Prevalence of Cryptosporidium-Associated Diarrhea in a High Altitude-Community of Saudi Arabia Detected by Conventional and Molecular Methods

  • Cryptosporidium diarrhea represents a relevant clinical problem in developing countries. In Al-Taif, a city of Saudi Arabia that lies at an altitude of an around 2 km above the sea level, Cryptosporidium infection seems to be undiagnosed in nearly all clinical laboratories. Furthermore, nothing was published regarding Cryptosporidium-associated diarrhea in this area. The objectives of this research were to (1) determine the Cryptosporidium prevalence among patients with diarrhea and (2) to estimate the performances of 3 different diagnostic methods. Total 180 diarrheal fecal samples, 1 sample per patient, were collected between January and August 2013. Samples were screened for Cryptosporidium with modified Zeihl Neelsen (ZN) microscopy, $RIDA^{(R)}$ Quick lateral flow (LF) immunotest, and a previously published PCR. The Cryptosporidium prevalence rate was 9.4% (17/180), 10% (18/180), and 11.6% (21/180) by microscopy, LF, and PCR test, respectively. Infection was significantly (P=0.004) predominant among children <5 years (22%) followed by children 5-9 years (11.1%). Although infection was higher in males than in females (16.2% males and 8.5% females), the difference was not statistically significant (P=0.11). Compared to PCR, the sensitivity of microscopy and the LF test were 80.9%, 85.7%, respectively. To conclude, high Cryptosporidium-associated diarrhea was found in this area especially in children ${\leq}9$ years. The PCR test showed the best performance followed by the LF test and ZN staining microscopy. The primary health care providers in Al-Taif need to be aware of and do testing for this protozoon, particularly for children seen with diarrhea.

Haematological and Immunological Response in Lambs Fed on Raw and Variously Processed Cottonseed Meal

  • An experiment was conducted with twenty crossbred male lambs to assess the effect of cotton (Gossypium) seed meal (CSM) on blood constituents and immunity. Lambs were randomly assigned to a reference diet (30% deoiled peanut meal, DPNM) and four test diets containing 40% of either raw, 45 minutes cooked, 1% $Ca(OH)_2$ and iron (1 free gossy-pol, FG : 0.3 Fe) treated CSM (replacing approximately 50%, reference concentrate mixture). These isonitrogenous and isocaloric concentrate mixtures were fed to meet 80% of protein requirements (NRC, 1985) along with ad lib maize hay for 180 days. Blood was collected at 60, 120 and 180 days post feeding. The lambs were sensitized with Brucella abortus S99 antigen after 140 days and were subjected to ELISA and delayed type hypersensitivity. Blood haemoglobin, erythrocyte count, leucocyte count, total protein, total albumin, total globulin, urea, creatinine concentration and aspartate aminotransferase activity in lambs fed on raw or processed CSM were comparable to the values of reference lambs. The higher (p<0.01) blood glucose levels observed in CSM fed lambs at 60 days of feeding was latter reduced to the levels comparable with those on reference diet at 120 and 180 days of feeding. The alanine amino transferase activity was lower in lambs fed raw and cooked CSM containing diets at 120 and 180 days of feeding. A marginal increase in serum iron and alkaline posphatase activity was observed in iron treated group and raw CSM fed lambs, respectively. The humoral immune response and DTH reactivity was lower (p<0.05) in lambs fed raw CSM (consuming 302.83 mg FG/day). Cooking, $Ca(OH)_2$ and iron treatment of raw CSM showed a positive response in alleviating the suppression of immune response owing to the reduced consumption of FG by 40.19, 17.40% and 26.73%, respectively in these diets. The present study thus indicated that consumption of 40% raw CSM (302.83 mg FG/day) though did not affect majority of the haematological and blood biochemical parameters, but markedly suppressed the immune mechanism of lambs.

Biological and Physicochemical Properties of Porcine Epidemic Diarrhea Virus Chinju99 Strain Isolated in Korea (국내 분리 돼지 유행성설사 바이러스 Chinju99주의 생물학적 및 물리화학적 성상)

  • A disease with severe diarrhea occurred in a herd of one thousand, 1-week-old piglets in Chinju, Korea, and was diagnosed as porcine epidemic diarrhea by the detection of N gene of porcine epidemic diarrhea virus (PEDV) from small intestines. A PEDV, named as Chinju99, was also isolated from the intestines after two blind-passages in Vero cells supplemented with trypsin (10 ug/ml). and the biological and physicochemical properties of the isolate were characterized. The virion was roughly spherical in shape and had spike peplomers on its outer surface. The virus exhibited cytopathic effects such as rounding degeneration at initiation of infection and syncytia formation later in Vero cells. The virus was labile to 20% ether and 5% chloroform but stable in acid with pH 4-7 at $4^{\circ}C$. The infectivity of the virus was maintained at $50^{\circ}C$ for 180 min, and the buoyant density of the virus in sucrose was 1.180 g/ml. All biological and physicochemical properties of the virus were typical features of coronaviruses.

