연료전지 산소환원반응 향상 위한 백금 촉매의 구조적 특성: 밀도범함수이론 연구 (Fundamental Mechanisms of Platinum Catalyst for Oxygen Reduction Reaction in Fuel Cell: Density Functional Theory Approach)

  • 연료전지에서의 전체 반응 속도는 산화전극에서 일어나는 수소산화반응에 비해 그 반응 속도가 현저히 느린 환원전극에서의 산소환원반응(oxygen reduction reaction, ORR)에 의해 결정된다. ORR 효율성 평가를 용이하게 하는 지표(descriptor)로서 촉매 표면에서의 산소원자 흡착강도를 활용하는데, 산소흡착강도는 촉매 표면의 기하학적 구조 변형에 따른 전자구조를 변형함으로써 조절할 수 있다. 이에 본 연구에서는 백금 표면의 원자모델을 이용하여 표면의 기하학적 구조가 산소흡착강도에 미치는 영향과 그 원인을 밀도범함수이론(density functional theory, DFT) 계산을 통해 분석하였다. 먼저, 기하학적 구조를 인위적으로 변형시킨 Pt(111) 표면에서의 산소흡착반응을 밀도범함수이론 계산을 이용해 분석함으로써 기하학적 구조변화가 산소흡착강도에 미치는 영향(strain effect)을 확인하였다. 최적화한 Pt 격자상수($3.977{\AA}$)에 ${\pm}1%$ 간격의 변화율을 적용하고 각 변화율마다의 산소흡착강도를 계산하였는데, Pt-Pt 원자 간 거리가 멀어질수록 산소흡착강도가 강해지는 것을 확인하였다. 이는 원자 간 거리가 증가할수록 d-band center가 페르미 준위(Fermi level)쪽으로 이동하게 되며, 이로써 일부 반결합 오비탈(anti-bonding orbitals)에 전자가 채워지지 않기 때문에 전체적으로 반결합 오비탈이 형성될 가능성이 적어지기 때문이다. 결과적으로, 순수한 백금이 가진 격자상수($3.9771{\AA}$) 보다 약 2~4% 작은 백금 표면 격자크기를 가질 수 있도록 유도할 수 있다면 산소흡착강도가 적절히 약하게 조절될 수 있으며, 이는 순수한 백금보다 더 향상된 ORR 성능을 가진 촉매물질 개발 연구를 위한 기초자료로서 활용할 수 있을 것이다.

Tox-Info 시스템의 중독정보 데이터베이스와 응급실에 내원하는 중독 환자 분포의 비교 (Comparison between Emergency Patient Poisoning Cases and the Tox-Info System Database)

  • Purpose: The Tox-Info system is a poisonous substance information database developed by the Korean National Institute of Food and Drug Safety Evaluation. The aim of this study was to estimate the coverage effectiveness of the Tox-Info system by comparing the toxic substances included in the database with the distribution of the toxic substances implicated in the cases of intoxicated patients presenting to emergency departments. The secondary aim of the study was to propose any additional substances that should be added to the database. Methods: We retrospectively reviewed the medical records of patients suffering with toxic exposure who had visited any of 12 selected emergency departments in Korea from January 2010 to December 2011. The identified toxic substances were classified into groups including prescription drugs, agricultural chemicals, household products, animals or plants, herbal drugs, and others. We calculated the coverage rate of the Tox-Info database relative to the number of intoxication cases and the type of toxic substances involved. Results: A total of 5,840 intoxicated patient records were collected. Their mean age was $46.6{\pm}20.5$ years and 56.2% were female. Of the total intoxication cases, 87.8% of the identified toxic substances were included in the Tox-Info database, while only 41.6% of all of the types of identified toxic substances were included. Broken down by category, 122 prescription drugs, 15 agricultural chemicals, 12 household products, 14 animals or plants and 2 herbal drugs involved in poisoning cases were not included in the Tox-info database. Conclusion: This study demonstrated the clinical usefulness of the Tox-Info system. While 87.8% of the substances involved in the cases were included in the Tox-Info database, the database should be continuously updated in order to include even the most uncommon toxic substances.

