Quantitative Structure-Activity Relationships for Radical Scavenging Activities of Flavonoid Compounds by GA-MLR Technique

  • The quantitative structure-activity relationship (QSAR) of a set of 35 flavonoid compounds presenting antioxidant activity was established by means of Genetic Algorithm-Multiple Linear Regression (GA-MLR) technique. Four-parametric models for two sets of data, the 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity $(R^2=0.788,\;Q^2_{cv}=0.699\;and\;Q^2_{ext}=0.577)$ and scavenging activity of reactive oxgen species (ROS) induced by $H_2O_2 (R^=0.829,\;Q^2_{cv}=0.754\;and\;Q^2_{ext}=0.573)$ were obtained with low external predictive ability on a mass basis, respectively. Each model gave some different mechanistic aspects of the flavonoid compounds tested in terms of the radical scavenging activity. Topological charge, H-bonding complex and deprotonation processes were likely to be involved in the radical scavenging activity.

Schisandra Chinensis Inhibits Oxidative DNA Damage and Lipid Peroxidation Via Antioxidant Activity

  • Schisandra chinensis have been traditionally used in Asia for the treatment of dyspnea, cough, mouth dryness, spontaneous diaphoresis, nocturnal diaphoresis, nocturnal emission, dysentery, insomnia and amnesia. The purpose of this study is to evaluate the protective effects of Schisandra chinensis on oxidative DNA damage and lipid peroxidation induced by ROS in non cellular and cellular system. DPPH radical, hydroxyl radical and hydrogen peroxide scavenging assay were used to measure the antioxidant activities. Phi X-174RF I plasmid DNA cleavage assay and intracellular DNA migration assay were used to evaluate the protective effect on oxidative DNA damage. MTT assay and lipid peroxidation assay were used for evaluating the protective effect on oxidative cell damage. It was found to scavenge DPPH radical, hydrogen peroxide and hydroxyl radical and it inhibited oxidative DNA damage, lipid peroxidation and cell death induced by hydroxyl radical. These data indicate that Schisandra chinensis possesses a spectrum of antioxidant and DNA-protective properties

Quantitative Determination of Acetone formed in the Thermal and Photochemical Decompositions of Azobisisobutyronitrile

  • Cyanoisopropyl radical derived from azobisisobutyronitrile (AIBN) by either thermolysis or photolysis reacts with oxygen to give cyanoisopropylperoxy radical which then was converted to acetone and cyano radical and/or acetyl cyanide and methyl radical. Of these products, acetone formed was quantitatively determined by the addition of thianthrene cation radical perchlorate to the reaction mixture. The results showed that 55.7 mmol, 16.9 mmol, and 16.0 mmol of acetone were formed for 7 hours from 1 mol of AIBN at $82{\pm}1^{\circ}C$ in acetonitrile, carbon tetrachloride, and benzene, respectively. However, 22.2 mmol of acetone was formed from photolysis of 1 mmol of AIBN in acetonitrile. The value decreased to 13.2 mmol by bubbling argon into the solvent prior to photolysis.

유산소운동능력의 차이가 로윙에르고미터 최대하 운동시 혈중 활성산소에 미치는 영향 (The Effects of Aerobic Exercise Capacity on Free Oxygen Radical in Blood during Submaximal Exercise in Rowing Ergometer)

  • 본 연구는 유산소운동능력의 차이를 보이는 두 집단이 각각 로윙에르고미터 최대하 운동을 실시한 후 혈중 활성산소의 변화에 어떠한 차이를 나타내는지 구명하는데 목적이 있으며 연구대상으로는 조정 선수집단 6명과 비선수집단 6명을 대상으로 최대유산소운동능력을 검사한 후 각각의 목표심박수 85~90%를 적용하여 로윙에르고미터 운동을 실시하였고 운동 전, 직후, 10분, 20분, 차30분에 각각 채혈하여 활성산소의 변화를 분석하였다. 그 결과는 유산소운동능력의 차이에 따른 활성산소의 변화에서 집단과 시기 간 상호작용 효과는 유의한(p<.05) 차이를 나타내어 시기에 따른 변화의 양상이 집단 간 차이가 있었고 사후검증 결과 회복기 10분에서 20분 그리고 20분에서 30분 구간에서 유의한 차이가 나타났다. 결론적으로 인체에 부정적인 영향을 미치는 활성산소로부터 유산소운동능력이 우수한 조정선수집단이 비선수집단에 비해 긍정적인 회복 패턴을 보였다.

Mechanism of Photodynamic Therapy using 9-hydroxypheophorbide-alpha on HeLa Cell Lines

  • Photodynamic therapy(PDT) is a treatment utilizing the generation of singlet oxygen and other reactive oxygen species(ROS), which selectively accumulate in target cells. The aim of present work is to investigate the photodynamic therapy mechanism of 9-HpbD-a-mediated PDT in HeLa cell lines. We studied the general reactive oxygen species(G-ROS) activation after 9-HpbD-a PDT using fluorescence stain with $H_2DCF-DA$. G-ROS activation observed after 9-HpbD-a PDT and higher activation condition was 1 hour after PDT at 0.5 ${\mu}g/ml$ 9-HpbD-a concentration. Sodium azide and reduced glutathione(the singlet oxygen quencher) could protect HeLa cells from cell death induced by 9-HpbD-a PDT. But D-mannitol(the hydroxyl radical scavenger) could not protect cell death. Singlet oxygen played a decisive role in 9-HpbD-a PDT induced HeLa cell death. Type II reaction was the main type of ROS formation at 9-HpbD-a PDT.

