Change of Fatty Acid Compositions during Hepatic Triacylglycerol Accumulation in Dietary Orotic Acid-induced Fatty Liver (오로토산 유발 지방산의 간장 트리아실글리세롤 축적간 지방산 조성의 변화)

  • Dietary orotic acid is known to induce the fatty liver. Fatty acid profiles in the lipid fraction of the liver and the serum in rats fed with or with orotic acid diet were analyzed. In all the hepatic lipid fraction of rats fed on the supplemented orotic acid diet, there was a significant increased in linoleic acid. In addition, linoleic acid was also increased in the triacylglycerol fraction of hepatic endoplasmic reticulum and the triacylglycerol and diacylglycerol fractions of hepatic Golgi apparatus of the orotic acid-feeding rats. In the time course study of the fatty acid profile in the hepatic triacylglycerol and diacylgycerol fractions, an increase of linoleic acid was observed similarly in the initial stages of orotic acid intake in the both fractions. However, linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats increased from day 1, but it began to decrease the increment from day 2, resulting in the lower level of linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats than that of rat fed a orotic acid-free diet after 10 days. Oleic acid (18:1) was increased in the only cholesteryl ester fraction of helpatic. However, oleic acid level in other fractions was not changed. The compositions of 14:0, 16:0 and 18:0 was reduced in the hepatic triacylogylcerol, diacylglycerol and cholesteryl ester fractions by orotic acid-feeding. However, these saturated fatty acids were significantly increased in the serum triacylglycerol fractions. The orotic acid indcued changes in linoleic acid level in hepatic triacylglycerol may be explained by the impaired fatty acid metabolism and limited excretion of this fatty acid from liver to serum.

  • It was generally known that over-ingestion of dietary orotic acid caused hepatic disorder by the lesion of fatty acid and cholesterol synthesis in rats. The present study was carried out to investigate the effects of dietary orotic acid on the lipid composition of serum, liver and kidney of rats. For the experiments, rats were fed with commercialized chow powder diet in the presence or absence of 1% orotic acid. The prepared diets were fed to male rats (Sprague-Dawley strain, $90{\sim}100$ g of body weight for 21 days. According to the results, orotic acid treated group showed that each concentration of serum cholesterol, triglyceride and phospholipid was significantly decreased (p<0.05). The centration of liver triglyceride was significantly decreased (p<0.05). In the presence of 1% orotic acid, the weight of liver inclosed while that of kidney decreased. The treatment of orotic acid seemed to have no effects on the phospholipid composition in liver and kidney, except the kidney lysophosphatidylcholine. In the comparison of the phospholipid composition between liver and kidney, the levels of sphingomyeline and phosphtidylethanolamine in the kidney were higher than those in the liver. Among the composition of fatty acid in kidney, the concentration of linoleic acid (18 : 2) was increased, and the concentration of arachidonic acid (20 : 4) was decreased with the addition of the orotic acid.

Effects of Taurine and Orotic Acid on Lipid Concentration of Hepatic and Serum in Mouse (마우스의 간장과 혈청 지질농도에 미치는 타우린 및 오로트산의 영향)

  • Male mice (ddY strain) were fed with the chow diet containing 10% sucrose supplemented with orotic acid at the 1% level or/and taurine at the 5% levels for 14 days. The concentrations of triacylglycerol and cholesterol in the liver were significantly lower in the orotic acid group than the control group. When taurine and orotic acid were administered simultaneously, the concentrations of triacylglycerol and cholesterol in the liver were higher and lower, respectively, compared to the orotic acid group. The concentration of triacylglycerol in the serum was higher in the taurine group than that of the control or the orotic acid groups, and the simultaneous supplementation of orotic acid and taurine further enhanced. There were no significantly difference in body weight gain, diary food intake, and the concentrations of serum cholesterol and hepatic phospholipid. These results suggest that dietary taurine stimulated the increasion of hepatic triacylglycerol by orotic acid in mouse.

  • Male mice (ddY strain) were fed a laboratory chow diet containing 10% sucrose supplemented with orotic acid, hesperetin or naringenin at the 1% level for 14 days. the concentrations of liver triacylglycerol and cholesterol were significantly lower in the OA group than in the control group. When both flavonoids and orotic acid were administered simultaneously, the orotic acid-dependent decrease in liver triacylglycerol and cholesterol were attenuated slightly. The concentration of serum cholesterol in the orotic acid group or the control group was lower than in the orotic acid groups supplemented with hesperetin or naringenin. There were no significantly difference in body weight gain, diary food intake, and the serum concentrations of triacylglycerol and high-density-lipoprotein cholestrol. It was concluded that the inducement of fatty liver in mice failed to feeding a laboratory chow diet containing 10% sucrose supplemented with 1% orotic acid for 14 days.

