The Use of Phenanthraquinone Monophenyl Thiosemicarbazone for Preconcentration, Ion Flotation and Spectrometric Determination of Zinc(II) in Human Biofluids and Pharmaceutical Samples

  • A rapid flotation methodology for zinc(II) separation and enrichment from human biofluids is established. At pH 6.0 and ambient temperature, using oleic acid (HOL) as a foaming reagent, zinc(II) was separated with phenanthraquinone monophenyl thiosemicarbazone (PPT) as a new flotation collector for Zn(II). The floated red colored 1 : 2 Zn(II)-PPT complex was measured spectrophotometrically at 526 nm with a molar absorptivity of $1.83 \;{\times}\; 10^5\; L$ mol $L ^{-1}\;cm ^{-1}$. Beer's law was obeyed over a concentration range 0.05-1.0 mg $L ^{-1}$ in the aqueous as well as in the scum layers. The proposed preconcentration flotation methodology was applied to determine Zn(II) in human biofluids. Application was, also, extended to determine Zn(II) in pharmaceutical samples and natural water samples spiked with known amounts of Zn(II) with a preconcentration factor of 100 and a detection limit of 10 ng m$L ^{-1}$. The method was verified by comparison of the spectrophotometric results with flame atomic absorption spectrometric (AAS) measurements. Moreover a postulation for the mechanism of flotation is proposed.

Acinetobacter sp. A54에 의한 Arabian Light 원유의 분해

  • Bacterial strains which degrade Arabian Light crude oil were isolated by enrichment culture from oil-spilled soil. The strain A54 was finally selected after testing emulsifying activity and oil conversion rate. Strain A54 was identified as a Acinetobacter sp. based on the morphological, biochemical and physiological characteristics. It appears to be highly specialized for growth on Arabian Light crude oil in minimal salts medium since it showed preference for oil or degradation products as substrates for growth. It was found that it could grow on at least fifteen different hydrocarbons. The optimum cultural and environmental conditions were as follows; 25$\circ$C for temperature, 7,5 for pH, 2.0% for NaCl concentration and 2.0% for crude oil concentration. Additionally, the optimal concentration of NH$_{4}$NO$_{3}$, and K$_{2}$HPO$_{4}$, were 12.5 mM and 0.057 mM, respectively. Cell growth and emulsifying activity as a function of time were also determined. Crude oil degradation and the reduction of product peaks were identified by the analysis of remnant oil by gas chromatography. Approximately 63% of crude oil were converted into a form no longer extractable by mixed organic solvents.

Conessine Treatment Reduces Dexamethasone-Induced Muscle Atrophy by Regulating MuRF1 and Atrogin-1 Expression

  • Conessine, a steroidal alkaloid, is a potent histamine H3 antagonist with antimalarial activity. We recently reported that conessine treatment interferes with $H_2O_2$-induced cell death by regulating autophagy. However, the cellular signaling pathways involved in conessine treatment are not fully understood. Here, we report that conessine reduces muscle atrophy by interfering with the expression of atrophy-related ubiquitin ligases MuRF-1 and atrogin-1. Promoter reporter assay revealed that conessine treatment inhibits FoxO3a-dependent transcription, $NF-{\kappa}B$-dependent transcription, and p53-dependent transcription. We also showed by quantitative RT-PCR and western blot assays that conessine treatment reduced dexamethasone-induced expression of MuRF1 and atrogin-1. Finally, we demonstrated that conessine treatment reduced dexamethasone-induced muscle atrophy using differentiated C2C12 cells. These results collectively suggest that conessine is potentially useful in the treatment of muscle atrophy.

간호대학생의 문화적 역량 측정도구 개발 (Development of a Cultural Competence Scale for Nursing Students)

  • Purpose: This methodological study was conducted to develop and test a cultural competence scale for nursing students. Methods: Based on the five constructs of cultural competency identified in the conceptual analysis of Suh, 76 items for the tool were derived initially. These items were reduced to 58 items after content validity tests (two times) by 6 multicultural experts. Data collected from 526 nursing students were utilized to test the validity and reliability of the preliminary tool. Item analysis, factor analysis, Pearson correlation coefficients, and Cronbach's alpha were used for the analysis. Results: Twenty-seven items were selected for the final scale, and categorized into 5 factors explaining 62.1% of the total variance. Cronbach's alpha was .91 and the reliability of the subscales ranged from .76 to .91. Criterion validity between the developed tool and empathy (r=.26, p <.001) was significant. Conclusion: The results show that this scale can provide scientific and empirical data when evaluating the effectiveness of school curriculums or multicultural empowerment programs regarding cultural competence of nursing students.

  • In this paper, three different types of commercially tools-P20, NC123K and ceramic-have been used to working austempered ductile iron(ADI). In the austempered condition the materials are hard, strong and difficult to machine. Thus, we selected a optimum tool material among three different types of used tools in machining of austempered ductile iron. It was used acoustic emission(AE) to know cutting characteristic for selected tool and flank wear land of the ceramic too. The obtained results are as follows; (1) The ceramic tool among three different types of tools is the best in machining austempered ductile iron. (2) In case of ceramic tool, the amplitude level of AE signal(AErms) is mainly affected bycutting speed in cutting speed in cutting condition and it is proportioned to cutting speed. (3) There have the relationship of direct proportion between the amplitude level of AE signal and flank wear land of the tool. (4) If it find the value of AErms at each cutting speed, the in-process detection to ceramic tool's wear is possible

