Reaction Characteristics of Oxidation Catalysts for HCCI Engine (HCCI 엔진용 산화촉매의 반응특성)

  • The Homogeneous Charge Compression Ignition (HCCI) engine concept allows for both NOx and particulate matter to be reduced simultaneously, and it is a promising way to meet the next environmental challenges. Unfortunately, HCCI combustion often increases CO and HC emissions. The development of oxidation catalyst (OC) requires high conversion efficiency for CO and HC at low temperature. Conventional oxidation catalyst technologies may not be able to convert these emissions because of the saturation of active catalytic sites. The OC used in this study was 600 cpsi cordierite. Three kinds of OC with different amounts of Pt and Pd were used. The influence of the space velocity (SV), $H_2O$ and $O_2$ concentration was also studied. All types of OCs were found to have over 90% CO conversion efficiencies at $170^{\circ}C$. When in the presence of water vapor, CO conversion was increased, but $C_3H_8$ conversion was decreased. The performance of the OC was not influenced by initial the HC concentration. The 2Pt/Pd catalyst was better in terms of thermal aging than the Pt-only catalyst. The $LOT_{50}$ of both fresh and aged OC was increased with increasing SV and with the presence of $H_2O$.

Convergence Study on Natural Preservatives from Various Native Plant Species in Jiri Mountain Area (지리산 지역 자생식물 활용 천연보존제 융합 연구)

  • The purpose of the study aimed to explore the possibility for natural preservative convergence materials by examining the antioxidant activity and antimicrobial effect of 19 wild plants in Jiri Mountain. Total polyphenols were higher in the order of CS(Camellia sinensis L., 87.9 mg GAE/ g), MP(Mentha piperascens Holmes., 85.1 mg), NN(Nelumbo nucifera Gaertn., 65.0 mg) and PD(Pinus densiflora Siebold & Zucc., 52.8 mg). Total flavonoids were high in NN(25.7 mg QUE / g) and MP (25.4 mg QUE / g). CS(58.1%), NN(47.9%), and MP(40.6%) showed high ABTS radical scavenging ability and the result was similar in DPPH radical scavenging ability. The extracts of HC(Hemerocallis coreana Nakai.), PD, and CO(Cornus officinalis Siebold et Zucc.) showed the highest inhibitory effect on the growth of E. coli. The extracts of PK(Pulsatilla koreana Nakai ex Nakai.), SC(Saururus chinensis Baill.), and MC(Smilax china L.) completely inhibited the proliferation of S. aureus, showing the possibility to be developed as natural preservatives and disinfectants.

Oxidative Modification of Neurofilament-L Induced by Endogenous Neurotoxin, Salsolinol

  • The endogenous neurotoxin, 1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline (salsolinol), has been considered a potential causative factor for the pathogenesis of Parkinson's disease (PD). In this study, we examined oxidative modification of neurofilament-L (NF-L) induced by salsolinol. When disassembled NF-L was incubated with salsolinol, the aggregation of protein was increased with the concentration of sasolinol. The formation of carbonyl compound was obtained in salsolinol-mediated NF-L aggregates. This process was protected by free radical scavengers, such as N-acetyl-L-cysteine and glutathione. These results suggest that the aggregation of NF-L is mediated by salsolinol via the generation of free radicals. We also investigated the effects of copper ion on salsolinol-mediated NF-L modification. In the presence of copper ions, salsolinol enhanced the modification of NF-L. We suggest that salsolinol might be related to abnormal aggregation of NF-L which may be involved in the pathogenesis of neurodegenerative diseases and related disorders.

Hydrogen separation of $V_{99.8}B_{0.2}$ Alloy Membrane in Water-gas shift Reaction (수성 가스 전이반응에서 $V_{99.8}B_{0.2}$ 합금 분리막의 수소분리)

  • The influence of co-existing gases on the hydrogen permeation without sweep gas was studied through a Pd-coated $V_{99.8}B_{0.2}$ alloy membrane. Membranes have been investigated in the pressure range 1.5-8.0 bar under pure hydrogen, hydrogen-carbon dioxide and hydrogen-carbon monoxide gas mixture without sweep gas at $400^{\circ}C$. Preliminary hydrogen permeation experiments without sweep gas have been confirmed that hydrogen flux was $40.7mL/min/cm^2$ for a Pd-coated $V_{99.8}B_{0.2}$ alloy membrane (thick : 0.5 mm) using pure hydrogen as the feed gas. In addition, hydrogen flux was $21.4mL/min/cm^2$ for $V_{99.8}B_{0.2}$ alloy membrane using $H_2/CO_2$ as the feed gas. The hydrogen permeation flux decreased with decrease of hydrogen partial pressure irrespective of pressure when $H_2/CO_2$and $H_2/CO$mixture applied as feed gas respectively and permeation fluxes were satisfied with Sievert's law in different feed conditions. It was found from XRD, SEM/EDX results after permeation test that the Pd-coated $V_{99.8}B_{0.2}$ alloy membrane had good stability and durability for various mixtures feeding condition.

