TNT 분해 세균 Stenotrophomonas sp. OK-5는 세 개의 다른 NAD(P)H-nitroreductase의 활성 fractions (NTR fractions I, II, III)을 갖고 있는 것으로 확인된 바 있다. 본 연구에서는 NTR fractions I, II, III에 대한 생리학적 특성과 분자생물학적 특성을 규명하고자 하였다. TNT에 대한 균주 OK-5의 NTR fractions I, II, 그리고 III의 활성은 억제 물질인 $\beta$-mercaptoethanol의 첨가 시에 효소의 활성 이 모두 억제되는 것으로 확인되었다. TNT와 그 유사 기질을 이용하여 균주 OK-5에서 분리된 NTR의 기질 특이성을 조사한 결과, nitrobenzene, 그리고 RDX에 대 해서는 비교적 활성 이 높게 나타났으나 2,6-DNT와 2,4-DNT에서는 낮은 활성을 나타내는 것으로 확인되었다. 균주 OK-5에서 정제된 NTR fraction I의 N-말단 아미노산 서 열은 $^1MSDLLNADAVVQLFRTARDS^20$로 분석되었고, Xanthomonas campestris의 NTR과 X. axonopodis의 NTR에서 각각 70%와 65%로 비교적 높은 유사성을 가지는 것으로 나타났다. 균주 OK-5의 NTR fraction I의 효소를 암호화하는 SmOK5nrI 유전자의 염기서열을 확인하고 분석된 유전자로부터 유추되는 아미노산 서열을 각각 비교한 결과 X. campestris의 NTR과 81%, X. axonopodis의 NTR과 75%,그리고 Streptomyces avermitilis의 NTR과 30%의 유사성이 있는 것으로 조사되었으나, Pseudomonas putida KT2440의 NTR (pnrB)과는 16%로 낮은 유사성이 있는 것으로 확인되었다.
폭약 2,4,6-trinitrotoluene (TNT)스트레스 조건하에서 토양세균 Pseudomonas sp. HK-6의 세포반응에 대하여 조사하였다. 다양한 농도의 TNT에 노출됨으로서 약 70-kDa DnaK와 60-kDa GroEL의 스트레스 충격단백질 (stress shock proteins, SSPs)이 단떠질이 유도되었다. 이들 SSPs의 존재는 SDS-PAGE과 anti-DnaK와 anti-GroEL monoclonal antibodies를 이용한 Western bolt을 통하여 확인되었다. SSPs은 0.5 mM TNT로 6-12 시간 처리된 세포에서 나타났으며, TNT에 노출 후8시간대 에서 최대의 단백질 유도가 관찰되었다. $30^{\circ}C$에서 $42^{\circ}C$로 열변환충격을 주었을 때의 SSPs는 TNT노출에서와 유사한 유도양상을 보여주었다. TNT에 노출된 Pseudomonas sp. HK-6세포에서 유도된 SSPs의 존재는 배양된 세포의 수용성 단백질 분획에 대하여 2-D PAGE를 통하여 확인되었다. Coomassie brilliant blue R25O로 염색된 젤로부터 pH 3-10 범위에서 약 450 개의 spots이 탐침되었으며, 이들 가운데 12 개의 spots이 TNT 스트레스에 대하여 현저하게 유도되었다. Gel상에서 가장 짙게 나타난 대표적 인 spot에 대한 N-말단 아미노산 서열을 분석한 결과, $^1XXAKDVKFGDSARKKML^17$로서, Pseudomonas putida의 GroEL의 N-말단 아미노산 염기서열인 $^1XXAKDVKFGDSARKKML^17$과 동일한 것으로 분석되었다.
