Effects of Manchurian Trout Gonadotropins on Sexual Maturation in Female Rainbow Trout

  • Manchurian trout (Brachymystax lenok) is an endangered fish species in East Asia including the Korean peninsula. To establish a method for artificial propagation and to improve our understanding of the reproduction in the species, we have produced recombinant gonadotropins, follicle-stimulating hormone (r-mtFSH) and luteinizing hormone (r-mtLH), which may play central roles in reproductive activities. In the present study, the biological activities of the recombinant hormones were analyzed by gonadosomatic index (GSI), ovarian follicle diameter, and sex steroid levels in mature rainbow trout (Oncorhynchus mykiss). In the 6th day post-injection, FSH-injected fish were slightly decreased in the GSI value, although there were no significant differences among those of control, r-mtFSH, and r-mtLH treatments. Injection of the r-mtFSH increased follicle diameters significantly as compared with those of control- and r-mtLH-injected fish. The plasma steroid levels showed wide differences in the groups at 1, 3, or 6th day post-injection. Despite the variable steroid levels, three individuals receiving either r-mtFSH or r-mtLH showed a great increase in a maturation-inducing steroid, $17{\alpha},20{beta}$-dihydroxy-4-pregnen-3-one, at 3 and 6 days. Taken together, these results suggest that biological efficacies of the recombinant FSH and LH should be further studied in the Manchurian trout.

Gonad Maturation Cycle of the Sea Urchin Strongylocentrotus nudus Population Inhabiting an Artificial Seaweed Forest, Samchuk, Korea

  • We determined the seasonal gonad maturation in Strongylocentrotus nudus sea urchins inhabiting an artificially enhanced seaweed forest along the Samchuk Coast of Korea from April 2006 to March 2007. A total of 30 sea urchins per month were collected from the study area, and gonadosomatic index (GSI), gonad index (GI), egg diameter, and RNA/DNA variation were measured for each specimen. GSI values of female and male urchins achieved maximums of 17.6 and 17.0, respectively, in June. Based on histological studies, maximum GI values occurred in July (4.6 for females and 4.8 for males). A mean ovarian egg diameter of $73.7\;\pm\;14.2\;{\mu}m$ was measured in August; during the main spawning period in September, mean egg diameter reached a maximum of $74.2\;\pm\;17.8\;{\mu}m$. The RNA/DNA ratio and RNA content for both males and females showed a distinct peak during the ripe stage in July, but another peak occurred in the spring season from March to April, when urchins deposit protein into the nutritive phagocytes of immature gonads prior to gametogenesis. The reproductive cycle of S. nudus is divided into five stages: early active (December-May), late active (March-July), ripe (July-September), spent and degenerative (August-November), and inactive (October-February). Our continuous removal of sea urchins from the study area did not influence the reproductive cycle, as populations quickly recovered, and achieved normal gonad development cycle in the site.

  • The condition factor (CF) were calculated by the morphometric data, and gonadal maturation and reproductive potential associated with fecundities of indoor cultured Hapalogenys nitens were investigated by microscopic and morphometric analysis from August 2011 to October 2012. Monthly changes in the GSI and CF were closely related to gonadal maturation and spawning. The ovary consisted of a pair of saccular structure with many ovarian lobules, the testis is a pair of lobular structure with many testicular lobules. The relationship between total fecundity (F) and total length (TL) of fish was expressed as $F=0.0449TL^{2.7926}$($r^2=0.0653$), and body weight (BW) was expressed as $F=1.4492BW^{0.8964}$($r^2=0.0863$). In this study, some exceptional special cases and phenomena were observed: absolute and relative fecundities were not increased with the increases of total length and body weight, respectively. Absolute and relative fecundities were no concerned with the increases of age classes of total length (cm) and of body weight (g).

