  • Bottled water companies submit monthly hydrologic data including periodical environmental effects investigation, daily water production capacity, water production, water level, water chemistry (pH, EC, temperature) per hour and strictly manage groundwater by periodical analyses. Thus few problems concerning drawdown due to excess intake of groundwater take place. Nevertheless, bottled water companies are imprinted as a contribution to civil affairs resulted regarding groundwater near the companies. Therefore, a new method is required during water balance analysis in environmental effects evaluation, which should be compatible with the evaluation by hydrologic experts as well more accessible to non-experts. In this study, water level of surface water and recharge rate in subcatchment where water production wells are located were measured and monthly baseflow rates were separated from normal streams. Besides, recharge properties of groundwater and surface water in the same catchment area were estimated using analyses of oxygen and hydrogen isotopes in groundwater (production well), surface water, and rainfall.

Substrate-Perfusion Studies on Coronary Circulation and Myocardial Energy Metabolism in Spontaneously Hypertensive Rat Hearts (발현성 고혈압쥐의 관상순환 기능과 심장근의 에너지 대사에 관한 생체외 에너지원의 관류 연구)

  • The effects of energy-yielding substrates on coronary circulation, myocardial oxygen metabolism, and intramyocytic adenylates of perfused Wistar control rat(WC) and spontaneously hypertensive rat(SHR) hearts were examined under basal and $\beta$-adrenergic stimulation conditions. The perfusion medium (1.0mM Ca2+) contained 5mM glucose (+5U/l insulin) in combination with 5mM pyruvate, 5mM lacate, 5mM acetate, or 5mM octanoate as energy substrates. Hearts were perfused with each substrate buffer for 20min under basal conditions. Coronary functinal hyperemia was induced by infusing for 20min isoproterenol (ISO, 1uM), a $\beta$-receptor agonist. Cardiac adenylates, glycolytic intermediates, and coronary venous lactate were measured by using an enzymatic analysis technique. Under basal conditions, acetate and octanoate significantly increased coronary flow(CF) of WC in parallel with myocardial oxygen consumption. However, CF of SHR was partly attenuated by coronary vasoconstriction despite metabolic acidosis. In addition, pyruvate and lactate depressd ISO-induced coronary functional hyperemia in SHR. It should be noted that octanoate exhibited coronary dysfunction under ISO conditions. On the other hand, fat substrates depleted myocardial high energy phosphate pool and accumulated breakdown intermediates. In SHR with coronary vasoconstriction under basal conditions, and with depressed coronary functional hyperemia, high energy phosphates were greatly depleted. These results suggest that energy substrates in the myocardium and coronary smooth muscle alter remarkably coronary circulation, and that coronary circulatory function is associated with a reserve of high energy phosphates and a balance between breakdown and nono synthesis of energy phosphates. These findings could be explained by alterations in the cytosolic redox state manipulated by LDH and hence in the cytosolic phosphorylation potential, which might be involved in hypertension of SHR.

  • A balance between production and degradation of reactive oxygen species (ROS) is critical for maintaining cellular homeostasis. Increased levels of ROS during oxidative stress are associated with disease conditions. Antioxidant enzymes, such as extracellular superoxide dismutase (EC-SOD), in the extracellular matrix (ECM) neutralize the toxicity of superoxide. Recent studies have emphasized the importance of EC-SOD in protecting the brain, lungs, and other tissues from oxidative stress. Therefore, EC-SOD would be an excellent therapeutic drug for treatment of diseases caused by oxidative stress. We cloned both the full length (residues 1-240) and truncated (residues 19-240) forms of human EC-SOD (hEC-SOD) into the donor plasmid pFastBacHTb. After transposition, the bacmid was transfected into the Sf9-baculovirus expression system and the expressed hEC-SOD purified using FLAG-tag. Western blot analysis revealed that hEC-SOD is present both as a monomer (33 kDa) and a dimer (66 kDa), as detected by the FLAG antibody. A water-soluble tetrazolium (WST-1) assay showed that both full length and truncated hEC-SOD proteins were enzymatically active. We showed that a potent superoxide dismutase inhibitor, diethyldithiocarbamate (DDC), inhibits hEC-SOD activity.

