Comparative susceptibility of different cell lines for culture of Toxoplasma gondii in vitro (톡소플라스마 곤디의 세포내 배양에 있어서 세포 주에 따른 감수성 비교)

  • In order to establish a useful cell culture system for T gondii we compared the degree of proliferation of T gondii tachyzoites among 8 different cell lines: 2 kinds of normal animal cells (MDCK-canine kidney cells; Vero-monkey kidney cells) and 6 kinds of human tumor cells (A 549, PC 14-lung cancer cells; SNU 1, SNU 16. Mlm 45-stomach cancer cells; HL-60-promyelocytic leukemia cells), through morphological observation and 3H-uracil uptake assay. The degree of susceptibility to infection with T gondii tachyzoites was highest in A 549 and PC 14 cells, medium in Vero, HL-60, MDCK and SNU 1, and lowest in SNU 16 and MBm 45 cells. The kinetics of T gondii multiplication during the post-Infection 60 hours were higllly dependent upon the dose of tachyzoites administered and the duration among the 8 tested fur the growth and multiplication of T gondii in vitro.

Protective Effect of Prunella spica Extracts against H2O2-Induced Cytotoxicity in PC12 Cells (Hydrogen peroxide가 유도하는 세포독성으로부터 PC12 세포를 보호하는 하고초(Prunella spica) 추출물의 영향)

  • The oxidative stress induced by reactive oxygen species (ROS) may play an important role in the pathogenesis of neurodegenerative diseases. In this study, we investigated the neuroprotective effects of methanolic extracts of Prunella Spica (PSE) against $H_2O_2$-induced oxidative stress in PC12 cells. The cells exposed to $H_2O_2$-induced oxidative stress were treated with various concentrations of PSE; this treatment resulted in the induction of a dose-dependent protective effect, which was evidenced by the results of MTT reduction assay, lactate dehydrogenase (LDH) release assay, morphological assay, and colony-formation assay. Interestingly, we also observed reduction of apoptotic bodies in the Hoechst staining and flow cytometric analysis. These data show that apoptosis was significantly suppressed in the PC12 cells that were exposed to $H_2O_2$-induced oxidative stress and treated with PSE. These results suggest that Prunella Spica could be a new potential protective agent against $H_2O_2$-induced oxidative stress.

Study of Apoptotic Effect on Hydrogen Peroxide-induced Rat PC-12 Cells by Aster tataricus Water Extract (자완 수추출물(水抽出物)이 $H_2O_2$에 의해 유도(誘導)된 PC-12 세포주(細胞株)의 세포사(細胞死)에 미치는 영향(影響))

  • Objective : Alzheimer's disease (AD) is a geriatric dementia that is widespread in old age. With an aging populace, AD is a looming problem in public health service. Alzheimer's disease is characterized by specific neuronal degeneration in certain areas of the brain. Mutations and abnormal expression of several genes are associated with ${\beta}-amyloid$ deposits and Alzheimer's disease; among them APP, PS1, and PS2, SOD, free radical, ROS. Methods:We studied herbal medicines that have a relationship to brain degeneration. From pre-modern times, although a variety of oriental prescriptions of Aster tataricus have been traditionally utilized for the treatment of AD, their pharmacological effects and action mechanisms have not yet been fully elucidated. Result : Based on morphological observations by phase-contrast microscope, TUNEL assay and MTT in the culture media, $H_20_2-induced$ cell death was significantly inhibited by Aticus. We examined by ROS formation, catalase activity and GSH activity. We studied the protective effect and inhibitory effects of neurotoxicity in $H_20_2-induced$ PC-12 cells by Aticus. Findings from our experiments show that Aticus inhibits apoptosis, which has neurotoxicities and cell damage in PC-12 cells. In addition, treatment with Aticus ($>25{\mu}g/ml$ for 6hrs) partially prevented $H_20_2-induced$ cytotoxicity in PC-12 cells, and induced a protective effect. Conclusion : As the result of this study, in the Aticus group, the apoptosis in the nervous system was inhibited, protected against the degeneration of PC-12 cells by $H_20_2$. Taken together, Aticus exhibited inhibition of $H_20_2-induced$ apoptotic cell death. Aticus was found to induce protective effect on GSH and catalase in PC-12 cells. Based on these findings, Aticus may be beneficial for the treatment of AD.