Neuroprotective Effect of l-Deprenyl Against 6-OHDA-Induced Dopamine Depletion in Rat Striatum and 6-OHDA-Induced Oxidative Stress in SH-SY5Y Cells (흰쥐 선조체에서 6-OHDA-유도 도파민 고갈 및 SH-SY5Y 세포주에서 6-OHDA-유도 산화적 스트레스에 대한 l-Deprenyl의 신경 보호효과)

  • A neurotoxin, 6-hydroxydopamine (6-OHDA) has long been used to form a Parkinson's disease (PD) model by inducing the lesion in catecholaminergic pathways, particularly the nigrostriatal dopamine (DA) pathway. Whereas l-deprenyl, a selective inhibitor of monoamine oxidase (MAO) type B, is now widely used in the treatment of PD, the precise action mechanism of the drug remains elusive. In this study, we investigated whether l-deprenyl shows protective effect against the DA depletion induced by 6-OHDA in rat brain, and against 6-OHDA-induced neurotoxicity and oxidative stress in catecholaminergic neuroblastoma SH-SY5Y cells that are known to lack MAO-B activity. Pretreatment of l-deprenyl significantly enhanced the striatal DA, 3,4-dihydroxyphenylacetic acid, homovanilic acid, and 3-methoxytyramine levels compared to the untreated 6-OHDA-lesioned rat, indicating that l-deprenyl pretreatment prevents 6-OHDA-induced depletion of not only striatal dopamine but also its metabolites. Treatment of 6-OHDA for 24hrs decreased the cell viability and increase the generation of ROS in dose-dependent manners. We further investigated whether caspase activity is involved in the action of l-deprenyl. Treatment of l-deprenyl $(0.1\~100{\mu}M)$ did not produce any changes in 6-OHDA-induced cleavage of poly (ADP-ridose) polymerase in SH-SY5Y cells. Our results suggest that the neuroprotective effect of l-deprenyl against 6-OHDA is due to its increased scavenger activity, but independent of inhibition of MAO-B or caspase-3 activation.

Effects of Adenophora triphylla Ethylacetate Extract on mRNA Levels of Antioxidant Enzymes in Human HepG2 Cells (인간 HepG2 Cell에서 항산화 효소의 mRNA 발현에 대한 잔대 에틸아세테이트 추출물 효과)

  • The root of Adenophora triphylla is widely used as traditional herbal medicine in Korea. We studied its effects on sodium nitroprusside (SNP) cytotoxicity and antioxidant genes expression in HepG2 cells. To study whether Adenophora triphylla ethylacetate extract (ATea) inhibited NO-induced cell death, HepG2 cells were preincubated for 24 hr with 50 and 100 $\mu$g/mL ATea followed by 24-hr exposure to 0.5 mM SNP (exogenous NO donor). No-induced cytotoxicity was inhibited by pretreatment of ATea, as assessed by mitochondrial dehydrogenase activity (MTT assay). We further investigated the effects of ATea on mRNA levels of various enzymes of the antioxidant system such as Cu, Zn superoxide dismutase (SOD 1), Mn SOD (SOD 2), glutathione peroxidase (GPx), catalase and several enzymes of the glutathione metabolism [glutathione reductase (GR), $\gamma$-glutamyl-cystein synthetase (GCS), glutathione-S-transferase (GST), $\gamma$-glutamyltranspeptidase ($\gamma$-GT), glucose-6-phosphate dehydrogenase (G6PD)] by RT-PCR. CAT, GCS, GR and G6PD mRNA levels were increased after treatment with ATea. The SOD 1, SOD 2, GPx, GST and $\gamma$-GT mRNA levels were not affected in ATea-treated HepG2 cells. We concluded that ATea have an indirect antioxidant effects, perhaps via induction of CAT, GCS, GR and G6PD.

Simultaneous Separation Analysis of Some Metal Ions in Piperidinedothiocarbamate Chelates by Reversed-Phase Liquid Chromatography (역상 액체 크로마토그래피에 의한 몇가지 금속 이온들의 Piperidinedithiocarbamate 킬레이트 동시분리분석)

  • Simultaneous separation and analysis of Ni(II), Pd(II), Co(II), Cu(II) and Hg(II) in peperidinedithiocarbamate (PDTC) chelates were investigated by reversed phase liquid chromatography. The optimum conditions for the separation of PDTC metal chelates were examined with respect to the pH, extraction solvent, and mobile phase strength on Novapak $C_{18}$ column using methanol/water mixture as mobile phase. All metal PDTC chelates were eluted in an acceptable range of capacity factor value ($0{\leq}log\;k^{\prime}{\leq}1$). The linear calibration curves were obtained in the concentration range of $0{\sim}1.2{\mu}g/mL$ for five metal ions, and also good precision in the range of 1.96~3.41% RSD was obseved. Under the optimum conditions, trace metat ions in the composite water sample were successfully separated and determined with relative error of ${\pm}2.0%$.

