Comparative assessment and uncertainty analysis of ensemble-based hydrologic data assimilation using airGRdatassim (airGRdatassim을 이용한 앙상블 기반 수문자료동화 기법의 비교 및 불확실성 평가)

  • Accurate hydrologic prediction is essential to analyze the effects of drought, flood, and climate change on flow rates, water quality, and ecosystems. Disentangling the uncertainty of the hydrological model is one of the important issues in hydrology and water resources research. Hydrologic data assimilation (DA), a technique that updates the status or parameters of a hydrological model to produce the most likely estimates of the initial conditions of the model, is one of the ways to minimize uncertainty in hydrological simulations and improve predictive accuracy. In this study, the two ensemble-based sequential DA techniques, ensemble Kalman filter, and particle filter are comparatively analyzed for the daily discharge simulation at the Yongdam catchment using airGRdatassim. The results showed that the values of Kling-Gupta efficiency (KGE) were improved from 0.799 in the open loop simulation to 0.826 in the ensemble Kalman filter and to 0.933 in the particle filter. In addition, we analyzed the effects of hyper-parameters related to the data assimilation methods such as precipitation and potential evaporation forcing error parameters and selection of perturbed and updated states. For the case of forcing error conditions, the particle filter was superior to the ensemble in terms of the KGE index. The size of the optimal forcing noise was relatively smaller in the particle filter compared to the ensemble Kalman filter. In addition, with more state variables included in the updating step, performance of data assimilation improved, implicating that adequate selection of updating states can be considered as a hyper-parameter. The simulation experiments in this study implied that DA hyper-parameters needed to be carefully optimized to exploit the potential of DA methods.

Development of a Rapid Enrichment Broth for Vibrio parahaemolyticus Using a Predictive Model of Microbial Growth with Response Surface Analysis (미생물 생장 예측모델과 반응표면분석법을 이용한 Vibrio parahaemolyticus의 신속 증균배지 개발)

  • In this study, we developed Rapid Enrichment Broth for Vibrio parahaemolyticus (REB-V), a broth capable enriching V. parahaemolyticus from 100 CFU/mL to 106 CFU/mL within 6 hours, which greatly facilitates the rapid detection of V. parahaemolyticus. Using a modified Gompertz model and response surface methodology, we optimized supplement sources to rapidly enrich V. parahaemolyticus. The addition of 0.003 g/10 mL of D-(+)-mannose, 0.002 g/10 mL of L-valine, and 0.002 g/10 mL of magnesium sulfate to 2% (w/v) NaCl BPW was the most effective combination of V. parahaemolyticus enrichment. Optimal V. parahaemolyticus culture conditions using REB-V were at pH 7.84 and 37℃. To confirm REB-V culture efficiency compared to 2% (w/v) NaCl BPW, we assessed the amount of enrichment achieved in 7 hours in each medium and extracted DNA samples from each culture every hour. Real-time PCR was performed using the extracted DNA to verify the applicability of this REB-V culture method to molecular diagnosis. V. parahaemolyticus was enriched to 5.452±0.151 Log CFU/mL in 2% (w/v) NaCl BPW in 7 hours, while in REB-V, it reached 7.831±0.323 Log CFU/mL. This confirmed that REB-V enriched V. parahaemolyticus to more than 106 CFU/mL within 6 hours. The enrichment rate of REB-V was faster than that of 2% (w/v) NaCl BPW, and the amount of enrichment within the same time was greater than that of 2% (w/v) NaCl BPW, indicating that REB-V exhibits excellent enrichment efficiency.

Screening of Biogenic Amine Non-Producing Yeast and Optimization of Culture Conditions Using Statistical Method for Manufacturing Black Raspberry Wine (복분자 와인 제조를 위한 바이오제닉 아민 비생성 효모의 선별 및 통계학적 기법을 이용한 배양조건 최적화)

