닥나무 추출물의 항산화 활성 및 성분 분석 (Antioxidative Activity and Component Analysis of Broussonetia kazinoki SIEB Extracts)

  • 본 연구에서는 닥나무 추출물의 항산화 활성과 성분 분석에 관한 연구를 실시하였다. 닥나무 추출물은 우수한 free radical(1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}=8.53{\mu}g/mL$)을 나타내었다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}-EDTA/H_2O_2$계에서 생성된 활성 산소종(reactive oxygen species, ROS)에 대한 에틸아세테이트 분획의 총항산화능(OSC50)은 $1.69 {\mu}g/mL$이었다. 닥나무 추출물에 대하여 $^1O_2$으로 유도된 사람 적혈구의 광용혈 실험 결과 에틸아세테이트분획은 $10{\mu}g/mL$의 낮은 농도에서 ${\tau}_{50}$이 183.3 min으로 대표적인 항산화 물질로 알려진 $\alpha$-tocopherol (${\tau}_{50}$ = 38.00 min)보다 매우 큰 세포보호 효과를 나타내었다. TLC, HPLC, LC/ESI-MS/MS 및 $^1H$-NMR을 이용하여 닥나무 추출물 에틸아세테이트 분획의 성분 분석을 수행하였다. 그 결과, 닥나무 추출물 에틸아세테이트 분획은 kazinol류의 kazinol J와 플라보노이드류인 luteolin을 함유하고 있음을 확인하였다. 이상의 결과들은 닥나무 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거함으로써, 또는 이에 대항하여 세포막을 보호함으로써, 생체계 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리키며 기능성 화장품 원료로서 응용 가능성이 있음을 시사한다.

들깨 잎 추출물의 Nitric Oxide Synthase 저해활성 및 Peroxynitrite 소거활성 (Inhibitory Activity of Nitric Oxide Synthase and Peroxynitrite Scavenging Activity of Extracts of Perilla frutescens)

  • Activated microglia by neuronal injury or inflammatory stimulation overproduce nitric oxide (NO) by inducible nitric oxide synthase (iNOS) and reactive oxygen species (ROS) such as superoxide anion, resulting in neurodegenerative diseases. The toxic peroxynitrite (ONOO$^-$), the reaction product of NO and superoxide anion further contributes to oxidative neurotoxicity. We tried to evaluate the effects of two kinds of varieties of Perilla frutescens var japnica Hara on the NO production in lipopolysaccharide (LPS)-activated microglia. The perilla cultivars of Namcheondeulkkae (NC) and Boradeulkkae (BR) were developed by pure line from the local variety and by a cross between 'deulkkae' and 'chajogi', respectively. Spirit, hexane, chloroform and butanol fractions of the leaves of NC and BR inhibited the production of NO in LPS-activated microglia. The fractions of BR showed stronger activity than NC and the spirit extracts was the most potent in both cultivars. The solvent fractions of BR suppressed the expression of protein and mRNA of iNOS in LPS-activated microglial cells. Moreover, the extracts of NC and BR showed the activity of peroxynitrite scavenging in cell free bioassay system. These results imply that Namcheondeulkkae and Boradeulkkae might have neuroprotective activity through the inhibition of NO production by activated microglial cells and peroxynitrite scavenging activity.

Euryale ferox Salisbury의 항산화효과 및 LDL 산화 억제효과 탐색 (Screening of Antioxidative Effect and Suppressive Effect of LDL Oxidation of Euryale ferox Salisbury)

  • Topical natural antioxidants are a useful strategy for the prevention of oxidative stress mediated cardiovascular disease including atherosclerosis. From the viewpoint of this underlying principle, the screening of natural plant extracts with scavenging activity for pro-oxidant reactive species is a primary requirement for the development of new topical antioxidant formulations. Euryale ferox Salisbury (EF) is botanical name and it's pharmaceutical name is EURYALES SEMEN (ES). The stems and branchs of EF have been used as a traditional herbal medicine for the treatment of dysentery, diarrhea, leucorrhoea, incontinence and paralysis of joint. In this study, the antioxidant activity of extract from EF was studied in vitro methods by measuring the antioxidant activity and free radical scavenging activity by TEAC and DPPH, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation. The EF extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation. In conclusion, the EF extracts have antioxidative effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and chronic degenerative disease such as atherosclerosis.

