The analysis of variational characteristics on water quality and phytoplankton by principal component analysis(PCA) in Kogum-sudo, Southwestern part of Korea (주성분분석에 의한 거금수도의 수질환경 및 식물플랑크톤 변동 요인 해석)

  • A study on the variational characteristics of water quality and phytoplankton biomass by principal component analysis(PCA) was carried out in Kogum-sudo from February to October in 1993. We analyzed PCA on biological factors such as chlorophyll a and phytoplankton cell numbers for centric and pennate diatoms, phytoflagellates, and total phytoplankton as well as physico-chemical factors as water temperature, salinity, transparency, dissolved oxygen(DO), saturation of DO, apparent oxygen utilization (AOU), chemical oxygen demand(COD), nutrient (ammonia, nitrite, nitrate, phosphate and silicate), N/P ratio and suspended solid(SS). The source of nutrients supply depended on the mineralization of organic matters and inputs of seawater from outside rather than runoff of freshwater. The phytoplankton biomass was changed within short interval period by nutrients change. And it was controlled by the combination of several environmental factors, especially of light intensity, ammonia and phosphate. The marine environmental characteristics were determined by the mineralization of organic matters in winter, by runoff of freshwater including high nutrients concentration in spring, by ammonia uptake and high phytoplankton productivity in summer, and phosphate supplied input seawater from outside of Kogeum-sudo in autumn. And Kogum-sudo was separated with 2 regions by score distributions of PCA. That is to say, one region was middle parts of straits which was characterized by the mixing seawater and the accumulated organic matters, other one region was Pungnam Bay and the water around Kogum Island which was done by high phytoplankyon biomass and productivity year-round.

Controlled Release Behavior of Bioactive Molecules from Photo-Reactive Hyaluronic Acid-Alginate Scaffolds

  • There are three important components in tissue engineering: the cells, signaling factors (cytokines and growth factors), and scaffolds. To obtain finely engineered tissue, all three components should perform their individual functions and be fully integrated with each other. For the past few years, we have studied the characteristics of photodimerizable HA (CHA)/alginate (CA) composite materials. CHA/CA complex hydrogels, which were irradiated under UV light and, then treated with calcium ions, were found to have good biocompatibility, mechanical properties and water resistance for implantable tissue scaffolds. In this study, we introduced a cell growth factor (basic fibroblast growth factor; bFGF) into the CHA/CA scaffolds and studied its release behavior. We also introduced tetracycline hydrochloride and flurbiprofen into the same scaffolds as model activation factors and evaluated their release behaviors from the scaffolds. The drug release rate from the materials was influenced by various parameters, such as the degree of crosslinking, the cross linker type, the physico-chemical properties of the drug, and the amount of the drug in the polymer. The results indicated that the negatively charged CHA/CA composite materials showed sustained release behavior and that HA has a particularly strong negative charge, making it attractive toward tetracycline hydrochloride and bFGF, but repulsive toward flurbiprofen.

  • A Study on the effect of the additives in lubricating oil was investigated on the basis of the thermal activated wear theory in terms of their wear behaviours, using four ballwear machine. The sample oils, which included diethyl-3, 5-di-t-butyi-4-hydroxy-benzyl phosphonate (DEP), ZDDP and TCP additives respectively, showed distinct wear characteristics depending upon the bulk oil temperature and the sliding velocity. The newly synthesized additive, viz., DEP showed excellent antiwear performance cornpared with the conventional additives, ZDDP and TCP. On the basis of the experimental results, it is reduced that the wear mechanism of the conventional additives, viz., ZDDP and TCP is the protective film formation and their antiwear capability is depending upon the shearing strength of the film formed. On the other hand, the new additive, DEP showed that the secondary activation energy was much eliminated and so, the thermal instability was reduced by the hydrogen scavenging reaction of the new additive, which was virtually an endothermic reaction process.In conclusion, a new concept of antiwear mechanism is estabilished and testified. And new chemical, which showed the function of hydrogen and free radical scavenging role, is synthesized and introduced as the new, highly antiwear effective lubricating oil additive.

Distribution of bacterial biomass in the water column of Soyang lake (소양호 수중 생태계에서의 세균 생체물질량의 분포)

  • Microbiological parameters such as bacterial biovolume and biomass in Soyang Reservoir was statistically analyzed with the physico-chemical enviromental factors. Analysis of correlation and multiple regression showed that temperature affects most of microbiological parameters. Variations of total bacterial number, total bacterial biovolume and saprophyte number were highly correlatd with the concentrations of chlorophyll a and pheophytin a. Bacterial production by the $^{3}H$-thymidine incorporation rate was largely affected by Seston. It suggests that microbiological factors such as bacterial biovolume and bacterial biomass were controled by the concentration of seston and distribution of phytoplankton which acts as carbon and energy source for the bacterial community in the water column of Soyang Reservoir.