  • Rubus coreanus is known as Korean black raspberry, native to Korea, Japan, and China. Preliminary studies evaluating their potential for cancer treatment in mammalian test systems are ongoing. In recent years, interest has been renewed due to their high levels of anthocyanins. Anthocyanins in black raspberry are important due to their potential health benefits as dietary antioxidant, anti-inflammatory compound, and as a chemopreventive agent. In the present study, Saccharomyces cerevisiae BA29 was isolated from black raspberry fruit and fruit juice as a biogenic amine non-producing strain for manufacturing of black raspberry wine, after which we investigated its characteristics: biogenic amine-producing ability, cell growth ability, alcohol-fermentation ability, and resistance to alcohol, glucose, and sulfur dioxide. Based on preliminary experiments, we optimized culture medium compositions for improving dried cell weight of S. cerevisiae BA29 by response surface methodology (RSM) as a statistical method. Design for RSM used a central composite design, and molasses with the industrial applicability was used as a carbon source. Through statistical analysis, we obtained optimum values as follows: molasses 200 g/L, peptone 30 g/L, and yeast extract 40 g/L. For the model verification, we confirmed about 3-fold improvement of dried cell weight from 6.39 to 20.9167 g/L compared to basal yeast peptone dextrose medium. Finally, we manufactured black raspberry wine using S. cerevisiae BA29 and produced alcohol of 20.33%. In conclusion, S. cerevisiae isolated from black raspberry fruit and juices has a great potential in the fermentation of black raspberry wine.

Enhancement of Excretory Production of an Exoglucanase from Escherichia coli with Phage Shock Protein A (PspA) Overexpression

  • Production of recombinant proteins by excretory expression has many advantages over intracellular expression in Escherichia coli. Hyperexpression of a secretory exoglucanase, Exg, of Cellulomonas fimi was previously shown to saturate the SecYEG pathway and result in dramatic cell death of E. coli. In this study, we demonstrated that overexpression of the PspA in the JM101(pM1VegGcexL-pspA) strain enhanced excretion of Exg to 1.65 U/ml using shake-flask cultivation, which was 80% higher than the highest yield previously obtained from the optimized JM101(pM1VegGcexL) strain. A much higher excreted Exg activity of 4.5 U/ml was further achieved with high cell density cultivation using rich media. Furthermore, we showed that the PspA overexpression strain enjoyed an elevated critical value (CV), which was defined as the largest quotient between the intracellular unprocessed precursor and its secreted mature counterpart that was still tolerable by the host cells prior to the onset of cell death, improving from the previously determined CV of 20/80 to the currently achieved CV of 45/55 for Exg. The results suggested that the PspA overexpression strain might tolerate a higher level of precursor Exg making use of the SecYEG pathway for secretion. The reduced lethal effect might be attributable to the overexpressed PspA, which was postulated to be able to reduce membrane depolarization and damage. Our findings introduce a novel strategy of the combined application of metabolic engineering and construct optimization to the attainment of the best possible E. coli producers for secretory/excretory production of recombinant proteins, using Exg as the model protein.

  • In this study, Mixed coordination method was selected to optimize the shape of the truss structures which takes multi-loading condition, allowable stress, buckling stress, displacement constraints into consideration. The structure was devided into substructures by Goal coordination method and the substructures were optimized by model coordination method which used two-level technique. Therefore the number of design variables and constrints can be decreased considerable. Under the condition of the same disign, the weight of truss structures can be decreased more considerable by means of optimizing even the shape of truss than by means of optimizing the section of truss while fixing geometrical configuration of it, even though there might be a little difference according to the early geomatrical shape of the truss and the design condition. Thus, the shape optimization of truss structures which utilize the results of this study can be helpful to the economical design of truss structures.

A study on characteristics of High Efficiency and Wideband Microstrip Band Pass Filter for Wireless Data Communication (무선데이터 통신을 위한 고효율 광대역 마이크로스트립 대역통과 필터 특성에 관한 연구)

  • This paper presents a compact, low insertion loss, sharp rejection and wide band microstrip band pass filter that is composed rectangular loop resonator and step-impedance-open-stub(SIOS) for wireless data communication. The SIOS can be reduce length about 30% more than general 0.25${\lambda}$ stub. And stubs can have the advantage of tuning impedance magnitude. In order to demonstrate agrement of this paper prove, the optimized wide band pass filters are realized and experimented. A transmission line model used to calculate the frequency response of the new filters shows good agreement with measurements. The filter with perturbation stubs has four poles at rejection band, the poles are excited 3.610GHz, 4.265GHz at low frequency band, 8.494GHz, 9.056GHz at high frequency band. And the filter has 3dB fractional bandwidth of 57%(3.695GHz), an insertion loss of better than 0.37dB from 4.549GHz to 8.244GHz, and two rejection of greater than 30dB within 237MHz(4.312GHz${\sim}$ 4.549GHz) at low frequency band, 234MHz(8.244GHz-8.491GHz) at high frequency band.