Pancreatic Carcinoma, Thrombosis and Mean Platelet Volume: Single Center Experience from the Southeast Region of Turkey

  • Background: The aim of this study was to investigate the general characteristics of patients with deep vein thrombosis (DVT) and pancreatic cancer as well as evaluate the relationship between mean platelet volume (MPV), DVT and survival. Materials and Methods: Seventy-seven patients with pancreatic cancer, who were admitted to Cukurova University Medical Faculty, Department of Medical Oncology, were enrolled in the study Results: The mean age was $59{\pm}20$. Forty-nine (63.6%) were men and 28 women (36.4%). Sixty-eight (88.3%) patients had adenocarcinoma and 9 (11.7%) had a malignant epithelial tumor. Thirty-six (46.7%) had liver metastasis at diagnosis. Twenty-six (33.8%) patients were alive, 20 (26%) were dead and in 31 (40.2%) the status was unknown. Only 14 (18.1%) patients had DVT. In 42 (54.5%) patients MPV values were normal, in 28 (36.4%) patients they were above normal, and in 7 (9.1%) patients they were below normal. There was no statistically significant difference between gender, tumour localization, chemotherapy and survival rates (p:0.56, p:0.11, p:0.21). There was no significant difference between DVT, gender, localisation, histological subtype, the presence of metastasis, stage and if the patient had been treated with chemotherapy (p:0.5, p:0.6, p:0.2, p:0.32, p:0.1, p:0.84). There was also no significant difference between MPV and DVT (p:0.57) but there was a significant difference between liver metastasis and DVT (p:0.02). Age, stage, the presence of metastasis and DVT were prognostic in pancreatic cancer patients. Conclusions: Cases of pancreatic cancer with liver metastasis should be studied more carefully as thrombosis is more common in these patients.

Leucaena leucocephala and Gliricidia sepium Supplementation in Sheep Fed With Ammonia Treated Rice Straw: Effects on Intake, Digestibility, Microbial Protein Yield and Live-Weight Changes

  • Two experiments were conducted to determine the effects of Leucaena leucocephala (leucaena) and Gliricidia sepium (gliricidia) supplementation on intake, digestion, outflow rates, microbial protein yield and live-weight changes in sheep fed with ammoniated rice straw (ARS). In experiment 1, three rumen cannulated Japanese Corriedale wether (mean body weight of 35.6 kg) in $3{\times}3$ Latin Square Design were used. Animals were fed ad libitum ARS alone, or supplemented with 200 g of either leucaena or gliricidia. In experiment 2, twenty-four growing native Philippine sheep with average body weight of $13.5{\pm}0.25kg$ were used in a completely randomized design (CRD) and offered similar diets to those of experiment 1. Supplementation increased total dry matter intake and nutrient digestibility except for fiber (p<0.05) without affecting ARS consumption. Nitrogen balance revealed that absorbed and retained N was significantly higher in leucaena and gliricidia. The significant improvement in N utilization and more digestible OM intake brought about by the inclusion of leucaena and gliricidia to ARS resulted in increased (p<0.05) microbial N yield. Efficiency of microbial N supply in supplemented group was not significantly different, but higher (p<0.05) than the 24.92 g N/kg DOMR for ARS group. Liquid outflow rate was 7.8 and 6.8 %/h, while the solid phase of rumen digesta was 4.4 and 3.8 %/h for the leucaena and gliricidia group respectively, which were significantly higher than 5.30 and 2.50 %/h in the control diet. The increase in total DMI resulted to higher (p<0.01) growth performance and efficient feed utilization. Average daily gain (ADG) was 19.3, 34.6 and 33.9 g/d for the ARS, leucaena and gliricidia respectively. It is therefore concluded that addition of leucaena and gliricidia to ARS in could increase nutrient intake and digestibility, subsequently improving N utilization and livestock performance.