High-pressure Compaction of Sawdust of Hyunsasi-poplar (Populus alba ${\times}$ P. glandulosa) for Densified Fuel (고밀화에 의한 현사시 톱밥의 고형연료화)

  • Recently, densified pellet fuel from wood biomass is widely used at North America and Europe as a regenerable and clean carbon neutral bioenergy. High-pressure compaction of sawdust of Hyunsasi-poplar (Populus alba ${\times}$ P. glandulosa) to form a densified fuel was studied. Calorific and elemental analysis were carried out to assess Hyunsasi-poplar clones as fuels. Hot-press process was adopted for compaction of sawdust and compaction was performed under temperature from 100 to 180$^{\circ}C$, at pressure of 250 to 1000 kgf/$\textrm{cm}^2$, and for 2.5 to 10 minutes. Densified fuels were evaluated by its oven-dry density and fines after 5-minute shaking test. The target density and fines of densified fuels were over 1.2 g/$\textrm{cm}^2$ and below 0.5%, respectively. When the press-temperature is over 160$^{\circ}C$, densified fuels with density eve. 1.2 g/$\textrm{cm}^2$ and with fines below 0.5% can be produced. And the pressure over 750 kgf/$\textrm{cm}^2$ was effective for this production. It was found that the optimum press condition for preparation of densified fuel was 180$^{\circ}C$ -1000 kgf/$\textrm{cm}^2$ minutes.

Migration Mechanism of Hazard Elements from Brass Kitchenwares (황동제 주방기구에서 유해중금속의 용출기작에 관한연구)

  • The migration mechanism of trace elements, Pb, Cd, Cu, Zn and Sn from brass food-contact utensiles was investigated. The migration of metals from brass was affected by the migration temperature (30, 60, 80 and $95^{\circ}C$), the simulant pH (2.5, 4.3, 6.0 and 7.0) and the migration time (30, 90, 180 and 360 min.). The amount of Pb migration was maximum at $95^{\circ}C$ with pH 2.5. This study indicated that the equilibrium of Pb migration was reached at 180 minutes. The mechanism of Pb migration was investigated with Scanning Electron Microscope (SEM) and Electron Probe Micro Analyzer (EPMA). To correlate the relations between the migration and the content of Pb in brass samples, ten brass samples having known content of Pb (the quantities of Pb in brass ; 0, 0.02, 0.09, 0.1, 0.2, 0.5, 0.9, 1.4, 5.4 and 9.2% (w/w)) were tested in the following conditions, 4% acetic acid, $95^{\circ}C$ and 30 minutes. The result represented that content of Pb in brass samples must be below 0.2% (w/w) to satisfy the 1.0 ppm (as Pb) of Korea Food Code.

Role of F/P Ratio on Curing Behavior for Phenolic Resol and Novolac Resins by FT-IR (FT-IR 분석에 의한 레졸과 노블락 페놀 수지의 경화거동에 미치는 F/P 몰비)

  • The curing behavior of a phenolic resin (F/p: 1.3, 1.9, 2.5 for resol resin, F/P: 0.5, 0.7, 0.9 for novolac resin) has been studied by FT-IR spectroscopy. In this study is to synthesis of resol and novolac type phenolic resin with different F/P molar ratios and to compare the level of cure at different curing temperature conditions ($130^{\circ}C$, $160^{\circ}C$, $180^{\circ}C$ for resol resin, $160^{\circ}C$, $170^{\circ}C$, $180^{\circ}C$ for novolac resin) for 3, 5, 7, 10, 20, and 60 (min.), respectively. The conversion (${\alpha}$) was determined by the ratio of the peak area with time to the peak area of non-baked phenolic QH ($3300cm^{-1}$) at spectra. It is concluded that the initial curing rate of resol and novolac resin was increased as the molar ratio of formaldehyde/phenol increased and as the curing temperature of resin increased. According to the analysis was by the homogenous first-order model, the initial curing rate of resol and novolac resin was increased as the molar ratio of formaIdehyde/phenol increased at specific curing temperature.

Antitumor Effect of Mugwort (Artemisia princeps Pampan.) in vivo

  • This study was devised to observe the antitumor activity of mugwort (Artemisia princeps Pampan.) against ICR mice inoculated with sarcoma-180 cells. The antitumor compounds were partially purified from petroleum ether extract of mugwort by silicic acid column chromatography. The active fraction used in in vivo test was obtained under the elution with acetone in silicic acid column chromatography. When the acetone fraction was intraperitoneally injected to the mice which had been subcutaneously inoculated on the left groin with sarcoma-180, the growth rate of tumor (sarcoma-180 mass) was inhibited by 30%. In case the acetone fraction was injected to the mice which had been inoculated intraperitoneally with sarcoma-180, the average life span was prolonged by 20%. After the injection of the active fraction, the spleen index and ${\gamma}-globulin$ ratio (%) were increased significantly (p<0.05). The administration of acetone fraction did not cause any abnormality in the body and the homeostasis of mice. Those observations suggest that the acetone fraction of mugwort extract has an antitumor effect in vivo.