  • In the modeling processes of 3D computer animation, methods to build optimal work conditions to realize real forms for more efficient works have been advanced. Digital sculpting software, published in 1999, ZBrush has been positioned as an essential factor in character model work requiring of realistic descriptions through different manufacturing methods from previous modeling work processes and easy shape realization. Their functional areas are expanding. So, in this production case paper, as a method to product more optimized animation character models, the efficiency of production method linking digital sculpting software (Z-Brush) and animation production software (Maya) was deliberated and its consequences and implications are suggested. To this end, first the technical features of polygon modeling and Retopology were reviewed. Second, based on it, the efficiency of animation character modeling work processes through step linking ZBrush and Maya suggested in this paper was analyzed. Third, based on the features drawn before, in order to prove the hypothesis on modeling optimization method suggested in this paper, the production process of character Dumvee from a short animation film, 'Cula & Mina' was analyzed as an example. Through this study, it was found that technical approach easiness and high level of completion could be realized through two software linked work processes. This study is considered to be a reference for optimizing production process of related industries or modeling-related classes by deliberating different modeling process linked systems.

Optimization of Multi-reservoir Operation with a Hedging Rule: Case Study of the Han River Basin (Hedging Rule을 이용한 댐 연계 운영 최적화: 한강수계 사례연구)

  • The major reason to construct large dams is to store surplus water during rainy seasons and utilize it for water supply in dry seasons. Reservoir storage has to meet a pre-defined target to satisfy water demands and cope with a dry season when the availability of water resources are limited temporally as well as spatially. In this study, a Hedging rule that reduces total reservoir outflow as drought starts is applied to alleviate severe water shortages. Five stages for reducing outflow based on the current reservoir storage are proposed as the Hedging rule. The objective function is to minimize the total discrepancies between the target and actual reservoir storage, water supply and demand, and required minimum river discharge and actual river flow. Mixed Integer Linear Programming (MILP) is used to develop a multi-reservoir operation system with the Hedging rule. The developed system is applied for the Han River basin that includes four multi-purpose dams and one water supplying reservoir. One of the fours dams is primarily for power generation. Ten-day-based runoff from subbasins and water demand in 2003 and water supply plan to water users from the reservoirs are used from "Long Term Comprehensive Plan for Water Resources in Korea" and "Practical Handbook of Dam Operation in Korea", respectively. The model was optimized by GAMS/CPLEX which is LP/MIP solver using a branch-and-cut algorithm. As results, 99.99% of municipal demand, 99.91% of agricultural demand and 100.00% of minimum river discharge were satisfied and, at the same time, dam storage compared to the storage efficiency increased 10.04% which is a real operation data in 2003.

Maximum rejection characteristics of the filter are -61.8 dB at low frequency and -76.3 dB at high frequency.

Optimization of Glycosyl Aesculin Synthesis by Thermotoga neapolitana β-Glucosidase Using Response-surface Methodology (반응표면분석법을 이용한 Thermotoga neapolitana β-glucosidase의 당전이 활성을 통한 glycosyl aesculin 합성 최적화)

  • Glycosyl aesculin, a potent anti-inflammatory agent, was synthesized by transglycosylation reaction, catalyzed by Thermotoga neapolitana ${\beta}-glucosidase$, with aesculin as an acceptor. The key reaction parameters were optimized using response-surface methodology (RSM) and $2{\mu}g$ of the enzyme. As shown by a statistical analysis, a second-order polynomial model fitted well to the data (p<0.05). The response surface curve for the interaction between aesculin and other parameters revealed that the aesculin concentration and reaction time were the primary factors that affected the yield of glycosyl aesculin. Among the tested factors, the optimum values for glycosyl aesculin production were as follows: aesculin concentration of 9.5 g/l, temperature of $84^{\circ}C$, reaction time of 81 min, and pH of 8.2. Under these conditions, 61.7% of glycosyl aesculin was obtained, with a predicted yield of 5.86 g/l. The maximum amount of glycosyl aesculin was 6.02 g/l. Good agreement between the predicted and experimental results confirmed the validity of the RSM. The optimization of reaction conditions by RSM resulted in a 1.6-fold increase in the production of glycosyl aesculin as compared to the yield before optimization. These results indicate that RSM can be effectively used for process optimization in the synthesis of a variety of biologically active glycosides using bacterial glycosidases.
