Incapability of Utilizing Galactose by pgs1 Mutation Occurred on the Galactose Incorporation Step in Saccharomyces cerevisiae

  • A Saccharomyces cerevisiae pgs1 nulI mutant, which is deficient with phosphatidyl glycerol (PG) and cardiolipin (CL) biosynthesis, grows well on most fermentable carbon sources, but fails to grow on non-fermentable carbon sources such as glycerol, ethanol, and lactate. This mutant also cannot grow on galactose medium as the sole carbon source. We found that the incorporation of $[^{14}C]-galactose$, which is the first step of the galactose metabolic pathway (Leloir pathway), into the pgs 1 null mutant cell was extremely repressed. Exogenously expressed PGS1 (YCpPGS1) under indigenous promoter could completely restore the pgs1 growth defect on non-fermentable carbon sources, and dramatically recovered $[^{14}C]-galactose$ incorporation into the pgs1 mutant cell. However, PGS1 expression under the GALl promoter $(YEpP_{GAL1}-PGS1myc)$ could not complement pgs1 mutation, and the GAL2-lacZ fusion gene $(YEpP_{GAL2}-lacZ)$ also did not exhibit its $\beta-galactosidase$ activity in the pgs1 mutant. In wild-type yeast, antimycin $A(1\;{\mu}g/ml)$, which inhibits mitochondrial complex III, severely repressed not only the expression of the GAL2-lacZ fusion gene, but also uptake of $[^{14}C]-galactose$. However, exogenously expressed PGS1 partially relieved these inhibitory effects of antimycin A in both the pgs1 mutant and wild-type yeast, although it could not basically restore the growth defect on galactose by antimycin A. These results suggest that the PGSI gene product has an important role in utilization of galactose by Gal genes, and that intact mitochondrial function with PGS1 should be required for galactose incorporation into the Leloir pathway. The PGS1 gene might provide a clue to resolve the historic issue about the incapability of galactose with deteriorated mitochondrial function.

  • The recent knowledge about the effects and the biological roles of prostaglandins(PGs) in the female reproductive system were reviewed on the following view points in detail and the authors also argued on the efficacy and reliability for the induction of labour and therapeutic abortions in midtrimester pregnancy. 1. The actions of PGs on the myometrium in vivo and in vitro and the effects of ovarian hormones on the actions of PGs. 2. The actions of PGs on the fallopian tube. 3. The role of PGs in the initiation of menstrual bleeding and dysmenorrhea. 4. The biological action of PGs on the mechanism of labour and parturition. 5. The efficacy and reliability of PGs for therapeutic abortion in the first and midtrimester pregnancy. 6. The biological role of PGs in the activity and life-span of corpus luteum.

Periplasmic glucans isolated from Proteobacteria

  • Periplasmic glucans (PGs) are general constituents in the periplasmic space of Proteobacteria. PGs from bacterial strains are found in larger amounts during growth on medium with low osmolarity and thus are often been specified as osmoregulated periplasmic glucans (OPGs). Furthermore, they appear to play crucial roles in pathogenesis and symbiosis. PGs have been classified into four families based on the structural features of their backbones, and they can be modified by a variety of non-sugar substituents. It has also recently been confirmed that novel PGs with various degrees of polymerization (DPs) and/or different substituents are produced under different growth conditions among Proteobacteria. In addition to their biological functions as regulators of low osmolarity, PGs have a variety of physico-chemical properties due to their inherent three-dimensional structures, hydrogen-bonding and complex-forming abilities. Thus, much attention has recently been focused on their physico-chemical applications. In this review, we provide an updated classification of PGs, as well as a description of the occurrences of novel PGs with substituents under various bacterial growth environments, the genes involved in PG biosynthesis and the various physico-chemical properties of PGs.

i-PGS 기반 선회베어링의 접촉피로강도 설계 (Contact Fatigue Strength Design of a Slewing Bearing Based on i-PGS)

