Effect of Natural Plant Mixtures on Behavioral Profiles and Antioxidants Status in SD Rats (자생식물 혼합 추출물이 SD 흰쥐에서의 행동양상 및 항산화 체계에 미치는 영향)

  • Caffeine, a psychoactive stimulant, has been implicated in the modulation of learning and memory functions due to its action as a non-selective adenosine receptors antagonist. On the contrary, some side effects of caffeine have been reported, such as an increased energy loss and metabolic rate, decrease DNA synthesis in the spleen, and increased oxidative damage to exerted on LDL particles. Therefore, the aim of this study was to develop a safe stimulant from natural plants mixture (Aralia elata, Acori graminei Rhizoma, Chrysanthemum, Dandleion, Guarana, Shepherd's purse) that can be used as a substitute for caffeine. Thirty SD rats were divided into three groups; control group, caffeine group (15.0 mg/kg, i.p.), and natural plants mixture group (NP, 1 mL/kg, p.o.). The effect of NP extract on stimulant activity was evaluated with open-field test (OFT) and plus maze test for measurement of behavioral profiles. Plasma lipid profiles, lipid peroxidation in LDL (conjugated dienes), total antioxidant capacity (TRAP) and DNA damage in white blood, liver, and brain cells were measured. In the OFT, immobility time was increased significantly by acute (once) and chronic (3 weeks) supplementation of NP and showed a similar effect to caffeine treatment. Three weeks of caffeine treatment caused plasma lipid peroxidation and DNA damage in liver cells, whereas there were no changes in the NP group. NP group showed a higher plasma HDL cholesterol concentration compared to the caffeine group. The results indicate that the natural plants mixture had a stimulant effect without inducing oxidative stress.

Effects of Sports Drink Including the Extract from Prunus mume on the Changes of Respiratory Variables, Heart Rate, and Blood Lactate Concentration in Submaximal Exercise (매실함유 음료섭취가 장시간 운동시 심박수, 호흡가스 변인 및 혈중 젖산농도 변화에 미치는 영향)

  • This study was designed to investigate the effects of sports drink including the extract from Prunus mume on the changes of oxygen uptake, minute ventilaton, heart rate, and blood lactate concentration during 1 hour treadmill running exercise corresponding to 75%VO$_2$max. Subjects were 16 male athletes(long distance runners and tennis players). The changing patterns of oxygen uptake and minute ventilation showed no significnat difference among all types of sports drink, but the minute vetilation of the placebo group showed a higher value than type I group during submaximal exercise. Although the changing patterns of heart rate and blood lactate concentration showed no significnat difference among all types of sports drink, but type E group showed a lower heart rate compared to Placebo group. And blood lactate concentration of placebo group showed a higher value compared to the value corresponding to lactate threshold during submaimal exercise. but the other types of sports drink showed a lower value of blood lactate concentration. Especially blood lactate threshold of type D and E groups showed a lower value (range from 1.44 to 2.00mM) during submaximal exercise. In these results, the sports drink including the extract from Prunus mume showed no significant effects to the changes of oxygen uptake, minute ventilaton, heart rate, and blood lactate concentration during 1 hour treadmill runnuing exercise corresponding to 75%VO$_2$max. But it could be suggested the positive effects of the intake of sports drink including the extract from Prunus mume on the inhibition of exercise-induced fatigue during submaximal exercise, because of the lower changing Patterns of heart rate, blood lactate concentration, and ventilation efficiency in this results.

The Influences of Additional Nutrients on Phytoplankton Growth and Horizontal Phytoplankton Community Distribution during the Autumn Season in Gwangyang Bay, Korea (가을철 광양만 식물플랑크톤의 수평 분포와 추가 영양염 공급이 식물플랑크톤 성장에 미치는 영향)

