산소희박환경에서 과산화수소를 이용한 디젤개질 가능성 탐구 (Study on Possibility of Diesel Reforming with Hydrogen Peroxide in Low-Oxygen Environments)

  • 잠수함 및 수중무인체계 등의 산소희박환경에서 연료전지를 통한 효과적인 전력생산을 위해서는 높은 수소저장밀도를 갖는 수소공급원이 필요하다. 디젤연료는 액체연료로서 저장 및 공급이 용이하며, 연료전지의 연료가 되는 수소의 단위질량 및 단위부피당 저장밀도가 높은 장점을 갖고 있다. 이러한 디젤연료의 장점을 기반으로 본 연구에서는 산소희박환경에서 수소생산을 위해 디젤연료의 개질반응을 이용하였으며, 산화제로 단위부피당 산소 저장밀도가 높고 액상으로 보관이 용이한 과산화수소 수용액을 기존의 산화제인 물과 산소의 대체산화제로 이용하는 방법을 제안하였다. 과산화수소 수용액의 디젤개질 산화제로써의 특성을 파악하기 위해 물, 공기 산화제와의 비교실험을 진행하였으며, 기존의 산화제와 디젤 개질반응 시 동일한 특성을 갖는 것을 실험적으로 확인하였다. 또한 상용디젤을 연료로 온도 및 과산화수소 수용액의 농도에 따른 개질성능을 평가하였으며, 49시간의 가속 열화실험을 통하여 디젤, 과산화수소 수용액을 이용한 수소생산의 가능성을 확인하였다.

촉매량의 Piperidine-1-oxyl과 NaOCl계에서 벤질 에테르 유도체들의 산화 반응 (Oxidation of Benzyl Ethers in Sodium Hypochlorite Mediated Piperidine-1-oxyl System)

  • 여러가지 비대칭 벤질 에테르들과 벤질 알킬 에테르들을 $CH_3CO_2Et$-NaOCI수용액(6.6 mol eq.)의 2상 용매계에서 4-methoxy-2, 2, 6, 6,-tetramethylpiperidine-1-oxyl(0.03 mol eq., 4-methoxy-TEMPO)을 이용하여 산화시키면 벤조에이트로 산화가 일어난다. 4-methoxy-TEMPO는 2차 산화제인 NaOCI에 의하여 본반응의 산화제인 N-oxo-4-2, 2, 6, 6, -tetramethyl-piperidium 염(N-oxoammonium 염)으로 변환된다. N-oxoammonium 염은 에테르를 산화시키고 N-hydroxy-4-methoxy-2, 2, 6, 6,-tetramethylpiperidine(hydroxyamine)으로 환원된다. Hydroxy-amine은 NaOCI에 의하여 N-oxoammonium 염으로 순환 재생되므로 4-methoxy-TEMPO는 촉매량 사용하였다. 이 반응은 또한 조촉매인 KBr(0.03 mol eq.)가 필수적이고 반응 중 pH는 8.0 이하로 유지되어야 한다. 0 - 5$^{\circ}C$의 반응 온도로 2.5시간 반응시키면 대부분 벤조에이트로 산화 되었다. 벤질 알킬 에테르들의 선택성 산화는 수소의 산도와 알킬기의 입체효과에 영향을 받음이 고찰되었다.

이산화티탄에 의한 삼염화에틸렌의 광촉매 분해반응 (Photocatalytic Degradation of Trichloroethylene over Titanium Dioxides)

  • 자외선 조사하에서 $TiO_2$를 촉매로 하여 trichloroethylene의 광분해반응을 행하였다. 여러 가지 $TiO_2$ 촉매의 활성을 비교하였고, 반응조건에 따른 광분해 활성을 조사하였다. 또한 빛의 세기에 따른 분해활성과 반응물에 첨가된 물의 영향을 조사하였다. 여러 가지 $TiO_2$ 촉매에서 P-25가 가장 높은 활성을 보였고, anatase형이 rutile형보다 높은 활성을 보였다. 또한 반응물의 유속이 느릴수록 또한 초기농도가 낮을수록 trichloroethylene의 분해 반응 활성은 증가하였고, 산화제로서 공기를 사용하는 것이 효과적이었다. 한편 반응물에 첨가된 물은 촉매의 활성을 감소시켰고, 빛의 세기가 증가할수록 분해 반응속도가 증가하였으나 태양광에 의해서는 분해율이 매우 낮았다. trichloroethylene의 농도가 낮을 경우에는 촉매의 활성저하가 거의 일어나지 않았다.

