Effect of dietary legumes on bone-specific gene expression in ovariectomized rats

  • In previous studies, we found that the consumption of legumes decreased bone turnover in ovariectomized rats. The purpose of the present study is to determine whether the protective effects on bone mineral density (BMD) and the microarchitecture of a diet containing legumes are comparable. In addition, we aim to determine their protective actions in bones by studying bone specific gene expression. Forty-two Sprague-Dawley rats are being divided into six groups during the 12 week study: 1) rats that underwent sham operations (Sham), 2) ovariectomized rats fed an AIN-93M diet (OVX), 3) ovariectomized rats fed an AIN-93M diet with soybeans (OVX-S), 4) ovariectomized rats fed an AIN-93M diet with mung beans (OVX-M), 5) ovariectomized rats fed an AIN-93M diet with cowpeas (OVX-C), and 6) ovariectomized rats fed an AIN-93M diet with azuki beans (OVX-A). Consumption of legumes significantly increased BMD of the spine and femur and bone volume of the femur compared to the OVX. Serum calcium and phosphate ratio, osteocalcin, expression of osteoprotegerin (OPG), and the receptor activator of nuclear factor ${\kappa}B$ ligand (RANKL) ratio increased significantly, while urinary excretion of calcium and deoxypyridinoline and expression of TNF-${\alpha}$ and IL-6 were significantly reduced in OVX rats fed legumes, compared to OVX rats that were not fed legumes. This study demonstrates that consumption of legumes has a beneficial effect on bone through modulation of OPG and RANKL expression in ovariectomized rats and that legume consumption can help compensate for an estrogen-deficiency by preventing bone loss induced by ovarian hormone deficiency.

The Effects of Artemisia Princeps var. Orientalis Extracts on Serum Lipids and Connective Tissues Collagen in Ovariectomized Rats (쑥이 갱년기 장애 유도 흰쥐의 혈중 지질 및 결합조직 중 Collagen 함량에 미치는 영향)

  • The aim of this study was to evaluate the effects of Artemisia Princeps Var. Orientalis (AP) on serum lipids and the collagen content of the connective tissues in ovariectomized rats. From day 2 until day 37 after the ovariectomy, Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), and ovariectomized rats supplemented with the AP 50 mg/kg bw/day (OVX-AP). The AP ethanol extracts were orally administrated 1 mL per day. The OVX rats were significantly heavier than the sham-operated rats at all time points, but supplementation with the AP extracts tended to gain weight less than OVX-control. Although total-cholesterol was increased at OVX-control, supplementation with the AP extracts tended to result in less than OVX-control. Triglyceride was significantly decreased after supplemented with the AP extracts (p<0.05). HDL-cholesterol is appeared higher AP extracts group than OVX-control. According to the results, we could know the fact that AP extracts were effective on serum lipids content throughout decreasing total-cholesterol, triglyceride and increasing HDL-cholesterol in ovariectomized rats. Supplementation with the AP extracts prevented a decrease in the collagen level in bone and cartilage tissues. These results are consistent with the conclusions based on the estrogenic activities of AP. Therefore, it may be used to possibly improve the quality of life in menopausal women.

The Effect of Aloe on the Bioactivity of Ovariectomized Rats (난소를 절제한 흰쥐의 생리활성에 미치는 알로에의 영향)

  • In order to observe the bioactivity of ovariectomized rats, ovariectomized group (Ovx), nonovariectomized group(Sham) and ovariectomized aloe-treat group(Ovx+Aloe) were made. Lipidperoxides of liver and kidney, serum AST.ALT, BUN, total cholesterol and HDL-cholesterol were investigated as follows. Lipidoxides of the liver and the kidney in Ovx group were 1.74 times and 1.38 times increased compared to Sham group respectively. But they were significantly decreased in Ovx+Aloe group compared to Ovx group. Serum AST and ALT in Ovx group were increased 1.49 times and 1.65 times respectively compared to Sham group respectively. But they were decreased compared to Ovx group. Serum total cholesterol in Ovx group was increased 1.48 times compared to Sham group. While it was increased 50% in Ovx+Aloe group. Serum HDL-cholesterol in Ovx group was decreasd 23$\%$ compared to Sham group. While it was increasd 58% in Ovx+Aloe group compared to Ovx group. Serum BUN in Ovx group was increased slightly compared to Sham group. In Ovx+Aloe serum BUN had no change compared to Ovx group.

