Paeparation and Properties of Epoxy Copolymers Containing Oxime-urethane Groups as Photobase Generators

  • Epoxy copolymers containing oxime urethane groups were prepared by the polymerization of glycidyl methacrylate and N-[5-(benzophenoneoximinocarbonylamino)pentanyl]maleimide (BOPM). Their physical properties were characterized by GPC, DSC and TGA analyses. Photochemical changes of the copolymers were studied by UV, IR spectroscopy, and contact angle measurements. A photoinduced cross-linking reaction in copolymer films was observed by measuring the insoluble fraction. Irradiation of the copolymers at 254 nm UV light leads to the formation of pendant amino groups by photodissociation of the oxime-urethane groups. Treatment of the amino groups with HCl resulted in the formation of ammonium salts, which changed the polymer surface to be hydrophilic. An insoluble fraction of the copolymers increased with irradiation time, heating time, and heating temperature. Cross-linking of the epoxy resin effectively catalyzed by the photogenerated pendant amines upon heating.

A Study on the Adsorption and Recovery of Metal Ions by Amberlite XAD Resins Impregnated with Oxime Compounds (Oxime 화합물을 침윤시킨 Amberlite XAD 수지에 의한 금속이온의 흡착 및 회수에 관한 연구)

  • The adsorption behaviors of some oxime compounds well known as metal chelating agents on the Amberlite XAD resins were compared by measuring their distribution coefficients (log Kd) in various media, respectively. Among the oxime compounds, salicylaldoxime (SAO) and $\alpha-benzoinoxime(${\alpha}$-BzO)$ which showed large log Kd values were chosen. The characteristics of XAD-4 resins impregnated with SAO and ${\alpha}$-BzO have been studied to apply them for the adsorption and recovery of minute quantities of metal ions in aqueous solution. The optimum conditions for adsorption of SAO and ${\alpha}$-BzO on the resin were 30% methanol media having pH range of 1~8(for SAO) and 1~9 (for ${\alpha}$-BzO), respectively. The distribution coefficients of two oxime compounds were decreased as temperature increased. From the adsorption enthalpy data of SAO and ${\alpha}$-BzO, ranging from 4.96 to 6.66 Kcal/mol, it is suggested that their adsorption mechanism on XAD-4 resin is likely due to molecular adsorption equivalent to dipole-dipole interaction. The impregnated resins were considerably stable in the aqueous solutions of pH 5.0~10.0 and in 0.1~5M hydrochloric acid solutions. The former is the medium for adsorption of metal ions, while the latter is for recovery of the adsorbed metal ions. The adsorption mole ratio of Mn(II), Co(II), Ni(II), Zn(II) ions on SAO-XAD-4 and ${\alpha}$-BzO-XAD-4 resins were about 1 : 2 at the optimum conditions, respectively.   • An enzyme-linked immunosorbent assay (ELISA) was established for the detection of zearalenone by using monoclonal antibodies produced by Z-M-26 hybridoma cells when injected into a mouse and zearalenone-oxime-OV A conjugate. Zearalenone-oxime-OV A conjugates were diluted with carbonyl buffer, coated to 96 well microtiter plates at $4^{\circ}C$ overnight and blocked with 1% BSA overnight. One thousand times diluted antibody solution together with standard zearalenone or sample was added to 96-well microtiter plates and stood overnight. A secondary antibody conjugated with HRP was added and an hour later, enzyme substrate (TMBZ) solution was added for color develpment. Mter 30 minutes, coloring reaction was terminated by adding 2 N $H_2S0_4$ and the O.D. was measured at 450 nm. Detection range of this method was about 0.1~100 ppb. The established indirect competitive ELISA method was suitable for a rapid and effective analysis of zearalenone in agricultural products.

Development of Enzyme Linked Immunosorbent Assay for Determination of Zearalenone in Animal Feeds (가축사료 중 Zearalenone 분석을 위한 Enzyme Linked Immunosorbent Assay법의 개발)

  • We examined to develop the enzyme linked immunosorbent assay (ELISA) for determination of zearalenone in animal feeds. Zearalenone was first converted to 6'-(carboxymethyl) zearalenone oxime(zearalenone oxime) to get a coupling site and then conjugated to bovine serum albumin(BSA) for use as immunogen and to horseradish peroxidase(HRP) for use as enzyme marker. Antibody against zearalenone was obtained after 11 weeks of immunization of rabbit with zearalenone oxime-BSA. Cross reactivity of the antibody with ${\alpha}-zearalanol,\;{\beta}-zearalenol,\;{\alpha}-zearalanol\;and\;{\beta}-zearalanol$ were 168, 46, 26 and 20% respectiviely. A simple procedure was devised for the screening of zearalenone in feeds using ELISA. Feeds samples(5g) were extracted by blending with 25 ml of methanol-phospate butTered saline-dimethylformate(70 : 29 : 1) and the extract was filtered and aqueous filterate analyzed. It took only 1 hours to do whole procedure for the analysis of zearalenone in feeds by the direct competitive ELISA, and detectable limit was 1-100 ppb. Using this procedure, only 4 of 24 feed samples showed positive results with 3.93-7.43 ppb levels.