양식산 농어, Lateolabrax japonicus 암컷의 난소발달과 혈중 스테로이드 호르몬 양상 및 난모세포 성숙 및 배란유도 (Relationship between Ovarian Development and Plasma Levels of Steroid Hormones, and Induction of Oocyte Maturation and Ovulation in the Cultured Female Korean Sea Bass, Lateolabrax japonicus)

  • 양식산 농어, Lateolabrax japonius 암컷의 난소와 혈액을 1997년부터 1999년까지 매년 10월부터 2월까지 2회 반복 채취하였다. Gonadosomatic index는 11월부터 증가하기 시작하여 12월(12.8$\pm$1.5)과 1월(14.8$\pm$3.5)에 최고 수준을 나타낸 후 2월(2.6$\pm$1.5)에는 급격히 감소하였다(P<0.05). 난소내 난모세포는 12월과 1월에 3차 난황구기까지 발달하고 성장이 완료되지만, 성숙 및 배란이 되지 않고 2월에는 퇴화되었다. 혈중 estradiol-l7$\beta$의 농도는 11월부터 증가하기 시작하여 12월 (1,152.3$\pm$107.2 pg/ml)과 1월(1,315.4$\pm$99.5 pg/ml)에 최고 수준을 나타낸 후 2월에는 급격히 감소하였다(P<0.05). 17 $\alpha$,20-dihydroxy-4-pregnen-3-one는 실험기간 동안 낮은 농도(86.6$\pm$6.5~93.8$\pm$2.8 pg/ml)를 유지하며 유의한 변화는 없었다(P<0.05). 성장이 완료된 난모세포를 갖고 있는 양식산 농어에 HCG를 주사(1,000 IU/kg과 2,000 IU/kg)하면 모든 개체에서 성숙 및 배란이 유도되었다. HCG 농도에 따른 수정율과 부화율은 차이가 없었으나, 부상란 수는 1,000 lU/kg에서 325,000$\pm$26,000개, 2,000 IU/kg에서 195,000$\pm$35,000개로 차이가 있었다. 이상의 결과를 종합하면, 양식산 농어 암컷은 양식환경 하에서도 난황형성 및 난모세포의 성장은 정상적으로 진행되지만, gonadotropin의 surge가 일어나지 않아 최종성숙과 배란이 되지 않는 것으로 추정되었다. 농어에서 난모세포의 인위적 성숙 및 배란 유도에 의한 난 생산은 HCG 1000 IU/kg가 효과적이었다.

누에에서 생산된 뱀장어 재조합 생식소자극호르몬이 암컷 뱀장어(Anguilla japonica)의 성성숙 유도에 미치는 영향 (Induction of Sexual Maturation in Female Eels Anguilla japonica by Recombinant Eel Gonadotropin Produced in Silkworm)

  • This study investigated the effects of recombinant eel gonadotropin hormone (rJeGTH) produced in silkworms, with and without a carboxyl-terminal peptide from equine chorionic gonadotropin (eCG), on the induction of sexual maturation in female eels Anguilla japonica. Experiments were conducted both in vivo and in vitro. In in vitro trials, germinal vesicle breakdown (GVBD) induction did not significantly differ between rJeFSH and $rJeFSH{\cdot}eCG$ treatments and the control group. However, previous studies did find that rJeLH and $rJeLH{\cdot}eCG$ treatments induced GVBD in female eels. Our in vitro exploration of $estradiol-17{\beta}$ ($E_2$) levels in immature ovarian tissues revealed significantly higher $E_2$ levels in the group treated with $rJeFSH{\cdot}eCG$ $1{\mu}g/mL$ than in the control group. In contrast, the in vivo experiments showed no effect of recombinant hormones on the sexual maturation of feminized eels. Previous studies and our own in vitro results have clearly shown that rJeGTH and $rJeGTH{\cdot}eCG$ have a positive effect on the sexual maturation of feminized eels. To develop the activity of rJeGTH in vivo, further studies should confirm circulation time and activity of these hormones in eels' bloodstream, modify the structure of the recombinant gene, and implement additional glycosylation.

