The Galactose-${\alpha}1$,3-galactose (${\alpha}1$,3Gal) epitope is responsible for hyperacute rejection in pig-to-human xenotransplantation. Human decay-accelerating factor (hDAF) is a cell surface regulatory protein that serves as a complement inhibitor to protect self cells from complement attack. The generation of ${\alpha}1$,3-galactosyltransferase (GGTA1) knock-out pigs expressing DAF is a necessary step for their use as organ donors for humans. In this study, we established GGTA1 knock-out cell lines expressing DAF from pig ear fibroblasts for somatic cell nuclear transfer. hDAF expression was detected in hDAF knock-in heterozygous cells, but not in normal pig cells. Expression of the GGTA1 gene was lower in the knock-in heterozygous cell line compared to the normal pig cell. Knock-in heterozygous cells afforded more effective protection against cytotoxicity with human serum than with GGTA1 knock-out heterozygous and control cells. These cell lines may be used in the production of GGTA1 knock-out and DAF expression pigs for xenotransplantation.

This study was conducted to determine the serum antibodies against toxoplasma in swine from breeding-pig farm, pig farm and abattoir by latex agglutination(LA) test. LA test was carried out with commercial Toxo-MT kit (Eiken chemical co.). The results obtained were summerized as follows : 1. The cut-off titer of positive and negative reactions by Toxo-MT antigen used in this experiment was determined as the serum dilution of 1 ; 32. 2. positive rates of toxoplasma antibodies in 823 swine sera were 17.0%(140 cases) by LA test. 3. The toxoplasma antibody detection rates against 194 swine sera in breeding-pig farm, 273 swine sera in pig farm and 356 swine in abattoir were 46.9%(91 cases), 8.4%(23 cases) and 7.3% (26 cases) , respectively. 4. In LA test serum antibody titers in 823 test sera were shown as 51 cases (36.4%) in 1 : 32, 40(28.6%) in 1；64, 17(12.1%) in 1:128, 14(10.0%) in 1：256, 10(7.1%) in 1:512, 5(3.6%) in 1：1,024, and 3(2.1%) in 1 : 2,048. 5. Positive rates of toxoplasma antibodies in swine sera from each breeding-pig farm were 20.0∼61.9%.

Transplantation is considered to be a very useful approach to improve human welfare and to prolong life-span. Heterologous organ transplantation using pig organs which are similar to human beings and easy to make mass-production has known as one of the alternatives. To ensure potential usage of the pig organ for transplantation application, it is essentially required to generate transgenic pig modifying immuno-related genes. Previously, we reported production of heterozygous ${\alpha}1,3$-galactosyltransferase (GalT) knock-out and human membrane cofactor protein (MCP) expressing pig ($GalT^{-MCP/+}$), which is enforced for suppression of hyperacute and acute immunological rejection. In this study, we reported generation of homozygous pig ($GalT^{-MCP/-MCP}$) by crossbreeding $GalT^{-MCP/+}$ pigs. Two female founders gave birth to six of $GalT^{-MCP/-MCP}$, and seven $GalT^{-MCP/+}$ pigs. We performed quantitative real-time PCR, western blot, and flow cytometry analyses to confirm GalT and MCP expression. We showed that fibroblasts of the $GalT^{-MCP/-MCP}$ pig do not express GalT and its product Gal antigen, while efficiently express MCP. We also showed no expression of GalT, otherwise expression of MCP at heart, kidney, liver and pancreas of transgenic pig. Taken together, we suggest that the $GalT^{-MCP/-MCP}$ pig is a useful candidate to apply xenotransplantation study.

The effect of nitric oxide synthase(NOS) inhibita, $N^G$-nitro-L-arginine-methyl ester(L-NAME) and prostanoid synthesis inhibiter, indomethacin on the photorelaxation, when was exposed to the long-wave length UV-light, was examined on the precontraction by the phenylephrine in the isolated pig renal artery. 1. UV-light relaxed both with-endothelium and without-endothelium in the pig renal arterial ring contracted by the phenylephrine. The magnitude of photorelaxation was dependent on the exposure time for UV-light. 2. UV-Iight induced relaxation was inhibited by L-NAME and indomethacin on the precontraction by the phenylephrine in the isolated pig renal artery. 3. UV-Iight induced relaxation was inhibited by methylene blue on the precontraction by the phenylephrine in the isolated pig renal artery. These results suggest that UV-light induced photorelaxation may be due to cGMP involved both nitric oxide and prostanoid on the precontraction by the phenylephrine in the isolated pig renal artery.

Recently, Korean food culture has westernized and the consumption of meat has been increased. As a result an enormous amount of disposal of bone is generated and most of them are buried without reutilization. By making bone ash using pig bone, the possibilities of application are examined. The purpose of this study is to establish a manufacturing process of bone ash for the celadon bone body using pig bone. The calcination of the pig bone was mostly to change to the calcium phosphate hydroxide $(Ca_5(PO_4)_3OH)$. The celadon body mixed with pig bone ash 8%, $CaCO_3$ 9%, when fired at $1240^{\circ}C$ under reduction atmosphere, shows 0.1% of water absorption, 65.23 MPa of bending strength.