Ecological Studies of the Lake Changjamot II. Primary Production in Lake Changjamot During Spring Season (장자못의 생태학적 연구 제II보 춘계 장자못의 기초생산)

  • A study was made on the primary production of Lake Changjamot during the spring season of 1973 by menas of the oxygen method. The stratification of temperature and dissolved oxygen were formed in May with the stratified structure of phytoplankton. The range of Secchi disc transparency was from 0.8m to 2.3m during the nine months of this investigation, which was begun in January, 1973. The value was lowest in early June when the phytoplankton blooming reached the peak. The concentration of PO4-P, NH3-N, NO3-N and NO3-N was reduced at the beginning of the phytoplankton blooming and increased again after May except PO4-P. It might have been caused by the inflow of the nitogenous fertilizer from the surrounding agricultural area since May when farming was started. The total amount of chlorophyll-a in the entire water column varied from 25mg/$m^2$ to 277mg/$m^2$ from January till September with the maximum value occurring in early June. These values show a considerable eutrophication of the in comparison with the data obtained in 1969. The daily gross production in the lake varied from a low of 655mgC/$m^2$ to a high of 2,859 mgC/$m^2$ during the spring season and this corresponds to the variation of the amount of chlorophyll. The total amount of daily respiration varied from 650mg C/$m^2$ in winter to 2,307 mg C/$m^2$ in late spring and exceeds gross primary production especially in late May showing the negative balance of daily production and consumption of organic material at that time. In conclusion, Lake Changjamot is a fairly productive and a moderately autotrophic lake and has been eutrophicated much during the past four years.

  • A new three-dimensional Gd-MOF, [Gd(p-XBP4)4(H2O)]·W(CN)8; (1; p-XBP4 = N,N'-p-phenylenedimethylenbis (pyridin-4-one)) has been synthesized by slow-evaporation and its crystal structure was characterized by single-crystal X-ray diffraction (SCXRD) analysis. For each GdIII ion, there are seven coordination sites, which are occupied by six oxygen atoms of six p-XBP4 ligands and one oxygen atom from the water molecule. The [W(CN)8]3- anion exists for charge balance with cationic framework. The GdII ions are interconnected by the p-XBP4 ligand to form the three-dimensional structure. Considering the magnetic property of lanthanide ions, magnetic studies of Gd-MOF were investigated by direct-current (DC) magnetic susceptibilities measurements.

  • Effects of operating variables on temperature profile and performance of 3 MWth chemical looping combustion system were estimated by mass and energy balance analysis based on configuration and dimension of the system determined by design tool. Air reactor gas velocity, fuel reactor gas velocity, solid circulation rate, and solid input percentage to fluidized bed heat exchanger were considered as representative operating variables. Overall heat output and oxygen concentration in the exhaust gas from the air reactor increased but temperature difference decreased as air reactor gas velocity increased. Overall heat output, required solid circulation rate, and temperature difference increased as fuel reactor gas velocity increased. However, overall heat output and temperature difference decreased as solid circulation rate increased. Temperature difference decreased as solid circulation rate through the fluidized bed heat exchanger increased. Effect of each variables on temperature profile and performance can be determined and these results will be helpful to determine operating range of each variable.

