Antioxidaitve and Differentiation Effects of Artemisia capillaris T. Extract on Hydrogen Peroxide-induced Oxidative Damage of MC3T3-E1 Osteoblast Cells (사철쑥(Artemisia capillaris T.) 추출물의 항산화 활성 및 H2O2로 산화적 스트레스를 유도한 조골세포의 활성과 분화에 미치는 영향)

  • In this study, the antioxidative activity of Artemisia capillaris T. extract on the proliferation and differentiation of MC3T3-E1 cells under $H_2O_2$-induced oxidative stress was investigated in order to determine its protective effect against oxidative stress as well as its availability as an antioxidant material related to treatment of bone diseases. As a result, the total polyphenol content of A. capillaris extract was 90.10 mg/g, whereas the flavonoid content was 4.45 mg/g. A. capillaris extract increased proliferation of MC3T3-E1 cells under $H_2O_2$-induced oxidative stress, and also increased the proliferation of differentiated osteoblast cells under oxidative stress. In addition, two differentiation markers, alkaline phosphatase activity and mineralization level, in A. capillaris extract tended to increase. These results indicate that A. capillaris extract suppresses the damage to osteoblasts caused by oxidative stress, which demonstrates its availability as an antioxidant material for preventing bone diseases.

Effects of Fermentation on the Metabolic Activities of Pine Needle Juice (발효과정이 솔잎 착즙액의 항산화, alpha-Glucosidase 및 Angiotensin Converting Enzyme 저해 활성에 미치는 영향)

  • The objective of this study was to compare the content and metabolic activities between fresh pine needle juice (PNJ) and fermented pine needle juice (FPNJ). A variety of factors were measured, including total phenolic content (TPC), antioxidant activity [DPPH radical scavenging activity (RSA), total radical-trapping antioxidant potential (TRAP), oxygen radical absorbance capacity (ORAC), cellular antioxidant capacity (CAC)], anti-genotoxic activity, ${\alpha}$-glucosidase inhibitory activity, and angiotensin converting enzyme (ACE) inhibitory activity. The TPC was $17.3{\pm}0.2$ and $4.6{\pm}0.0$ mg GAE/g in PNJ and FPNJ, respectively. The DPPH RSA, TRAP, and ORAC values increased in a dose-dependent manner for both PNJ and FPNJ, with significantly higher activities in PNJ than FPNJ. The CAC against AAPH-induced oxidative stress in HepG2 cells was protected by both PNJ and FPNJ. Pretreatment with PNJ and FPNJ in human leukocytes produced significant reductions in $H_2O_2$-induced DNA damage at a concentration of $50{\mu}g/mL$. ${\alpha}$-Glucosidase inhibitory activity was significantly higher in FPNJ than PNJ. The ACE inhibitory activity was about 87.1% and 60.0% in 1:1 diluted PNJ and FPNJ, respectively. This study suggests that the fermentation of PNJ could enhance the regulation of blood glucose metabolism and both PNJ and FPNJ might be a new potential source of natural antioxidant, anti-diabetic, and anti-hypertensive agents applicable to food.

A Qualitative Study of Physicians' Perspectives on Non-Cancer Hospice-Palliative Care in Korea: Focus on AIDS, COPD and Liver Cirrhosis (국내의 비암성 질환의 호스피스 완화의료 적용에 대한 전문가의 인식에 관한 질적 연구: 후천성 면역결핍 증후군, 만성 폐쇄성 폐질환, 간경화를 중심으로)