Nutritional status of patients treated with radiotherapy as determined by subjective global assessment

  • Purpose: The purpose of this prospective multi-institutional study was to evaluate the nutritional status of patients undergoing radiotherapy (RT) for treatment of head and neck, lung, or gastrointestinal cancer. Materials and Methods: A total of 1,000 patients were enrolled in this study at seven different hospitals in Seoul, Korea between October 2009 and May 2010. The nutritional status of patients after receiving 3 weeks of RT was evaluated using subjective global assessment (SGA). The nutritional status of each patient was rated as well nourished (A), moderately malnourished (B), or severely malnourished (C). Results: The mean age of patients in this study was $59.4{\pm}11.9$ years, and the male to female ratio was 7:3. According to the SGA results, 60.8%, 34.5%, and 4.7% of patients were classified as A, B, or C, respectively. The following criteria were significantly associated with malnutrition (SGA B or C; p < 0.001): loss of subcutaneous fat or muscle wasting (odds ratio [OR], 11.473); increased metabolic demand/stress (OR, 8.688); ankle, sacral edema, or ascites (OR, 3.234); and weight loss ${\geq}5%$ (OR, 2.299). Conclusion: SGA was applied successfully to assess the nutritional status of most patients. The prevalence of malnutrition in a radiation oncology department was 39.2%. The results of this study serve as a basis for implementation of nutrition intervention to patients being treated at radiation oncology departments.

Growth and Nutrient Utilization in Buffalo Calves Fed Urea-ammoniated Wheat Straw and Hydrochloric Acid plus Urea Treated Wheat Straw

  • Ten growing male buffalo calves (aged 6-8 months, average body weight $88.2{\pm}0.57$ kg were divided randomly into two groups. Animals in group I were fed on concentrate mixture A (CP 20.2%, TDN 77.4%) and urea- ammoniated wheat straw (UAWS) while the animals in group II were fed on concentrate mixture B (CP 17.9%, TDN 77.6%) and HCl plus urea treated wheat straw (HCl UAWS) to meet their nutrient requirement for 500 g gain/d as per Kearl (1982). This feeding practice lasted for 120 days, during which fortnightly body weight were recorded to assess their growth rate. A metabolism trial was conducted after 90 days of experimental feeding to compare the digestibility of nutrients, their balance, plane of nutrition and relative cost of feeding in two groups of animals. Results revealed a significant increase in the CP content of ammoniated wheat straw due to addition of HCl viz 12.1% from 7.5%. There was a decrease in the intake of DM (p<0.05), OM (p<0.05), EE (p<0.05), NDF (p<0.01), ADF (p<0.01), cellulose (p<0.01) and hemicellulose (p<0.01) in group II as compared to group I. The digestibility (%) of DM, OM and CP was significantly (p<0.01) more in group II, whereas the digestibility (%) was significantly more for NDF (p<0.05) and hemicellulose (p<0.01) in group I than group II. There was no significant difference in the N, Ca and P balance in two groups. Intake of total DM (g/d) or (g/kg $W^{0.75}$) was significantly (p<0.01) more in group I as compared to group II, whereas the intake of DCP and TDN (g/d or g/kg $W^{0.75}$) was alike in two groups. The total body weight gain (kg), average daily gain (g/d) and feed conversion efficiency were significantly (p<0.01) more in UAWS fed group as compared to HCl UAWS fed group. Feeding cost (Rs./kg. weight gain) was significantly (p<0.05) more in group II as compared to group I. It is concluded that HCl UAWS is not suitable for the feeding of growing buffalo calves as it reduced the growth rate in comparison to UAWS fed buffalo calves.