Air Fuel Ratio Determination Method for Alternative Fuel Based on Carbon Balance and Linear Equation (탄소 균형과 1차식에 의한 대체 연료의 공연비 산정법)

  • This paper is to compare the carbon-balanced and liner air-fuel ratio determination methods for alternative fuels. In the previous work, expansion of Eltinge chart, unburned hydrocarbon compensation, comparison of the results from various methods were discussed. It has been also concluded that Eltinge method might be regarded as the most general equation of AFR determination among the existing ones. In the recent years, however, increasing demand for the environmental preservation, including global warming-up protection, and energy conservation lead to introduce the alternative fuel to the internal combustion engine. Therefore, the exact calculations of AFR for these fuels are needed. Especially, for the fuel that contains oxygen, all AFR calculation equations except Eltinge have to be re-formulated. In this paper, the AFR for alternative fuel were calculated by re-formulated carbon balance, accuracy of which was already confirmed, and linear equations, which are newly proposed by statistical method for each fuel. The results show that AFRs based on carbon balance have a little more error compared with gasoline, however, the accuracy is enough for this formula to apply to various fuel. The proposed linear equation also have excellent accuracy below $\lambda=1.2$.

  • Fuel cells are devices that convert chemical energy directly into electrical energy. Owing to the high efficiency of the fuel cells, a large number of research work have been done during these years. Among many kinds of the fuel cells, a polymer electrolyte membrane fuel cell is such kind of thing which works under low temperature. Because of the specialty, it stimulated intense global R&D competition. Most of the major world automakers are racing to develop polymer electrolyte membrane fuel cell passenger vehicles. Unfortunately, there are still many problems to be solved in order to make them into the commercial use, such as the thermal and water management in working process of PEMFCs. To solve the difficulites facing the researcher, the analysis of the inner mechanism of PEMFC should be implemented as much as possible and mathematical modeling is an important tool for the research of the fuel cell especially with the combination of experiment. By regarding some of the assumptions and simplifications, using the finite element technique, a two-dimensional electrochemical mode is presented in this paper for the further comparison with experimental data. Based on the principals of the problem, the equations of electronic charge conservation equation, gas-phase continuity equation, and mass balance equation are used in calculating. Finally, modeling results indicate some of the phenomenon in a unit cell, and the relationships between potential and current density.

Respirometry for the Assessment of Organics Biodegradability in Municipal Wastewater: I. Respirometry (호흡률법에 의한 하수의 생분해 특성 평가: I. 호흡률법)

  • Organics may be divided into biodegradable and nonbiodegradable fractions on the basis of biodegradability. Biodegradable organics may be subdivided into readily and slowly biodegradable fractions. As this biodegradability of organics in municipal wastewater has a great influence on the efficiency of a biological nutrient removal process, it has been assessed by respirometry. The respirometer, which consisted of a respiration chamber and a respiration cell, was used to measure the respiration rate of biomass utilizing the readily biodegradable organics. The readily biodegradable organics are about 10% of the COD in municipal wastewater. The adequate ratio of wastewater to sludge volume and the concentration of sludge are required in measuring the respiration rate due to the readily biodegradable organics. By using a biochemical oxygen demand test, the slowly biodegradable organics including biomass are estimated about 66% of COD. The soluble inert organics are about 11% of COD. On the basis of mass balance, the particulate inert organics are estimated about 13% of COD.

Estimation of Nitrite Concentration in the Biological Nitritation Process Using Enzymatic Inhibition Kinetics

  • Recently, interests to remove nitrogen in the nitritation process have increased because of its economical advantages, since it could be a short-cut process to save both oxygen for nitrification and carbon for denitrification compared to a typical nitrification. However, the kinetics related with the nitritation process has not yet been fully understood. Furthermore, many useful models which have been successfully used for wastewater treatment processes cannot be used to estimate effluent nitrite concentration for evaluating performance of the nitritation process, since the process rate equations and population of microorganisms for nitrogen removal in these models have been set up only for the condition of full nitrification. Therefore, the present study was conducted to estimate an effluent nitrite concentration in the nitritation process with a concept of enzymatic inhibition kinetics based on long-term laboratory experiments. Using a nonlinear least squares regression method, kinetic parameters were accurately determined. By setting up a process rate equation along with a mass balance equation of the nitrite-oxidizing step, an effluent nitrite concentration in the nitritation process was then successfully estimated.