불임 여성의 난소로부터 회수된 미성숙 난자의 체외 성숙과 배양에 관한 연구 (Study on In Vitro Maturation and Culture of Immature Oocytes Collected from Ovaries of Infertile Women)

  • Objective: This study was performed to examine the maturation and the development to the blastocyst stage of immature oocytes collected from patients with high risk of ovarian hyperstimulation syndrome (OHSS). Materials and Methods: Cumulus-oocyte complexes (COCs) were collected following only HCGpriming for non stimulated IVF-ET cycles of the patients. At the time of oocyte collection, COCs were classified into three groups in accordance with their appearance (Group I: oocytes with dispersed cumulus cells; Group II: oocytes with compacted cumulus cells; Group III: oocytes with sparse cumulus cells). The in vitro maturation and blastocyst development rates of the COCs were compared among these groups. From August 2001 to June 2002, 48 IVM/IVF-ET cycles from 42 patients (mean age: $32.4{\pm}3.8$ years) were performed. To prevent the occurrence of OHSS, the patients were primed with 10, 000 IU HCG alone 36 h before oocyte collection without gonadotropin stimulation. Oocytes were aspirated on cycle days from 7 to 13. The normal COCs were classified into three groups according to their appearance. The aspirated immature oocytes were cultured in YS maturation medium containing 30% (v/v) human follicular fluid (HFF), 1 IU/ml FSH, 10 IU/ml HCG and 10 ng/ml rhEGF. Fertilization was induced by intracytoplasmic sperm injection (ICSI). All zygotes were co-cultured with cumulus cells in $10{\mu}l$ YS medium containing 10% HFF until day 7 after oocyte collection. Blastocyst transfer was performed on day 5 after ICSI. Results: Th e mean number of oocytes cultured in the IVM/IVF cycles was $24.7{\pm}10.6$. Of 1185 COCs, those assigned to Group I, II and III were 470 (39.7%), 414 (35.0%) and 301 (25.4%), respectively. The maturation rate (94.5%, 444/470, p<0.05) in Group I was significantly higher than those of Group II (62.8%, 260/414) and Group III (73.1%, 220/301). Especially, 30.9% of COCs in Group I (145/470) was matured on the day of oocyte aspiration. There were no differences in the rates of fertilization and cleavage among the three groups. The development rate to the blastocyst stage in Group I (54.6%, 206/377, p<0.05) was also significantly higher than those in Group II (33.0%, 68/206) and Group III (30.1%, 52/173). Twenty-four clinical pregnancies (50.0%) was obtained and 22 pregnancies (45.8%) are ongoing. Implantation rate in the present study was 24.6%. Conclusion: These results suggest that there is a positive correlation between the appearance of COCs and the developmental competence of the immature oocytes in non stimulated IVM/IVF cycles.

異種動物의 眼前房에 이식된 濾胞卵子의 成熟에 관한 연구 (Studies on the Maturation of the Follicular Oocytes by Xenoplastical Transplantation in the Anterior Chamber of the Eye)

  • 안전방안에서 여포난자가 성숙한다는 것이 몇가지 동물을 이용한 실험에서 밝혀진 바가 있었거니와 여포 난자가 異種의 안전방내에서도 성숙이 가능한 가를 보기 위하여 본 실험을 행하였다. 토끼의 경우 자체의 안전방내에 이식한 여포난자가 다른 동물 즉 흰쥐나 흰생쥐의 안전방에 이식하였을 때보다 낮은 성숙률을 보여 주고 있을 뿐, 흰쥐, 흰생쥐들은 각각 상호간의 안전방에 이식하였을때 同種의 다른 개체의 안정방에서 얻은 결과보다 훨씬 좋은 성숙률을 나타내고 있다. 특히 흰쥐의 여포난자는 숫컷 흰쥐의 안전방에서 24시간 사이에 76%가 제 1및 제 2차 분렬의 증기에 있으며 같은 종류의 수컷 생쥐의 안전방에서 55%가 성숙한 것 보다 훨씬 높은 율을 보여주고 있는 것이다. 안전방의 전방수가 함유단백질의 양이거나 삼투압에 있어서 일반체액이거나 표준배양액의 것들과는 크게 다르며 세포나 조직의 배양액으로는 그다지 적당한 것으로 보이지 않으나 여포난자의 활성획득에는 큰 영향이 없다할 것이다. 특히 異種간의 이식실험에서 얻은 결과가 자체거나 同種간의 이식실험과 거의 같거나 오히려 더 나은 성과를 보여주고 있는데, 는 그 이유에 대해 더 밝혀야 할 문제로 본다. 단지 난자는 성숙분렬을 위한 활성을 갖는데 난자자체의 구성성분 즉 난황물질의 양에 크게 좌우되는 것같으며 또한 난자가 배양될 안전방의 환경요인이 성숙률을 결정하는 데 큰 영향을 주는 것으로 보인다.

