Four ruminally cannulated Holstein steers (BW $482.9{\pm}8.10kg$), fed low protein TMR (CP 11.7%) as a basal diet, were used to investigate changes in rumen fermentation and blood metabolism according to protein fraction, cornell net carbohydrates and protein system (CNCPS), and enriched feeds. The steers, arranged in a $4{\times}4$ Latin square design, consumed TMR only (control), TMR supplemented with rapeseed meal (AB1), soybean meal (B2), and perilla meal (B3C), respectively. The protein feeds were substituted for 23.0% of CP in TMR. Ruminal pH, ammonia-N, and volatile fatty acids (VFA) in rumen digesta, sampled through ruminal cannula at 1 h-interval after the morning feeding, were analyzed. For plasma metabolites analysis, blood was sampled via the jugular vein after the rumen digesta sampling. Different N fraction-enriched protein feeds did not affect (p > 0.05) mean ruminal pH except AB1 being numerically lower 1 - 3 h post-feeding than the other groups. Mean ammonia-N was statistically (p < 0.05) higher for AB1 than for the other groups, but VFA did not differ among the groups. Blood urea nitrogen was statistically (p < 0.05) higher for B2 than for the other groups, which was rather unclear due to relatively low ruminal ammonia-N. This indicates that additional studies on relationships between dietary N fractions and ruminant metabolism according to different levels of CP in a basal diet should be required.
Mugwort was successively fractionated with n-hexane, chloroform, ethyl acetate, n-butanol and water and the fractions were evaluated by their growth-promoting activites for Bifidobacterium sp. in vitro experiments. The growths of Bifidobacterium adolescentis, B. bifidum, B. infantis and B.longum were enhanced with the addition of the water fraction, while the fractions of chloroform and ethylacetate inhibited Clostridium perfringens. When the wate fraction was added to media at a concentration of 0.01-0.5%(w/v), the growhts of Bifidobacterium sp. were increased according to the concentration of water fraction used. The water fraction stimulated also the growth of lactobacillus acidophillus, whereas those of E. coli and Enterococcus faecalis were not affected. The growth-promoting activity of water fraction was stable at the range of pH 2 to pH 10 and kept in thermal treatment at 10$0^{\circ}C$ for 30 minutes.
A protein-polysaccharide fraction was prepared from the carpophores of Laetiporus sulphureus. This fraction suppressed growth of sarcoma 180 in A-strain mice when administered i. p. To investigate the mechanism of antitumor action of this fraction, plaque assay was conducted by administrating i. p. to the mise at a dose level of 50mg/kg for five days. Ten days later, the mice were immunized with 1 * 10$^{7}$ sheep red blooc cells. The number of hemolytic plaque forming cells was significantly greater than that of the control mice. Three monosaccharides and fifteen amino acids were identified in the protein-polysaccharide fraction.

A sopA protein (41K) encoded by plasmid pXX288 was observed in the cytoplasm, whereas a sopB protein (37K) encoded by plasmid pXX157 was observed in the membrane fraction. Most of the sopB protein was solubilized from the crude membrane by treatment with Sarkosyl, which suggested that the protein may be located in the inner membrane. The sopA protein was precipitated at the concentration of 30 to 60% ammonium sulfate. The sedimentation profile of the crude membrane fraction showed a little difference according to culture media used, and the sopB protein existed in all fractions of inner membrane. The DNA of plasmids, pXX157, pXX300, and pXX167 co-sedimented with inner membrane fraction.
