Effects of Heat Stress and Dietary Tryptophan on Performance and Plasma Amino Acid Concentrations of Broiler Chickens

  • Two experiments were conducted to investigate the impact of high temperature and dietary tryptophan (Trp) on performance, selected organ weights and plasma free amino acid (AA) concentrations in broiler chickens. In Experiment 1, exposure to $27-33^{\circ}C$ of chickens for 2 weeks from 2 weeks of age did not affect growth and plasma free AA concentration except for a decrease in the concentration of plasma tyrosine (Tyr). In Experiment 2, 2-week-old birds were allocated to one of three temperature treatments; $24^{\circ}C$ (control), $36^{\circ}C$ (heat stress, HS) and $24^{\circ}C$ pair-fed (24PF) for 2 weeks and fed on diets containing 50, 100 and 300% of NRC requirement for Trp. Heat stress caused a reduction of weight gain and feed intake irrespective of dietary Trp levels compared with control counterparts, while feeding of 300% Trp diet did not attenuate the reduced performance by HS exposure. In groups fed the 100% Trp diets, plasma aromatic AA (AAA) and Tyr concentrations were decreased in the HS birds compared with the 24PF group. Plasma concentrations in most of AA groups were increased by HS in chickens fed the 50% Trp diet, while those were not changed by HS in chickens fed the 300% Trp diet, compared with 24PF counterparts. The plasma Trp/LNAA (LNAA=large neutral AAs, which are comprised of BCAA, AAA and Trp) ratio was increased by HS in chickens fed the 100% Trp diet, while it was decreased in chickens fed on 50% Trp diet as compared with 24PF group. From these results, it is suggested that performance and plasma amino acid profile deranged by heat stress are modulated, at least, to be relieved from the heat stress by feeding 50% Trp diet but not at all by feeding 300% Trp diet. The involvement of altered plasma AA profiles, in particular plasma Tyr concentrations and Trp/LNAA ratio, is discussed in association with the performance characteristics of HS chickens.

Whole-genome association and genome partitioning revealed variants and explained heritability for total number of teats in a Yorkshire pig population

  • Objective: The study was designed to perform a genome-wide association (GWA) and partitioning of genome using Illumina's PorcineSNP60 Beadchip in order to identify variants and determine the explained heritability for the total number of teats in Yorkshire pig. Methods: After screening with the following criteria: minor allele frequency, $MAF{\leq}0.01$; Hardy-Weinberg equilibrium, $HWE{\leq}0.000001$, a pair-wise genomic relationship matrix was produced using 42,953 single nucleotide polymorphisms (SNPs). A genome-wide mixed linear model-based association analysis (MLMA) was conducted. And for estimating the explained heritability with genome- or chromosome-wide SNPs the genetic relatedness estimation through maximum likelihood approach was used in our study. Results: The MLMA analysis and false discovery rate p-values identified three significant SNPs on two different chromosomes (rs81476910 and rs81405825 on SSC8; rs81332615 on SSC13) for total number of teats. Besides, we estimated that 30% of variance could be explained by all of the common SNPs on the autosomal chromosomes for the trait. The maximum amount of heritability obtained by partitioning the genome were $0.22{\pm}0.05$, $0.16{\pm}0.05$, $0.10{\pm}0.03$ and $0.08{\pm}0.03$ on SSC7, SSC13, SSC1, and SSC8, respectively. Of them, SSC7 explained the amount of estimated heritability along with a SNP (rs80805264) identified by genome-wide association studies at the empirical p value significance level of 2.35E-05 in our study. Interestingly, rs80805264 was found in a nearby quantitative trait loci (QTL) on SSC7 for the teat number trait as identified in a recent study. Moreover, all other significant SNPs were found within and/or close to some QTLs related to ovary weight, total number of born alive and age at puberty in pigs. Conclusion: The SNPs we identified unquestionably represent some of the important QTL regions as well as genes of interest in the genome for various physiological functions responsible for reproduction in pigs.

Sex-Related Differences of EEG Coherences between Patients with Schizophrenia and Healthy Controls (조현병 환자와 건강한 대조군에서 성별에 따른 뇌파 동시성의 차이)

  • Objectives Alteration of epigenetic effects of testosterone during early development was suggested as an ancillary mechanism for the genesis of schizophrenia. EEG coherence was thought to be a marker for cerebral laterality of which important determinant was testosterone during early development. We studied sex-related differences of EEG coherences between patients with schizophrenia and controls to examine the sex effects in the genesis of schizophrenia. Methods EEG was recorded in 35 patients with schizophrenia and 46 healthy controls in the eyes closed resting state. Pair-wise EEG coherences were calculated over delta, theta, alpha, beta and gamma frequency bands. To examine the differences of EEG coherence according to sex in each group, ANCOVA was performed using Statistical Analysis system (SAS, Ver 9.3) and R (Ver 2.15.2). Results Healthy control males showed more increased right intrahemispheric coherences than healthy control females in delta, theta, alpha and beta frequency bands. In patients with schizophrenia, this male dominant pattern in right intrahemispheric coherences was attenuated especially in alpha and beta bands. Healthy control females showed more increased interhemispheric coherences than healthy control males in delta, theta, beta and gamma frequency bands. In patients with schizophrenia, these female dominant patterns in interhemispheric coherences were attenuated especially in delta, theta, and beta bands, which were commonly observed in frontal to central areas. Conclusion Sex differences in resting EEG coherences were attenuated in schizophrenia patients. These results imply that sex-related aberrant cerebral lateralization might exist in patients with schizophrenia, which are partly due to sex hormones via epigenetic mechanisms.

