalesced into 8,246 unique gene clusters, showing a significant similarity to known genes in the human RefSeq, human mRNAs and UniGene database. In addition, among 8,246 gene clusters, 4,287 genes ($52\%$) were found to contain full-length cONA clones. To facilitate the extraction of useful information in collected these human brain ESTs, we developed a user-friendly interface system, the Korea Brain Unigene Database (KBUD). The KBUD web interface allows access to our human brain data through three major search modes, the BioCarta pathway, keywords and BLAST searches. Each result when viewed in KBUD offers comprehensive information concerning the analyzed human brain ESTs provided by our data as well as data linked to various other publiC databases. The user-friendly developed KBUD, the first world-wide web interface for human brain EST data with ESTs of human brain disorders as well as normal brains, will be a helpful system for developing a better understanding of the underlying mechanisms of the normal brain well as brain disorders. Oyster mushrooms are widely cultivated and consumed in Korea. P. ostreatus 'Suhan(ASI 2504)' is an ideal cultivar for mushroom farmers due to its dark pileus and thick stipe; however, as it is very sensitive to environmental conditions, an alternative cultivar is required. To develop a new cultivar, parental strains 'Suhan(ASI 2504)' and 'ASI 0665 (Heuktari)' were selected from various collected strains according to morphological characteristics. P. ostreatus 'Soltari' was developed by DiMon crossing between the dikaryotic strain 'Suhan' and the monokaryotic strain derived from 'Heuktari'. Thirty-eight of the 100 crossed strains were selected following analysis of mitochondrial genetic characteristics, and 'Soltari' was ultimately selected by continuous cultivation tests. The mitochondrial DNA profile of 'Soltari' was found to be the same as that of 'Heuktari, and a nuclear DNA profile of 'Soltari' was similar as those of the parental strains, 'Suhan' and 'Heuktari.' 'Soltari' mycelium grows adequately in moderate to high temperatures of $12-20^{\circ}C$, although its optimum temperature was found to be $30^{\circ}C$. Fruiting body production per 1.1-L cultivation bottle was approximately 158.8 g. Its stipe length and thickness were comparable to those of diameter and thickness were somewhat lower (42.72 vs. 51.33 mm and 18.18 vs. 22.46 mm, respectively). 'Soltari' was found to be more resistant to high $CO_2$ atmosphere than 'Suhan', and the color of the pileus of 'Soltari' was dark gray at high temperature. Therefore, it is suggested that this new cultivar 'Soltari' is a good alternative cultivar and will contribute to energy saving in oyster mushroom farms.

Porphyra pulchella sp. nov. Ackland, West, Scott and Zuccarello was obtained at Mimosa Rock National Park, New South Wales; Westgate Bridge, Victoria, Australia; and Waihau Bay, North Island, New Zealand. It occurs mainly in mangrove habitats and is very small (± 1 mm) in field collections. In laboratory culture at 21 ± 2°C tiny blades (0.5-3.0 mm) reproduced exclusively by archeospores liberated from vegetative cells of the upper sector of the blades. The archeospores displayed amoeboid and gliding motility once discharged. At 14 ± 2°C the blades grew to 25 mm and produced longitudinal spermatangial streaks mixed with ‘phyllosporangial’ streaks. The discharged ‘phyllospores’ showed amoeboid motility and germinated forming asexual blades. A conchocelis phase with typical bangiophycidean pit connections was observed in blade cultures after 8-10 weeks at 14 ± 2°C. Conchocelis filaments produced conchosporangia and these released amoeboid conchospores that developed into archeosporangiate blades. Molecular data indicate that all 3 isolates are genetically identical.
To develop new variety of oyster mushroom, 63 intra-specific hybrids between the strain Suhan and #Nongi201 were developed using hyphal anastomosis technique in 2004. The Po2008-275 hybrid between the dikaryon strain 04-154(Suhan x #Nongi201) and the monokaryon strain derived from ASI2487 were developed using hyphal anastomosis in 2008. The Po2008-275 was shown the best cultural characteristics, selected to be a new variety and named as 'Guseol'. The new commercial strain, 'Guseol' had dark grey pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Guseol' were of an excellent quality in that not only the stipe was thick and long but also the pileus was small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $10{\sim}16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body was about 3 to 5 days depending on the temperatures. The shape of fruiting body was thin funnel shape. Fruiting body production per box($43{\times}43{\times}12cm$) was about $1545{\pm}400.9g$ which was almost 137% quantity compared to that of parental strain 04-154. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Guseol' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the mycelial growth of this mushroom were much faster in mushroom complete medium. Similar results were observed with other variety '#Chunchu2'. Analysis of the genetic characteristics of the new commercial strain 'Guseol' showed a major DNA profile as that of the parental 04-154 when primer URP 1, primer URP 2 and primer URP 5 were used, but different to '#Chunchu2' that was used as a control. This new variety of the dark grey oyster mushroom had smart and high quality image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.