  • In order to estimate the effect of additional nutrients on phytoplankton growth and horizontal phytoplankton community distribution during the autumn season in 2010 and 2011, we investigated the abiotic and biotic factors of surface and bottom waters at 20 stations of inner and offshore areas in Gwangyang Bay, Korea. Also, nutrient additional experiments were conducted to assess additional nutrient effects on phytoplankton assemblage using the surface water. In both years, the total nutrients were high at the enclosed inner bay and the mouth of Seomjin River, whereas it was low at the St.15~20 where in influenced by the surface warm water current from offshore of the bay. On the other hand, nano- and pico-sized Chl. a were gradually increased towards the outer bay and their trends were significant in 2011 than in 2010. The cryptophyta species occupied more than 85% of total phytoplankton assembleges in 2010, whereas their abundance in 2011 remainds to be 1/10 levels of 2010. Following the cryptophata species, the diatom Chaetoceros spp. and Skeletonema-like spp. were found to be dominant species. Further the biosaasy experimental results shows that the phytoplankton biomass in the +N and +NP treatments was higher compared to control and +P treatments and its trend was significant at St.8 and St.20 where nutrient concentration were low. Based on the bioassay and field survey, providing the high nutrients may have stimulated to phytoplankton growth such as S. costatum-like spp.. In particular, opportunistic micro-algae such as Cryptomonas spp. were able to achieve the high biomass under the relatively mid nutrient condition from bottom after break down of seasonal stratification in the Gwangyang Bay.

Effect of Dietary Supplementation of CLA-Containing Oil (CLAzen 80) on fatty Acid Composition of Egg Yolk in Laying Hens (산란계 사료내 CLA 함유 Oil (CLAzen 80) 첨가가 난황내 지방산 조성에 미치는 영향)

  • The objectives of the present study were to investigate the effects of varying levels of dietary oil containing conjugated linoleic acid (CLA) on the egg production and fatty acid composition of egg yolk. Seventy-two 59-wk-old ISA Brown laying hens were randomly allotted to four dietary treatments, each consisting of three replicates with six birds per replicate. There were four treatments that consist of diets containing 0, 1, 2, or $3\%$ commercial CLA-containing oil. Egg production was not significantly different among the dietary treatments at 0, 2, 4, and 6 week. The proportion of saturated fatty acids such as C16:0 and C18:0 in egg yolk were increased, but that of monounsaturated fatty acid C18:1 was decreased by feeding CLA-containing oil supplementation. However, the proportion of polyunsaturated fatty acids such as C18:2 and C18:3 in egg yolk were not different among dietary treatments at 2 and 4 wk of the experiment. At 6 week, the proportion of C18:2 in egg yolk was decreased by feeding CLA-containing oil compared with the control. Polyunsaturated fatty acid:saturated fatty acid (P:S) ratio and n-6:n-3 polyunsaturated fatty acid ratio were similar across the treatments between 2 and 4 week. The P:S ratio was decreased by dietary CLA-containing oil supplementation at 6 week. The proportion of CLA in egg yolk was linearly increased with increasing levels of CLA-containing oil supplementation. In conclusion, dietary supplementation of CLA-containing oil to laying hens increased beneficially increased CLA content in their egg yolk.

Effect of Calvarial Cell Inoculated Onto the Biodegradable Barrier Membrane on the Bone Regeneration (흡수성 차폐막에 접목된 두개관골세포의 골조직 재생에 미치는 영향)