Effect of Tower Climbing Exercise, Aerobic Exercise & Alpha Lipoic Acid Administration on Bone Metabolism Factor in Ovariectomized Rats (자율적 등반운동과 유산소성 운동시 alpha lipoic acid 처치가 난소절제 흰 쥐의 골 대사 변인에 미치는 영향)

  • The purpose of this study was to investigate the change of bone metabolic parameters between climbing exercise training and treadmill running training with alpha lipoic acid supplementation in ovariectomized rats. Fifty-six Sparague-Dawley, 8 weeks of age, were assigned into nine groups: normal control at pre-test (Pre-NC), normal control at post-test (NC), sham-sedentary (SS), ovariectomized-control (OVX-Con), ovariectomized-ALA inject (OVX-ALA), ovariectomized-climbing exercise (OVX-CE), ovariectomized-ALA inject+climbing exercise (OVX-ACE), ovariectomized-treadmill exercise (OVX-TE) and ovariectomized-ALA inject+treadmill exercise (OVX-ATE). The climbing exercise group voluntarily climbed the 200 cm tower to drinking water from the bottle set at the top of it. The treadmill exercise group was performed the 60 min treadmill running with grade 6-8%, and velocity 10-15 m/min. The frequency of exercise training was performed 6 days per wk in both training intervention. For the comparison of bone metabolic responses following to different training intervention, the strength, length and weight of femur, total bone density, bone mineral contents, deoxypyridioline, osteocalcin and parathyroid hormone, estradiol and lutenizing hormone were measured in this study. The body weight was higher in the OVX-Con, OVX-ALA and OVX-TE groups as compared to NC group. The total bone mineral density of OVX-ACE showed a higher value than SS, OVX-Con, OVX-ATE and OVX-TE. But urine and blood metabolic parameters showed no significance among groups. In conclusion, this study results suggested that climbing exercise training and alpha lipoic acid supplementation may an effective intervention for prevention of osteoporosis in ovriectomized rats.

Effects of Nanocalcium Supplemented Milk on Bone Calcium Metabolism in Ovariectomized Rats

  • This study examined effects of calcium supplemented milk on bone loss in ovariectomized rats. Twenty four Sprague-Dawley female rats, 7 weeks-old, were divided into 4 groups, ovariectomized and fed diets containing: 1) control, no Ca supplemented milk, 2) ovx 1, Ca carbonate supplemented milk, 3) ovx 2, ionized Ca supplemented milk, and 4) ovx 3, nano Ca supplemented milk. All rats were fed 1 ml of milk containing 20 mg supplemented Ca. After 18 wk feeding, body weight gain and food efficiency ratio were significantly different between ovx 1 and ovx 3. Serum concentration of calcium and phosphorus were not different among groups. However, there was a significant difference in calcium content of dry femoral weight in ovx 3 compared with the control and ovx 2. In addition, femoral bone mineral density ($g/cm^2$) was significantly greater in ovx 3 than in other groups (p<0.05). The ovx 3 group showed the highest stiffness (N/mm), maximum energy (N) in femur and trabecular bone area (%). The present study indicated that nano Ca supplementation in milk may be an effective way to enhance bone calcium metabolism for ovariectomized rats.

Effects of Punica granatum L. Extracts on Serum Lipids Level in Ovariectomized Rats (석류가 갱년기 장애를 유도한 흰쥐의 혈 중 지질 함량에 미치는 영향)

  • This study investigated the effects of Punica granatum L. (PG) ethanol extracts on serum lipid levels in ovariectomized rats. In order to elucidate the effects of PG ethanol extracts on the change of lipid concentration in rats, Sprague-Dawley female rats were randomly assigned to groups as follows: sham-operated rats (sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with ethanol extracts of PG seed at 50 mg/kg bw/day (OVX-PS50), ovariectomized rats supplemented with ethanol extracts of PG seed at 200 mg/kg bw/day (OVX-PS200), ovariectomized rats supplemented with ethanol extracts of PG peel at 50 mg/kg bw/day (OVX-PS50), ovariectomized rats supplemented with ethanol extracts of PG peel at 200 mg/kg bw/day (OVX-PP200). PG ethanol extracts were orally supplemented at the level of 1mL per day for 7 weeks. The body weight gain, food intake and food efficiency ratio were not significantly different between groups by t-test. The levels of total-cholesterol and triglyceride in serum were not changed by supplementation of PG ethanol extracts. The level of HDL-cholesterol on serum of the OVX-PS50 and OVX-PS200 groups were similar to the OVX-control group, but the OVX-PP50 and OVX-PP200 groups were higher than sham group. These results suggest that ethanol extracts of PG peel were more effected than those of PG seed.