  • Effect of Jeongkicheonhyangtang and Kamijeongkicheonhyangtang have been experimentally studied. The results have been obtained as follows : 1. It is proved that analgecic effect on sample I group and sample II group are clearly decreased than control group. Thus, the statistically significant decrease is p<0.005. 2. It is proved that anti-inflammatory effect on sample I group and sample II group are decreased compare to control group. Thus, the statistically significant decrease of sample I group is p<0.05 in 30 and 60-minute, p<0.01 in 90-minute, and p<0.005 in 120 and 180-minute. And those of sample ll group is p<0.05 in 30 and 60-minute and p<0.005 in 90, 120 and 180-minute. 3. It is proved that appearance of gastric ulcer is decreased compare to the control group and the statistically significant decrease is p<0.05. 4. It is proved that plasma epinephrine contents of sample I and II group are clearly decreased compare to the Control Group and the statistically significant decrease is p<0.005. 5. It is proved that plasma norepinephrine contents of sample I group is drastically decreased compare to the control group and the statistically significant decrease is p<0.005. 6. It is proved that plasma dopamine contents of sample I and II group are decreased than the contort group and statistically significant decrease of the sample I group is p<0.005, and those of the sample II group is p<0.05. As the above mentioned results of experiment, It is belived that Jeongklcheonhyang and Kamijeongkicheonhyang have effect for stress.

Molecular Cloning and Expression of a Cellulolytic Xylanase Gene from Bacillus circulans in Escherichia coli (Bacillus circulans 기원의 Cellulolytic Xylanase 유전자의 대장균에서의 클로닝 및 발현)

  • A gene for cellulolytic xylanase of Bacillus circulnns ATCC21365 was cloned on pUC 19 in Eschwichia coli. The recombinant plasniid pXLI80 contained an 1.8 id, inselt composed of0.5 kb and 1.3 kb PslI fragments derived from B, circulans. The 0.5 kh fragment in the upstream region of 1.3 kb one was confirmed lo be indispensable for not only expression but also hyperexpression of the cloned gene. The transformant overproduced the xylanase 135 times greater than that produced by the orlginal B circulnns. The optimum pH and temperature of the cloned enzyme we]-e pH 5.2 and $60^{\circ}C$, respectively. Heal pretl-eatment at TEX>$55^{\circ}C$C for 1 Indid not cause inhibition of the activity of this enzyme. The elm.ynie could hydl-olyre CMC and lichenan as well as xylan to produce xylose(or GI), xylohiose(or G2) and xylolnose(or G3) as inah products. Hence We defined the cloned enzyme as a cellulolytic xylanase. The SDS-PAG electrophoretic mobility and zyiiogram of this enzyme derived from whole cell extracts or c~~lture supematants or E. coli(pXL180) indicated a molecular weight of 45,000 and nonprocessing of the enzyme in the peilplasln of E. coli.

Effects of Blood Biochemistry and Tumors' Weights of Artemisia capillaris Methanol Extract in Mice Bearing Cancer Cells (인진쑥 methanol 추출물의 투여가 암이 유발된 마우스에서 보인 혈액생화학적 및 종양 무게에 미치는 영향)

  • The Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and probably the most common plant among the various herbal folk remedies being used in the treatment of abdominal pain hepatitis chronic liver disease, jaundice and coughing in Korea. Recently the biological and pharmacological actions of herb have been studied well such as antibacterial, antidiabetic and antitumor activities. This experiment was conducted to investigate antitumor and immunomodulatory effects of Artemisia capillarix extracts against Hepa-1c1c7 and Sarcoma 180 cancer cells on in vivo experimental tests. On in vivo experimental tests using 280 ICR mice the gain of body weight in the control-group mice bearing Sarcoma 180 ascites tumor was 1.5 times more than that of the normal-group mice after 33 days. However, the gain of body weight in all experimental groups administered with Artemisia capillaris extracts was significantly lower than that of the control-group mice (P<0.05). The mean survival times of mice administered with Artemisia capillaris extracts of 25 and 100 mg/kg for 28 days were shown to be 25.39% and 15.39% longer than that of the control-group mice injected with saline (P<0.05). Artemisia capillaris extracts showed the highest tumor inhibition effects, which were 42.4% and 27.2% when intraperitoneally injected with doses of 25 and 100 mg/kg once a day for 28 days in inoculated ICR mice with Sarcoma 180 solid tumor cells (P<0.05). The results suggest that Artemisia capillaris methanol extracts have prominent antitumor effects on the cancer cell lines Hepa-1c1c7 and Sarcoma 180.