자색고구마 신자미로부터 분리한 안토시아닌 분획물의 항산화 활성과 산화스트레스에 대한 간세포 보호 효과 (Antioxidant Activity and Protective Effects of Anthocyanins-Rich Fraction from Korean Purple Sweet Potato Variety, "Shinjami" against Oxidative Stress in HepG2 Cell)

  • Anthocyanins, a class of flavonoids, are natural water-soluble pigments, which are mainly found in vegetables and fruits. Anthocyanins have attractive pharmacological activities, such as anti-oxidant, anti-inflammatory, anti-cancer, and anti-diabetic. The purpose of this study was to investigate the protective effects of anthocyanins-rich fraction (ANF) from Korean purple sweet potato variety, "Shinjami", against hydrogen peroxide ($H_2O_2$)-induced oxidative stress. In our results, pre-treatment of HepG2 cells with ANF ($10{\mu}g/mL$) significantly prevented cell death and maintained cell integrity, following exposure to 0.9 mM hydrogen peroxide. The $H_2O_2$-dependent production of intracellular ROS was also significantly decreased by pre-treatment with ANF (6 h, $10{\mu}g/mL$). In addition, ANF increased the mRNA levels of antioxidant enzymes, catalase and glutathione level in $H_2O_2$-treated HepG2 cells. These results indicated that ANF protected HepG2 cells against $H_2O_2$-induced oxidative stress by inducing protective system.

Acanthopanax sessiliflorus stem confers increased resistance to environmental stresses and lifespan extension in Caenorhabditis elegans

  • BACKGROUND/OBJECTIVES: Acanthopanax sessiliflorus is a native Korean plant and used as a traditional medicine or an ingredient in many Korean foods. The free radical theory of aging suggests that cellular oxidative stress caused by free radicals is the main cause of aging. Free radicals can be removed by cellular anti-oxidants. MATERIALS/METHODS: Here, we examined the anti-oxidant activity of Acanthopanax sessiliflorus extract both in vitro and in vivo. Survival of nematode C. elegans under stress conditions was also compared between control and Acanthopanax sessiliflorus extract-treated groups. Then, anti-aging effect of Acanthopanax sessiliflorus extract was monitored in C. elegans. RESULTS: Stem extract significantly reduced oxidative DNA damage in lymphocyte, which was not observed by leaves or root extract. Survival of C. elegans under oxidative-stress conditions was significantly enhanced by Acanthopanax sessiliflorus stem extract. In addition, Acanthopanax sessiliflorus stem increased resistance to other environmental stresses, including heat shock and ultraviolet irradiation. Treatment with Acanthopanax sessiliflorus stem extract significantly extended both mean and maximum lifespan in C. elegans. However, fertility was not affected by Acanthopanax sessiliflorus stem. CONCLUSION: Different parts of Acanthopanax sessiliflorus have different bioactivities and stem extract have strong anti-oxidant activity in both rat lymphocytes and C. elegans, and conferred a longevity phenotype without reduced reproduction in C. elegans, which provides conclusive evidence to support the free radical theory of aging.