  • To overcome the large ring gear manufacturing problems seen in slewing bearings and girth gears, pin gear drive units have been developed. Among them, a novel slewing bearing with an internal pinwheel gear set (i-PGS) is introduced in this paper. First, we consider the exact cam pinion profile of i-PGS with the introduction of a profile shift coefficient. Furthermore, a new root relief profile modification for the i-PGS cam pinion is presented. Then, the contact stresses are investigated to determine the characteristics of the surface fatigue by varying the shape design parameters. The results show that the contact stresses of i-PGS can be reduced significantly by increasing the profile shift coefficient. In addition, the contact ratio, a measure of teeth overlapping action, decreases with the decrease of the allowable pressure angle.

  • 초고주파에서 집적회로용 연결선(interconnect)에 PGS(Patterned Grouned Shield)를 적용하는 실험을 하였다. PGS는 신호선으로부터 비절연 실리콘 기판을 차폐시킴으로써 광대역에 걸쳐 전송선의 실리콘기판을 통한 전력손실을 크게 줄일 수 있음을 측정결과를 통해 보였다. 또한 PGS를 이용한 전송선의 특성을 분석하고 PGS가 전송선의 파장을 줄여주는 효과가 있음을 확인하였다.

The Urban Parks and Rivers Contribute to the Citizen Satisfaction and Utilization in Uijeongbu City

  • This research aimed at measuring Park and Green Satisfaction (PGS) using subjective indicators of 'surface, line and spot' green evaluated by citizens. Also frequency of visits to park and green measured using objective indicators (number of visits) to find the relationship with PGS. A conceptual model of PGS was developed to relate evaluation to satisfaction and finally to utilization of open spaces. A sample of 500 questionnaire survey was employed for Uijeongbu City in Korea. A Structual Equation Modeling (AMOS) techniques was used to test the hypothesized relationship among factors (construct). As a result, first, PGS was explained by three latent factors of 'urban park' (${\gamma}=0.54$), 'linear facilities' (${\gamma}=0.25$), and 'surface green' (${\gamma}=0.15$) respectively. These three exogenous construct was found very useful classification system for open spaces of cities. Second, PGS (${\gamma}=0.34$) was found as a mediating variable to utilization of open spaces and also PGS was closely related to citizens Environmental Quality Satisfaction (EQS), such concept as, 'livability' and 'aesthetic quality'. The more satisfied with park and green the more people use the space. The PGS was an important QOL indicator together with the subjective indicator of 'livability'. Third, jogging and walking trails and bike ways along the river corridor was the most important green facilities contribute to the PGS and EQS. The near the distance (within 500m) the more number of visit to river corridor (green way). The river corridor promote accessibility to nature and other parks.

인삼사포닌의 급성및 아급성 독성에 관한 연구 (Studies on the Acute and Subacute Toxicity of Ginseng Saponin)

  • Acute toxicities of purified ginseng saponin (PGS) in mice, and sbacute toxicities of PGS in rats were investigated. Average lethal doses ($LD_{50}$) of PGS in male mice were 270mg/kg (i.v.), 342mg/kg (i.p.), 505mg/kg (i.m.), 950mg/kg (s.c.), and more than 5,000mg/kg (p.o.), respectively. Results of subacute toxicity of PGS was as follows. Body weight was markedly increased by administration of PGS 7.7mg/kg but side effects were shown by administration of 77mg/kg and above dose. Especially administration of PGS 240mg/kg caused a marked decrease of albumin/globulin ratio, and 28% increase of urea nitrogen in serum, as well as 33% increase of liver weight/body weight ratio.

