Strength Properties of Light-Weighted Soils Mixed with E.P.S (발포스티로폴을 이용한 경량혼합처리토의 강도특성)

  • The strength properties of Light-Weighted Soils(LWS) mixed with Expanded Polystrene(E.P.S) are studied. Test results show that when the initial water contents of dredged soils are under 135% and the cement contents are above 1%, Light-Weight Soils are in the appropriate strength range of 2.0 to 4.0kg/㎠. However, E.P.S contents had a little affects on the strength properties of LWS. In the view of E.P.S diagram's effects on LWS, which have expand ratio of 25, 35, 45 and 60, further studies considering of physical properties of original E.P.S beads are needed.

  • The purpose of this study is to expand businesses' adoption of the Enterprise Resource Planning System (E.R.P.S.). In order to accomplish the objective, the following conditions need to be fulfilled. 1) Clearness of the goal and expected effect of E.R.P.S. adoption 2) Definiteness of the extent of E.R.P.S. construction 3) The management's will to E.R.P.S. adoption If the E.R.P.S is adopted, the following effects are expected. 1) Speedy quality decision-making is possible. 2) Time taken from the order of the product to its release can be curtailed (from 40days to 10days). 3) Time taken for the development of the product can be shortened (from 24 months to 10 months). 4) Cost reduction is possible by using necessary information in real time.

  • The influence of porosity (P) on Young's modulus (E) and Poisson's ratio $(\upsilon)$ of sintered steels produced from four types of steel powders was investigated. The values of E and $(\upsilon)$ depend mainly on the value of P, and those were a little affected by alloying elements. The relationships between E, $(\upsilon)$, and P were described as following equations: $E\;=\;E_0{\cdot}(1\;-\;k_E{\cdot}P)^2$ and $\upsilon\;=\;({\upsilon}_0\;-\;\upsilon_{sub}){\cdot}(1\;-\;k_{\upsilon}{\cdot}P)2+\upsilon_{sub}$, where subscript 0 means P = 0, and $k_E,\;k_{\upsilon}$ and ${\upsilon}_{sub}$ are empirical constants. These approximate equations showed good agreement with empirical results.

Direct characterization of E2-dependent target specificity and processivity using an artificial p27-linker-E2 ubiquitination system

  • Little attention has been paid to the specificity between E2 and the target protein during ubiquitination, although RING-E3 induces a potential intra-molecular reaction by mediating the direct transfer of ubiquitin from E2 to the target protein. We have constructed artificial E2 fusion proteins in which a target protein (p27) is tethered to one of six E2s via a flexible linker. Interestingly, only three E2s (UbcH5b, hHR6b, and Cdc34) are able to ubiquitinate p27 via an intra-molecular reaction in this system. Although the first ubiquitination of p27 (p27-Ub) by Cdc34 is less efficient than that of UbcH5b and hHR6b, the additional ubiquitin attachment to p27-Ub by Cdc34 is highly efficient. The E2 core of Cdc34 provides specificity to p27, and the residues 184-196 are required for possessive ubiquitination by Cdc34. We demonstrate direct E2 specificity for p27 and also show that differential ubiquitin linkages can be dependent on E2 alone.

Characterization of plasmids of Zymomonas mobils and Construction of E. coli-Zymomonas shuttle Vector (Zymomonas mobilis플라스미드의 특성연구 및 E.coli-Zymomonas셔틀 벡터 제조)

  • We have characterized the plasmids of zymomonas, and constructed E. coli-Zymomonas shuttle vector. Plasmids have been detected in four strains of Zymomonas mobilis. All strains tested had at least one plasmid ranging in size from about 1.7 to 46kb. Antibiotics resistances of Z. mobilis were tested to select the host strain. All strains were very sensitive to tetracycline and chloramphenicol. Homology tests between the plasmids in four strains showed that the plasmids of ATCC10988 is highly homologous to those of ZM1, and that there is no homology between plasmids of ZM4 and Agll. The 1.7kb plasmid of ATCC10988, named as pZM886, also has no homology with plasmids of ZM4. A hybrid plasmid, designated to pBZ41, was constructed from pZM886 and pBR322. A restriction map of pBZ41 was established. Replicon of pZM886 didn't operate in E.coli and pBR322 seemed not to replicate in Zymomonas. pBZ41 was transfered from E. coli to Zymomonas by conjugal mobilization. The transconjugants were resistant to tetracycline and maintained pBZ41 stably.