Effect of Marbling Score on Carcass Grade Factors, Physico-chemical and Sensory Traits of M. Longissimus Dorsi in Hanwoo (근내지방도가 한우 도체등급 요인, 이화학적 특성 및 관능적 특성에 미치는 영향)

  • The objective of this research was to investigate the effect of marbling scores on carcass grade factors, physico-chemical and sensory traits of Hanwoo. Data used in this study were collected from 73,316 carcasses obtained at the Nonghyup Seoul slaughterhouse in 2009 and 271 cuts of M. longissimusdorsi were analyzed to estimate beef qualities. As the marbling scores increased, backfat thickness was linearly increased (p<0.05) in cows (9.51 mm-14.69 mm) and steers (8.71 mm-14.23 mm). As the marbling scores increased, maturity of cows was increased and meat color, texture and quality grade improved for all genders. With increase of the marbling scores, crude fat contents (3.07%-26.70%), water holding capacity (52.09%-58.66%), $L^*$ value (34.08-41.97) were significantly increased (p<0.05) and Warner-Bratzler shear force was linearly decreased (p<0.05) from 8.58 kg to 2.60 kg. The increase of crude fat contents had the similar interval with the increase of marbling scores from 1 to 6 but it is sharply increased from marbling score 7. There was no difference observed on $pH_{24}$ (5.50-5.66), protein contents (19.57%-21.15%) among the marbling scores. Marbling score was significantly correlated with live weight (r=0.29) in cows, loineye area (r=0.35) in steer, and overall acceptability of panel scores (r=0.67) (p<0.01).

Incubation Conditions and Physico-Chemical Factors Affecting Aflatoxin B1 Binding of Lactic Acid Bacteria (Aflatoxin B1에 대한 유산균의 결합력에 영향을 미치는 배양조건과 물리화학적 인자)

  • The purpose of this study was to investigate the aflatoxin $B_1$ binding of lactic acid bacteria (LAB) isolated from Korean traditional soybean paste and to evaluate the effect of incubation conditions and physico-chemical factors on the binding ability of LAB to this mutagen. The amount of aflatoxin $B_1$ bound by Enterococcus faecium DJ22, Lactobacillus fermentum DJ35, Lactobacillus rhamnosus DJ42, and Lactobacillus pentosus DJ47 was strain specific with the percent bound ranging from 19.3% to 52.1%. However, Enterococcus faecalis DJ14, Lactobacillus panis DJ29, and Pediococcus halophilus DJ50 strains did not exhibit any of the binding ability to aflatoxin $B_1$. For most strains, the binding ability was significantly affected by the environmental conditions such as the aflatoxin $B_1$ level, incubation time and temperature, and the initial cell count of LAB. The stability of the aflatoxin $B_1$-bacteria complexes was significantly more unstable after washing. In addition, the binding stability between viable and nonviable cells was not statistically significant. Treatment with heating, acidic pH, ${\alpha}$-amylase, protease, lysozyme, or sodium metaperiodate caused a significant (P<0.05) decrease in aflatoxin $B_1$ binding for the tested strains, suggesting that carbohydrates or proteins in the cell walls may be involved in aflatoxin $B_1$ binding ability. Since the aflatoxin $B_1$ binding of LAB was significantly reduced (P<0.05) by the pretreatment of the urea, the binding force observed in this study may have resulted from hydrophobic interaction.

Seasonal Variation of Picoplankton Community in Lake Juam (주암호에서 미세조류의 계절적 군집 변화)

  • The purpose of this study is to investigate the seasonal variation of picoplankton community in Lake Juam depending on the change of physico-chemical factors such as rainfall, water depth, DO and pH. The concentration of chlorophyll-a was most high as 18.03 mg/$m^3$ in July when the rainfall and water temperature were highest. The concentration was gradually decreased in October, April and that of January was decreased most low as 1.86 mg/$m^3$. The highest concentration of the Chl-a was shown at 2 and 5 m of water depth than surface, and the concentration was gradually decreased when the water depth becomes deep. Overall, microplankton was the highest rate as 33.9~54.2%, nanoplankton was 24.3~30.5% and picoplankton was 21.6~41.2%. Picoplankton was included as considerable concentration in the water of Juam lake. Therefore it is necessary to remove thoroughly the picoplankton in the water treatment processes such coagulation·sedimentation and sand filtration. The protoplasm released from destruction of picoplankton by chlorine has high possibility to cause regrowth of bacteria and pathogenic microorganism in the distribution system by playing the role of the assimilable organic carbon.