배양 혈관 내피세포에서 Hydrogen Peroxide에 의한 투과성 증가에 미치는 성향정기산의 효과 (Protection by Sunghyangchungisan against Hydrogen Peroxide-induced Increase in Endothelial Permeability)

  • Objectives : Hindered barrier function of vascular endothelium has been implicated in the initiation and progression of degenerative vascular diseases such as atherosclerosis. In this study, the effect of Sunghyangchungisan(SHCS) as a protectant against oxidant-induced destruction of endothelial barrier function was assessed. Methods : Toward this end, endothelial cells derived from the human umbilical vein were cultured as monolayers on permeable membrane filters. Endothelial permeability was monitored by measuring transendothelial electrical resistance and movement of low density lipoprotein (LDL) across the endothelial monolayer. Results : Along with increased movement of LDL, $H_2O_2$-induced increase in endothelial permeability was paralleled by a decrease in transendotheliaI electrical resistance. The effect of $H_2O_2$ was mimicked by phorbol 12-myristate 13-acetate (PMA), a potent activator of proteinkinase C. Calphostin-C, a protein kinase C inhibitor, effectively blocked the increase in endothelial permeability induced by $H_2O_2$ or PMA, indicating that activation of protein kinase C is associated with the $H_2O_2-induced$ permeability change. SHCS effectively protected the endothelial monolayer against $H_2O_2-induced$ increase in permeability, whereas, it did not affect PMA-induced change. Forskolin, a potent activator of adenylyl cyclase, antagonized $H_2O_2$ to increase endothelial permeability. In addition, in ${H_2O_2}-treated$ cens, intracenular cAMP concentration was significantly decreased, indicating that impaired cAMP production as well as activation of proteinkinase C is a mechanism underlying ${H_2O_2}>-induced$$H_2O_2$ with regard to its effect on intracellular cAMP content. However, SHCS itself did not affect resting cAMP concentration in endothelial cells. Conclusions : These results suggest that SHCS might operate as an effective protectant against oxidant-induced destruction of endothelial barrier function. The mechanism does not appear to involve direct interaction with protein kinase C- or cAMP-associated signaling mechanism.

L-histidine and L-carnosine exert anti-brain aging effects in D-galactose-induced aged neuronal cells

  • BACKGROUND/OBJECTIVES: Brain aging is a major risk factor for severe neurodegenerative diseases. Conversely, L-histidine and L-carnosine are known to exhibit neuroprotective effects. The aim of this study was to examine the potential for L-histidine, L-carnosine, and their combination to mediate anti-brain aging effects in neuronal cells subjected to D-galactose-induced aging. MATERIALS/METHODS: The neuroprotective potential of L-histidine, L-carnosine, and their combination was examined in a retinoic acid-induced neuronal differentiated SH-SY5Y cell line exposed to D-galactose (200 mM) for 48 h. Neuronal cell proliferation, differentiation, and expression of anti-oxidant enzymes and apoptosis markers were subsequently evaluated. RESULTS: Treatment with L-histidine (1 mM), L-carnosine (10 mM), or both for 48 h efficiently improved the proliferation, neurogenesis, and senescence of D-galactose-treated SH-SY5Y cells. In addition, protein expression levels of both neuronal markers (β tubulin-III and neurofilament heavy protein) and anti-oxidant enzymes, glutathione peroxidase-1 and superoxide dismutase-1 were up-regulated. Conversely, protein expression levels of amyloid β (1-42) and cleaved caspase-3 were down-regulated. Levels of mRNA for the pro-inflammatory cytokines, interleukin (IL)-8, IL-1β, and tumor necrosis factor-α were also down-regulated. CONCLUSIONS: To the best of our knowledge, we provide the first evidence that L-histidine, L-carnosine, and their combination mediate anti-aging effects in a neuronal cell line subjected to D-galactose-induced aging. These results suggest the potential benefits of L-histidine and L-carnosine as anti-brain aging agents and they support further research of these amino acid molecules.

