Osteoporosis is a metabolic bone disease associated with an imbalance of bone remodeling. Osteoporosis is characterized by decreased bone mass and increased bone fractures. In this study, we investigated the effects of horse bone extracts (HBEs) in vivo. Horse bone was extracted with 80% alcohol (HBE-A) at $100^{\circ}C$ or water (HBE-W) at $120^{\circ}C$. Animal model of postmenopausal osteoporosis was used, in which osteoporosis was induced by ovariectomy of female S.D. rats (female rats were divided into 5 groups), and HBEs were administered to ovariectomized rats every day for 8 weeks. After 8 weeks, the rats were sacrificed and the following osteoporotic factors were measured: body weight, bone mineral density (BMD), uterine/body weight ratio, serum estradiol (E2), and serum alkaline phosphatase (ALP). The results showed that the administration of HBE-W decreased the changes of body weight in ovariectomized rats. HBE-W increased the uterine/body weight ratio and BMD. In addition, HBEs decreased the ALP. Therefore, HBEs may be used for the prevention or treatment of bone disease.

In the present study, the correlation between the improvement of morphological characteristics and the effect of WBV was investigated and analyzed in OVX rats. The rats were randomly assigned to 5 groups: control, sham, WBV 17Hz, WBV 30Hz and WBV 45Hz. The WBV groups were exercised on a vibration platform (magnitude : $1mm_{peak-peak}$, frequency : 17Hz, 30Hz and 45Hz, 30 minutes/day for 5 days/week). The 4th lumbar on rats was scanned by In-vivo Micro-CT at the week 0(just before WBV) and the week 8(after WBV). Structural parameters of the 4th lumbar, based on two dimensional (2D) scan image data, were investigated and analyzed. The quantitative decrement rate of trabecular bone on WBV groups with 30Hz and 45Hz were lower than control and sham groups. The results showed the positive effect of WBV on osteoporotic bones of OVX rats.

The purpose of this study was to determine the effects of aqua-acupuncture of Laennec N HO (Hominis placenta Cervi pontotrichum cornu Carthami semen). on trabecular area and physiological change in ovariectomized osteoporotic rats. This experiment was performed to .investigate an antiosteoporosis. effects of Laennec N HO(Hominis placenta Cervi pontotrichum cornu Carthmi semen) aqua-acupuncture on the osteoporosis induced by both ovariectomy in Rats. So rats were divided into Sham group and OVX group. OVX groups were divided into control group, Exp. I group, Exp II group and Exp III group). The Ratios of trabecular area seen in ephiphysis and diaphysis of tibia and lumbar vertebral body and physiological change were as follows. 1. In the proximal ephiphysis of tibia, Exp I II III groups were significantly increased compared to that in control group (p<0.05). 2. In the diaphysis of tibia, Exp III group were significantly increased compared to that in control group (p<0.05). 3. In the first lumbar vertebral body, Exp II group were significantly increased compared to that in control group. 4. In the cortical thickness index of tibia, Exp III III groups were significantly increased compared to that in control group. 5. Osteoca1cin was increased in the Exp II group than those of other Experiment groups and control group, and Phosphorus was decreased in the Exp I II group than that in control group, But a significant difference was not seen. GOT, GPT and Alkaline phosphatase were decreased in all Experiment groups than that in control group. But a significant difference was not seen. From the above results, it might be suggested that aqua-acupuncture Laennec N HO(Hominis placenta Cervi pontotrichum cornu Carthami semen) prevented the increase in bone turnover and the decrese in bone mass induced by OVX in rats.