Fungicidal activities of Cassia obtusifolia extracts and their active principles were tested against Botrytis cinerea, Erysiphe graminis, Phytophthora infestans, Puccinia recondita, Pyricularia grisea, and Rhizoctonia solani, and compared with synthetic fungicides and two dihydroxyanthraquinones. At 1 g/l, the chloroform fraction of C. obtusifolia extracts showed fungicidal activity against B. cinerea, E. graminis, P. infestans, and Py. grisea, and the ethyl acetate fraction showed fungicidal activity against E. graminis and P. infestans. Danthrone was chromatographically isolated from the chloroform fraction and showed fungicidal activity against B. cinerea, E. graminis, P. infestans, and Py. grisea with 68, 100, 78, and $91\%$ control values at 0.5 g/l, respectively. Specifically, alizarin and quinizarin inhibited E. graminis, P. infestans, and Py. Grisea, but did not inhibit the growth of P. recondita and R. solani. These results indicate at least one of the fungicidal actions of danthrone.
Yung Nok Lee (Dept. of Biology, Korea University) : Studies on the phosphate metabolism in Chlorella, with special reference to polyphosphate. Kor. J. Microbiol., Vol.2, No.1, p1-11 (1964). 1. Uniformly $^{32}P$-labeled Chlorella cells which were irradiated with Cobalt-60 gamma-rays of about 70, 000 $\gamma$ dose, were further grown in a standard "cold" medium ("hot".rarw."cold"), and some portions of the algae were taken out at the begining of, and at intervals during the culture, and subjected to analyze the contents of $^{32}P$- and total P in various fractions of the cell materials. Results obtained were compared with those of nonirradiated normal cells. 2. Amounts of phosphate in various fractions of the nonirradiated normal Chlorella cells were measured using uniformly $^{32}P$--labeled cells. Analysis of the $^{32}P$--labeled algal cells showed that the highest value in P-content was the fraction of RNA followed by those of lipid, polyphosphate "C" polyphosphate "B", DNA, nucleotidic labile phosphate compounds, polyphosphate "A" and protein. It was observed that content of total polyphosphates in a single Chlorella cell was almost equal to RNA-P content in the cell, and the amount of RNA-P was almost equal to ten times of DNA-P content. 3. When the $^{32}P$--labeled algae which were irradiated with gamma-rays were grown in a normal "cold" medium, phosphate contents in the fraction of DNA, nucleotidic labile phosphate compounds and protein decreased markedly, while the contents of phosphate in the fractions of polyphosphate "C" and potyphosphate "B" increased in comparison with those of unirradiated normal cells. So, it was considered that the pretreatment of above mentioned dose of gamma-ray inhibited DNA and protein synthesis from polyphosphate in Chlorella cells. 4. Proceeding the culture of $^{32}P$--labeled Chlorella in a "cold" standard medium, whose synthetic activity of DNA and protein from polyphosphate was disturded by gamma-ray irradiation, the amounts of $^{32}P$-in the fraction of polyphosphate "C" increased, in contrast with those of polyphosphate "B" fraction. Objectives: Dyslipidemia in diabetes mellitus is a significant risk factor for the development of cardiovascular complications. The aim of this study was to evaluate the effect of the ethyl-acetate fraction of an ethanolic extract from Streospermum suaveolens on lipid metabolism in streptozotocin (STZ)-induced diabetic rats. Methods: Diabetes was induced by intraperitonial injection of STZ (50 mg/kg). Diabetic rats were treated with an ethyl-acetate fraction orally at doses of 200 and 400 mg/kg daily for 14 days. On the $15^{th}$ day, serum lipid profiles, such as total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL), and high-density lipoprotein (HDL), were estimated in experimental rats. The atherogenic (AI) and the coronary risk (CRI) indices were also evaluated. Results: The ethyl-acetate fraction at doses of 200 and 400 mg/kg significantly (P < 0.001) and dose-dependently reduced serum cholesterol, triglycerides and LDL, but increased HDL towards near normal levels as compared to diabetic control rats. The fraction also significantly (P < 0.001) lowered the atherogenic index (AI) and coronary risk index (CAI) in a dose-dependent manner. Conclusion: The present study demonstrated that the ethyl-acetate fraction of Stereospermum suaveolens exhibits a potent antihyperlipidemic activity in hyperglycemic rats and suggests that the plant may have therapeutic value in treating the diabetic complication of hyperlipidemia.