  • Purpose: From August 2017, hospice-palliative care (HPC) will be provided to patients with acquired immunodeficiency syndrome (AIDS), chronic obstructive pulmonary disease (COPD), and liver cirrhosis in Korea. To contribute to building a non-cancer (NC) hospice-palliative care model, NC specialists were interviewed regarding the goals, details, and provision methods of the model. Methods: Four physicians specializing in HPC of cancer patients formulated a semi-structured interview with questions extracted from literature review of 85 articles on NC HPC. Eleven NC disease specialists were interviewed, and their answers were analyzed according to the qualitative content analysis process. Results: The interviewees said as follows: It is difficult to define end-stage NC patients. HPC for cancer patients and that for NC patients share similar goals and content. However, emphasis should be placed on alleviating other physical symptoms and emotional care rather than pain control. Timing of the care provision should be when patients are diagnosed as "end stage". Special issues should be considered for each NC disease (e.g., use of anti-retroviral drugs for AIDS patients, oxygen supply for COPD patients suffering from dyspnea, liver transplantation for patients with liver cirrhosis) and education should be provided to healthcare professionals. NC patients tend to negatively perceive HPC, and the government's financial assistance is insufficient. Conclusion: It is necessary to define end-stage NC patients through in-depth discussion to minimize issues that will likely accompany the expansion of care recipients. This requires cooperation between medical staff caring for NC patients and HPC givers for cancer patients.

Antioxidative Activities of Dried and Fresh Citrus Peels in Jeju (제주산 감귤류 진피와 과피의 항산화 활성)

  • This study was carried out to investigate the total polyphenol and flavonoid contents and antioxidative activities of the methanol extracts of Citrus sunki, C. unshiu and C. natsudaidai peels before and after drying. Total polyphenolic content was high in the citrus peels before drying compared to the dried peels, and highest in the order of C. sunki, C. natsudaidai and C. unchiu. On the other hand, flavonoid content was high in the dried citrus peels than in the citrus peels before drying, and was highest in the order of C. unshiu, C. natsudaidai, C. sunki, but in dried peels it was highest in the order of C. natsudaidai, C. unshiu, C. sunki. DPPH radical scavenging activity was highest in the order of C. natsudaidai, C. unchiu, C. sunki, and that in the citrus peels before drying was higher than in the dried peels. Inparticular, C. natsudaidai peels before drying showed higher activity than vitamin C in 0.5 or 1.0 mg/100 g. Hydrogen peroxide scavenging activity was highest in the order of C. unshiu, C. natsudaidai, and C. sunki in the citrus peels before drying. The dried peels were highest in the order of C. unshiu, C. natsudaidai, C. sunki at 0.5 mg/100 g, but high in order of C. natsudaidai, C. unshiu and C. sunki at more than 1.0 mg/100 g. Inparticular, all C. natsudaidai peels displayed high activity more than 87% at 2.0 mg/100 g. Clear patterns in alkyl radical scavenging activity could not be confirmed in Citrus species whether before or after drying of peels. Hydroxyl radical scavenging activity was highest in the order of C. natsudaidai, C. unshiu and C. sunki but was relatively low compared to the scavenging activity of other activated oxygen species.

The Early-Stage Changes of Water Qualities after the Saemangeum Sea-dike Construction (새만금 방조제 체절 이후 초기의 수질변화에 관한 연구)

  • Saemangeum salt-water Lake has been created by the completion of the sea-dike in April 2006. To monitor the water qualities of the lake during the sea-dike construction, salinity, SS, nutrients(DIN, DIP, DISi), and chlorophyll-$\alpha$ was analyzed for the surface water from 1999 to 2007. Due to the dike construction, weaker tidal current and lesser resuspension of bottom sediment resulted in the marked decrease of the concentrations of SS in the lake water. Consequently the clearer lake water has provided better condition for primary production with deeper penetration of sunlight into the water column and sufficient nutrient content in the water. Finally the chlorophyll-$\alpha$ content became approximately double in the concentration after the dike construction. Highly stimulated algal production with the marked decrease of the concentrations of SS was decreased the concentration of DIP in the surface water. On the other hand the concentration of DIN and DISi in surface water was increased after dike construction due to the expansion of the freshwater and the supply from bottom layer. As a result, the lake revealed an extremely high NIP ratio and a DIP-limited ecosystem. The lake has been transformed from a typical coastal ecosystem to a brackish one. Since the dike completion, the lake has shown a similar change pattern to the Geum River estuary. Due to the salt-wedge intrusion of seawater, it is highly probable to expect the formation of low-oxygen zone at the bottom layer near the river-mouth area of the lake during the summer. Therefore we need a continuous sentinel monitoring of bottom water qualities in the near future.