  • Biodegradable barrier membrane has been demonstrated to have guided bone regeneration capacity on the animal study. The purpose of this study is to evaluate the effects of cultured calvarial cell inoculated on the biodegradable barrier membrane for the regeneration of the artificial bone defect. In this experiment 35 Sprague-Dawley male rats(mean BW 150gm) were used. 30 rats were divided into 3 groups. In group I, defects were covered periosteum without membrane. In group II, defects were repaired using biodegradable barrier membrane. In group III, the defects were repaired using biodegradable barrier membrane seeded with cultured calvarial cell. Every surgical procedure were performed under the general anesthesia by using with intravenous injection of Pentobarbital sodium(30mg/Kg). After anesthesia, 5 rats were sacrificed by decapitation to obtain the calvaria for bone cell culture. Calvarial cells were cultured with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. The number of cell inoculated on the membrane were $1{\times}10^6$ Cells/ml. The membrane were inserted on the artificial bone defect after 3 days of culture. A single 3-mm diameter full-thickness artificial calvarial defect was made in each animal by using with bone trephine drill. After the every surgical intervention of animal, all of the animals were sacrificed at 1, 2, 3 weeks after surgery by using of perfusion technique. For obtaining histological section, tissues were fixed in 2.5% Glutaraldehyde (0.1M cacodylate buffer, pH 7.2) and Karnovsky's fixative solution, and decalcified with 0.1M disodium ethylene diaminetetraacetate for 3 weeks. Tissue embeding was performed in paraffin and cut parallel to the surface of calvaria. Section in 7${\mu}m$ thickness of tissue was done and stained with Hematoxylin-Eosin. All the specimens were observed under the light microscopy. The following results were obtained. 1 . During the whole period of experiment, fibrous connective tissue was revealed at 1week after surgery which meant rapid soft tissue recovery. The healing rate of defected area into new bone formation of the test group was observed more rapid tendency than other two groups. 2 . The sequence of healing rate of bone defected area was as follows ; test group, positive control, negative control group. 3 . During the experiment, an osteoclastic cell around preexisted bone was not found. New bone formation was originated from the periphery of the remaing bone wall, and gradually extended into central portion of the bone defect. 4 . The biodegradable barrier membrane was observed favorable biocompatibility during this experimental period without any other noticeable foreign body reaction. And mineralization in the newly formed osteoid tissue revealed relatively more rapid than other group since early stage of the healing process. Conclusively, the cultured bone cell inoculated onto the biodegradable barrier membrane may have an important role of regeneration of artificial bone defects of alveolar bone. This study thus demonstrates a tissue-engineering the approach to the repair of bone defects, which may have clinical applications in clinical fields of the dentistry including periodontics.

Natural killer cell activity in mice infected with Acanthamoeba culbertsoni (Acanthamoeba culbertsoni 감염에 있어 자연살세포의 활성)

  • The natural killer cell activity of splenocytes and TBC, active NK cells, recycling capacity of natural killer cells were observed by means of both the 51Cr-release cytotoxicity assay and single cell cytotoxicity assay against YAC-1. CSH/HeJ mice were infected intranasally with $1{\times}10^4{\;}or{\;}1{\times}10^5$ trophosoites of pathogenic Acanthamoeba culbertsoni. The infected mice showed mortality rate of 34% in $1{\times}10^4$ group and 65% in 1{\times}10^5 group, and mean survival time was $16.40{\pm}3.50$ {\;}and{\;}3.20{\pm}4.09$ days respectively. The cytotoxic activity of natural killer cells of the 2 groups was significantly higher than that of non-infected mice from the 12th hour to the 2nd day after infection, showing the highest on the first day. On the l0th day after infection, the cytotoxic activity of natural killer cells was significantly suppressed as compared with that of the control. There was no significant difference in NK cell cytotoxicity between two infected groups. The targetbinding capacity and active NK cells of natural killer cells in $1{\times}10^5$ trophosoite infected mice was significantly increased on the 12th hour and the first day after infection as compared with the control group. Maximal recycling capacity (MRC) was not changed during the observation period. The present results indicated that the elevation of natural killer cell activity in the mice infected with A. culbertsoni was due to elevation of target.binding capacity and increased active NK cells of natural killer cells, and not due to the maximal recycling capacity of the individual NK cell, and there was no difference between two experimental dose groups.

  • Beans are acknowledged to be food resources, which have more abundant proteins and fats. The constituent parts of beans (i.e. aspartic aid, glycine, arginine) are effective against diabetes, and dietary fiber contained in the beans has an important property to maintain insulin sensitivity. Based on these, using streptozotocin (STZ)-induced diabetic rats, this study examined how the rat-eye soybean, which is principal products of the Imsil province, is effective to attenuate and/or prevent the development of diabetes mellitus. We divided rats into the non-diabetic and diabetic group, and diabetic group was further subdivided into six experimental groups [DC, diabetic control; DI, diabetes with insulin treatment (4∼6 IU/rat); DB, diabetes with black bean; DY, diabetes with yellow soybean; DS, diabetes with rat-eye soybean; DSS, diabetes with vinegar-fermented rat-eye soybean. All bean treatment (1.5 mg/l g body weight).]. Food efficiency ratio (FER), body weight and insulin sensitivity in diabetic rats were significantly reduced compared to those in normal control animals. These reductions were obviously attenuated by administration of a variety of beans used in this study (20∼30%), and the recovery effects were comparable to the results obtained by insulin treatment. Taken together, this study suggests that all beans used may have an essential property to improve and/or attenuate the development of diabetes mellitus in rats.