  • Effects of 1% dietary orotic acid on triglyceride metabolism were examined in SD-rats and Kud: ddY mice. When rats were fed semisynthetic diet containing 1% orotic acid and n-6 polyunsaturated fatty acid (linoleic acid), the hepared diet. In contrast to rats which respond to orotic acid consumption with increases in hepatic triglyceride content, mice did not so respond. The rats-limiting step in triglyceride synthesis is catalyzed by the enzyme phosphatic acid phosphohydrolase (EC3.1.3.4) which is present in the liver cytosol and microsomes of rats fed oroic acid diet. This finding suggests that the activity of this enzyme may play a tole in the fatty liver formation in rats.

  • Biopbysical investigation has been carried out for the interaction of riboflavin tetraacetate and an orotic acid derivative to know the binding mechanism between dihydroorotate dehydrogenase and dihydroorotic acid which are concerned with the oxidoreduction in vivo. Infrared and nuclear magnetic resonance spectra were measured for the chloroform solution of riboflavin tetraacetate, orotic acid butyl ester and its same mixture. Orotic acid butyl ester has been found to associate with riboflavin tetraacetate. They form a strong complex with each other. The complex is an 1:2 cyclic hydrogen bonded trimer through the imino and 2-C carbonyl groups of the isoalloxazine ring and the imino groups of the orotic acid derivative.

Improvement Effect of Fermented Silkworm (Bombyx mori L.) Powder against Orotic Acid-induced Fatty Liver in Rats (발효누에분말 투여가 orotic acid 유발 흰쥐에 미치는 지방간 개선 효과)

  • Current study was to investigate the potential effects of silkworm, Bombyx mori L. fermented with Bacillus subtilis (BFSP) and Aspergillus kawachii (AFSP) at the 5% (w/w) levels in Sprague-Dawley rats, which was intoxicated with 1% (w/w) orotic acid (OA) for 10 days. The rat group administered silkworm powder showed improvements in fatty liver condition. Consumption of fermented silkworm powder reduced triglyceride concentrations in the liver tissues and serum and increased the serum lipid concentrations to normal levels, thereby aiding in improving fatty liver conditions. These effects were more pronounced in the BFSP than that in SP or AFSP in orotic acid-induced hepatotoxicity and oxidative stress. Based on these results, fermented silkworms are considered to be a material with significant potential for development into a functional health food that can improve fatty liver conditions.

  • It has been reported in the technical literature that orotic acid can be used in order to induce improved crystallization of biodegradable and biocompatible polymers like poly(L-lactic acid), polyhydroxybutyrate and poly(hydroxybutyrate-co-hydroxyhexaonat). The expected advantage of the changed crystalline structure is a reinforcing effect of the polymers. A lot of papers reported about the application of inorganic and organic agents for acceleration of heterogeneous nucleation. This study reports on an attempt to use orotic acid as appropriate non-toxic nucleating agent for improving mechanical properties of isotactic polypropylene. Special attention is given to demonstrate the effect of nucleation in a typical melt spinning process in order to improve the mechanical properties. The effects were demonstrated using rheology, thermal analysis and tensile testing.

  • Pyrimidine auxotrophic basidiospores of Pleurotus sajor-caju were selected using positive selection method. Wild type basidiospores could not grow on minimal medium containing the pyrimidine analog 5'-fluoro-orotic acid (5'-FOA) whereas pyrimidine auxotrophs grew normally. After treatment of basidiospores with ultraviolet light, a total of 13 pyrimidine auxotrophic basidiospores were isolated among 24 5'-FOA resistant mutants. These mutants require the pyrimidine such as uracil, cytosine, thymine. Mating type group and growth rate of their mutants were determined.

  • Two new orotic acid derivatives orotyl-DL-alanine and orotyl-L-tyrosine were synthesized. They were obtained as high melting crystalline masses by condensing DL-alanine and L-tyrosine each with orotyl chloride in aqueous sodium hydroxide solution, followed by acidifying the reaction mixture.