Changes in Antioxidant and Antiobesity Activities of Cirsium setidens Nakai Ethanolic Extract Depending on Different Harvest Time (수확시기별 고려엉겅퀴 주정추출물의 항산화 및 항비만 활성 비교)

  • This study was conducted to provide basic data of Cirsium setidens Nakai in different harvest time that will be applied for development of functional foods and ingredients. We investigated pectiolinarin and pectolinarigenin content, total flavonoids content and antioxidant effects (DPPH radical scavenging activity and ORAC assay) of C. setidens Nakai. Our results showed that the pectolinarin and total flavonoids contents of C. setidens Nakai in harvesting time ranged from $43.13{\pm}0.22$ to $95.65{\pm}0.34mg/g$ and from $32.81{\pm}1.68$ to $40.43{\pm}0.35mg$ rutin equivalent (RE)/g, respectively. The DPPH radical scavenging activity of C. setidens Nakai did not show differences in harvesting time. The oxygen radical absorbance capacity (ORAC) value was highest in August (2016) extracts ($827.72{\mu}mole\;TE/g$). In addition, C. setidens Nakai exthanolic extract in harvesting time did not show any cytotoxicity up to $200{\mu}g/mL$. During adipocyte differentiation, C. setidens Nakai extract in harvesting time significantly inhibited lipid accumulation and ROS production, compared with the controls. These results suggest that C. setidens Nakai extract could be considered as a non-toxic natural resources of functional food ingredients and natural antioxidants.

Antioxidative Activities of the Codonopsis lanceolata Extract in vitro and in vivo (더덕(Codonopsis lanceolata) 추출물의 in vitro 및 in vivo 항산화 효과)

  • In vitro activities of Codonopsis lanceolata (CL) 70% ethanol extract and its fractions (hexane, chloroform, ethyl acetate, butanol and water) were examined by total polyphenol content, reducing power, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate) (ABTS), 2,2-diphenyl-$\beta$-picrylhydrazyl (DPPH), and oxygen radical absorbance capacity (ORAC) assays. The ethyl acetate fraction from CL ethanol extract (CLEA) showed the highest total polyphenol content (22.7 mg/g) among five fractions, and also exhibited an excellent reducing power (0.42~1.27 at $250\sim1,000\;{\mu}g/mL$). CLEA at $100\sim400\;{\mu}g/mL$ concentrations had 27.7~70.3% of ABTS radical scavenging activity and the highest DPPH radical scavenging activity (81.6% at $400\;{\mu}g/mL$). CLEA had dominantly higher $ORAC_{{ROO}{\cdot}}$activity compared to other fractions. CLEA and butanol fraction had significantly higher $ORAC_{{OH}{\cdot}}$ activities than 70% ethanol extract, hexane, chloroform and water fractions. The CLEA exhibited the highest antioxidant activity in CL 70% ethanol extract and its fractions. Thus, effect of CLEA treatment on antioxidant gene expression under the oxidative stress conditions by a high fat diet in animal model was studied by microarray and RT-PCR methods. The 31 antioxidant genes were expressed but the genes were not up-regulated at least a two-fold by CLEA treatment. We concluded that CLEA does not have an indirect antioxidant effect but a direct antioxidant effect by up-regulation of antioxidant genes in high fat diet-induced obese mice.

Effect of 5 Week Long High-Fat Diet on Energy Metabolic Substrate Utilization and Energy Content Evaluation of Dietary Fat (5주간의 고지방식이 섭취시 흰쥐의 에너지 대사 기질 이용과 식이지방에너지 평가에 관한 연구)