겉보리에서 배양한 영지버섯 추출물의 항산화 및 항염증 효능 평가 (Evaluation of Anti-Oxidant and Anti-Inflammatory Activities of Ganoderma lucidum Cultured on Hulled Barley)

  • Background: Ganoderma lucidum cultured on hulled barley was investigated as a potential natural source of antioxidants and anti-inflammatory agents. Methods and Results: The yields from Ganoderma lucidum cultured on hulled barley water and ethanol extract were 17.69% and 25.77%, respectively. The antioxidant activity of Ganoderma lucidum cultured on hulled barley extracts was confirmed by various methods including assayss of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis-(3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS), nitrite radical scavenging, and $Fe^{3+}$ to $Fe^{2+}$ reducing power activity. The ethanol extract of Ganoderma lucidum cultured on hulled barley showed improved DPPH, ABTS and nitrite radical scavenging activity compared with the water extract. After treatment of RAW264.7 cells with Ganoderma lucidum cultured on hulled barley ethanol extracts, the cell viability compared with the control was 92.82%, even at a concentration of $3,000{\mu}g/m{\ell}$. The ethanol extract inhibited reactive oxygen species (ROS) generation in RAW264.7 cells stimulated with $H_2O_2$, even at low concentrations. In addition, the ethanol extract showed an inhibitory effects on the production of lipopolysaccharide-induced nitric oxide (NO) in RAW264.7 cells. Conclusions: This study suggests that the extract of Ganoderma lucidum cultured on hulled barley is a potential source of natural antioxidants and anti-inflammatory agents.

생리활성 펩타이드의 피부미용학적 특성 및 활용 (Characteristics and Applications of Bioactive Peptides in Skin Care)

  • Bioactive peptides (BAP) showed excellent cosmetic activity than bio-materials such as caffeic acid (CA), gallic acid (GA), and nicotinic acid (NA). Caffeoyl tripeptide-1 (CT-1) is a BAP that is stabilized with Gly-His-Lys (GHK) tripeptide and CA by using Fmoc solid phase peptide synthesis. Digalloyl tetrapeptide-19 (DT-19) is stabilized by combining Lys-Glu-Cys-Gly with GA and nicotinoyl tripeptide-1 (NT-1) is synthesized by GHK and NA. According to experiments, CT-1 has an excellent anti-oxidant function even with a very small amount of 10 ppm CT-1. DT-19's tyrosinase inhibition activity has the better effect of about 28.57% in 0.01% and 33.33% in 0.005% of concentration and about 7.89% in 0.001% concentration than vitamin-C. In addition, NT-1 is safer than the NA. Almost BAPs like pal-KTTKS, acetyl hexapeptide, and copper tripeptide-1 have the anti-wrinkle effect while DT-19 and NT-1 are applicable for potential BAPs focused on the whitening effect. The three kinds of BAPs like CT-1, DT-19, and NT-1 consisting of amino acids are safe to the skin, and have more excellent stability than bio-materials which are found to be unstable and cause skin irritation. Due to the high biological activity of BAP in the field of skin care, its utilization will increase constantly.

함초 추출물 발효액의 기능성 (The Functionality of the Saltwort (Salicornia herbacea L.) Extract Fermented Juice)

  • 각종 미네랄과 무기염류의 함량이 풍부한 것으로 알려진 함초를 이용하여 발효 후 그에 따른 기능성을 검토해 보고자 하였다. 유산균 접종 후 pH는 초기 4.50에서 발효 후 2.84로 변했으며, 산도는 초기 0.29에서 발효 후 0.55로 증가하였다. 한편 생균수를 측정한 결과 초기 $1.2{\times}10^{11}CFU/mL$이었던 생균수는 발효 72시간 후 $1.4{\times}10^{13}CFU/mL$를 보였다. 함초 추출물 발효액을 동결건조시킨 후 그에 따른 기능성을 검토하였으며 SOD-유사활성과 DPPH-라디칼 소거능 측정결과, 농도 의존적인 경향을 보였으며 혈전분해능 측정결과 발효 전의 함초 추출물에서는 나타나지 않았던 혈전분해능을 발효 후 동결건조분말 25%의 시료 농도에서 plasmin $3.84{\mu}g/mL$의 농도와 동일한 수준의 혈전용해능을 보였다.