Structure and Magnetic Properties of Ho and Ni Co-doped BiFeO3 Ceramics

  • Recently, multiferroic materials gain much attention due to their fascinating fundamental physical properties. These materials offer wide range of potential applications such as data storage, spintronic devices and sensors, where both electronic and magnetic polarizations can be coupled. Among single-phase multiferroic materials, $BiFeO_3$ is typical because of the room-temperature magnetoelectric coupling in view of long-range magnetic- and ferroelectric-ordering temperatures. However, $BiFeO_3$ is well known to have large leakage current and small spontaneous polarization due to the existence of oxygen vacancies and other defects. Furthermore the magnetic moment of pure $BiFeO_3$ is very weak owing to its antiferromagnetic nature. Recently, various attempts have been performed to improve the multiferroic properties of $BiFeO_3$ through the co-doping at the A and the B sites, by making use of the fact that the intrinsic polarization and magnetization are associated with the lone pair of $Bi^{3+}$ ions at the A sites and the partially-filled 3d orbitals of $Fe^{3+}$ ions at the B sites, respectively. In this study, $BiFeO_3$, $Bi_{0.9}Ho_{0.1}FeO_3$, $BiFe_{0.97}Ni_{0.03}O_3$ and $Bi_{0.9}Ho_{0.1}Fe_{0.97}Ni_{0.03}O_3$ bulk compounds were prepared by solid-state reaction and rapid sintering. High-purity $Bi_2O_3$, $Ho_2O_3$, $Fe_2O_3$ and $NiO_2$ powders with the stoichiometric proportions were mixed, and calcined at $500^{\circ}C$ for 24 h to produce the samples. The samples were immediately put into an oven, which was heated up to $800^{\circ}C$ and sintered in air for 1 h. The crystalline structure of samples was investigated at room temperature by using a Rigaku Miniflex powder diffractometer. The field-dependent and temperature-dependent magnetization measurements were performed with a vibrating-sample magnetometer and superconducting quantum-interference device.

Development of PCR Primers for Specific Identification and Detection of Botrytis cinerea on Tomato (잿빛곰팡이병균(Botrytis cinerea)의 종 동정과 PCR 검출을 위한 종 특이적 Primer의 개발)

  • Botrytis cinerea, gray mold pathogen, causes serious losses in greenhouse tomato crop. In this study, a primer set was developed for identification and specific PCR detection of B. cinerea from tomato plants. The primer pair (BTF1/BTR1) was designed from polymorphic sequence region in pyruvate carboxylase gene (pyc) of B. cinerea. A PCR product (112 bp) was amplified on genomic DNA of 13 B. cinerea isolates from 10 different host plants, but not on those from 6 other Botrytis spp., 4 Botryotinia spp., 5 Sclerotinia spp. and 16 other genus of phytopathogenic fungi. The sensitivity limit of the primer set was 2 pg of genomic DNA of B. cinerea, approximately. The PCR assay using species-specific primer set was specifically able to detect the pathogen on naturally infected tomato plants and artificially infected plants. These results suggest that the sensitivity and specificity of this primer set can be applied in a rapid and accurate diagnosis of tomato disease caused by B. cinerea.

A Molecular Dynamics Simulation Study of Hydroxyls in Dioctahedral Phyllosilicates (분자동역학 시뮬레이션을 이용한 이팔면체 점토광물 수산기 연구)

  • Clay minerals are a major player to determine geochemical cycles of trace metals and carbon in the critical zone which covers the atmosphere down to groundwater aquifers. Molecular dynamics (MD) simulations can examine the Earth materials at an atomic level and, therefore, provide detailed fundamental-level insights related to physicochemical properties of clay minerals. In the current study, we have applied classical MD simulations with clayFF force field to dioctahedral clay minerals (i.e., gibbsite, kaolinite, and pyrophyllite) to analyze and compare structural parameters (lattice parameter, atomic pair distance) with experiments. We further calculated vibrational power spectra for the hydroxyls of the minerals by using the MD simulations results. The MD simulations predicted lattice parameters and interatomic distances respectively deviated less than 0.1~3.7% and 5% from the experimental results. The stretching vibrational wavenumber of the hydroxyl groups were calculated $200-300cm^{-1}$ higher than experiment. However, the trends in the frequencies among different surface hydroxyl groups of each mineral was consistent with experimental results. The angle formed by the surface hydroxyl group with the (001) plane and hydrogen bond distances of the surface hydroxyls were consistent with experimental result trends. The inner hydroxyls, however, showed results somewhat deviated from reported data in the literature. These results indicate that molecular dynamics simulations with clayFF can be a useful method in elucidating the roles of surface hydroxyl groups in the adsorption of metal ions to clay minerals.