We evaluated the effects of organic Ca supplements chelated with milk protein (CaMP) in ovariectomized osteoporotic rats. Eight week-old Sprague-Dawley female rats were ovariectomized and fed a low $CaCO_3$ diet (0.1%) for 4 weeks to create an osteoporotic model. At that point, L4-$CaCO_3$ rats were sacrificed and the rest of the rats were divided into 4 groups, each of which was fed an experimental diet for 4 weeks: low-$CaCO_3$ (0.1%; L8-$CaCO_3$) and CaMP at 3 Ca levels: low (0.1%; L8-CaMP), normal (0.5%; N8-CaMP), and high (1.5%; H8-CaMP). Daily weight gain, serum ALP, weight and breaking force of femurs, Ca content of the lumbar, and Ca absorption were measured. Daily weight gain increased in the N8-CaMP and H8-CaMP groups compared to the low Ca groups. The ALP activity in the CaMP-fed rats was significantly lower than in the $CaCO_3$-fed rats. Both breaking force and femur weight were higher in the N8-CaMP and H8-CaMP groups compared to the L8-$CaCO_3$ group. Ca content of the lumbar increased dose-dependently with Ca intake levels of CaMP. Ca absorption rates of the CaMP-fed rats increased more than that of the rats fed low Ca levels of $CaCO_3$. These results demonstrate that the CaMP supplement had positive effects on bone metabolism and Ca bioavailability in ovariectomized osteoporotic rats. Therefore, CaMP may be recommended as a useful Ca supplement to prevent bone loss in osteoporosis.
To investigate the effect of dietary Ca levels on metabolic changes of Ca and skeleton in postmenopausal women, 10-month-old ovariectomized female rats were compared with 2 month old rats. The rats were fed either 0.2% or 1.2% Ca diets for 16 weeks. Food intake and weight gain as higher in rats fed high Ca diets and in ovariectomized rats. Apparent Ca absorption as higher, and Ca balance was lower in the low Ca groups. Vertebrae density was higher in old rats or those fed a high Ca diets. The old rats and ovariectomized rats showed decreased bone formation, increased bone resorption and kidney function deterioration resulting in increased urinary Ca excretion. Contradictory to the above observation, old rats and ovariectomized rats still showed higher bone mass and bone ash content. Therefore aging was not fully onging in 10-month-old rats. Bone weights, mineral contents, and mineral/wt ratio were lower in ovariectomized rats. Dietary Ca level did not affect urinary Ca excretion, urinary protein excretion, GFR, serum alkaline phosphatase, or urinary hydroxyporline excretion. This means that dietary Ca level did not influence kidney function or bone turnover. However Ca content and the ash content of femur, 4th vertebra, and scapula were increased in high Ca groups. Therefore, it is considered that decreased bone formation and accelerated bone resorption may account for the increased osteoporotic risk in women in menopause after middle age. However, Ca metabolism can be improved and bone components can be maintained if Ca is supplemented.
Objectives & Methods : The purpose of this study is to observe the effects of Drynariae Rhizoma Herbal-acupuncture infusion solution at Umgok($KI_{10}$) on Osteoporotic Rats Induced by Ovariectomy. The author performed several experimental items to analyze cytotoxic, osteoporosis evaluation, change of ALP, osteocalcin, Ca and histological change of tibia. Results : 1. BMD were increased meaningly in DR-HA than control group. 2. ALP was not inhibited by DR-HA, osteocalcin was decreased meaningly in DR-HA. 3. Osteoclast like cell in tibia was increased meaningly in control group, decreased meaningly in DR-HA. 4. In the histological study in tibia, TBV and TBT were significantly increased, GPL was significantly decreased in the DR-HA than control group. Conclusions : From the result above, the results suggest that DR-HA at $KI_{10}$ has significant effect on osteoporosis, and to be put to practical use in the future osteoporosis clinic.

The preventive effect of Salvia miltiorrhiza extracts (SMEs) on the progress of bone loss induced by ovariectomy (OVX) was studied in rats. We measured body weight and bone histomorphometry in sham, OVX or SMEs-administered OVX rats. From light microscopic analyses, a porous or erosive appearances were observed on the surface of trabecular bone of tibia in OVX rats, whereas those of the same bone in sham rats and in SMEs-administered rats were composed of fine particles. The trabecular bone area and trabecular thickness in OVX rats decreased by 50% from those in sham rats, these decreases were completely inhibited by administration of SMEs for 7 weeks. In this study, the mechanical strength in femur neck was significantly enhanced by the treatment of SMEs for 7 weeks. In OVX rats, free T3 was normal in all cases, whereas free T4 was significantly increased. Although there was no difference between OVX and SMEs-administered rats in T3 level, we have found significant difference between them in T4 level. These results strongly suggest that SMEs are effective in preventing the development of bone loss induced by OVX in rats.

The model rats of osteoporsis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with DBE, extract of Dioscorea batatas, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of DBE. Osteoporetic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns. DBE showed inhibitory effect on bone loss in osteoporetic condition, and reduced the increase of ALP activity and osteocalcin level in serum, and reduced the increase of OH-proline level in urine. But, DBE had no effect on cell proliferation and ALP activity in rat calvarial cell culture.
