연잎에서 DPPH 라디칼 소거활성을 가지는 페놀성 화합물의 분리 (Phenolic Compounds from the Leaves of Nelumbo nucifera Showing DPPH Radical Scavenging Effect)

  • Nelumbo nucifera (Nymphaeaceae) has been widely used in a traditional oriental medicine to treat bleeding, fever, diarrhea, hemorrhoid, sunstroke, dysentery and dizziness. The leaves of this plant were refluxed with methanol, and then fractionated with organic solvents to screen the antioxidant activity using DPPH radical. Ethyl acetate and n-butanol fractions showed good DPPH radical scavenging effects and were carried out column chromatographies to isolate nine compounds. Their chemical structures were characterized as p-hydroxybenzoic acid (1), uracil (2), luteolin (3), quercetin $3-O-{\beta}-D-glucopyranoside$ (4), $rhamnetin 3-O-{\beta}-D-glucopyranoside$ (5), $isorhamnetin 3-O-{\beta}-D-glucopyranoside$ (6), $quercetin 3-O-{\beta}-D-glucuropyranoside$ (7), $quercetin 3-O-{\beta}β-D-xylofuranosyl(1{\rightarrow}2)-{\beta}-D-galactopyranoside$ (8), and adenine (9) by comparison NMR spectral data and with those in references. Compounds 1, 2, 5 and 9 were firstly isolated from this plant. Compounds 1, 3 and 4 showed potent DPPH radical scavenging activity. Especially, compound 3, luteolin showed the higher effect than ascorbic acid used as a positive control.

Towards Methionine Overproduction in Corynebacterium glutamicum - Methanethiol and Dimethyldisulfide as Reduced Sulfur Sources

  • In the present work, methanethiol and dimethyldisulfide were investigated as sulfur sources for methionine synthesis in Corynebacterium glutamicum. In silico pathway analysis predicted a high methionine yield for these reduced compounds, provided that they could be utilized. Wild-type cells were able to grow on both methanethiol and dimethyldisulfide as sole sulfur sources. Isotope labeling studies with mutant strains, exhibiting targeted modification of methionine biosynthesis, gave detailed insight into the underlying pathways involved in the assimilation of methanethiol and dimethyldisulfide. Both sulfur compounds are incorporated as an entire molecule, adding the terminal S-$CH_3$ group to O-acetylhomoserine. In this reaction, methionine is directly formed. MetY (O-acetylhomoserine sulfhydrylase) was identified as the enzyme catalyzing the reaction. The deletion of metY resulted in methionine auxotrophic strains grown on methanethiol or dimethyldisulfide as sole sulfur sources. Plasmid-based overexpression of metY in the ${\Delta}$metY background restored the capacity to grow on methanethiol or dimethyldisulfide as sole sulfur sources. In vitro studies with the C. glutamicum wild type revealed a relatively low activity of MetY for methanethiol (63 mU/mg) and dimethyldisulfide (61 mU/mg). Overexpression of metY increased the in vitro activity to 1,780 mU/mg and was beneficial for methionine production, since the intracellular methionine pool was increased 2-fold in the engineered strain. This positive effect was limited by a depletion of the metY substrate O-acetylhomoserine, suggesting a need for further metabolic engineering targets towards competitive production strains.

FIA에 의한 유기산의 해리상수 측정에 관한 연구 (A Study on the Measurement of Dissociation Constant of Organic Acid with FIA)

  • 빠른 시간내에 적은 시료로(mg 단위) FIA와 Diode Array Spectrophotometry를 이용하여 해리상수를 구할 수 있는 방법을 개발하는 데 실험의 비중을 두었고, 그 대상으로 Bromo Cresol Green을 사용하였다. 소량의 산 염기를 가할 때 제일 sensitive한 발색반응을 보이는 지시약의 pK는 7.0임이 컴퓨터에 의해 계산되었다. Spectrophotometry를 이용한 Bromothylmol Blue와 Bromocresol Green은 Kielland 방법에 의한 Activity를 보정해 준 후 pKa가 각각 7.31과 4.82를 얻었다(Reported value by Bishop 7.30, 4.79). FIA에 의한 Bromocresol Green의 pKa값은 4.78을 얻어 다른 방법과 유사함을 알 수 있었다.