This study was performed to identify the optimum fractionation method and conditions to obtain exopeptidase-active fractions from octopus hepatopancreas (HP) crude extracts (CEs) using four techniques: solid ammonium sulfate fractionation, polyethylene glycol (PEG) fractionation, anion exchange chromatography, and gel filtration chromatography. The fractions with the highest total activity toward L-leucine-p-nitroanilide (Leu-pNA) were fraction IV from the ammonium sulfate and PEG fractionation, and fraction II in ion exchange and gel filtration chromatography. The total exoprotease activity of these fractions was highest in fraction IV (4,050.20 U) of ammonium sulfate fractionation, followed by fraction II (3,600.28 U) from gel filtration chromatography, fraction IV (2,861.30 U) from PEG fractionation, and fraction II (2,576.28 U) from ion exchange chromatography. These results suggest that ammonium sulfate fractionation using 60-80% ammonium sulfate was the most efficient method for separating the exoprotease active fractions from CEs of octopus HP.

Five different plant cell wall degrading enzymes were tested for their ability to release p -hydroxybenzoic acid from carrot alcohol insoluble residue (AIR) and cellulose fraction. Phenolics of AIR from cell wall materi민 (CWM) in carrot were found to consist primarily of p-hydroxybenzoic acid (1,977 $\mu$g/g AIR) with minor contribution from vanillin (55.9 $\mu$g/g AIR), ferulic acid (13.6 $\mu$g/g AIR) and p-hydroxybenzaldehyde (10.6 $\mu$g/g AIR). The contents of ferulic acid in Driselase, Cellulase, Macerozyme R-200, Macerozyme R-10 and Sumyzyme MC were 2,319, 2,060, 391, 95.2, 34.1 $\mu$g/g, respectively. Incubation of Driselase with AIR released only 2.8$\%$ of the total 4 M NaOH extractable p-hydroxybenzoic acid. These results indicate that commercial five plant cell wall dograding enzymes can not release P-hydroxybenzoic acid from carrot AIR and cellulose fraction.
In the process of the incorporation of orthophsphate into protein and other cell constituents, the role of inorganic polyphosphate and RNA-polyphosphate complex and the correlation between them were pursued by analyzing the contents of $^{32}P$ and total P in various fractions of Chlorella cells, which had been uniformly labeled with $^{32}P$ before the inoculation in a normal "cold" medium or P-free medium during the culture. The effects of ionizing radiation and various micronutritional-element deficiencies on the phosphate incorporation into, and biosynthesis of, protein and other introgenus compounds in the cells were also observed. When the uniformly $^{32}P$-labeled algae were grown in a normal "cold" medium the contents of $^{32}$ P in the fractions of protein, DNA and RNA-polyphosphate complex increased, but those in the fraction of acid-insoluble polyphosphate decreased. On the other hand, amount of $^{32}P$in the fraction of RNA was almost unchanged in spite of rapid increase of the total P. In the growing period of $^{32}P$-labeled algae in a P-free medium, amounts of $^{32}P$ in the fractions of DNA, protein and lipid increased, while those in the fractions of RNA-polyphosphate and inorganic polyphosphates decreased. When the algal cells were irradiated with about 70, 000r of gamma-rays before the inoculation in the medium, amounts of phosphate in the fractions of DNA, RNA, nucleotides and protein decreased during the culture, compared with those of the control. However, the phosphate content in the fraction of acid-insoluble polyphosphate of the irradiated cells increased than those of the control. In the growing period of the algae in a Mo-free, medium, amounts of acid-soluble total phosphate and nucleotides of the cells increased, while the amounts of residual protein and RNA decresed compared with those of the normal cells. Amounts of alkali-labile protein and phospholipid of the cells grown in a B-free medium decreased, whereas amount of phosphate in acid-soluble fraction increased compared with the control. In general, the contents of protein and RNA in each microelement deficient cells decreased more or less, compared with those in the normal cells.in the normal cells.
