Basic Informations on the Characteristics of Strains of Oyster Mushroom (느타리버섯 계통별(系統別) 특성(特性)에 관한 기초(基礎) 조사(調査))

  • Thle experiments were carried out to select the highest yield and good quality of the oyster mushroom (Pleurotus sp.) strain for brooding material sources. The strains used covered 6 from 4 countries, No. 2063, 2078 and 2093 were collected from Korea, No. 2068 introduced from Japan, No. 2091 from Germany and No. 2095 from Taiwan. Some characteristics of the strains are as follows. 1) The optimum temperature for strains, 2091, 2093 and 2063 of P. ostreatus was $30^{\circ}C$, it ranged from 25 to $30^{\circ}C$ for the others. 2) The optimum moisture content were 70% for strain 2068, 80% for strains 2093, 2095 and 2091, 75% for other strains. 3) The optimum pH of the mycelial growth were at pH 6.0 for strains 2093 and 2068, pH 6.0 and pH 8.0 for other strains. 4) The days from spawning to budding was the shortest in strain 2095 and the yield was the highest in 2091, $64.4kg/3.3m^2$on the rice straw cultivation.

Production of Yeast from the Acid Hydrolyzate of the Waste Composts of Oyster Mushroom (느타리버섯 폐상퇴비(廢床堆肥) 산가수분해액(酸加水分解液)을 이용(利用)한 효모생산(酵母生産))

  • Culture conditions for yeast production from the acid hydrolyzate of the 2nd waste composts of oyster mushroom (Pleurotus ostreatus) were determined. Among the yeast strains tested, Candida quilliermondii JAFM 215, which was culture at $30^{\circ}C$, pH 5.0, showed good culture yield. Yeast production was the highest yield with the medium composition of 0.3% $NH_4Cl$, 0.15% $KH_2PO_4$, 0.02% $MgSO_4{\cdot}7H_2O$, and 0.05% $CaCl_2$. Yeast growth was increased at the concentration of 0.001 to 0.01% furfural, but at the higher concentration the yeast growth was inhibited. Utilization rate of sugar was 86.2%, and yield of yeast from sugar was 50.45%. Crude protein of yeast ranged from 50 to 52%.

Studies on the Development of Casing Materials Using Sawdust Bottle Culture in Cultivated Mushroom, Agaricus bisporus (병재배 폐톱밥을 이용한 양송이 복토재료 개발에 관한 연구)

  • After bottle culture of Pleurotus ostreatus, sawdust was taken out from the bottle and accumulated in the middle of March, and then composted. As the result, Y value was decreased rapidly 30 days after composting, and it was decreased slowly after 30 days. It is considered that 118 days is required for composting, however, it is possible to use for casing material after at least 48 days composting. The pH and total nitrogen content of sawdust based on composting period had tendency to increase as composting was processed. Total carbon and C/N rate had tendency to decrease as time went on. Based on the rate of 10, 30 and 50%, each sawdust was added to clay loam used as casing material for culturing A. bisporrus. Among various treatments, the mycelial growth of A. bisporus was more favorable in the treatment of 30% sawdust than in the single treatment of clay loam. Based on the date necessary for primodium formation of A. bisporus, the primodium formation in the treatment of 30% sawdust was reduced to about 5 days as compared with that of any other treatments When 30% sawdust was added to clay loam used as casing material for culturing A. bisporus, the yield of its fruiting body was increased to 28%.

  • The retrotransposon marY1 is a gypsy family retroelement, which is detected ubiquitously within the fungal taxonomic groups in which mushrooms are included. To utilize marY1 as a molecular marker for the DNA fingerprinting of mushrooms, oligonucleotides marY1-LTR-L and marY1-LTR-R were designed on the basis of highly conserved regions from the multiple sequence alignment of 30 marY1 sequences retrieved from a nucleotide sequence database. In accordance with $\underline{Re}trotransposon$ $\underline{M}icrosatellite$ $\underline{A}mplified$ $\underline{P}olymorphism$ (REMAP) fingerprinting methodology, the two oligonucleotides were utilized together with the short sequence repeat primers UBC807 and UBC818 for polymerase chain reaction using templates from different mushroom genomic DNAs. Among the tested oligonucleotides, the marY1-LTR-L and UBC807 primer set yielded the greatest amount of abundance and variation in terms of DNA band numbers and patterns. This method was successfully applied to 10 mushroom species, and the primer set successfully discriminated between different commercial mushroom cultivars of the same strains of 14 Pleurotus ostreatus and 16 P. eryngii. REMAP reproducibility was superior to other popular DNA fingerprinting methodologies including the random amplified polymorphic DNA method.

Development of Optimal Culture Media for the Stable Production of Mushroom (버섯의 안정적 생산을 위한 최적배지의 개발)

  • Several antagonistic bacteria, SD-1, 4, 10, 11, 14, 15, and 16, which have strong CMCase and amylase activities, were isolated from the fermented mushroom media. Among them, SD-1, 10, 11, and 15 have strong antibacterial activities against the mushroom pathogenic bacteria Pseudomonas sp., and SD-1, 10, 11, 14, and 16 have strong antifungal activities against the mushroom pathogenic fungi, Trichoderma sp. SD-14, 15, and 16 did not inhibit the growth of mushroom Pleurotus eryngii ASI-2302, and Pleurotus ostreatus ASI-2042 and ASI-2180. When the culture broth mixture of the seven bacterial strains was applied to the mushroom media, the growths of pathogens, Pseudomonas sp. and Trichoderma sp., were inhibited.