Alteration of Gas Exchange in Rice Leaves Infected with Magnaporthe grisea

  • Infection with rice blast fungus (Magnaporthe grisea) significantly reduced foliar net photosynthesis (A) of rice cultivars: Ilpoom, Hwasung, and Choochung in greenhouse experiments. By measuring the amount of diseased leaf area with a computer image analysis system, the relation between disease severity (DS) and net photosynthetic rate was curvilinearly correlated (r=0.679). Diseased leaves with 35% blast symptom can be predicted to have a 50% reduction of photosynthesis. The disease severity was linearly correlated (r=0.478) with total chlorophyll (chlorophyll a and chlorophyll b) per unit leaf area(TC). Light use efficiency was reduced by the fungal infection according to the light response curves. However, dark respiration (Rd) did not change after the fungal infection (p=0.526). Since the percent of reduction in photosynthesis greatly exceeded the percent of leaf area covered by blast lesions, loss of photosynthetic tissue on an area basis could not by itself account for the reduced photosynthesis. Quantitative photosynthetic reduction can be partially explained by decreasing TC, but cannot be explained by decreasing Rd. By photosynthesis (A)-internal CO$_2$ concentration (C$_i$ curve analysis, it was suggested that the fungal infection reduced ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activity, ribulose-1,5-bisphosphate (RuBP) regeneration, and inorganic phosphate regeneration. Thus, the reduction of photosynthesis by blast infection was associated with decreased TC and biochemical capacity, which comprises all carbon metabolism after CO$_2$ enters through the stomata.

Component Characteristics of Xanthoceras sorbifolium Seeds for Bioenergy Plant Utilization

  • Xanthoceras sorbifolium is considered as bio-energy crops owing to the high oil content from kernel. This study was performed to analyze calorific value, crude ash content, ultimate ratio, crude lipid and fatty acid composition among seed sources. Calorific values ranged from $4,526.0\;cal\;g^{-1}$ to $7,377.2\;cal\;g^{-1}$ in seeds and kernels showed the highest value. Calorific values and crude ash contents were observed as significant difference among plantations and/or individuals (p>0.05). Kernel from SD-F plantation showed the highest calorific value and lower crude ash content. C content comprised 63.4%, the highest levels was detected from SD-F (64.8%). Crude lipid content in kernel observed as 54.5 g $100\;g^{-1}$ from SD-F. In contrast it was determined the lowest value from LN-JARS as 46.5 g $100\;g^{-1}$. The fatty acid composition of kernel was determined to those of oleic acid (31.3%) and linoleic acid (38.1%) from SD-F and LN-JARS. These results will be offered to useful information for breeding materials selection.

  • This paper presents an improved algorithm of optimal measurement system design with a reliability evaluation method for a large power system. The proposed algorithm is developed to cosider the observability and to achieve highest accuracy of the state estimator as well with the limited investment cost. When the effect on these dummy bus measurements is considered in the proposed algorithm the other errors in the power system is also detected and then analyzed until to achieve the limited values. By taking advantage of the matrix sparsity and the optimal bus ordering the memory and the time are successfully reduced in the P/C's and workstation's model. The improved program is successfully tested for IEEK sample system and KEPCO system with PSS/E lineflow calculated data package.

Quenching Mechanisms and Kinetics of $\alpha$-, $\beta$-, $\gamma$-, and $\delta$-Tocopherol in Photosensitized Oxidation of Lard

  • Quenching mechanisms and kinetics of $\alpha$-, $\beta$-, $\gamma$-, and $\delta$-tocopherol in photosensitized oxidation of lard were studied. Lard at 0.03, 0.07, 0.11, and 0.3 M in methylene chloride containing $4.4{\times}10^{-6}\;M$ chlorophyll and 0, 0.1, 0.3, and 0.6 mM $\alpha$-, $\beta$-, $\gamma$-, and $\delta$-tocopherol were stored under light for 4 hr, respectively. Oxidation was determined by headspace oxygen and peroxide value. Tocopherols prevented the photosensitized oxidation of lard (p<0.05). Steady state kinetic study showed that $\alpha$-, $\beta$-, $\gamma$-, and $\delta$-tocopherol prevented the photosensitized oxidation of lard by quenching singlet oxygen. Singlet oxygen quenching rates of $\alpha$-, $\beta$-, $\gamma$-, and $\delta$-tocopherol by headspace oxygen depletion were 1.86, 2.39, 2.47, and $2.11{\times}10^7/M/sec$, respectively. The quenching rates of $\alpha$-, $\beta$-, $\gamma$-, and $\delta$-tocopherol by peroxide value were 1.42, 1.11, 0.97, and $0.42{\times}10^7/M/sec$, respectively. The quenching rates of tocopherols were slightly different depending on the measurements of oxidation.

Optimum Alcohol Fermenting Conditions for Kiwi (Actinidia chinensis) Wine

  • The objective of this study was to establish the optimum alcohol fermenting conditions for the processing of kiwi wine and vinegar products. Six yeast strains were examined for their alcohol production from kiwi at $30^{\circ}C$ for 72 hr with continuous shaking at 100 rpm. Under these conditions, Saccharomyces kluyveri DJ97 produced the highest alcohol content of 10.2%. As the fermentation time extended to 96 hr, the alcohol content reached a maximum of 12.75%. The optimum alcohol fermenting conditions for kiwi fruit were accomplished when kiwi was added to an equal amount of water, inoculated with S. kluyveri DJ97 and fermented at $30^{\circ}C$ for 96 hr with continuous shaking. The content of soluble solids decreased as the alcohol concentration increased, whereas little change was observed in the pH and titratable acidity during the low temperature aging process. Other alcoholic compounds, such as methanol, isopropanol, n-propanol, isobutanol, and isoamylalcohol, tended to increase as fermentation progressed.
