A simple method to determine viable cell numbers of each species in mixed culture was developed. The oxygen uptake rate (OUR) equals to the product of the specific OUR and the size of the microbial population. In a mixed culture, the OUR is a result of the respiration activities of each sub-population. The OUR was determined from the slope of the linear relationship between time and the decrease of dissolved oxygen concentration when aeration was stopped. The specific OUR was calculated from the slope of the viable cell number versus OUR curve. These values for C. lusitaniae at 20 and $30^{\circ}C$ were $1.36{\times}10^{-9}$ and $3.90{\times}10^{-9}$ and those for P tannoPhilus at 20 and $30^{\circ}C$ were $0.59{\times}10^{-9}$ and $1.86{\times}10^{-9}$ [(%/s)/(cells/ml)J. respectively. Using these values, viable cell numbers were calculated after the OURs of mixed culture at two temperatures were measured. A good agreement between the viable cell numbers determined by this method and by plate count was obtained.
In order to clarify the receptor types and mechanisms underlying the positive inotropic effect of dopamine on the mammalian ventricular myocardium, the action potential, its first derivatives and isometric contraction of the rabbit papillary muscle were recorded using a force transducer and glass capillary microelectrodes filled with 3M KCl. The results were as follows; (1) In normal Tyrode solution, the contractile force was increased and duration of action potential was shortened with increments of dopamine concentration ($10^{-6}-10^{-4}M$). (2) The dose-response curve was markedly shifted to the right by pretreatment with reserpine (5mg/kg i.p., 24hrs prior to the experiment). (3) In 19mM $K^+-Tyrode$ solution, the duration of action potential, maximum rate of rise (V_{max}) of action potential and overshoot were significantly increased with increments of dopamine concentration ($10^{-6}-10^{-4}M$). (4) The inotropic effect of dopamine on the rabbit papillary muscle pretreated with reserpine was antagonized by atenolol ($10^{-6}M$), but not by phentolamine ($3{\times}10^{-6}M$). (5) In rabbit papillary muscle partially depolarized by 19mM $K^+-Tyrode$ solution, slow electrical response (calcium mediated action potential) as well as contraction were restored by dopamine ($10^{-4}M$); this restoration was blocked by calcium antagonists ($3{\times}10^{-5}M$$LaCl_3{\cdot}6H_2O$, $3{\times}10^{-6}M$ diltiazem) or ${\beta}-adrenoceptor$ antagonist ($3{\times}10^{-6}M$ atenolol), but not affected by ${\alpha}-adrenoceptor$ antagonist ($10^{-5}M$ phentolamine, $3{\times}10^{-6}M$ yohimbine) or vascular dopaminergic receptor antagonist ($10^{-5}M$ haloperidol). The above results may be interpreted as that the positive inotropic effect of dopamine through both direct and indirect action are caused by increase in slow inward current ($Ca^{2+}$ influx into themyocardial cell), and the direct action is mainly due to the stimulation of ${\beta}-adrenoceptors$ in the rabbit papillary muscle.

NiTi alloy has been used for orthodontic wire due to good mechanical properties, such as elastic strength, friction resistance, and high corrosion resistance. Recently, these wire were coated by polymer and ceramic materials for aesthetics. The purpose of this study was to investigate electrochemical characteristics of tooth colored NiTi wire using various instruments. Wires (round type and rectangular type) were used, respectively, for experiment. Polymer coating was carried out for wire. Specimen was investigated with optical microscopy (OM), field emission scanning electron microscopy (FE-SEM) and energy dispersive x-ray spectroscopy (EDS). The corrosion properties of the specimens were examined using potentiodynamic tests (potential range of -1500 ~ 2000 mV) and electrochemical impedance spectroscopy (frequency range of 100 kHz ~ 10 mHz) in a 0.9 % NaCl solution by potentiostat. From the results of polarization behavior, the passive region of non-coated NiTi wire showed largely, whereas, the passive region of curved NiTi wire showed shortly in anodic polarization curve. In the case of coated NiTi wire, pitting and crevice corrosion occurred severely at interface between non-coated and coated region. From the results of EIS, polarization resistance(Rp) value of non-coated round and rectangular NiTi wire at curved part showed $5.10{\times}10^5{\Omega}cm^2$ and $4.43{\times}10^5{\Omega}cm^2$. lower than that of coated NiTi wire. $R_p$ of coated round and rectangular NiTi wire at curved part showed $1.31{\times}10^6{\Omega}cm^2$ and $1.19{\times}10^6{\Omega}cm^2$.

