Isolation of Bacillus amyloliquefaciens H41 Producing Growth Inhibition Factor against Vibrio anguillarum (어병균 Vibrio anguillarum 생육 저해 인자를 생산하는 Bacillus amyloliquefaciens H41의 분리)

  • To investigate the possible use of probiont in fish farming industry, a bacterium with inhibitory effect against Vibrio anguillarum was isolated from gastrointestinal tract of the marine fish of yellow tail. It was identified to be Bacillus amyloliquefaciens H41 based on biochemical and physiological characterization. The optimal growth conditions of the isolated strain were 1% peptone, 1.5% yeast extract, 1% sucrose, 0.5% NaCl, 0.05% $MgSO_4{\cdot}7H_20$, pH 7.0-8.0, and 20 hr of incubation between $28-35^{\circ}C$ under aeration. The culture supernatant of the isolated strain showed inhibition activity against V. anguillarum. Inhibition activity was cleared by forming a clear zone by a paper-disk method. The maximal production of growth inhibition factor was induced by cultivation under 1% peptone, 1.5% yeast extract, 1% sucrose, 1% NaCl, 0.05% $MgSO_4{\cdot}7H_20$, pH 7.5 and at $35^{\circ}C$. The highest growth inhibition factor production was observed after 16-24 hr cultivation under aeration. The culture supernatant of the isolated strain showed inhibition activity whereas no inhibition activity was shown from the standard B. amyloliquefaciens KCTC 1724 strain. The growth inhibition affected only against V. anguillarum among other pathogenic Vibrios tested here.

Optimization of an Industrial Medium and Culture Conditions for Probiotic Weissella cibaria JW15 Biomass Using the Plackett-Burman Design and Response Surface Methodology

  • The objective of this study was to optimize industrial-grade media for improving the biomass production of Weissella cibaria JW15 (JW15) using a statistical approach. Eleven variables comprising three carbon sources (glucose, fructose, and sucrose), three nitrogen sources (protease peptone, yeast extract, and soy peptone), and five mineral sources (K2HPO4, potassium citrate, ⳑ-cysteine phosphate, MgSO4, and MnSO4) were screened by using the Plackett-Burman design. Consequently, glucose, sucrose, and soy peptone were used as significant variables in response surface methodology (RSM). The composition of the optimal medium (OM) was 22.35 g/l glucose, 15.57 g/l sucrose, and 10.05 g/l soy peptone, 2.0 g/l K2HPO4, 5.0 g/l sodium acetate, 0.1 g/l MgSO4·7H2O, 0.05 g/l MnSO4·H2O, and 1.0 g/l Tween 80. The OM significantly improved the biomass production of JW15 over an established commercial medium (MRS). After fermenting OM, the dry cell weight of JW15 was 4.89 g/l, which was comparable to the predicted value (4.77 g/l), and 1.67 times higher than that of the MRS medium (3.02 g/l). Correspondingly, JW15 showed a rapid and increased production of lactic and acetic acid in the OM. To perform a scale-up validation, batch fermentation was executed in a 5-l bioreactor at 37℃ with or without a pH control at 6.0 ± 0.1. The biomass production of JW15 significantly improved (1.98 times higher) under the pH control, and the cost of OM was reduced by two-thirds compared to that in the MRS medium. In conclusion, OM may be utilized for mass producing JW15 for industrial use.

  • A sequential optimization strategy, based on statistical experimental designs, is employed to enhance the production of alkaline protease by a Bacillus pseudofirmus local isolate. To screen the bioprocess parameters significantly influencing the alkaline protease activity, a 2-level Plackett-Burman design was applied. Among 15 variables tested, the pH, peptone, and incubation time were selected based on their high positive significant effect on the protease activity. A near-optimum medium formulation was then obtained that increased the protease yield by more than 5-fold. Thereafter, the response surface methodology(RSM) was adopted to acquire the best process conditions among the selected variables, where a 3-level Box-Behnken design was utilized to create a polynomial quadratic model correlating the relationship between the three variables and the protease activity. The optimal combination of the major medium constituents for alkaline protease production, evaluated using the nonlinear optimization algorithm of EXCEL-Solver, was as follows: pH of 9.5, 2% peptone, and incubation time of 60 h. The predicted optimum alkaline protease activity was 3,213 U/ml/min, which was 6.4 times the activity with the basal medium.

  • The main objectives of the study were to find optimum fermentation conditions of Propionibacterium acidipropionici for the production of organic acids. Three strains of Propionibacterium acidipropionici were selected for batch fermentation. Nutrients and environmental conditions on cell growth were defined by series of experiments. The optimum conditions of peptone, yeast extract, pH were determined to be 1.5%, 0.75%, $5.5{\sim}7.5$, respectively. Among the straines of P. acidipropionici selected for the experiment, ATCC4965 was the best in terms of total productivity and cell yield, which values were 0.29g total acids/L/h and 0.26 g dry cell/g glucose, respectively.