A Study on the Characteristics of MMPI Profile and Personality in Patients with Hwa-byung (화병환자의 MMPI 프로파일과 성격특성 연구)

  • Objectives : This study was to analyze characteristics of MMPI profile, personality and defensive mechanism on 32 Hwa-byeng patients. Methods : 32 Hwa-byeng patients and 28 normal control group were administered the MMPI. The characteristics of MMPI profile and the clinical scales of personality and defensive mechanism were analyzed statistically. Results : 1. Half of female Hwa-byeng patients are in their forties, twelve patients are high school graduates, housewives seem to have Hwa-byeng more than workers, and married ones have Hwa-byeng more than singles, divorcees or separated one 2. 60.6% patients are getting stressed from their husbands(the specific reasons are infidelity(55%), incapability(25%) and violence(20%)). And the rest are from in-law(28.8%), children(6.4%) and their own parents(3.2%). 3. MMPI profile of Hwa-byeng patients appeared as a type of 3-1 elevated by the scale of 3(Hy) and 1 (Hs). And Hwa-byeng patients appeared significantly high in the rest of scales except L, K, Mf and Ma scales comparing to control group. 4. Hwa-byeng patients were classified into 3 groups. First group is "V" type which is high in K scale and low in 3 (Hy), 4(Pd), 6(Pa). Second group is "V" type which is high in K scale and high in 3(Hy), 4(Pd), 6(Pa). Third group is high in F scale and high in 1(Hs), Z(D), 3(Hy), 4(Pd), 6(Pa), 7(Pt), 8(Sc) and 0(Si). 5. The analysis on MMPI profiles indicates that the thirties are highier in F scale than the fifties, the fifties are highier in Hy scale than the forties, workers are highier in Pd scale than housewives, singles or divorcees are highier in Pa scale than the married. 6. The characteristics of personality in Hwa-byeng patients are as follows. They are significantly lower in HST, NAR and very higher in CPS, PAG, DEP, STY, AVD, SZD comparing to control group. Especially in AVD and SZD scales are appeared the highest.

5-Hydroxytryptophan Reduces Levodopa-Induced Dyskinesia via Regulating AKT/mTOR/S6K and CREB/ΔFosB Signals in a Mouse Model of Parkinson's Disease

  • Long-term administration of levodopa (L-DOPA) to patients with Parkinson's disease (PD) commonly results in involuntary dyskinetic movements, as is known for L-DOPA-induced dyskinesia (LID). 5-Hydroxytryptophan (5-HTP) has recently been shown to alleviate LID; however, no biochemical alterations to aberrant excitatory conditions have been revealed yet. In the present study, we aimed to confirm its anti-dyskinetic effect and to discover the unknown molecular mechanisms of action of 5-HTP in LID. We made an LID-induced mouse model through chronic L-DOPA treatment to 6-hydroxydopamine-induced hemi-parkinsonian mice and then administered 5-HTP 60 mg/kg for 15 days orally to LID-induced mice. In addition, we performed behavioral tests and analyzed the histological alterations in the lesioned part of the striatum (ST). Our results showed that 5-HTP significantly suppressed all types of dyskinetic movements (axial, limb, orolingual and locomotive) and its effects were similar to those of amantadine, the only approved drug by Food and Drug Administration. Moreover, 5-HTP did not affect the efficacy of L-DOPA on PD motor manifestations. From a molecular perspective, 5-HTP treatment significantly decreased phosphorylated CREB and ΔFosB expression, commonly known as downstream factors, increased in LID conditions. Furthermore, we found that the effects of 5-HTP were not mediated by dopamine1 receptor (D1)/DARPP32/ERK signaling, but regulated by AKT/mTOR/S6K signaling, which showed different mechanisms with amantadine in the denervated ST. Taken together, 5-HTP alleviates LID by regulating the hyperactivated striatal AKT/mTOR/S6K and CREB/ΔFosB signaling.

Evaluation of Nivolumab Use and Factors related to Treatment Outcomes in a Cancer Center of a Top Tier General Hospital (상급종합병원 암센터에서 Nivolumab 사용평가와 치료성과에 미치는 영향인자)

  • Background: We strived to evaluate the status of nivolumab use and associated factors on the clinical efficacy of the drug. Methods: The study was retrospectively conducted in patients who had been administered nivolumab at least once at the cancer center of Seoul National University Hospital from June 2015 to April 2017. Data were collected from electronic medical records. A medication-use evaluation was performed based on the American Society of Health-System Pharmacists mediation-use guidelines. Results: Sixty-six of the 74 patients (89.2%) showed indications approved for nivolumab use by the Korean Ministry of Food and Drug Safety (MFDS; n=55) or the US Food and Drug Administration (FDA; n=11). Approximately 73.0% of the patients were administered the approved dose of 3 mg/kg but 25.7% were administered an unapproved fixed dose of 100 mg. The overall response rate was 21.7%, and the response rate of non-small cell lung cancer patients, who accounted for the largest number of indications, was 18.8%. Adverse reactions were found in 90.1% of the patients and were mostly mild (86%). The expression of programmed death-ligand 1 (PD-L1) was analyzed as a factor affecting treatment response (p=0.028, odds ratio [OR]=11.331). Conclusion: PD-L1 expression was found to affect treatment response. However, caution is required while using an unapproved dosage and in the absence of monitoring for effectiveness and safety. Therefore, an effective protocol or instruction manual for the proper use of nivolumab should be considered.