  • Forest area in Korea ocupies as much as 68 percent of the total land area, but forest production figure in the statistics is rather trifling: that is about 2 percent of Gross National Production (G.N.P.), on the average. In view of the primary industrial sector, its production only weighs no more than 5 percent of this whole sector. Forest production written above refers only to direct forest income of the whole forest income. For the primary forest products they are in many cases used as raw materials for other interrelated industries. The added value there-from, which arises from round about production Process, in other word, indirect income is of most singnificance. Nevertheless, until nowadays forest production has been merely refered to timber production i, e, direct production but indirect income has never been looked upon. In this regard, calculated indirect forest income by means of input ratio method. The material used were Leontiefls tables of two 1963 and 1966 fiscal years, surveyed and analysed by The Bank of Korea. Indirect forest income calculated were 42,688,200,000 won in 1963 and 74,789,800,000 won in 1966 compared direct forest income of 14,361,000,000 won in 1963 and 17,709,000,000 won in 1966. So far as indirect forest income is considered total forest production indices composed of direct and indirect forest income amount to 8.23% in 1963 and 10.12% in 1966 of Gross National Production. Invisuable forest income which originates from, what we cal, indirect benefit of forestry such as land conservation, flood and drought control, soil run off control, scenic beauty and many others is naturall, not included in the calculation. As already mentioned, primary forest products are, for the most part, utilized as raw materials for other industries, therefore indirect forest income is rather appreciable than direct forest income, contributing for the growth and development of other connected industries. In these points of view, forestry must not be evaluated trifling in deciding industrial importances.

Effects of Supplementary $Safmannan^(R)$(Beta-Glucan & MOS) and $World-Labs^(R)$ (Multiple Probiotics) on the Performance, Nutrient Availability Small Intestinal Microflora and Immune Response in Broiler Chicks (Beta-Glucan과 MOS의 복합제($Safmannan^(R)$)와 복합 생균제($World-Labs^(R)$)가 육계의 생산성과 영양소 이용율 소장내 미생물 균총 및 면역 체계에 미치는 영향)

  • In order to study the effects of supplementary $Safmannan^(R)$(Beta-glucan & MOS complex) and $World-Labs^(R)$ (multiple probiotics) on the performance, nutrient availability, small intestinal microflora and immune response in broiler chicks, one thousand hatched broilers ($Ross^(R)$ were assigned to 4 treatments: control(basal diet), $BMD^(R),\;Safmannan^(R)\;and\;World-Labs^(R)$. There were no significant differences in the performance and in serum IgG, ND titre. However parameters of leukocytes and erythrocytes were significantly different among treatments (p<0.05). Leukocytes and RBC of $World-Labs^(R)\;and\;Safmannan^(R)$ were mostly lower than $BMD^(R)$ and control whereby MCH and MCHC of $World-Labs^(R)\;and\;Safmannan^(R)$ were higher than other treatments. The cfu of intestinal microflora had no significant differences among treatments. The $BMD^(R)$ treatment was higher than others in amino acid and crude fat availability and $World-Labs^(R)$ was higher than others in crude fiber availability. It was concluded that supplements used in the present experiment did not significantly affect the production parameters. However, significant impact on blood parameters, especially on leucocytes, may need further investigation.

A survey of the serving temperature control practices in hospital dietetics -Comparison between centralized and decentralized tray assembly systems (병원급식의 적온관리 실태조사 -배선방법별 비교 연구-)

  • The serving temperature control practices were assessed in 20 general hospital's dietetics utilizing centralized or decentralized tray assembly systems. The results of the study were summarized as follows : 1) All of the surveyed hospitals were utilizing conventional foodservice system. The number of dietary employees per bed was very low when comparing with that in America. Working hours of employees per week were approximately two times greater than those in America. 2) When comparing two tray assembly systems, dietary labor hours and costs in centralized system were less than those in decentralized system. 3) When comparing serving temperature practices between two tray assembly systems, the temperatures of meals utilizing centralized tray service were significantly lower than those in decentralized system, and only the steamed rice served in decentralized system was within the acceptable temperature range. 4) There was no significant difference in sensory acceptance scores marked by patients served by two different types of tray assembly systems. The serving temperature was significantly correlated to the patient's overall acceptability.