Bioreporter 균주는 복잡한 환경매체의 특정 오염원 탐지를 위해 유용하게 사용되고 있다. 특히 발광 유전자 재조합 균주는 민감하고 배경에 의한 영향을 받지 않는 장점이 있다. 사용한 유전자 재조합 균주(Pseudomonas putida mt-2 KG1206)는 TOL 플라즈미드와 pUCD615 벡터에 $P_{m}\;promoter$가 삽입된 재조합 플라즈미드를 함유하고 있으며, 톨루엔 계열 및 중요 분해산물에 대해 분해와 함께 발광을 생산하는 특성을 갖고 있다. 본 연구에서는 균주 동결 및 동결건조 준비 및 적용과정에 필요한 다양한 조건들을 조사하여, 향후 환경매체에 적용하기 위한 최적 방법에 대한 프로토콜을 작성하였다. 조사한 최적 조건들은 다음과 같다. 동결보호시약(24% sucrose), 동결건조 시간(12시간), 균주 농도($OD_{600}=0.6$), 동결균주 활성회복($35^{\circ}C$에서 빠르게 해동), 동결건조 균주 활성회복(LB배지에 $3{\sim}6$시간 노출), 현장 운반 조건(활성 회복 후 $20^{\circ}C$ 정도의 실온). 본 연구 결과는 재조합 균주 환경 적용을 위해 필요한 균주 동결 및 동결 건조에 대한 중요한 자료들을 제시하고 있다.
산업폐수로부터 분리한 Pseudomonas sp. BCNU 106은 다양한 유기용매를 이용하여 인돌로부터 인디고를 합성할 수 있다. BCNU 106은 높은 수준의 인돌과 다량의 p-xylene, propylbenzene 그리고 mesitylene의 존재 하에서 효과적으로 인디고를 생산했다. 본 연구에서 인디고의 최대수율 조건은 20% (v/w)의 p-xylene과 4 g/l의 인돌인 것으로 확인되었다. 이러한 조건에서 인디고의 수율과 인돌의 전환효율은 각각 315.5 mg/l와 97%로 조사되었다. 그 결과 Pseudomonas sp. BCNU 106은 산업적으로 중요한 인디고 생산을 위한 잠재적인 균주가 될 것으로 판단된다.
Polyhydroxyalkanoates (PHAs) are emerging as alternatives to plastics by replacing fossil fuels with renewable raw substrates. Herein, we present the construction of engineered Escherichia coli strains to produce short-chain-length PHAs (scl-PHAs), including the monomers 4-hydroxyvalerate (4HV) and 3-hydroxyvalerate (3HV) produced from levulinic acid (LA). First, an E. coli strain expressing genes (lvaEDABC) from the LA metabolic pathway of Pseudomonas putida KT2440 was constructed to generate 4HV-CoA and 3HV-CoA. Second, both PhaAB enzymes from Cupriavidus necator H16 were expressed to supply 3-hydroxybutyrate (3HB)-CoA from acetyl-CoA. Finally, PHA synthase (PhaCCv) from Chromobacterium violaceum was introduced for the subsequent polymerization of these three monomers. The resulting E. coli strains produced four PHAs (w/w% of dry cell weight): 9.1 wt% P(4HV), 1.7 wt% P(3HV-co-4HV), 24.2 wt% P(3HB-co-4HV), and 35.6 wt% P(3HB-co-3HV-co-4HV).
Arylsulfate sulfotransferase (ASST) transfers a sulfate group from a phenolic sulfate ester to a phenolic acceptor substrate. In the present study, the gene encoding ASST was cloned from a genomic library copy of Citrobacter freundii, subcloned into the vector pGEM3Zf(-) and sequenced. Sequencing revealed two contiguous open reading frames (ORF1 and ORF2) on the same strand and based on amino acid sequence homologyl they were designated as astA and dsbA, respectively. The amino acid sequence of astA deduced from C. freundii was highly similar to that of the Salmonella typhimurium, Enterobacter amnigenus, Klebsiella, Pseudomonas putida, and Campylobacter jejuni, encoded by the astA genes. However, the ASST activity assay revealed different acceptor specificities. Using p-nitrophenyl sulfate (PNS) as a donor substrate, $\alpha$-naphthol was found to be the best acceptor substrate, followed by phenol, resorcinol, p-acetaminophen, tyramine and tyrosine.