Enterotoxigenic Escherichia coli (ETEC) is the major pathogenic E. coli that causes diarrhea and edema in post-weaning piglets. In this study, we describe the morphology and characteristics of ØCJ19, a bacteriophage that infects ETEC, and performed genetic analysis. Phage ØCJ19 belongs to the family Myoviridae. One-step growth curve showed a latent phase of 5 min and burst size of approximately 20 phage particles/infected cell. Phage infectivity was stable for 2 h between 4℃ and 55℃, and the phage was stable between pH 3 and 11. Genetic analysis revealed that phage ØCJ19 has a total of 49,567 bases and 79 open reading frames (ORFs). The full genomic sequence of phage ØCJ19 showed the most similarity to an Escherichia phage, vB_EcoS_ESCO41. There were no genes encoding lysogeny, toxins, virulence factors, or antibiotic resistance in this phage, suggesting that this phage can be used safely as a biological agent to control ETEC. Comparative genomic analysis in terms of the tail fiber proteins could provide genetic insight into host recognition and the relationship with other coliphages. These results showed the possibility to improve food safety by applying phage ØCJ19 to foods of animal origin contaminated with ETEC and suggests that it could be the basis for establishing a safety management system in the animal husbandry.
LMC X-4 is an eclipsing high-mass X-ray binary exhibiting a superorbital modulation with a period of ~ 30:5 days. We present a detailed study of the variations of the superorbital modulation period with a time baseline of ~ 18 years. The period determined in the light curve collected by the Monitor of All-sky X-ray Image (MAXI) significantly deviates from that observed by the All Sky Monitor (ASM) onboard the Rossi X-ray Timing Explorer (RXTE). Using the data collected by RXTE/ASM, MAXI, and the Burst Alert Telescope (BAT) onboard Swift, we found a significant period derivative, $\dot{P}=(2.08{\pm}0.12){\times}10^{-5}$. Furthermore, the O{C residual shows complex short-term variations indicating that the superorbital modulation of LMC X-4 exhibits complicated unstable behaviors. In addition, we used archive data collected by the Proportional Counter Array (PCA) on RXTE to estimate the orbital and spin parameters. The detected pulse frequencies obtained in small time segments were fitted with a circular orbital Doppler shift model. In addition to orbital parameters and spin frequency for each observation, we found a spin frequency derivative of $\dot{v}=(6.482{\pm}0.011){\times}10^{-13}Hz{\cdot}s^{-1}$. More precise orbital and spin parameters will be evaluated by the pulse arrival time delay technique in the future.
PbTiO$_{3}$-PbZrO$_{3}$-Pb(Ni$_{1}$3/Nb $_{2}$3/O$_{3}$)(PZT-PNN) thin films were prepared from corresponding metal organics partially stabilized with diethanolamine by the sol-gel spin coating method. Each mol rates of PT:PZ:PNN solutions were #1(50:40:10), #2(50:30:20), #3(45:35:20) and #4(40:40:20), respectively. The spin-coated PZT-PNN films were sintered at the temperature from 500.deg. C to 600.deg. C for crystallization. The P-E hysteresis curve was drawn by Sawyer-Tower circuit with PZT-PNN film. The coercive field and the remanent polarization of #4(40:40:20 mol%) PZT-PNN film were 28.8 kV/cm and 18.3 .mu.C/cm$^{2}$, respectively. Their dielectric constants were shown between 128 and 1120, and became maximum value in MPB(40:40:20 mol%). The leakage currents of PZT-PNN films were about 9.4x 10$^{-8}$ A/cm$^{2}$, and the breakdown voltages were about 0.14 and 1.1 MV/cm. The Curie point of #3(45:35:20 mol%, sintered at 600.deg. C) film was 330.deg. C.
The article presents a theoretical-experimental approach developed for modeling the coefficient of sliding friction in the dynamic system tool-workpiece in slide diamond burnishing of low-alloy unhardened steels. The experimental setup, implemented on conventional lathe, includes a specially designed device, with a straight cantilever beam as body. The beam is simultaneously loaded by bending (from transverse slide friction force) and compression (from longitudinal burnishing force), which is a reason for geometrical nonlinearity. A method, based on the idea of separation of the variables (time and metric) before establishing the differential equation of motion, has been applied for dynamic modeling of the beam elastic curve. Between the longitudinal (burnishing force) and transverse (slide friction force) forces exists a correlation defined by Coulomb's law of sliding friction. On this basis, an analytical relationship between the beam deflection and the sought friction coefficient has been obtained. In order to measure the deflection of the beam, strain gauges connected in a "full bridge" type of circuit are used. A flexible adhesive is selected, which provides an opportunity for dynamic measurements through the constructed measuring system. The signal is proportional to the beam deflection and is fed to the analog input of USB DAQ board, from where the signal enters in a purposely created virtual instrument which is developed by means of Labview. The basic characteristic of the virtual instrument is the ability to record and visualize in a real time the measured deflection. The signal sampling frequency is chosen in accordance with Nyquist-Shannon sampling theorem. In order to obtain a regression model of the friction coefficient with the participation of the diamond burnishing process parameters, an experimental design with 55 experimental points is synthesized. A regression analysis and analysis of variance have been carried out. The influence of the factors on the friction coefficient is established using sections of the hyper-surface of the friction coefficient model with the hyper-planes.