Hepatitis C Virus Core Protein Activates p53 to Inhibit E6-associated Protein Expression via Promoter Hypermethylation (C형 간염바이러스 코어 단백질에 의한 p53 활성화와 프로모터 과메틸화를 통한 E6AP 발현 억제)

  • The E6-associated protein (E6AP) is known to induce the ubiquitination and proteasomal degradation of HCV core protein and thereby directly impair capsid assembly, resulting in a decline in HCV replication. To counteract this anti-viral host defense system, HCV core protein has evolved a strategy to inhibit E6AP expression via DNA methylation. In the present study, we further explored the mechanism by which HCV core protein inhibits E6AP expression. HCV core protein upregulated both the protein levels and enzyme activities of DNA methyltransferase 1 (DNMT1), DNMT3a, and DNMT3b to inhibit E6AP expression via promoter hypermethylation in HepG2 cells but not in Hep3B cells, which do not express p53. Interestingly, p53 overexpression alone in Hep3B cells was sufficient to activate DNMTs in the absence of HCV core protein and thereby inhibit E6AP expression via promoter hypermethylation. In addition, upregulation of p53 was absolutely required for the HCV core protein to inhibit E6AP expression via promoter hypermethylation, as evidenced by both p53 knockdown and ectopic expression experiments. Accordingly, levels of the ubiquitinated forms of HCV core protein were lower in HepG2 cells than in Hep3B cells. Based on these observations, we conclude that HCV core protein evades ubiquitin-dependent proteasomal degradation in a p53-dependent manner.

Antimicrobial Activity of Flos Syzygii Aromatici Extracts against Mice Infected with Escherichia coli O157:H7 (Escherichia coli O157:H7에 감염된 마우스에 대한 정향 추출물의 항균효과)

  • The present study evaluated the antibacterial effect of Flos syzygii Aromatici methanolic extracts (FSAE). In addition, the effectiveness of FSAE against Escherichia coli O157:H7 infection was studied using ICR female mice. At 24 h after incubation of E. coli O157:H7, FSAE at the concentration of 0.269 (p < 0.05), 0.538 (p < 0.001) and 1.075 mg/mL (p < 0.001) significantly inhibited the growth of E. coli O157:H7 compared to the control group. After single challenge with E. coli O157:H7, forty female ICR mice were divided into four experimental groups which were orally administered with saline (control), 0.538 (group 1), 1.075 (group 2) and 2.15 mg/mL (group 3) of FSAE, respectively. On the 3rd day, the number of fecal E. coli O157:H7 in group 2 (p < 0.05) and group 3 (p < 0.01) was significantly decreased compared to that in the control group. On the 7th day post-treatment, the number of fecal E. coli O157:H7 in all FSAE-treated groups was significantly decreased compared to that in the control group (group 1, p < 0.05; group 2 and 3, p < 0.001). According to the results of the present study, administration of FSAE to mice can reduce the severity of E. coli O157:H7 infection. Therefore, the current study suggests that FSAE could be a good candidate for the treatment of enteric infections in domestic animals.

  • In the polythylene film mulching(P.E. mulching) culture, soil temperature ranked in the order of clear P.E.-, black P.E.-, and non-mulching. The difference in temperature between P.E mulching and non-mulching conditions was greatest in maximum temperature in fine day. Under the dry season, soil water content ranked in the order of black P.E.-, clear P.E.- and non-mulched soil. Under the rainy season, however, the content in non-mulched soil was higher than in the mulched soils, while there was little difference between the two colored films. In three phases of soil, liquid phase ratio was higher and gaseous phase ratio was lower in mulched soil than in non-mulched soil under the dry season. However, the opposite result was observed under the rainy season. The content of soil organic matter in red pepper field applied with the compost and mixed-fertilizer ranked in the order of black P.E.-, clear P.E.- and non-mulching conditions. However, the content between mulching and non-mulching differed little in peanut field applied with mixed-fertilizer. In red pepper field, soil nitrogen content in mulching conditions slightly differed from that in non-mulching conditions during the dry season. The soil nitrogen content decreased rapidly 86 days after fertilizer application during the rainy season. In peanut field, there was little difference in the content between the two conditions. The nitrogen content in the leaves of red pepper and peanut was much higher in P.E mulching than in non-mulching.
