Streptomyces rimosus가 생산하는 Protease의 정제와 특성 (Purification and Properties of Extracellular Protease from Streptomyces rimosus)

  • Oxytetracycline 생산균주인 Streptomyces rimosus를 maltose 2%, NH$_4$Cl 0.5%, yeast extract 0.4 %, MGSO$_4$.7$H_2O$ 0.2% 조성의 배지를 배양초기 PH 6.5로 하여 3$0^{\circ}C$, 72시간 진탕배양하여 얻은, 세포외 protease를 유안분획, Sephadex A-50 이온교환, Sephadex G-100 gel 여과를 행하여 정제하였다. 효소의 최적온도는 5$0^{\circ}C$이며, 최적 pH는 8.0이었으며, Co$^{2+}$ 이온에 의해 활성화되며 Hg$^{2+}$, Fe$^{2+}$ 및 EDTA에 의해 저해를 받으며 casein 분자량을 23,600으로 추정하여 구한 Km값은 2.7$\times$$10^{-4}$M이었다.

Purification and Properties of Cyclodextrin Glucanotrnsferase Synthesizing $2-O-{\alpha}-D-Glucopyranosyl{\;}_{L}-Ascorbic$ Acid from Paenibacillus sp. JB-13

  • A Gram-positive bacterium (strain JB-13) that was isolated from soil as a producer of cyclodextrin glucanotransferase (CGTase) [EC 2.4.1.19] was identified as Panibacillus sp. JB-13. This CGTase could catalyze the transglucosylation reaction from soluble starch to L-ascorbic acid (AA). A main product formed by this enzyme with ${\alpha}-glucosidase$ was identified as $2-O-{\alpha}-D-glucopyranosyl{\;}_{L}-ascorbic$ acid (AA-2G) by the HPLC profile and the elemental analysis. CGTase was purified to homogeneity using ammonium sulfate fractionation, ion-exchange chromatography on DEAE-Seohadex A-50, and gel chromatography on Sephacryl S-200HR. The molecular weight was determined to be 66,000 by both gel chromatography and SDS-PAGE. The isoelectric point of the purified enzyme was 5.3. The optimum pH and temperature was PH 7.0 and $45^{\circ}C$ respectively. The enzyme was stable in the range of pH 6-9 and at temperatures of $75{\circ}C$ or less in the presence of 15 mM ${CaCl_2}.\;{Hg^2+},\;{Mn^+2},{Ag^+},\;and\;{Cu^2+}$ all strongly inhibited the enzyme's activity.

Clostridium thermocellum으로 부터 새로운 type I I 제한효소 Cth I 의 분리 (A New Restriction Endonuclease from Clostridium thermocellum)

  • 새로운 제한효소인 Cth I을 Clostridium thermocellum ATCC 27405로부터 분리하여, 그 효소의 생화학적 특성을 연구하였다. 이 효소는 Bcl I endonuclease의 isoschizomer로서 5'-TGATCA-3'를 인식한다. Cth I endonuclease는 섭씨 60도의 높은 온도에서 잘 작용하며, 10mM까지의 저농도의 NaCl과 MG$^{2+}$ 이온의 존재하에서 pH7.5와 10.5사이에서 최적의 활성을 보여주었다. Cth I endonuclease의 활성은 DNA 기질이 dam methylation 되었을 때 저해를 받았다.

습식법에 의한 초미립 $SrTiO_3$ 분말 합성 (Synthesis of Submicron $SrTiO_3$ Powders by Wet Process)

  • Pure submicron $SrTiO_3$ powders had been synthesized with chemical wet process that $5N-NH_4OH$ solution was sprayed into the mixed solution of $SrTiO_3$, $TiCl_4$ and $H_2O_2$ with $N_2$ carrier gas. The characteristic properties of powders obtained from this experiment were as follows. The optimum synthesis condition in reaction bath was above PH 8.5 and under $25^{\circ}C$ The particle size of precipitated SrTiO(OH) powders dried at 6$0^{\circ}C$ was under 0.01${\mu}{\textrm}{m}$ and uniform. Amorphous precipitated complex powders emitted adsorbed water at 15$0^{\circ}C$ less that and crystalline $SrTiO_3$powders was produced from calcining the complex at 30$0^{\circ}C$. Sintered body of SrTiO3 fired at 133$0^{\circ}C$ showed that relative dielectric constant was 228 at 1MHZ and bulk density was 4.73g/$cm^3$.

Exopolysaccharide생산 유산균주의 분리 및 배양조건 (Isolation Of Latobacillus Producing Exopolysaccharide and Optimization of its Production)

  • 원유로부터 다당류를 생산하는 유산균을 분리, 선발하여 균의 배양조건을 조사하고 Lactobacillus ssp. SCU-M으로 동정, 명명된 최적배지의 탄소원은 galactose 1.5% 첨가시 553 mg/L의 EPS를 생성하였으며 whey의 이용성 검토를 위한 deproteinized whey와 lactic acidified whey를 탄소원으로 사용하였을 때 각각 639, 550 mg/L를 생성하였다. 질소원으로 yeast extract 1.5%, peptone 0.5%를 첨가한 경우 934 mg/L, 미량성분으로 , 0.15% $K_2$HPO, 0.15% 첨가시 1,076 mg/L의 다당 생산성을 나타내었다. 따라서 이들 결과를 종합하여 galactose 1.50%, yeast extract 1.00%, peptone 0.25%, MgSO$_4$, 7$H_2O$ 0.15%, $K_2$HPO$_4$0.15%, tween 80, 0.10%를 적정 배지로 하여 PH 6.5, 배양온도 37$^{\circ}C$에서 배양시 1,680 mg/L의 EPS를 생성하였다.