Protective effect of Cordyceps militaris against hydrogen peroxide-induced oxidative stress in vitro

  • BACKGROUND/OBJECTIVES: Excessive production of reactive oxygen species (ROS) such as hydroxyl (${\cdot}OH$), nitric oxide (NO), and hydrogen peroxide ($H_2O_2$) is reported to induce oxidative stress. ROS generated by oxidative stress can potentially damage glial cells in the nervous system. Cordyceps militaris (CM), a kind of natural herb widely found in East Asia. In this study, we investigated the free radical scavenging activity of the CM extract and its neuroprotective effects in $H_2O_2$-induced C6 glial cells. MATERIALS/METHODS: The ethanol extract of CM ($100-1,000{\mu}g/mL$) was used to measure DPPH, ${\cdot}OH$, and NO radical scavenging activities. In addition, hydrogen peroxide ($H_2O_2$)-induced C6 glial cells were treated with CM at $0.5-2.5{\mu}g/mL$ for measurement of cell viability, ROS production, and protein expression resulting from oxidative stress. RESULTS: The CM extract showed high scavenging activities against DPPH, ${\cdot}OH$, and NO radicals at concentration of $1,000{\mu}g/mL$. Treatment of CM with $H_2O_2$-induced oxidative stress in C6 glial cells significantly increased cell viability, and decreased ROS production. Cyclooxygenase-2 and inducible nitric oxide synthase protein expression was down-regulated in CM-treated groups. In addition, the protein expression level of phospho-p38 mitogen-activated protein kinase (p-p38 MAPK), phospho-c-Jun N-terminal kinase (p-JNK), and phospho-extracellular regulated protein kinases (p-ERK) in $H_2O_2$-induced C6 glial cells was down-regulated upon CM administration. CONCLUSION: CM exhibited radical scavenging activity and protective effect against $H_2O_2$ as indicated by the increased cell viability, decreased ROS production, down-regulation of inflammation-related proteins as well as p-p38, p-JNK, and p-ERK protein levels. Therefore, we suggest that CM could play the protective role from oxidative stress in glial cells.

함초(나문재 및 퉁퉁마디) 추출물의 항산화 작용에 관한 연구(I) (Antioxidative Properties of Extract/Fractions of Suaeda asparagoides and Salicornia herbacea Extracts(I))

  • 본 연구에서는 함초 추출물(나문재 및 퉁퉁마디)의 항산화능을 조사하였다. 나문재 추출물의 free radical(1,1-diphenyl-2-picrylhyazyl, DPPH) 소거활성($FSC_{50}$)은 100 % ethanol 추출물(329.33 ${\mu}g/mL$) < 50% ethanol 추출물(40.73) < ethylacetate 분획(13.87) < ethylacetate 분획에서 당을 제거시킨 aglycone 분획(7.80) 순으로 증가하였다. 퉁퉁마디의 경우, free radical 소거활성은 ethylacetate 분획 및 aglycone 분획이 각각 23.21 및 28.50 ${\mu}g/mL$이었다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}-EDTA/H_2O_2$ 계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 나문재 추출물의 총항산화능은 50 % ethanol 추출물($OSC_{50}$, $0.99{\mu}g/mL$) < ethylacetate 분획(0.05) < aglycone 분획(0.03)순으로, aglycone 분획에서 가장 큰 활성을 나타내었다. 퉁퉁마디의 경우, ethylacetate 분획 및 당을 제거시킨 aglycone 분획의 ROS 소거활성은 각각 0.10 및 0.20 ${\mu}g/mL$이었다. 나문재 및 퉁퉁마디 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 억제 효과를 측정하였다. 100 % ethanol 나문재 추출물의 경우 농도 의존적($1{\sim}100{\mu}g/mL$)으로 광용혈을 억제하였다. 특히 ethylacetate 분획에서 당을 제거시킨 aglycone 분획은 50${\mu}g/mL$ 농도에서 $\tau_{50}$이 310 min으로 매우 큰 세포보호 효과를 나타내었다. 퉁퉁마디의 경우는 ethylacetate 분획에서 비교적 큰 세포보호 활성을 나타내었다. 이상의 결과들은 나문재 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거함으로써 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리킨다.