A Study on the Epilithic Algae in a stream drained from Hot Springs (온천 주변 소형하천에 서식하는 부착조류군집에 관한 연구)

  • Attached algal community and several physico-chemicai characters were investigated at Seokjong hot spring in Chollabukdo in February and in April, 1997. The results of physico-chemicai factors are as follows : water temperatures were varied from 6.5$^{\circ}$C to 32.0$^{\circ}$C; pH range was 6.9 to 7.7; The levels of electric conductivity were 105 $\mu$mhos/cm to 477 $\mu$mhos/cm; chemical oxygen demand(COD) range was 1.5 mg/l to 21.2 mg/l; ammonia concentration was 0.20 mg/l to 8.74 mg/l; nitrate concentrations were 0.36 mg/l to 2.43 mg/l; phosphate concentration was N.D. to 0.52 mg/l; sulfur concentration was 14.6 mg/l to 66.1 mg/l. Attacched algal species were identified. Totally, 63 taxa were classified and composed of 4 phylum, 4 class, 10 order, 17 family and 31 genera. The total biomass of attacched algae was $1.036 \times 10^{7}$ cells/l that composed of $5.39 \times 10^{6}$ cells/l of blue-green algae, $4.05 \times 10^{6}$ cells/l of diatom and $0.93 \times 10^{6}$ cells/l of green algae. The succession of dominant species was Synechocystis thermalis. This could be due to the thermal wastewaters. in biotic indices tests, the values of dominance index(DI) were 0.33 to 0.67 in winter, 0.18 to 0.68 in spring. The values of diversity index(H') were 1.44 to 2.69 in winter, 1.62 to 2.89 in spring. And the values of eveness index (J) were 0.31 to 0.61 in winter, 0.37 to 0.65 in spring.

Improving the Reliability of the National Database for Chemical Hazard Information (국가 화학물질 유해성정보 데이터베이스 구축 과정의 신뢰도 제고 방안에 관한 연구)

  • Objectives: According to the Act on Registration, Evaluation, Etc. of Chemicals, new and existing chemicals must be registered by 2030. In addition, industries need to submit hazard data as an attachment during the registration process. Therefore, we constructed a nationwide chemical database to support small industry by providing hazard data and original sources. During the process, we developed a new standard procedure for minimizing errors and increasing reliability. Methods: We analyzed the categories of errors and the cause of the errors through the verification results of the 2019 project. We present an improved database construction methodology and system. Results: Errors are categorized according to their causative factors into simple, technical, and structural type errors. Simple errors arise simply because of decreased concentration or negligence in following the instructions. Technical errors are caused by a discrepancy between the professional field and the type of data. Structural errors indicate systemic errors such as incomplete forms on the excel database or ambiguity in the guidelines. Lessons from the errors collected in the 2019 project are used to update the procedures for database authorization and technical guidelines. The main update points are as follows; 'supplementation of review process', 'giving regular training to external reviewers', 'giving additional information to authors, like physico-chemical properties of substances, degradability, etc.', 'amendment of excel form', and 'guideline upgrades'. Conclusions: We conducted this study with the aim of improving the accuracy and reliability of the database of hazard information for chemical substances. The new procedures and guidelines are now being used in the 2020 project for construction of a hazard information database for Korea.

Properties of Organically Practiced Paddy Soils (유기농 실천 논토양의 이화학적 특성)

  • To produce the primary data for best management of soil nutrient in organically practiced soils, 75 leading organic farms whose paddy fields were certified as organic were selected. Soil samples were collected from the paddy fields before plowing, and then analyzed for the determination of physico-chemical properties. Soil pH, organic matter and available phosphate were analyzed and averaged 6.2, $25.6mg\;kg^{-1}$ and $88.4mg\;kg^{-1}$, respectively. Contrary to the national-scale-surveyed paddy soils including organic and conventional farming, pH was higher, available phosphate was lower in the organically practiced soils, but organic matter was similar. With the increasing cultivation period in organic, soil pH and porosity were also increased, EC, available phosphate, bulk density and soil hardness were lower than those from the national survey. Organic matter, however, was not significantly changed. The bulk density was negatively correlated with the organic matter content for both surface topsoil ($R^2=-0.5424$) and subsoil ($R^2=-0.6429$) (p<0.05). Soil quality is improved in most of soil chemical and physical composition factors excluding organic matter and available phosphate. However, it is necessary to establish the counter measure plan for organic matter management and to develop phosphate-containing materials which can be used as organic agricultural material in the future.