  • A gram-negative bacterium was isolated from spent substrate of Agaricus bisporus and showed marked antagonistic activity against Pseudomonas tolaasii. The bacterium was identified as Pseudomonas azotoformans by based on the cultural, biochemical and physiological characteristics, and 16S rRNA gene sequence. The isolated bacterium was saprophytic but not parasitic nor pathogenic to cultivation mushroom. The isolated bacterium for P. tolaasii cell was not sufficient for inhibition in vitro. Control efficacy of Pseudomonas azotoformans HC5 to brown blotch of P. tolaasii was 73, 78, and 71% on A. bisporus, Flammulina velutipes, and Pleurotus ostreatus, respectively. In the future, the suppressive bacterium may be useful for development of a biocontrol system.

Comparative Molecular Phylogenetic Relationships in Different Strains of Pleurotus spp. (느타리속 버섯 계통의 분자생물학적 유연관계의 비교연구)

  • Pleurotus spp. have been used for edible and medicinal purposes in Asian countries for a long time. The fruiting bodies of the Pleurotus ostreatus, Pleurotus citrinopileatus and Pleurotus salmoneostramineus contained many physiologically beneficial substances for human health. Therefore, it is necessary to study the genetic diversity of Pleurotus mushroom cultivars commercially cultivated in Korea. Eleven strains of Pleurotus spp. were collected from different geographical regions in South-East Asia and ITS regions of rDNA and RAPD of genomic DNA were analyzed. The size of the ITS1 and ITS2 regions of rDNA from the different strains varied from 167 to 254 bp and 156 to 213 bp, respectively. The sequence of ITS1 was more variable than that of ITS2, and the 5.8S sequences were identical. A phylogenetic tree based on the ITS region sequences indicated that selected strains could be classified into 4 clusters. Eleven Pleurotus species were also analyzed by RAPD with 20 arbitrary primers. Ten of these primers were efficiently amplified the genomic DNA. The number of amplified bands varied with the primers and strains, with polymorphic fragments in the range from 0.1 to 2.0kb. The results revealed that genetic diversity of selected strains of P. ostreatus, P. citrinopileatus and P. salmoneostramineus is low.

Psychrophilic oyster mushroom variety 'Seolan' (느타리버섯 신품종 육성 연구 -저온성 느타리버섯 신품종 '설안'-)

  • 'Seoln', a new variety of oyster mushroom was developed by Mushroom Research Institute, Gyonggi Province Agricultural Research and Extension Services in 2005. It was bred with mating between monokaryotic strains isolated from KME35124 and $F{\cdot}X3$. The major characteristics of the mushroom was shown a lot of primordia, the gray-colored and bunch pileus. The optimum temperature for the mycelial growth was around $26{\sim}30^{\circ}C$ and that for the primordia and growth of fruit-body was around $10{\sim}15^{\circ}C$. Incubation period was required around 25 days with bottle culture and about 23 days in Poly Prophylene(P.P) plastic bag culture. The yield was shown high by 138.8g/bottle and 250.9%/P.P bag.

Identification of Varieties by Biochemical Methods in Pleurotus spp. (느타리 버섯류(Pleurotus spp.)의 생화학적 방법에 의한 품종구분)

  • To identify genetic difference of 13 strains in three Pleurotus species, analyses of rDNA, AP-PCR and RFLP were carried out. IGRI and $ITSI{\sim}II$ regions of rDNA amplified by PCR were about 0.9 and 0.7 kb, respectively. These PCR products were digested with six restriction enzymes to analyse polymorphism. Especially, treatment of HaeIII enzyme on $ITSI{\sim}II$ regions showed specific bands in three Pleurotus sajor-caju strains. Genetic differences among three species were classified by similarity analyses based on rDNA polymorphism. Various band patterns of $2,500{\sim}150\;bp$ were showed by AP-PCR. Identification of species and varieties in 13 Pleurotus strains was possible according to primers used in AP-PCR. In order to develop genetic markers, RFLPs using IGRI and $ITSI{\sim}II$ probes derived from ASI 2180 and 2070 were carried out on eight Pleurotus varieties. RFLP patterns using IGRI probe were more various than that of $ITSI{\sim}II$ probe.

  • 'Dajoa', a new variety of oyster mushroom was developed by Mushroom Research Institute, Gyonggi Province Agricultural Research and Extension Services in 2004. It was bred with mating between monokaryotic strains isolated from Boonli 89-1 and ASI 2018-249. The major characteristics of the mushroom are showing a lot of pinheadings, the gray-colored and infundibuliform pileus. The optimum temperature for the mycelial growth was around $26{\sim}28^{\circ}C$ and that for the pinheading and growth of fruitbody was around $15{\sim}18^{\circ}C$. Incubation period was required around 24 days with bottle culture and about 21days in Poly Prophylene(P.P) plastic bag culture. The yields were shown high by 140.7g/850cc bottle and 260.3g/1kg P.P. plastic bag.