이스라엘 잉어(Cyprinus carpio) 난모세포의 계절에 따른 조직학적 변화 및 호르몬 수준의 변화 (Histological Changes of Oocytes Development and Hormone Levels in the Israeli carp, Cyprinus carpio, according to Season)

  • This study was undertaken to understand development stage of ovary and changes of hormones concentration from February to May in the Israeli carp, Cyprinus carpio. The results obtained in these experiments are as follows ; 1. Serum LH levels began to increase sharphy in March, coinciding with the onset of rapid ovarian development. 2. LH levels were well correlated with changes in gonadosomatic index. 3. Dramatic increase in gonadosomatic index occurred during the months of March. 4. Ripe stage(Stage Ⅴ) rapidly increase in March. 5. Early perinucleolus oocyte radpily develop into late perinucleolus oocytes in March. 6. The vitellogenic phase begins as these late perinucleolus oocytes become transformed into early maturing oocytes through the accumulation of yolk. 7. The cytoplasm completely fills with yolk as oocytes reach the late maturing stage. 8. Changes in the microscopic appearances of the ovaries were well correlated with changes in both gonadosomatic index and macroscopic appearance. 9. It is concluded from these observations that LH plays a major role in sexual maturation of the Israeli carp.

Ketamine과 Pentobarbitone이 생쥐 난자의 퇴화 및 과립세포의 자연세포사에 미치는 영향 (Effects of Ketamine and Pentobarbitone on Degeneration of Oocyte and Apoptosis of Granulosa Cells in Mouse Ovary)

  • In mammal, lots of follicles start simultaneously their growth but only a few oocytes are ovulated in every sexual cycles. Most of matured and grown oocytes are destined to degenerate by atresia. However, the molecular and physiological mechanisms are not elucidated yet. The present study was designed to establish an induction method of follicular atresia with ketamine or pentobarbitone and evaluate the effect of these anesthetics on oocyte maturation and granulosa cell apoptosis of the mouse ovarian follicle. The percentages of degenerated oocyte and apoptotic granulosa cell in ketamine treated groups were significantly higher than that in controls (58.9% vs 33.5%, p<0.01, degeneration; 44.9% vs 26.6%, p<0.01, apotosis). Futhermore, it was revealed that the concentrations of progesterone in both groups were markedly higher than that in control. In cunclusion, it is considered that ketamine induce an atresia as pentobarbitone, and may be useful for inducing follicular atresia.

번식기에 개구리 여포의 스테로이드 생성변화와 난자의 성숙능 획득 (Steroidogenic Shift and Acquisition of Meiotic competence by Cultured Ovarian follicles of Rana at Breeding Season)

  • 3 종의 한국산개구리를 사용하여 여포들이 생성하는 스테로이드와 난자의 성숙능이 번식기 근처에서 어떻게 변하는가를 조사하였다. 개구리의 여포를 생체외 배양하면서 이들이 뇌하수체추출물(frog pituitary homogenate, FPH) 혹은 CAMP의 자극으로 생성하는 progesterone(Pa)과 testosterone(T)의 양을 측정하여 이로부터 P4yT의 비율을 구하였으며 여포난자들이 FPH. P4 혹은 12-0-tetrsdecanoyl phorbol-13-acetate(TPA)에 반응하여 성숙을 일으키는 시기를 조사하였다. 북방산개구리의 여포난자는 11월 부터 호르몬에 반응하여 성숙을 일으켰으며 2월에 PJT의 비율이 급격히 증가하였다 참개구리는 5월에 가서of PJT의 비율이 현저히 증가하고 FPH에 반응하여 성숙을 일으켰다. 그러나 P4에 의한 난자성숙은 3월부터 시작하였다 옴개구리는 7월에 P4/T 비율의 증가가 있었으며 이때부터 또한 TPA에 반응하여 난자의 성숙을 일으켰다 따라서 3종의 개구리에서 모두 각각의 번식기 근처에서 PJT의 비가 급격히 증가하고 또한 성숙능이 생긴다는 것을 알 수있었다.