The Effects of Cellulose, Pectin and Starch on Standardized Ileal and Apparent Total Tract Amino Acid Digestibilities and Bacterial Contribution of Amino Acids in Feces of Growing Pigs

  • Eight ileally cannulated pigs (BW $35.9{\pm}0.9kg$) were randomly allotted according to a $4{\times}3$ Latin square design to determine the effects of cellulose, pectin and starch on standardized ileal digestibility (SID) and apparent total tract digestibility (ATTD) of crude protein (CP) and amino acids (AA) as well as on the bacterial AA contribution in feces. The pigs were fed the control diet (20.2% CP, % dry matter (DM)) or one of the three experimental diets in which 25% of the control diet was substituted by cellulose, starch or pectin. Due to this substitution, dietary CP levels were lower in the cellulose (15.5% CP, % DM), pectin (15.4% CP, % DM) and starch diet (15.2% CP, % DM). Following a 15-d adaptation period, feces were collected for 5 d and ileal digesta for a total of 24 h. Starch increased SID of CP, while cellulose and pectin had no significant effect on the digestibility of CP. Overall, starch supplementation resulted in higher (p<0.05) SID values of histidine, isoleucine, threonine, alanine, aspartic acid, cysteine, glycine and serine compared with cellulose, while pectin decreased (p<0.05) SID of valine and proline compared with the starch and control diet. Both cellulose and pectin reduced (p<0.05) the ATTD of CP and AA, while starch decreased (p<0.05) ATTD of phenylalanine, alanine, proline and serine compared with the control. With regard to bacterial AA composition of the fecal mixed bacterial mass (MBM), cellulose supplementation increased (p<0.05) its content of N and almost all AA, except for valine, while pectin caused higher contents of arginine, histidine and proline compared with the control (p<0.05). The bacterial contribution of arginine in feces was higher (p<0.05) in the cellulose treatment, while pectin reduced (p<0.05) the bacterial contribution of leucine, alanine, glutamic acid and proline in feces compared with the control. In conclusion, the effects of cellulose, starch and pectin on SID were rather small. Bacterial activity in the large intestine can only explain the reduced ATTD values for arginine in the cellulose treatment, but not for the other AA in the cellulose and pectin treatments, suggesting higher endogenous losses of these AA in the large intestine.

Down-regulation of SENP1 Expression Increases Apoptosis of Burkitt Lymphoma Cells

  • Objective: To investigate the effect of down-regulation of Sentrin/SUMO-specific protease 1 (SENP1) expression on the apoptosis of human Burkitt lymphoma cells (Daudi cells) and potential mechanisms. Methods: Short hairpin RNA (shRNA) targeting SENP1 was designed and synthesized and then cloned into a lentiviral vector. A lentiviral packaging plasmid was used to transfect Daudi cells (sh-SENP1-Daudi group). Daudi cells without transfection (Daudi group) and Daudi cells transfected with blank plasmid (sh-NC-Daudi group) served as control groups. Flow cytometry was performed to screen GFP positive cells and semiquantitative PCR and Western blot assays were employed to detect the inference efficiency. The morphology of cells was observed under a microscope before and after transfection. Fluorescence quantitative PCR and Western blot assays were conducted to measure the mRNA and protein expression of apoptosis related molecules (caspase-3, 8 and 9). After treatment with $COCl_2$ for 24 h, the mRNA and protein expression of hypoxia inducible factor -$1{\alpha}$ (HIF-$1{\alpha}$) was determined. Results: Sequencing showed the expression vectors of shRNA targeting SENP1 to be successfully constructed. Following screening of GFP positive cells by FCM, semiqualitative PCR showed the interference efficiency was $79.2{\pm}0.026%$. At 48 h after transfection, the Daudi cells became shrunken, had irregular edges and presented apoptotic bodies. Western blot assay revealed increase in expression of caspase-3, 8 and 9 with prolongation of transfection (P<0.05). Following hypoxia treatment, mRNA expression of HIF-$1{\alpha}$ remained unchanged in three groups (P>0.05) but the protein expression of HIF-$1{\alpha}$ markedly increased (P<0.05). However, in the sh-SENP1-Daudi group, the protein expression of HIF-$1{\alpha}$ remained unchanged Conclusion: SENP1-shRNA can efficiently inhibit SENP1 expression in Daudi cells. SENP1 inhibition may promote cell apoptosis. These findings suggest that SENP1 may serve as an important target in the gene therapy of Burkitts lymphoma.