The following results were obtained from the observation on the change of plasma cortisol concentration in the experiment of intravenous administration of Haengsoyeum Water Extract in the rabbit. And the effects of Haengsoyeum extract on the contractile force of the isolated guinea pig trachea smooth muscle. 1. In intravenous administration the plasma cortisol concentration increased significantly about 1 hours after with a does of 0.2 ml/kg. 2. In intravenous administration the plasma cortisol concentration increased significantly about from 1 to 3 hours after with a does of 0.4 ml/kg. 3. The contractile response of the trachea smooth muscle of isolated guinea pig to histamine $10^{-4}\;M$ was significantly inhibited by Haengsoyeum extract. 4. The contractile response of the trachea smooth muscle of isolated guinea pig to acetylcholine $10^{-4}\;M$ was considerably inhibited by Haengsoyeum extract. 5. The contractile response of the trachea smooth muscle of isolated guinea pig to 5-hydroxytryptamine $10^{-4}\;M$ was inhibited by Haengsoyeum extract.

The objective of this study was to determine effects of humic acid (HA) and blueberry leaf powder (BLP) supplementation in pig feed on productivity, blood profiles, and meat quality characteristics of longissimus muscle. The experimental design included six treatments: 1) CON, no addition; 2) T1, BLP 0.1%; 3) T2, BLP 0.2%; 4) T3, humic acid 2%; 5) T4: humic acid 2%+BLP 0.1%; and 6) T5: humic acid 2%+BLP 0.2%. HA and BLP supplementation in pig feed significantly increased average daily feed intake (ADFI) values (p<0.05). HA supplementation in pig feed had beneficial effects in lipid profiles without altering feed efficiency rate (FER). HA and BLP co-supplementation in pig feed decreased pH in longissimus thoracis (p<0.05). In addition, sensory characteristics were enhanced when pig feed was supplemented with HA and BLP without causing adverse effects in meat quality. Taken together, addition of HA and BLP in pig feed may produce functional meat products.

The aims of this study were to investigate the effect of adenosine triphosphate (ATP), which has been known as the neurotransmitter of nonadrenergic, noncholinergic nerves, and the source of $Ca^{\sharp}$ in the effect of ATP on the isolated renal artery of pig. The results of this study were summarized as follows: 1. ATP caused the contraction and the contractile responses were increased in a dose-dependent manner between the concentration of ATP $2{\times}10^{-3}M$ and $10^{-2}M$ on the isolated renal artery of pig. 2. The contractile responses induced by ATP $(5{\times}10^{-3}M)$ were not blocked by pretreatment with cholinergic receptor blocker (atropine, $10^{-6}M$), $\alpha$-adrenergic recptor blocker(phentolamine, $10^{-6}M$) or $\beta$-adrenergic receptor blocker (propranolol, $10^{-6}M$), and $H_1$-receptor blocker (pyrilamine, $10^{-6}M$) or $H_2$-receptor blocker (cimetidine, $10^{-6}M$) on the isolated renal artery of pig. 3. The contractile responses induced by ATP $(5{\times}10^{-3}M)$ were not appeared in $Ca^{\sharp}$-free medium. As the concentration of $Ca^{\sharp}$ in $Ca^{\sharp}$-free medium was increased, the contractile responses induced by ATP $(5{\times}10^{-3}M)$ were enhanced but were completely inhibited by pretreatment with $Ca^{\sharp}$-channel blocker, papaverine $(5{\times}10^{-5}M)$ or verapamil $(5{\times}10^{-5}M)$ on the isolated renal artery of pig.

Background: Patients with pyogenic lung abscess who do not respond to medical therapy are usually candidates for thoracotomy with pulmonary resection. Percutaneous tube drainage, used routinely and with good results before the antibiotic era, has nearly been forgotten. Methods: We treated 8 patients who had poor cadidates for a lobectomy and didn't respond to medical management. A pig-tail catheter (6 to 10 Fr.) was inserted into the abscess under the fluoroscopic guidance. Results: All patients brought about dramatic clinical responses with prompt closure of cavity and radiographic improvement. Conclusion: Percutaneous pig-tail catheter drainage provides excellent clnical results with ease and safety, and avoids unnecessary loss of functioning lung parenchyma. But lobectomy should be considered in patients who major life-threatening bleeding or massive pulmonary necrosis.

This study was conducted to assess performances of the developed environmental control systems under various seasons of Korea. In all trials for the environmental control systems, the manure pit ventilation system in the windowless pig-housing with partly slatted floor was used. Consequently, under all seasons of Korea, the complex environmental control systems could comfortably maintain the indoor temperature (14.8~27.2$^{\circ}C$) , concentrations of noxious gases (CO2 gas : 631~1,874ppm, NH3 gas : 0.3~3.2ppm), air velocity (0.11~0.23m/s), air movement, and so on. Therefore, the performances of the complex environmental control systems were evaluated as proper as the intended.