  • [99mTc]Tc-Hexamethylpropylene amine oxime (HMPAO) is currently used as a regional cerebral blood flow imaging agent for single photon emission computed tomography (SPECT). The HMPAO ligand exists in two isomeric forms: d,l and meso showing different properties in vivo. Later studies indicated that brain uptake patterns of 99mTc-complexes formed from separated enantiomers differed. Separation of enantiomers is difficult by fractional crystallizations method. Usually, the substance is obtained in low chemical yield in a time-consuming procedure. Furthermore, the final product still contains some impurity. So we have developed new efficient route for synthesis of R,R- and S,S-HMPAO enantiomeric compounds in 6-steps. Nucleophilic substitution (SN2) reactions of 2,2-dimethylpropane-1,3-diamine either with S- (1a) or R-methyl2-chloropropanoate (1b) were performed to produce compounds R,R- (2a) or S,S-isomer (2b) derivatives protected with benzylchloroformate (Cbz), respectively. And then Weinreb amide and methylation reaction using Grignard reagent, oxime formation with ketone group and deprotectiion of Cbz group by hydrogenolysis gave S,S- (7a) or R,R-HMPAO (7b), respectively. Entaniomeric compounds were synthesied with high yield and purity without any undesired product. The 7a or 7b kits containing 10 ㎍ SnCl2-2H2O were labeled with 99mTc with high radiolabeling yield (90%).

Stability of Mono- and Bis-pyridinium Oximes in Aqueous Systems (Monopyridinium Oxime과 Bispyridinium Oxime 화합물의 수용액 중 안정성 연구)

  • The stability of three oximes, Hl-6 [(4-carbamoyl-2'[(hydroxyimino)-methyl]- 1,1'-oxydimethylenedi-(pyridinium chloride)], Hl-CN [(4 cyano-2'-[(hydroxyimino)-methyl] -1,1'-oxydimethylene-di-(pyridinium chloride)], and 2-PAM [pralidoxime chloride] in aqueous solutions was evaluated by HPLC assay. The rate of degradation is dependent on the pH as well as the temperature at which the solution is stored. The optimum pH for the stability of these oximes was pH 2 to 3. The degradation rate constant for 2-PAM ($k\;at\;70^{\circ}C$, $2.07{\times}10^{-4}/hr;\;E_a\;value$, 27.2 kcal/mol) was smaller than those for bis-pyridiniumoximes, Hl-6 ($k\;at\;70^{\circ}C$, $3.38{\times}10^{-3}/hr$) and Hl-CN ($k\;at\;70^{\circ}C$, $8.66{\times}10^{-3}/hr;\;Ea\;value$, 20.7 kcal/mol). In mechanistic analyses, it was found that Hl-CN was decomposed through not only the hydrolysis of nitrile group but also the cleavage of methylene ether bridge, in contrast to Hl-6 which was degraded mainly through the cleavage of methylene ether bridge.

A One-Component Negative Photoresist Based on an Epoxy Terpolymer Containing Oxime-Urethane Groups as a Photobase Generator

  • For their application as one-component photoresists, we prepared epoxy terpolymers containing oxime-urethane and benzophenone groups by the radical polymerization of glycidyl methacrylate (GMA), metha-cryloxyethyl benzophenoneoxime urethane (MBU), and N-(4-benzoyl)phenylmaleimide (BPMI). The terpolymer composition was optimized to provide the most photosensitive photoresist. The photo-decomposition reaction of the oxime-urethane groups in the terpolymer was monitored by UV absorption spectroscopy, and the photo-crosslinking reaction of the epoxy terpolymer was observed by measuring the normalized thickness. The photosensitivity of the epoxy terpolymer increased as the amount of BPMI and MBU units increased up to 16 and 24 mol%, respectively. Among the terpolymers we prepared, terpolymer T-II(contents of GMA, MBU, BPMI are 75, 19, 6.1 mole%, respectively) exhibited the highest photosensitivity ( $D_{c}$ $^{0.5}$ = 430 mJ/$\textrm{cm}^2$) and had a moderate contrast (${\gamma}$$^{p}$ = 1.23). Negative-tone micropatterns having a line width of ca. 10 ${\mu}{\textrm}{m}$ were obtained by developing the system with chloroform.m.

  • Oxime ethers of ${\beta}$-oxo-${\gamma}$-phenylbutanoic acids were prepared to develop more effective PPAR ${\alpha}$ and ${\gamma}$ dual agonists. Among them, compound 11k exhibited potent $in$ $vitro$ activities with $EC_{50}$ of 2.5 nM and 3.3 nM in PPAR ${\alpha}$ and ${\gamma}$, respectively. It showed better glucose lowering effects than rosiglitazone 1 and improved the lipid profile like plasma triglyceride in db/db mice model.