Production of Hyaluronic Acid from Streptococcus zooepidemicus (Streptococus zooepidemicus에 의한 히아루론산의 생산)

  • An optimal composition of medium for hyaluronic acid production and some characteristics of its from Streptococcus zooepidemicus were investigated. The hyaluronic acid from S. zooepidemicus was reached maximum level in the BY-medium containing 0.1% beef extract, 0.1% yeast extract, 3.0% glucose, 2.0% peptone, 0.1% NaCl and $0.5%CaCO_3$ (pH 7.5) at $37^{\circ}C$ for 36 hours with shaking. Addition of $CaCO_3$ to the medium was necessary to neulralize the lowered pH which was resulted from hyaluronic acid production. Molecular weights of extracelluar and cellular hyaluronic acid produced by the strain were $1-1.4{\times }10^6$ and $5{\times}10^6$, respectively. The amount of extracellular hyaluronic acid was 91.9% of total hyaluronic acid produced and the vest was all intracellular.

  • The present researches were carried out to examine the favorable nutritional conditions for mycelial growth of Cercospora kikuchii, and inhibition of bacterial growth by the fungus at various cultural conditions. As the nitrogen source, the fungus grew better on yeast-extract than the other sources of nitrogen tested. Sucrose, glucose, maltose and fructose were good sources of carbon for mycelial growth of the fungus. Two isolates of C. kikuchii, wild types A and D, inhibited the growth of bacteria on the medium containing yeast-extract or peptone as the nitrogen source, and sucrose, glucose, maltose or fructose as the carbon source, at pH 5.0. However, the albino mutant showed very little inhibitory effect under the same conditions.

  • The ligninolytic basidiomycete, Pleurotus ostreatus K-2946, was produced a manganese peroxidase (MnP) activity when grown in liquid culture with glucose-yeast-peptone (G-Y-P) medium. However, lignin peroxidase (LiP) was not detected in this culture medium. The purification progress of MnP was purified that included chromatography on Sepharose CL-6B, Superdex 75 prep grade and Mono-Q. MnP purified by column chromatography, was 36400 dalton and a pI of 3.95. The optimal pH and temperature of the purified MnP activity were 5.0 and $55^{\circ}C$. The characteristics of MnP produced was quite similar to those of MnP 3 isoenzyme produced by other strains of P. ostreatus.

Characteristics of Metacordyceps yongmunensis, a New Species from Korea

  • Metacordyceps yongmunensis is a newly reported species from Korea, which is very similar to Cordyceps species in morphological characters. It grows on large lepidopteran pupa, and numerous white stromata grow on a single host. Mycelial growth characteristics of M. yongmunensis isolates were studied in different media and at different temperatures. Also, different carbon sources, nitrogen sources, and mineral salts were tested for mycelial growth of M. yongmunensis. Schizophyllum (mushroom) genetics complete medium plus yeast extract, Schizophyllum (mushroom) genetics minimal medium, and Martin's peptone dextrose agar produced longer colony diameters and more compact mycelial density than other media. The optimum temperature for mycelial growth was $25^{\circ}C$. Carbon sources such as sucrose, soluble starch, dextrose, glucose, dextrin, maltose, and fructose showed better mycelial growth, whereas peptone, yeast extract and tryptone resulted in the best mycelial growth of all of the nitrogen sources tested. All of the mineral salts tested showed similar growth as the control, except $K_2HPO_4$ which showed longer colony diameter and more compact mycelial density. The compact colonies were white and cottony with a greenish margin. The results showed that M. yongmunensis is an easy fungus to growas it grew from 30 to more than 50 mm in 2 wk.

Optimization of Hydroxyl Radical Scavenging Activity of Exopolysaccharides from Inonotus obliquus in Submerged Fermentation Using Response Surface Methodology

  • The objectives of this study were to investigate the effect of fermentation medium on the hydroxyl radical scavenging activity of exopolysaccharides from Inonotus obliquus by response surface methodology (RSM). A two-level fractional factorial design was used to evaluate the effect of different components of the medium. Corn flour, peptone, and $KH_2PO_4$ were important factors significantly affecting hydroxyl radical scavenging activity. These selected variables were subsequently optimized using path of steepest ascent (descent), a central composite design, and response surface analysis. The optimal medium composition was (% w/v): corn flour 5.30, peptone 0.32, $KH_2PO_4$ 0.26, $MgSO_4$ 0.02, and $CaCl_2$ 0.01. Under the optimal condition, the hydroxyl radical scavenging rate (49.4%) was much higher than that using either basal fermentation medium (10.2%) and single variable optimization of fermentation medium (35.5%). The main monosaccharides components of the RSM optimized polysaccharides are rhamnose, arabinose, xylose, mannose, glucose, and galactose with molar proportion at 1.45%, 3.63%, 2.17%, 15.94%, 50.00%, and 26.81%.