  • In this paper, Wireless sensor network technology applied to various greenhouse agro-industry items such as horticulture and local specialty etc., we was constructed automatic control system for optimum growth environment by measuring growth status and environmental change. existing monitoring systems of greenhouse gather information about growth environment depends on the temperature. but in this system, Can be efficient collection and control of information to construct wireless sensor network by growth measurement sensor and environment monitoring sensor inside of the greenhouse. The system is consists of sensor manager for information processing, an environment database that stores information collected from sensors, the GUI of show the greenhouse status, it gather soil and environment information to soil and environment(including weather) sensors, growth measurement sensor. In addition to support that soil information service shows the temperature, moisture, EC, ph of soil to user through the interaction of obtained data and Complex Growth Environment information service for quality and productivity can prevention and response by growth disease or disaster of greenhouse agro-industry items how temperature, humidity, illumination acquiring informationin greenhouse(strawberry, ginseng). To verify the executability of the system, constructing the complex growth environment measurement system using wireless sensor network in greenhouse and we confirmed that it is can provide our optimized growth environment information.

Control of Odor Emissions Using Biofiltration: A Case Study of Dimethyl Disulfide

  • A laboratory- scale dual-column biofilter system was used to study the biofiltration of dimethyl disulfide (DMDS). The gas flow rate and DMDS concentration to the biofilter were varied to study their effect on the remov-al of dimethyl disulfide. Operating parameters such as pH, temperature, and water content were monitored during the biofilter operation and necessary precautions were taken to keep these parameters within the acceptable limits. It was observed that the removal efficiency of DMDS was optimal at neutral pH values. After five month op-eration, the neutralization of the filter beds with sodium carbonate became necessary for the optimum operation of the biofilters. The microbial population already present in the compost mixtures was found to be adequate in treat-ing DMDS. The compost mixtures were found to be similar in terms of biofiltration efficiency of DMDS. However, pressure drops observed in the first column compost mixture (compost/ peat mulch) was extremely high, making this compost economically not feasible. The second mixture (compost/bark) provided pressure drops within accept-able limits. A minimum residence time of 30 seconds at the optimal operating conditions appeared to be adequate for achieving high removal efficiencies (>90%).

Proteus sp. ST-1에 의한 Azo계 색소의 분해에 관한 연구 (The Study on Degradation of Azo Dyes by Proteus sp. ST-1)

  • Direct Sky Blue-5B is an Azo dye known as general for staining of textile and leather, etc., and as materials which are difficult to be biodegraded in nature. The bacterium strain which could degrade direct Sky Blue-5B was isolated from activated sludge of dyeing factory and identified as Proteus sp. by experiment on morphological, cultural and biochemical characteristics, and so named Proteus sp. ST-1. The optimum condition of the strain for degradation of Sky Blue-5B were at about 35$^{\circ}C$ and PH 7~8. The strain had been capable of degradation with organic nitrogen effectively and had completely degraded 200mg/1 of the dye within 12hrs at 37$^{\circ}C$. The enzyme system related to degradation of Azo dye may be intracellular, and so degraded the dye after absorption into cell. The degradation products of Sky Blue-5B by Proton sp. 57-1 were analyzed by Gas Chromatography /Mass Spectrometry and Spectrophotomer, from this observation, it may be infered that the strain degraded the dye directly without any mediate.

  • 효모세포의 원형질체 최적형성을 위한 stabilizer의 종류 및 농도, pH 그리고 lysis 방법을 조사하는 한편, intact cell과 protoplast사이의 효소활성도 및 poly-P 생합성율을 비교하였다. 그 결과 protoplast 형성에 있어 snail gut enzyme은 5시간, drisielase는 3시간 정도의 incubation 시간이 필요했으며, stabilizer로는 0.8 M mannitol, 6 M KCl이 좋았다. Protoplast는 intact cell에 비해 ALPase 활성은 22-27%, ACPase는 4-15% 정도 감소하였으며, poly-P 형성은 protoplast에서 유의한 증가가 일어나지 않았다.

흡착법에 의한 Lipase의 고정화 (Studies on the Immobilization of Lipase by Adsorption Method)

  • Lipase를 유지분해에 이용하기 위한 기초연구로서 Candida cylindracea에서 추출된 lipase를 흡착법에 의해 고정화하고 그의 반응특이성을 본 결과는 다음과 같다. Lipase 흡착에 적합한 흡착제로는 silica gel이 선정되었으며, Silica gel 1.6g에 lipase 47.5 units를 $5^{\circ}C$, PH 7.0에서 100분간 흡착시키는 것이 좋았다. Silica gel에 고정화 시킨 lipase를 유지방과 olive oil의 분해에 적합한 최적온도 및 최적pH는 가용성효소와 비교시 $37^{\circ}C$, pH 7.0으로 변하지는 않았으나 활성의 범위는 넓어졌다. 또한 열안정성 및 pH안정성도 가용성 효소에 비하여 활성의 범위가 넓어졌다. 유지의 분해에 적합한 고정화 효소의 최적효소농도는 유지방의 경우 30g이었으며 올리브유의 경우 80g으로 선정하였다. 이 때 최적기질농도는 유지방 및 올리브유 모두 20%였다. 반응시간에 따른 반응률은 유지방을 이용하여 조사한 결과 가용성 효소는 반응 4시간까지는 급격한 분해를 나타냈으나 고정화 효소는 8시간 까지 급격한 증가를 나타내고 그 이상은 거의 일정하였다. 또한 유리되는 지방산의 profile은 가용성 효소와 비교시 capric acid의 생성은 모두 높았으며, myristic acid의 함량은 높고 butyic acid의 함량은 적었다.