The yield criterion, or called yield function, plays an important role in the study of plastic working of a sheet because it governs the plastic deformation properties of the sheet during plastic forming process. In this paper, we propose a novel anisotropic yield function useful for describing the plastic behavior of various anisotropic sheets. The proposed yield function includes the anisotropic version of the second stress invariant J2 and the third stress invariant J3. The anisotropic yield function newly proposed in this study is as follows. F(J2)+ αG(J3)+ βH (J2 × J3) = km The proposed yield function well explains the anisotropic plastic behavior of various sheets by introducing the parameters α and β, and also exhibits both symmetrical and asymmetrical yield surfaces. The parameters included in the proposed model are determined through an optimization algorithm from uniaxial and biaxial experimental data under proportional loading path. In this study, the validity of the proposed anisotropic yield function was verified by comparing the yield surface shape, normalized uniaxial yield stress value, and Lankford's anisotropic coefficient R-value derived with the experimental results. Application for the proposed anisotropic yield function to aluminum sheet shows symmetrical yielding behavior and to pure titanium sheet shows asymmetric yielding behavior, it was shown that the yield curve and yield behavior of various types of sheet materials can be predicted reasonably by using the proposed new yield anisotropic function.

Background: Due to multiple similarities in the structure and physiology of human and pig skin, the pig model is extremely useful for biological drug testing after subcutaneous administration. Knowledge of the differences between subcutaneous injection sites could have a significant impact on the absorption phase and pharmacokinetic profiles of biological drugs. Objectives: This study aimed to analyze the impact of administration site on pharmacokinetics and selected biochemical and hematological parameters after a single subcutaneous administration of ustekinumab in pigs. Drug concentrations in blood plasma were analyzed by enzyme-linked immunosorbent assay. Pharmacokinetic analyses were performed based on raw data using Phoenix WinNonlin 8.1 software and ThothPro v 4.1. Methods: The study included 12 healthy, female, large white piglets. Each group received a single dose of ustekinumab given as a 1 mg/kg subcutaneous injection into the internal part of the inguinal fold or the external part of the inguinal fold. Results: The differences in absorption rate between the internal and external parts of the inguinal fold were not significant. However, the time of maximal concentration, clearance, area under the curve calculated between zero and mean residence time and mean residence time between groups were substantially different (p > 0.05). The relative bioavailability after administration of ustekinumab into the external part of the inguinal fold was 40.36% lower than after administration of ustekinumab into the internal part of the inguinal fold. Conclusions: Healthy breeding pigs are a relevant model to study the pharmacokinetic profile of subcutaneously administered ustekinumab.
To evaluate if the apoptotic fragment assay could be used to estimate the dose prediction after radiation exposure, we examined apoptotic mouse crypt cells per 1,000 cells after whole body $^{60}Co$$\gamma$-rays and 50MeV ($p{\rightarrow}Be^+$) cyclotron fast neutron irradiation in the range of 0.25 to 1 Gy, respectively. The incidence of apoptotic cell death rose steeply at very low doses up to 1 Gy, and radiation at all doses tigger rapid changes in crypt cells in stem cell region. These data suggest that apoptosis may play an important role in homeostasis of damaged radiosensitive target organ by removing damaged cells. The curve of dose-effect relationship for the data of apoptotic fragments was obtained by the linear-quadratic model $y=0.18+(9.728{\pm}0.887)D+(-4.727{\pm}1.033)D^2$ ($r^2=0.984$) after $\gamma$-rays irradiation, while $y=0.18+(5.125{\pm}0.601)D+(-2.652{\pm}0.7000)D^2$ ($r^2=0.970$) after neutrons in mice. The dose-response curves were linear-quadratic, and a significant dose-response relationship was found between the frequency of apoptotic cell and dose. These data show a trend towards increase of the numbers of apoptotic crypt cells with increasing dose. Both the time course and the radiation dose-response curve for high and low linear energy transfer (LET) radiation modalities were similar. The relative biological effectiveness (RBE) value for crypt cells was 2.072. In addition, there were significant peaks on apoptosis induction at 4 and 6h after irradiation, and the morpholoigcal findings of the irradiated groups were typical apoptotic fragments in crypt cells that were hardly observed in the control group. Thus, apoptosis in crypt cells could be a useful in vivo model for studying radio-protective drug sensitivity or screening test, microdosimetric indicator and radiation-induced target organ injury. Since the apoptotic fragment assay is simple, rapid and reproducible in the range of 0.25 to 1 Gy, it will also be a good tool for evaluating the dose response of radiation-induced organ damage in vivo and provide a potentially valuable biodosimetry for the early dose prediction after accidental exposure.