Phenolic Compounds from Antioxidant Plant Materials and their Protective Effect on PC12 cells (항산화 식물의 페놀성화합물에 의한 PC12 세포보호 효과연구)

  • This study was designed to evaluate the antioxidant activities and protective effects on PC12 cells of the extract of Epimedium koreanum and its main constituents icariin and icariside I. After screening the seven identified flavonoid glycosides from E. koreanum through DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) assay, E. koreanum, Icariin and Icariside I exhibited significant effect on radical scavenging activity. E. koreanum, icariin and icariside I were examined using DPPH, ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) and FRAP (ferric reducing ability power) assay. In all antioxidant assays, E. koreanum, icariin and icariside I showed high radical scavenging activities in a dose-dependent manner. Protective effects against $H_2O_2-induced$ PC12 cells were assessed with MTT assay. The results indicated that cell viability and protection on PC12 cells of icariside I and icariin increased dose dependently. These study results suggest that E. koreanum, icariin and icariside showed high antioxidant capacities and cell protective effects. Icariside I, one of the metabolites of icariin, may be a new and effective flavonoid compound as a functional component.

Antioxidant Effects of Gagam-jangwon-hwan(jiajianzhuangyuanwan) on Hydrogen Peroxide-Induced Oxidative Stress in PC 12 Cell Lines ($H_2O_2$로 유도된 산화적 스트레스에 대한 장원환가감방(壯元丸加減方)의 PC 12 cell 에서의 항산화 효과)

  • Objectives : Antioxidant effects of Gagam-jangwonhwan(LMK01 and 02) water extract against $H_2O_2$-induced oxidative damage and cell death were investigated in rat pheochromocytoma line PC 12. Methods : The cells were treated with LMK01 and 02 water extract and $H_2O_2$, oxidative damage-inducing materials for 24 h. The cellular viability was assessed by WST-1 assay, oxidative damages of the cells by 8-OHdG quantitation, apoptosis by Hoechst 33342 staining assay and activity of antioxidant enzymes by catalase and glutathione peroxidase assay. Results : 1. LMK01 and LMK02 water extracts improved significantly cell viability in $H_2O_2$-treated groups than $H_2O_2$-alone treated cells 2, LMK02 suppressed significantly oxidative damage in $H_2O_2$-treated groups than $H_2O_2$-alone treated cells but LMK01 didn't. Meanwhile, difference of oxidative damages in conditions treated with LMK01 or LMK02 was not significant, 3. The $H_2O_2$ induced-apoptosis in PC 12 cell lines was inhibited effectively by LMK01 and LMK02, and especially the features of apoptosis were obviously reduced in LMK02-treated cells. 4. LMK01 and LMK02 increased significantly activities of both catalase and glutathione peroxidase than those of $H_2O_2$-alone treated group and moreover, LMK02 showed significantly higher activities than those of LMK01. Conclusions : As shown, LMK01 and LMK02 suppressed $H_2O_2$-induced oxidative damage and cell death in PC 12 cell effectively. And they increased activity of major antioxidant enzymes in PC 12 cell line. Therefore, this study suggests the possibility of clinical usage over oxidative stress-induced neurodegenerative disease such as Alzheimer's disease.

  • The effects of liriodenine, an aporphine isoquinoline alkaloid, on dopamine biosynthesis and L-DOPA-induced neurotoxicity in PC12 cells were investigated. Treatment of PC12 cells with liriodenine at 10 $\mu\textrm{M}$ showed 33.6% inhibition of dopamine content decreased at 3 h and reached a minimal level at 12 h after the exposure to liriodenine at 10 $\mu\textrm{M}$. Neuroprotective Effect according to Reactive Oxygen Species Scavenging Activity from Extracts of Cudrania tricuspidata Leaves (활성산소 소거활성에 따른 꾸지뽕잎 추출물의 신경세포 보호 효과)

  • In an attempt to identify the neuroprotective effect of Cudrania tricuspidata (CT) leaves against ROS (reactive oxygen species)-induced oxidative stress in neuronal cells, the extracts from CT leaves were investigated using PC12 cells and N18-RE-105 cells. The methanolic and ethanolic extracts from CT were denoted as CTM (Cudrania tricuspidata Leaves methanolic extracts) and CTE (Cudrania tricuspidata Leaves ethanolic extracts), respectively. The neuroprotective effects of the extracts were measured by DCF-DA assay, MTT reduction assay, and LDH release assay. The PC12 cells exposed to $H_2O_2$-induced oxidative stress and the N18-RE-105 cells exposed to glutamate-induced oxidative stress were treated with various concentrations of CTM and CTE. The results, CTM treatments resulted in the induction of a dose-dependent protective effect in PC12 cells and N18-RE-105 cells. Interestingly, CTE also showed neuroprotective effect in PC12 cells and N18-RE-105 cells. Therefore, these results suggest that CTM and CTE could be a new potential candidate as neuroprotective agents against ROS-induced oxidative stress in neuronal cells.