Zinc Nutritional Status in Korean Adults from Rural, Urban and Metropolitan Areas

  • The zinc intake and status of South Koreans from rural, urban and metropolitan areas were compared to evaluate the zinc nutritional status in different regional areas in South Korea. The dietary habits of 721 healthy adult subjects (271 from rural, 240 from urban, 210 from metropolitan city) with an age range 30 ∼ 64 (mean age 54$\pm$18) were assessed using a food frequency questionnaire. Mean daily Zn intake for rural, urban, and metropolitan areas was 6.5 mg, 7.3 mg, and 11.4 mg (p<0.05), respectively, which was 54%,61% and 95% of the Korean RDA for man (12 mg/d). Mean phytate: zinc molar ratios for rural, urban, and metropolitan city were 41, 34, and 30, respectively (p < 0.05), which were higher than the cutoff level of 20 for poor zinc status. The zinc intake and phytate: Zn molar ratio in the rural area were 0.5 and 1.3-fold compared to those of the metropolitan city, which can cause poor zinc nutriture in the rural area. Most of the zinc biomarkers were lower in the rural area than in the metropolitan city (p < 0.05) (mean rural and metropolitan values for plasma Zn: 80.8 $\mu\textrm{g}$/dL and 119.8 $\mu\textrm{g}$/dL, respectively; RBC Zn: 7.8 $\mu\textrm{g}$/dL and 8.8 $\mu\textrm{g}$/dL, respectively; plasma alkaline phosphatase (ALP) activity: 87 mU/mL and 100.4 mU/mL, respectively). It seems that a lower zinc intake in the rural area decreased zinc biomarker levels, such as plasma and RBC zinc, and plasma alkaline phosphatase activity, and caused the poor zinc nutritional status in this area. Most of the zinc biomarkers, such as RBC zinc and urinary and plasma zinc levels, in the subjects from the three localities, were within the normal range even when zinc intake of rural and urban subjects was low. The exception was plasma ALP activity in the rural area, which was lower than the reference level. Thus, marked zinc deficiency in these subjects were not observed, however, the potential for marginal zinc deficiency should be considered, especially for the rural area, because of the low zinc intake and the biomarker levels for marginal zinc deficiency.

Carcass characteristics of lambs fed spineless cactus as a replacement for sugarcane

  • Objective: Fresh sugarcane has been a new roughage source for ruminant's in semiarid regions, a function of the decline of sugar and alcohol industry in recent years. However, there is little data published regarding lambs fed sugarcane associated with spineless cactus. This study evaluated the effect of sugarcane replacement with spineless cactus (0%, 33%, 66%, and 100%) in the diet of Santa $In\hat{e}s$ lambs on carcass characteristics. Methods: Thirty-six non-castrated Santa Ines lambs at four months of age and an initial body weight of $22{\pm}2.3kg$ were assigned in a randomized block design and slaughtered after 70 days of confinement. The effects of spineless cactus as a replacement for sugarcane in the diet of the lambs on the carcass characteristics, commercial cut weight and yield, leg tissue composition, and carcass measurements were studied. Results: The study revealed quadratic behavior in slaughter body weight, and hot and cold carcass weight, with maximum values of 38.60, 18.60, and 18.11 kg and replacement levels of 40.18%, 44.42%, and 43.14%, respectively. The cold carcass yield presented an increasing linear behavior. The compactness index of carcass and leg presented a quadratic effect, with estimated maximal values of 0.28 and 0.57 kg/cm and replacement levels of 43.37% and 45.5%, respectively. The weights of commercial cuts of leg, loin, shoulder, and breast showed quadratic behavior, with maximum values of 2.79, 0.852, 1.46, and 1.30 kg and replacement levels of 49.5, 45.32, 39.0, and 40.7, respectively. For tissue composition, quadratic behavior was verified for leg weight, subcutaneous fat, and total fat. Conclusion: The replacement of sugarcane by spineless cactus at level 44% is recommended for finishing lambs considering that this level improved most of the carcass characteristics, weights, and yields of commercial cuts and leg tissue composition.