GaAs MMIC상에서 주기적 접지구조를 가지는 결합선로의 절연특성에 관한 연구 (A Study on Characteristics of Coupled Line Employing Periodical Ground Structure on GaAs MMIC)

  • In this study, using a periodical ground structure(PGS) on GaAs monolithic microwave integrated circuit(MMIC), transmission line with a high isolation characteristic was developed for application to compact signal/bias lines of highly integrated MMIC. And the origin of the high isolation characteristic was theoretically investigated. The high isolation characteristic was originated from a resonance between adjacent microstrip lines employing PGS. With only a spacing of $20{\mu}m$, the coupled microstrip line employing PGS showed an isolation value of -47 dB at 60 GHz. The frequency range for high isolation was easily controlled by changing the PGS structure. Above results indicate that microstrip lines employing PGS are very useful for application to compact signal/bias lines of highly integrated MMIC requiring a high isolation characteristics between lines. In addition, equivalent circuit employing closed-form equation for the coupled line with PGS was also extracted.

Surface-Displayed IL-10 by Recombinant Lactobacillus plantarum Reduces Th1 Responses of RAW264.7 Cells Stimulated with Poly(I:C) or LPS

  • Recently, poly-γ-glutamic acid synthetase A (pgsA) has been applied to display exogenous proteins on the surface of Lactobacillus casei or Lactococcus lactis, which results in a surface-displayed component of bacteria. However, the ability of carrying genes encoded by plasmids and the expression efficiency of recombinant bacteria can be somewhat affected by the longer gene length of pgsA (1,143 bp); therefore, a truncated gene, pgsA, was generated based on the characteristics of pgsA by computational analysis. Using murine IL-10 as an exogenous gene, recombinant Lactobacillus plantarum was constructed and the capacity of the surface-displayed protein and functional differences between exogenous proteins expressed by these strains were evaluated. Surface expression of IL-10 on both recombinant bacteria with anchorins and the higher expression levels in L. plantarum-pgsA'-IL-10 were confirmed by western blot assay. Most importantly, up-regulation of IL-1β, IL-6, TNF-α, IFN-γ, and the nuclear transcription factor NF-κB p65 in RAW264.7 cells after stimulation with Poly(I:C) or LPS was exacerbated after co-culture with L. plantarum-pgsA. By contrast, IL-10 expressed by these recombinant strains could reduce these factors, and the expression of these factors was associated with recombinant strains that expressed anchorin (especially in L. plantarum-pgsA'-IL-10) and was significantly lower compared with the anchorin-free strains. These findings indicated that exogenous proteins could be successfully displayed on the surface of L. plantarum by pgsA or pgsA', and the expression of recombinant bacteria with pgsA' was superior compared with bacteria with pgsA.

Phylogenetics and Gene Structure Dynamics of Polygalacturonase Genes in Aspergillus and Neurospora crassa

  • Polygalacturonase (PG) gene is a typical gene family present in eukaryotes. Forty-nine PGs were mined from the genomes of Neurospora crassa and five Aspergillus species. The PGs were classified into 3 clades such as clade 1 for rhamno-PGs, clade 2 for exo-PGs and clade 3 for exo- and endo-PGs, which were further grouped into 13 sub-clades based on the polypeptide sequence similarity. In gene structure analysis, a total of 124 introns were present in 44 genes and five genes lacked introns to give an average of 2.5 introns per gene. Intron phase distribution was 64.5% for phase 0, 21.8% for phase 1, and 13.7% for phase 2, respectively. The introns varied in their sequences and their lengths ranged from 20 bp to 424 bp with an average of 65.9 bp, which is approximately half the size of introns in other fungal genes. There were 29 homologous intron blocks and 26 of those were sub-clade specific. Intron losses were counted in 18 introns in which no obvious phase preference for intron loss was observed. Eighteen introns were placed at novel positions, which is considerably higher than those of plant PGs. In an evolutionary sense both intron loss and gain must have taken place for shaping the current PGs in these fungi. Together with the small intron size, low conservation of homologous intron blocks and higher number of novel introns, PGs of fungal species seem to have recently undergone highly dynamic evolution.