Effect of Donganme (Sorghum bicolor L. Moench) against oxidative stress in vitro and in a cellular system in glial cells

  • In this study, we investigated the protective effects of 'Donganme' (Sorghum bicolor L. Moench) against oxidative stress under in vitro conditions and in a cellular system using C6 glial cells. The radical scavenging activities were observed using the substrates 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl (•OH) radicals. The Donganme extract had an •OH radical scavenging activity of 82.66% at a concentration of 100 ㎍·mL-1. Additionally, when DPPH was used as the substrate, the Donganme extract exhibited a strong radical scavenging activity in a concentration-dependent manner with an IC50 value of 28.56 ㎍·mL-1. Furthermore, treating C6 glial cells with hydrogen peroxide (H2O2) reduced the cell viability and generated reactive of oxygen species (ROS) and lactate dehydrogenase (LDH) compared to the normal levels, indicating that H2O2 induced oxidative stress. However, Donganme extracts increased the cell viability and inhibited ROS and LDH production against oxidative stress by H2O2 in the C6 glial cells. In particular, it showed effective cell protection with the cell viability, ROS production, and LDH release at 83.50, 88.06, and 14.87%, respectively, which were lower than the control or similar to the normal levels even at a low concentration of 100 ㎍·mL-1. The present study suggests that the Donganme extract was effective in protecting against oxidative stress in C6 glial cells through its antioxidant activity. Thus, Donganme could be a promising therapeutic agent for neurodegenerative diseases due to oxidative stress.

식물성 유지 및 수중유적형 유화계에서 육두구 종자 에탄올 추출물의 항산화활성 평가 (The antioxidant ability of nutmeg ethanolic extract in bulk oil and oil-in-water emulsion matrices)

  • 본 연구는 육두구 종자가 천연 산화방지제로서 효과적으로 에멀젼 제조 시 안정성을 가지는지를 확인하고자, 80% 에탄올로 추출한 육두구 종자(NM80)의 항산화 활성과 유지 산화안정성을 in vitro system과 실제 유지 및 유화액 산화 시스템에서 평가하고자 하였다. 결과적으로 DPPH 및 ABTS 양이온 라디칼 소거 활성과 ORAC가 평가에서 NM80은 농도 의존적으로 항산화 활성이 증가하였으며, TPC, TFC, FRAP 환원력은 각각 33.74 μmol tannic acid equivalent/g extract, 0.13 μmol quercetin equivalent/g extract, 295.27 μmol ascorbic acid equivalent/g extract로 관찰되었다. 또한, 유지 산화안정성을 측정하기 위하여 옥수수 기름을 180℃에서 90분 산화시켰을 때 NM80 200 ppm 첨가 시 일차산화물인 CDA가가 0.69%로 나타나 대조군보다 3.26% 감소하였으며, 동일한 조건에서 이차산화물인 ρ-AV와 TBA는 대조군보다 각각 16.94, 17.34% 낮았다. 또한, 유화액을 제조하여 NM80 200 ppm 첨가 후 60℃로 4일 동안 산화하였더니, 헤드스페이스 산소 함량은 20.44%로 대조군보다 산소 소비율이 6.29% 감소하였으며, CDA의 양은 대조군보다 82.85% 낮았다. 이러한 항산화 활성과 유지 산화 안정성에서 우수했던 NM80은 향기 성분으로 알릴 페녹시아세테이트, 유제놀 아세테이트, 유제놀 등이 검출되었다. 이와 같은 결과로 항산화 활성과 유지 및 유화액의 산화 안정성의 효과가 있는 NM80을 식품가공 시 첨가한다면 천연 항산화제로서 에멀젼의 품질 향상에 기여할 것으로 판단된다.