Rhus verniciflua Stokes Extract and Its Flavonoids Protect PC-12 Cells against H2O2-Induced Cytotoxicity

  • Rhus verniciflua Stokes (RVS), an herbal medicine found in East Asia, was extracted and further fractionated to investigate its antioxidant capacity and neuroprotective effects. The RVS ethyl acetate (EtOAc) fraction had the highest level of total phenolics and antioxidant capacity among all solvent fractions tested. Pretreatment of PC-12 cells with the EtOAc fraction effectively attenuated $H_2O_2$-induced oxidative damage. Furthermore, the EtOAc fraction significantly attenuated caspase-3 activity, resulting in inhibition of $H_2O_2$-induced apoptosis. We identified and quantified fustin, sulfuretin, and butein in the EtOAc fraction using accurate mass quadrupole time-of-flight mass spectrometry and reversed-phase high-performance liquid chromatography. The intracellular antioxidant capacity and superoxide dismutase (SOD) activity were significantly increased in PC-12 cells treated with the EtOAc fraction and with individual flavonoids. When cells were pretreated with the EtOAc fraction or individual flavonoids and then co-incubated with diethyldithiocarbamic acid (an inhibitor of SOD activity), cell viability against $H_2O_2$-induced oxidative stress was attenuated. These results suggest that the RVS EtOAc fraction and its flavonoid constituents protect PC-12 cells against $H_2O_2$-induced neurotoxicity through their antioxidant properties.

Protective Effect of Sophorae Subprostratae Radix and Each Fractions on PC12 cell Damage Induced by Hypoxia/Reperfusion (PC12 세포의 허혈모델에 있어 광두근 분획물의 항산화효과연구)

  • This research was performed to investigate protective effect of Sophorae subprostratae Radix and each fractions against ischemic damage using PC12 cells. To observe the protective effect of Sophorae subprostratae Radix on ischemia damage, vibility and changes in activities of Superoxide dismutase (SOD), Glutathione Peroxidase (GPx), Catalase and Production of Malondialdehyde (MDA) were observed after treating PC12 cells with Sophorae subprostratae Radix during ischemic insult. Groups were divided into five groups: no treated (Normal), hypoxia chamber for 48hrs followed by 6h at normoxic chamber (H/R), Sop horae subprostratae Radix total phase treated group with H/R (Total), Sophorae subprostratae Radix water phase treated group with H/R (Water), Sophorae subprostratae Radix BuOH phase treated group with H/R (BuOH), Sophorae subprostratae Radix alkaloid phase treated group with H/R (Alkaloid). The results showed that (1) in hypoxiajreperfusion model using PC12 cell, the Sophorae subprostratae Radix has the protective effect against ischemia in the dose of 0.2 ㎍/㎖, 2 ㎍/㎖ and 20 ㎍/㎖, (2) Sophorae subprostratae Radix increased the activities of glutathione peroxidase and catalase. (3) the activity of Superoxide Diamutase(SOD) increased by ischemic damage, which might represent the self protection. This study suggests that Sophorae subprostratae Radix has neuroprotective effect against neuronal damage following hypoxiajreperfusion cell culture model using PC12 cell and dose dependency effects. In conclusion, Sophorae subprostratae Radix has protective effects against ischemic oxidative damage at the early stage of ischemia.

Antioxidative and Protective Effects of Ulmus davidiana var. japonica Extracts on Glutamate-Induced Cytotoxicity in PC 12 Cells (느릅나무 추출물의 항산화 효과 및 L-glutamate 유래 PC12 세포독성 보호효과)

  • Antioxidative and protective effects of Ulmus davidiana var. japonica against oxidative damages induced by glutamate in PC 12 cells were investigated. Inhibitory activity against $FeSO_{4}-H_{2}O_{2}$-induced oxidative stress and DPPH radical-scavenging activity were detected in ethyl acetate and butanol fractions of ethanol extracts from stems and roots. Ethyl acetate and butanol fractions of ethanol extracts from roots significantly inhibited glutamate-induced cytotoxicity and reactive oxygen species in PC 12 cells. These results demonstrate ethyl acetate and butanol fractions of ethanol extracts of U. davidiana var. japonica have potent protective effect against glutamate-induced oxidative stress.