Whole Genome Sequence of a Korean Isolate (strain 51) of Helicobacter pylori

  • Substantial genomic diversity has been expected among clinical isolates of H. pylori. We have suggested that the two complete H. pylori genomes already sequenced may be insufficient for providing a discriminatory tool for typing clinical isolates as well as an insight into the genomic diversity, which enable to establish strategy for control of H. pylori infection. In this study, we determine the nucleotide sequence of the entire genome of Korean strain 51 and compare it with two reported genomic sequences to suggest validity for extensive genomic sequencing of H. pylori. The genome of H. pylori 51 consists of a circular chromosome with a size of 1,591,297 bp, which is corresponding to $95.4\%\;and\;96.8\%$ of the 26695 and J99 chromosome length, respectively. We predict that there are 1,454 open reading frames (ORFs) in 51, representing $91.4\%\;and\;97.2\%$ of the reported numbers of ORF of 26695 and J99, respectively. In contrast to 26695 and J99 that have 123 and 65 strain-specific genes, respectively, of the 1,454 genes, only 39 genes are unique to 51. Differences in genomic organization between 51 and each foreign strain were greater than between 2 foreign strains in pair wise entire sequence alignments by BLASTN. Particularly, the extent of genomic rearrangement observed between 51 and 26695 is higher than between 51 and J99. Multiple sequence alignment of orthologous genes among 3 strains showed that 51 is genetically closer to 26695 rather than J99. Phylogenetic analysis of nonsynonymous and synonymous mutation indicated J99 has the longest branch length in the unrooted phylogenetic tree, suggesting that J99 has higher mutation rate than the other 2 strains.

  • Recently the web service area is rapidly growing as the next generation IT paradigm because of increase of concern about SOA(Services-Oriented Architecture) and growth of B2B market. Since a service discovery through UDDI(Universal Description, Discovery and Integration) is limited to a functional requirement, it is not considered an effect on frequency of service using and reliability of mutual relation. That is, a quality as nonfunctional aspect of web service is regarded as important factor for a success between consumer and provider. Therefore, the web service selection method with considering the quality is necessary. This paper suggests the agent-based quality broker architecture and selection process which helps to find a service providing the optimum quality that the consumer needs in a position of service consumer. A theory of agent is accepted widely and suitable for proposed system architecture in the circumstance of distributed and heterogeneous environment like web service. In this paper, we considered the QoS and CoS in the evaluation process to solve the problem of existing researches related to the web service selection and used PROMETHEE(Preference Ranking Organization MeTHod for Enrichment Evaluations) as an evaluation method which is most suitable for the web service selection among MCDM approaches. PROMETHEE has advantages that solve the problem that a pair-wise comparison should be performed again when comparative services are added or deleted. This paper suggested a case study with the service composition scenario in order to verify the selection process. In this case study, the decision making problem was described on the basis of evaluated values for qualities from a consumer's point of view and the defined service level.

  • Because of the shortage of construction workers due to The rising labor costs and an aging labor force, construction time has been extended. As a solution, The construction time of high-rise buildings can be reduced by adopting precast concrete construction methods. Most relevant studies have focused on the development and structural analysis of such methods and not on their construction management. Therefore, this study focused on the construction management of the hollow precast concrete column (HPC) method. The objective of this study was to evaluate the performance of HPC formulations through the analytic hierarchy process and benefit-cost analysis. After a gap analysis of the available literature and expert interviews, the evaluation criteria were selected. A questionnaire survey was administered to professionals with ample experience in precast concrete construction for the pair-wise evaluation of the benefit and costs of the HPC method. The results show that the benefits of the HPC method outweighed its costs. Therefore, the HPC method is a suitable substitute for the half-slab method.

Design of Sensor Network for Estimation of the Shape of Flexible Endoscope (연성 대장내시경의 형상추정을 위한 센서네트워크의 설계)

  • In this paper, a method of shape prediction of an endoscope handling robot that can imitate a surgeon's behavior using a sensor network is suggested. Unit sensors, which are composed of a 3-axis magnetometer and 3-axis accelerometer pair comprise the network through CAN bus communication. Each unit of the sensor is used to detect the angle of the points in the longitudinal direction of the robot, which is made from a flexible tube. The signals received from the sensor network were filtered using a low pass Butterworth filter. Here, a Butterworth filter was designed for noise removal. Finally, the Euler angles were extracted from the signals, in which the noise was filtered by the low path Butterworth filter. Using this Euler angle, the position of each sensor on the sensor network is estimated. The robot body was assumed to consist of links and joints. The position of each sensor can be assumed to be attached to the center of each link. The position of each link was determined using the Euler angle and kinematics equation. The interpolation was carried out between the positions of the sensors to be able to connect each point smoothly and obtain the final posture of the endoscope in operation. The experimental results showed that the shape of the colonoscope can be visualized using the Euler angles evaluated from the sensor network suggested and the shape of serial link estimated from the kinematics chain model.