갈근(葛根) 추출물이 항산화에 미치는 영향 (Effects of Puerariae Radix extract on the activity of antioxidant)

  • Objective The objective of this study was to investigate the antioxidative effects of Puerariae Radix extract. Method Total antioxidant capacity (TAC), Total antioxidant response (TAR), Total phenolic content, Reactive oxygen species (ROS), 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities, lipid peroxidation were examined. Result Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. TAC and TAR of Puerariae Radix extract at the concentration of 5 mg/ml were 2.02 and 1.50 mM Trolox equivalents, respectively. Total phenolic content of Puerariae Radix extract at the concentration of 5 mg/ml was 2.29 mM gallic acid equivalent. Concentration of Puerariae Radix extract at which DPPH radical scavenging activity was inhibited by 50% was 5.91 mg/ml as compared to 100% by pyrogallol solution as a reference. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeSO4/ascorbic acid. Puerariae Radix extract at the concentration of 1 mg/ml slightly but significantly decreased TBARS concentration. The extract further prevented lipid peroxidation in a dose-dependent manner. The effect of Puerariae Radix extract on reactive oxygen species (ROS) generation was examined using cell-free system induced by hydrogen peroxide/FeSO4. Addition of 1 mg/ml of Puerariae Radix extract significantly reduced dichloroflurescein (DCF) fluorescence. The extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the extract significantly prevented ROS generation in vitro. Thus antioxidant effects of Puerariae Radix extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation, followed by inhibition of lipid peroxidation. Conclusion As a result, Puerariae Radix seems to have antioxitative effect and antioxidant compount.

가감온비탕합오산(加減溫脾湯合五散) 완제(完製)(HWW$^{(R)}$)의 항산화 효과에 대한 연구 (Antioxidative Activities of Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro)

  • Objectives: The objective of this study was to investigate the antioxidant effects of manufactured Wen-pi-tang-Hab-Wu-ling-san (WHW$^{(R)}$) in vitro. Methods: WHW$^{(R)}$ was prepared by the pilot manufacture of WHW water extract from a GMP system appointed company. Antioxidative activities were determined by in vitro tests as follows: the scavenging activities of oxygen free radicals including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide anion, hydrogen peroxide and nitric oxide radicals, as well as ferrous ion chelating capacity and Trolox equivalent antioxidant capacity (TEAC). Results: WHW$^{(R)}$ significantly scavenged oxygen free radicals such as DPPH (IC$_{50}$=115.28 $\pm$ 0.25 $\mu$g/$m\ell$), superoxide anion (IC$_{50}$=8.56 $\pm$ 0.08 $\mu$g/$m\ell$), hydrogen peroxide (IC$_{50}$=240.36 $\pm$ 3.41 $\mu$g/$m\ell$) and nitric oxide (IC$_{50}$=162.28 $\pm$ 0.21 $\mu$g/$m\ell$) radicals. WHW$^{(R)}$ also showed ferrous ion chelating activity (IC$_{50}$=543.19 $\pm$ 4.85 $\mu$g/$m\ell$) and Trolox equivalent effects (IC$_{50}$=45.311 $\mu$g/$m\ell$) in TEAC and ORAC assay, respectively. Conclusion: This study demonstrates that WHW$^{(R)}$ has strong antioxidative properties through free radical scavenging activity. These data suggest that WHW$^{(R)}$ be used as an antioxidant agent.

Paraquat 저항성 망초의 protective 효소 (Protective Enzymes of Paraquat-Resistant Conyza bonariensis)

  • 망초(Conyza bonariensis)에서 제초제 paraquat 저항성을 구명하기 위해 paraquat가 생성하는 superoxide 라디칼과 과산화수소 등의 유해 산소 물질을 제거하는 데에 관련된 효소의 활성을 저항성 종과 감수성 종의 망초에서 측정하였다. 경작지 부근에 자라는 망초는 비경작지에 자라는 망초에 비해 paraquat 저항성이 강하였다. 국내에서 paraquat를 자주 살포하는 지역에서 저항성 종의 망초가 출현하고 있음을 처음으로 보고한다. 저항성 종의 superoxide dismutase의 활성, ascorbate peroxidase의 활성, 그리고 glutathione reductase의 활성은 감수성의 그것에 비해 각각 약 20%, 44%, 그리고 64% 높게 나타났다. 이러한 결과로부터 paraquat에 대한 망초의 저항성은 superoxide dismutase, ascorbate peroxidase, 그리고 glutathione reductase 등으로 구성된 효소의 유해 산소 물질을 제거하는 효율성에 부분적으로 달려 있을 수 있다고 사료된다.