The Effect of Sea Tangle Extract on Serum Lipid Level in Ovariectomized Rats (다시마 추출물이 난소를 절제한 흰 쥐의 혈 중 지질 함량에 미치는 영향)

  • This study was performed to investigate the effects of sea tangle (ST) extracts on serum lipid concentration in ovariectomized rats. Weight-matched female Sprague-Dawley strain rats were assigned to four groups. Three groups were surgically ovariectomized (OVX). The fourth group was sham operated. Rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with ST at 50 mg/kg body wt (OVX-ST50) and ovariectomized rats supplemented with ST at 200 mg/kg body wt (OVX-ST200). The activities of serum GOT and GPT were increased by ovariectomized. The serum GOT levels in OVX-ST50 and OVX-ST200 groups were decreased than OVX-control group. But GPT level was only decreased in the serum of OVX-ST200 group. Serum total cholesterol and triglyceride contents decreased in sham group compared with OVX-control group by ovariectomy. Six weeks feeding of ST extract resulted in a significant lowering of serum triglyceride and lowering tendency of total cholesterol Level. The serum HDL-cholesterol was higher in ST groups than OVX-control group. These results suggest that the beneficial effect of ST extract may be used to possibly improve on the lipid metabolic syndrome of menopausal women.

Effects of Mulberry fruits Extracts on Serum Lipid Level in Ovariectomized Rats (난소를 절제한 흰쥐에서 오디가 혈 중 지질 함량 변화에 미치는 영향)

  • The aim of this study was to evaluate the effects of three different mulberry cultivars extracts on serum lipid contents in ovariectomized estrogen-deficient rats. Sprague-Dawley female rats were randomly assigned to the following groups: sham-operated rats (sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with $ 80\% $ ethyl alcohol extracts from various kinds of Tajikistan mulberry (OVX-TM), Korea mulberry (OVX-KM) and China mulberry (OVX-CM) at 200mg/kg bw/day, respectively. The mulberry extracts were orally administrated at 1mL per day. The body weights of OVX rats were significantly heavier than the sham-operated rats at all values (p <0.05). The ovariectomy caused an expected increasing in the levels of serum total cholesterol and triglyceride. The serum HDL-cholesterol level on the OVX-TM, OVX-KM and OVX-CM groups were higher than in the OVX-control group.

Effects of L-carnitine on the ovariectomized rat (L-carnitine이 난소가 제거된 흰쥐에 미치는 영향)

  • This was conducted to determine the effects of body weight, organ weight, hematological values and biochemical parameters by L-carnitine in the ovariectomized (OVX) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. OVX group (n=10) received operation and no treatment. OVX+Carn group (n=10) received operation and L-carnitine. Body weight was significantly lower in OVX+Carn group than in all other groups. Also, organ weight such as heart, liver, spleen and kidney was measured. The heart and spleen weight were significantly lower in the OVX+Carn group than in the Intact and Sham group. The liver weight in the OVX+Carn group was significantly differences in comparison with those in the other groups. Also, there was significantly differences in the organ weight of kidney between in the OVX+Carn group and in the other groups. The hematological values of WBC, RBC, MCV, MCH and MCHC were no significant differences in any other groups. The total cholesterol, triglyceride and high density lipoprotein decreased significantly in the OVX+Carn group as compared to those in the OVX group. But, there were no significant differences in low density lipoprotein in any other groups. We conclude that L-carnitine enhanced the body weight in the ovariectomized rats. Our findings suggest that L-carnitine may influence the process of absorption of fat in the ovariectomized rats.

The Effect of Capsosiphon fulvecense extract on Inhibition of Platelet Aggregation and Serum Lipid Level in Ovariertomized Rats (매생이 추출물이 난소를 절제한 흰쥐의 혈소판 응집과 혈청 내 지질 조성에 미치는 영향)

  • Estrogen deficiency in peri-and postmenopausal women results in variety of neurovegetative, psychic and somatic symptoms, and may contribute to severe diseases within the aged female population. Capsosiphon fulvecense (CF), green algae, loaded with large amount of proteins, calcium and ion. This seaweed is distributed in the Southern coast in Korea and widely throughout the world. Our objective was to investigate the effects of Capsosiphon fulvecense (CF) extract on serum lipid concentration and platelet aggregation in ovariectomized rats. Three groups were surgically ovariectomized (OVX). The fourth group was sham operated. Sprague-Dawley female rats were randomly assigned to the following groups : sham-operated rats (Sham), ovariectomized control rats (OVX-Control), ovariectomized rats supplemented with CF at 50mg/kg bw/d (OVX-CF5O) and ovariectomized rats supplemented with CF at 200mg/kg bw/d (OVX-CF200). The serum triglyceride level was significantly decreased after supplemented with the CF extracts. The serum HDL-cholesterol level in the OVX-CF group was significantly higher than the level in the OVX-control group. The ability of platelet aggregation treated with CF extract (CF group) was $7\%$ less than OVX-control group. Taken these together, CF extract may be used to improve the quality of life in menopausal women.