  • This study was undertaken to examine the effect of Cheongahwan(CAH), being known to reinforce Kidney-yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the kidney tissues. Alterations in enzyme activities were observed after in vivo treatment in rats. CAH caused a significant increase in the activities of superoxide dismutase (SOD), glutathione peroxidase and glutathione S-transferase. But catalase activity was not significantly altered by CAH. Treatment in vitro of CAH decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that CAH stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of CAH may contribute to prevent the oxygen free radical-induced impairment of cell function.

산소부화공기가 난류 확산 평면화염의 연소에 미치는 영향 (Effect of Oxygen Enriched Air on the Combustion of a Turbulent Diffusion Flat Flame)

  • Combustion using oxygen enriched air is an energy saving technology that can increase thermal efficiency by the improvement of burning rate and by the high temperature flame. Flame figures, OH radical intensities, temperature distributions and emission concentrations were measured according to oxygen enriched concentration and swirl number in a turbulent diffusion flat flame. It appeared that flame figure became flat and NO concentration decreased with increase of swirl number, and that the flame temperature increased high with increase of oxygen enriched concentration. In particular, it was most significant between oxygen concentration $40{\sim}60%$.

The Activity of Xanthine Oxidase (Type O) in Some Partial Portions of Rat Skin

  • To evaluate the physiological significance of xanthine oxidase (XO) in rat skin, the activity of XO (type O) in skin was compared with that of small intestine or liver. Concomitantly, XO activities in some partial portions (scalp, leg, dorsal and ventral part) of skin were determined and then compared with each partial portion. XO activity of skin was lower than that of small intestine and rather higher than that of liver. Furthermore, the activity of XO in skin, after clipping of hairs and then in 5 days, was more increased than that of rat which was clipped before having been sacrificed. As for activities of free radical scavenging system (GPx, GST, SOD), skin is lower than liver and small intestine. Although it is hewn that the oxygen free radical generated by XO system lead to injurious effect on the cell, the XO activity of ventral part which is to be less exposed to xenobiotics and biological agents was the lowest among those of ventral, dorsal, leg and scalp parts in skin. In conclusion, it may be hypothesized that XO system in skin act on defence mechanism.

택사 메탄올 추출물과 주성분의 항산화작용 (Antioxidant Effect of Alisma plantago-aquatica var. orientale and Its Main Component)

  • Reactive oxygen species(ROS) or free radicals are produced in the pathogenesis of human diseases including atherosclerosis, diabetes, cancer, and aging. Antioxidants are associated with the prevention of ROS-induced tissue and cellular damage in the various diseases. This study investigated the antioxidative activities of the methanol extract of Alisma plantagoaquatica var. orientale and its main component under conditions of radical generation using allophycocyanin and ferric-thiocyanate assay. Alisol B 23-acetate as a main component was isolated from the methanol extract of Alisma plantago-aquatica var. orientale. In results, the extract of Alisma plantago-aquatica var. orientale showed inhibitory activity on AAPH [2,2'-azobis(2-amidinopropane)dihydrochloride]-induced protein oxidation. Also, the extract of Alisma plantago-aquatica var. orientale and alisol B 23-acetate inhibited lipid peroxidation. These results indicate that Alisma plantago-aquatica var. orientale and alisol B 23-acetate show promise as therapeutic agents for various damages involving free radical reactions.

Protective Activities of Fractions of Water Extract Obtained from Artemisia iwayomogi Kitamura against Oxidative Stress-induced Mutagenesity: Correlation with Their Reactive Oxygen Scavenging Activity

  • Water extracts of injinssuk (Artemisia iwayomogi Kitamura) (WE) were obtained from the dried and ground leaves and stems of injinssuk. The WE was further fractionated into crude polysaccharide (C-PS) and nonpolysaccharide fractions (N-PS). The protective activities against the tert-butyl hydro peroxide induced mutangenecity on Escherichia coli PQ37 and reactive oxygen species scavenging activity of the WE, C-PS, and N-PS were studied. The WE obtained from leaves showed a significantly higher inhibitory effect on the mutagenicity than WE from stem. The WE obtained from the leaves having higher crude polysaccharide content but lower content of total carbohydrates had significantly higher antimutagenicity than that from the leaves with lower crude polysaccharide but higher total carbohydrate contents. Further study showed that C-PS fraction showed markedly stronger antimutagenic effect than N-PS. C-PS was also more effective than N-PS for hydroxyl radical scavenging activity, but was similar to N-PS in superoxide radical scavenging activity.