  • This study investigated the effect of a long-term high-fat diet on energy metabolic substrate utilization in resting rats in order to revalue source fat energy efficiency during a high-fat diet and its effect on energy expenditure and body fat accumulation. Sprague-Dawley male rats at 4 weeks of age were bought from Orient Bio Con. The rats were divided into a control (CON) group and a high-fat diet (HF) group. Rats ate a high-fat diet (w/w 40%, kcal/kcal 64.9%) ad libitum for 5 weeks. Food intake and body weight were measured every day at 09:00 throughout the experimental period. Energy expenditure was measured using an animal energy metabolism chamber after 4 weeks. The final body weight did not change between the CON and HF groups, but caloric intake was significantly higher in the HF group than in the CON group (p<0.05). There was no difference between the groups in oxygen uptake, however carbon dioxide production was significantly higher in the HF group. Also, the respiratory exchange ratio was higher in the HF group. Carbohydrate oxidation was lower in the HF group than in the CON group, but fat oxidation in the HF group was greater. These results mean that energy substrate oxidation at rest is affected by diet composition, especially dietary fat content. Abdominal fat fad weights were significantly higher by 33% in the HF group than in the CON group even though the calorie intake in the HF group was higher by 6%. These results suggested that the dietary fat calorie value might have a higher Atwater value of 9 kcal/g, which mean that dietary fat calorie values could be reconsidered in body weight control scenarios such as which the obese or weight class athletes.

Physical and Chemical Characteristics of Cotton Waste Substrate According to Fermentation Conditions for Oyster Mushroom Bed Cultivation (느타리버섯 폐면배지의 발효조건별 이화학적 특성)

  • We have surveyed the variation of physical and chemical characteristics of aerobic and anaerobic outdoor fermentation of cotton wastes using for oyster mushroom cultivation. The inner temperature of cotton wastes fermented aerobically covered with thin cloth and setting pallet at bottom was higher than that of anaerobic fermented cotton wastes covered with P.E vinyl and the maximum temperature was $75^{\circ}C$ at 5th day after fermentation. pH of cotton wastes fermented aerobically was increased up to 8.9 after fermentation of $9{\sim}12$ days, but that of anaerobically fermented was decreased up to 5.0. Total carbon content was decreased but total nitrogen content was increased when fermentation was in progress. Oxygen concentration of cotton wastes fermented aerobically was decreased until 6 days after fermentation but increased after 9 days of fermentation. Ammonia concentration of cotton wastes fermented aerobically and anaerobically was below 10 ppm and $20{\sim}85\;ppm$ respectively. In anaerobic condition the cotton wastes was contaminated with mold ($15{\sim}50%$), where no contamination was found in aerobic condition during spawn running stage. Yields of mushroom grown on cotton wastes aerobically fermented for $6{\sim}9$ days was $23.0{\sim}23.6\;kg$ per $3.3\;m^2$ area.

Variations in antioxidant activity in Protaetia brevitarsis larvae depending on the feeding source (먹이원에 따른 흰점박이꽃무지(Protaetia brevitarsis) 유충의 항산화활성)

  • The objective of this study was to evaluate the antioxidant activity of extracts of Protaetia brevitarsis larvae fed on fermented oak sawdust (FOS) or spent mushroom substrates (SMS, Pleurotus eryngii). Total polyphenol content was 32% higher in extracts of larvae fed on SMS (P. eryngii) (75.33±0.43 mg GAE/g) than in extracts of larvae fed on FOS (57.02±1.73 mg GAE/g). The flavonoid content of extracts of larvae grown on FOS and SMS (P. eryngii) was 24.6±0.28 mg/g and 25.4±0.75 mg/g, respectively. DPPH radical scavenging activity increased in an extract concentration-dependent manner, and the DPPH radical scavenging capacity of the extract of larvae produced on SMS (P. eryngii) was higher than that of the larvae produced on FOS. The reducing power of the larval extracts produced on FOS and SMS (P. eryngii) increased in an extract concentration-dependent manner, but there was no significant difference between them. The extract of larvae fed on SMS (P. eryngii) (66.55±0.99 uM TE/g) had a higher oxygen radical absorbance capacity (ORAC) than extracts of larvae grown on FOS (76.32±0.48 uM TE/g). The effect of larval extracts on cell proliferation was investigated using a WST-1 (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulfonate) assay on RAW 264.7 cells. When cells were treated with larval extracts produced on FOS and SMS (P. eryngii) at concentrations of 0, 2, 4, 8, 16, 32, 40, and 64 mg/ml, RAW 264.7 cells proliferated at 90% or more. Therefore, larval extracts produced on FOS and SMS (P. eryngii) were not toxic to RAW 264.7 cells.