2-Hydroxy-6-oxo-6phenylhexa-2,4-dienoate (HOPDA) hydrolase catalyzes the hydrolytic cleavage of HOPDA to bemzpate and 2-hydroxypenta-2, 4-dienoate (HPD) during microbial catabolism of biphenyl and polychlorinated biphenyls. A HOPDA hydrolase gene (pcbD) was isolated from the genomic library of Pseudomonas sp. P20 and designated as pCNUO1201; a 7.5-kb XbaI DNA fragment from Pseudomonas sp. P20 was inserted into the pBluescript SK(+) XbaI site. E. coli HB101 harboring pCNU1201 exhibited HOPDA hydrolase activity. The open reading frame (ORF) corresponding to the pcbD gene consisted of 855 base pairs with an ATG initiation codon and a TGA termination codon. The ORF was preceded by a rebosome-binding sequence of 5'-TGGAGC-3' and its G+C content was 55 mol%. The pcbD gene of Pseudomonas sp. P20 was located immedeately downstream of the pcbC gene encoding 2,3- dihydroxybiphenyl 1,2-dioxygenase, and approximately 4-kb upstream of the pcbE gene encoding HPD hydratase. The pcbK gene was able to encode a polypeptide with a molecular weight of 31,732 containing 284 amino acid residues. The deduced amino acid sequence of the HOPDA hydrolase of Pseudomonas sp. P20 exhibited high identity (62%) with those of the HOPDA hydrolases of P. putida KF715, P. pseudoalcaligenes KF707, and Burkholderia cepacia LB400, and also significant homology with those of other hydrolytic enzymes including esterase, transferase, and peptidase.
A biological filter for treatment of toluene among volatile organic compounds was studied. The investigation was conducted using specially built stainless steel columns packed with granular activated carbon and cold for removal of toluene. The G.A. and mold as filter material was also coated with Pseudomonas putida microorganisms.The biofilter unit was operated in the condition of moisture content vairation at gas loading rate of 12.5 l/min. Gaseous toluene taken from tedlar bag was analyzed by the use of G.C equipped with F.I.d detector. The removal efficiency of gaseous toluene was 95% at average inlet concentration of 950 ppm during bio-degradation operating condition. Effective removal efficiency was obtained with moisture content 27.5% at activated carbon and 32% at mold in this study. The effective operating condition were obtained with pH 6-8, temperature 28-42℃ for microbial degradation at gas loading rate of 12.5 l/min in packed material.
The purpose of this study was to develop a pilot scale bio-filter system removing ammonia gas with microorganisms. The system consisted of chaff(filter medium), a blower, a temperature sensor, a moisture sensor, a solenoid valve, and a heating system. Temperature and moisture contents were controlled via a PC to provide the microorganisms with proper environment. The microorganisms used in this study were Bacillius. coagulans NLRI T-6 and Pseudononas. putida NLRI S-21 of bacilli. Performance tests were performed to evaluate gas removal rate during 20 days. The result was shown that the removal rate was high in early days and gradually dropped below 90% without injecting the microbes. However, it was shown that when injecting the microbes, the removal rate was almost 100% and pH value was maintained at between 7 and 9 during the whole twenty-day period.
Six strains of an obligate predatory bdellovibrio isolate that preys on Burkholderia glumae in rice paddy field water and rhizosphere soil, were identified and characterized. The numbers of Bdellovibrio cells varied from $3.2{\times}10^3$ to $9.2{\times}10^3$ plaque-forming unit/g after enrichment in cells of B. glumae. Prey range tests with six Bdellovibrio strains and 17 prey strains of rice-pathogenic, antibiosis-related, or nitrogen-fixing bacteria resulted in unique predation patterns in related prey cells. Strain BG282 had the widest prey range on 7 plant pathogenic bacteria among the 17 prey strains tested. However, no predation occurred with strains of Azospirillum brasilense, Paenibacillus polymyxa, Pseudomonas fluorescens, P. putida, and Serratia marcescens that are associated with antibiosis or nitrogen fixation in the rice ecosystem. Identification was confirmed by the presence of typical bdelloplast in the prey cells of B. glumae and by a PCR assay using B. bacteriovorus-specific primers. Furthermore, 16S rDNA sequencing of the six bdellovibrio strains showed a homology range of 97.2% to 99.2% to the type strain of B. bacteriovorus.