Study on the biodegradation of alternatives (four species including C8H8F9KO3S) for perfluorooctane sulfonate

  • Objectives The objective of this study was to evaluate the biodegradation potential of four perfluorooctane sulfonic acid (PFOS) alternatives that were developed at Changwon National University. While PFOS has been used widely in industrial and consumer products, it is known to be a persistent organic pollutant. Therefore, greener alternatives are highly desirable. Methods Biodegradation tests were run for 28 days using standard test protocols. The biochemical oxygen demand was measured daily throughout the experimental period, and the data were used to calculate the biodegradation rates. Microorganisms were isolated from the some of the tests that showed evidence of biodegradation. Results $C_8H_8F_9KO_3S$, which has the same number of carbons as the parent compound PFOS but a reduced number of fluorines, showed the highest biodegradation rate followed by $C_{10}H_8F_{13}KO_3S$. Chemical alternatives with lower number of carbons did not biodegrade readily in the experiments. Conclusions Together, these results suggest that it may be advantageous to develop PFOS alternatives with 8 carbons, the same as PFOS, but a reduced number of fluorines; as such, chemicals are more susceptible to biodegradation than the parent compound.

Screening System for Chitin Synthase II Inhibitors from Natural Resources and its Inhibitor Prodigiosin

  • Chitin synthases are identified as key enzymes of chitin biosynthesis in most of the fungi. Among them, chitin synthase II has been reported to be and essential enzyme in chitin biosynthesis, and exists as a membrane-bound form. To search and screen new antifungal agents from natural resources to inhibit chitin synthase II, the assay conditions were established using the enzyme isolated from Saccharomyces cerevisiae ECY38-38A(pAS6) that overproduces only chitin synthase II. This enzyme was activated only by partial proteolysis with trypsin. Its actibity reached the maximum at $80{\;}\mu\textrm{g}/ml$ of trypsin and was strongly stimulated by 2.0 mM $Co^{2+}$, 1.0 nM UDP-[$^{14}C$]-GicNAc, and 32 mM free-GlcNAc. Under these assay conditions, the highest chitin synthase II activity was observed by incubation at $30^{\circ}C$ for 90 min. However, and extremely narrow range of organic solvents up to as much as 25% of DMSO and 25% of MeOH was useful for determining optimal assay conditions. After a search or potent inhibitors of chitin synthase II from natural resources, prodigiosin was isolated from Serratia marcescens and purified by solvent extration and silica gel column chromatographies. The structure of prodigiosin was determined by UV, IR, Mass spectral, and NMR spectral analyses. Its molecular weight and formula were found to be 323 and $C_{20}H_{25}N_{3}O$, respectively. Prodigiosin ingibited chitin synthase II by 50% at the concentration of $115{\;}\mu\textrm{g}/ml$.

수도권 초미세먼지 농도모사: (V) 북한 배출량 영향 추정 (PM2.5 Simulations for the Seoul Metropolitan Area: (V) Estimation of North Korean Emission Contribution)

  • Quantitative assessment on the impact from North Korean emissions to surface particulate matter(PM) concentration in the Seoul Metropolitan Area (SMA), South Korea is conducted using a 3-dimensional chemistry transport model. Transboundary transport of air pollutants and their precursors are important to understand regional air quality in East Asian countries. As North Korea locates in the middle of main transport pathways of Chinese pollutants, quantifiable estimation of its impact is essential for policy making in South Korean air quality management. In this study, the Community Multiscale Air Quality Modeling System is utilized to simulate regional air quality and its sensitivity, using the Comprehensive Regional Emissions inventory for Atmospheric Transport Experiment 2015 and the Clean Air Policy Support System 2013 emissions inventories for North and South Korea, respectively. Contributions were estimated by a brute force method, perturbing 50% of North and South Korean emissions. Simulations demonstrate that North Korean emissions contribute $3.89{\mu}g/m^3$ of annual surface PM concentrations in the SMA, which accounts 14.7% of the region's average. Impacts are dominant in nitrate and organic carbon (OC) concentrations, attributing almost 40% of SMA OC concentration during January and February. Clear seasonal variations are also found in North Korean emissions contribution to South Korea (and vice versa) due to seasonal characteristics of synoptic weather, especially by the change of seasonal flow patterns.

토양 내 철의 이동특성에 관한 연구 (A Study on Transport Characteristics of Fe in Soil)

  • 이류-확산 국부적 평형흡착모델(convection-dispersion local equilibrium sorption model)과 Two-site 비평형 흡착모델(non-equilibrium sorption model)을 이용하여 철(Fe)의 토양 내 흡착 및 이동특성을 살펴보았다. 회분실험에서는 철의 주입농도를 변화시키면서 시간에 따른 농도변화를 측정함으로써 철의 흡착특성을 선형흡착등온선과 Freundlich 흡착등온선으로 살펴보았으며, 칼럼실험에서는 주입유속, 유기물함량, 철의 농도를 변화시키면서 주입액과 유출액의 철의 농도를 측정하고, CXTFIT 프로그램을 이용하여 two-site 비평형 흡착 모델에 사용된 매개변수(D, R, ${\beta}$, ${\omega}$)를 구하였다. CXTFIT 프로그램에 의해 구하여진 매개변수들을 이용하여 철의 토양 내 이동특성을 살펴보았으며, 철과 같은 흡착성 물질의 이동특성은 two-site 비평형 흡착모델에 의해 예측할 수 있음을 알 수 있었다.

Trichlorophenoxy alkylene ammonium 유도체(誘導體)의 합성(合成) 및 살균작용(殺菌作用)에 관(關)한 연구(硏究) (The Bacteriostatic Action of Trichlorophenoxy Alkylene Ammonium Derivatives and their Synthesis)

  • The bacteriostatic action of trichlorophenoxy alkylene ammonium derivatives, 2(2,4,5-trichlorophenoxy) ethylene N-triethyl ammonium bromide; (2,4,5-TETB), 2(2,4,6-trichlorophenoxy) ethylene N-triethyl ammonium bromide; (2,4,6-TETB), 2,4,5-triphenoxy ethylene pyridinium bromide; (2,4,5-TEPB), and 2,4,6-trichlorophenoxy ethylene pyridinium bromide:(2,4,6-TEPB) were observed by Jackson and Finland's and Finland's and Pak's on Staphylococcus aureus, E. coli, Salmonella typhi, Shigella flexneri, and Bacillus subtilis, comparing with those of phenol and trichlorophenol. The following results were obtained. 1) Phenol is bacteriostatic in a concentration of $10^{-3}$ for all above species. 2) Trichlorophenol is bacteriostatic in concentration of $10^{-4}$ to $5{\times}10^{-5}$ for all above species(Table 7), 3) 2,4,5-TETB and 2,4,6-TETB are bacteriostatic in a concentration of $10^{-4}$ and $5{\times}10^{-5}$ for Staphylococcus aureus, Salmonella typhi, and Shigella flexneri, but do not for E. coli and Bacillus subtilis(Table 8 and 9). 4) 2,4,5-TEPB and 2,4,6-TEPB are bacteriostatic in a concentrationof $10^{-4}$ and $5{\times}10^{-5}$ only for Staphylococcus aureus among four species(Table 10 and 11).

Microbial Differentiation on Chemical Properties of Paddy Soils in Reclaimed Tidal Lands at Western-coast Area of Korea

  • The scientific information on the microbial differentiation according to the changes in chemical properties of paddy soil in reclaimed tidal lands is not enough to understand the reclamation processes. The changes in microflora based on the chemical properties of paddy soils at the same sites of reclaimed tidal lands (21 samples from Nampo, Ewon, Sukmoon and Shihwa sites) were investigated in 2013 and 2015. In general, organic matter in paddy soils increased whereas pH decreased with the reclamation time. The electrical conductivities (EC) of soil samples were closely related to the exchangeable $Na^+$. With an increases in EC of paddy soils from 0.39 to $48.9dS\;m^{-1}$, the ratios (%) of halotolerant and halophilic bacteria to mesophilic bacteria proportionally increased from 0.2% to 102,000%. The population of halotolerant and halophilic bacteria in total microflora was also differentiated with the changes in EC of the same sites from reclaimed tidal soils within 2 years. The population of mesophilic bacteria decreased with an increase in EC above $5dS\;m^{-1}$. The microflora including halotolerant and halophilic bacteria could be a candidate as a biological parameter in evaluating the reclamation processes in addition to the chemical index of EC.

전라북도 장수군 오미자 재배과원 토양 특성 (Characteristics of Schizandra chinensis Baillon Orchard Soils Located in Jangsu-gun, Jeollabuk-do)

  • 전라북도 장수군 오미자 과원을 대상으로 토양의 이 화학적 특성을 조사하여 오미자의 품질향상, 수량증대 및 유해물질 종합관리 시스템 구축을 위한 기초조사를 수행하였다. 오미자 재배과원 토양은 대부분 양질사토였으며, 토양입단화도는 약 33.12%로 입단의 발달이 부족한 상태였으며, 토양공극율은 약 60% 수준으로 적당한 것으로 나타났다. 토양 pH는 평균 5.51이었으며, 양이온교환용량은 평균 4.11 $cmol_c\;kg^{-1}$으로 매우 낮게 나타났다. 식물영양성분은 적정 수준으로 유지되고 있었으나, 장수군 오미자 재배지 토양의 이화학적 특성 개량을 위해서는 유기물 및 석회처리 등의 토양관리가 필요한 것으로 나타났다. 조사대상 중금속의 함량은 자연함유량 수준으로 중금속 오염으로 인한 작물 안정성은 확보된 것으로 조사되었다.
