PEM fuel cell vehicles have been getting much attraction due to a sort of highly clean and effective transportation. The onboard fuel processor, however, is inevitably required to supply the hydrogen by conversion from some fuels since there are not enough available hydrogen stations nearby. A lot of studies have been focused on analyses of ATR reactor under the assumption of thermo-neutral condition and those of the optimized process for the minimization of energy consumption using thermal efficiency as an objective function, which doesn't guarantee the maximum hydrogen production. In this study, the analysis of optimization for 100 kW PEMFC onboard fuel processor was conducted targeting various fuels such as gasoline, LPG, diesel using newly defined hydrogen efficiency and keeping simply synthesized heat exchanger network regardless of external utilities leading to compactness and integration. Optimal result of gasoline case shows 9.43% reduction compared to previous study, which shows the newly defined objective function leads to better performance than thermal efficiency in terms of hydrogen production. The sensitivity analysis was also done for hydrogen efficiency, heat recovery of each heat exchanger, and the cost of each fuel. Finally, LPG was estimated as the most economical fuel in Korean market.
An optimal condition for the continuous production of ${\delta}-aminolevulinate$(ALA) was investigated using high concentrated resting cells of Rhodocyclus gelatinosus KUP-74. The increase of the amount of extracellular ALA versus the concentration of resting cells showed rectangular hyperbolic pattern until 20 mg cells/ml, but no further increase in the ALA amount by increasing its concentration was occurred. The highest yield of the extracellular ALA was observed after 3 hr of incubation of 1 ml reaction system containing 20 mg cells, 4 mM levulinate and 5 mM L-glutamate. On the other hand, the immobilized cells prepared by Ca-alginate inclusive method needed to incubate for 6 hr with 6 mM levulinate and 10 mM L-glutamate to give maximal yield of the extracellular ALA. In addition, under these conditions the resulted continuous productivities of the ALA by immobilized cells and highly concentrated resting cells were appeared 50 percent decreases after incubations for 185 hr and 100 hr, respectively, and the method of the cells to be immobilized was more efficient to recover the extracellular ALA produced.

The purpose of this study is to investigate how ozone-dissolution-tank structure affects micro-ozone-bubble distribution, energy consumption and water treatment efficiency. The partition walls inside the ozone-dissolution-tank generate pressure changes, shear forces, and swirling flows, which change the size of the bubble diameter. The size of the bubble diameter differs by 10.5% depending on the partition walls. Changes in ozone-bubble diameter are related to energy consumption. As the ozone-bubble becomes smaller, the bubble generation energy increases, but the ozone production energy decreases as the dissolution efficiency increases. Therefore, an ozone-dissolution-tank should be determined by means of an optimal condition producing a micro-ozone-bubble with a minimum sum of bubble generation energy and ozone production energy. The energy consumed to inject the same amount of ozone into the effluent differs by 2.5% depending on the partition walls. However, considering the water treatment efficiency, the conditions for selecting the ozone-dissolution-tank are variable. This is because the free radicals that increase as the ozone-bubble gets smaller are very efficient for water treatment. Even at the same ozone injection concentration, the water treatment efficiency differs by 10.4% according to the partition walls. Therefore, we have studied ozone-dissolution-tank structure which produces reasonable ozone-bubble considering water treatment efficiency and energy efficiency.
This study is focused on establishing the optimal conditions to enhance the production of antilisterial substances by Lactobacillus paracasei BK57 isolated from Baikkimchi. In addition, the growth and in situ lactic acid and bacteriocin production of this strain were investigated during the manufacture of fresh cheese. And then the efficacy of using Lactobacillus starter as a protective culture to improve the safety of fresh cheese against Listeria monocytogenes KCTC 3569 was estimated. Maximum growth rate and activity of antibacterial substances were obtained in Lactobacilli MRS broth at $37^{\circ}C$ with controlled pH 6.0 after 30 h of incubation under aerobic condition. However, the growth rate and antimicrobial activity of bacteriocin produced in whole milk supplemented with yeast extract (2.0%) as a substrate were lower than those obtained in MRS broth. Live cells and cell-free culture supernatant of BK57 strain were effective in the suppression of L. monocytogenes in milk, whereas the inhibitory of the bacteriocin obtained from BK57 strain was higher in BHI broth than in milk. During storage at $4^{\circ}C$ and $15^{\circ}C$ for 6 days, no significant difference was found in the cell viability and antimicrobial activity of BK 57 strain in fresh cheese. In samples held at two temperatures, there was at least a 15% reduction in the numbers of the pathogen in fresh cheese artificially contaminated with approximately $10^5CFU/ml$ of L. monocytogenes within 6 days. Our results demonstrated the usefulness of L. paracasei BK57 having antilisterial activity as a biopreservative in the cheese making process.

Gluconacetobacter hansenii TF-2, an isolate from black tea fungus, was statically cultivated to ferment cellulose gel from citrus juice. The juice prepared by press filtering of peeled citrus fruit contained 135.5 mg of total sugar/mL, 1.23% of total acid, and average pH of the juice was 3.98. The bacterium produced cellulose gel optimally on the surface of culture broth containing 17% of citrus juice and 10$^{\circ}$Brix of total sugar. The optimum temperature was 3$0^{\circ}C$ for producing acetic acid and gel formation. The bacterium could not produce acetic acid on gel formation at 4$0^{\circ}C$. The optimum pH was 3.0∼4.0 but was not significantly different between pH 3.0∼4.0. The cultivation for 18 days under optimal conditions produced gel as 14.2$\pm$0.6 mm of thickness and acids equivalent to 1.90$\pm$0.22% of acetic acid. The pH of culture broth was stabilized at 2.6∼2.8 during the cultivation. Remaining sugar content was 27.1$\pm$4.2 mg/mL of total sugar and 6.9 mg/mL of reducing sugar. The gel productivity was 137.8$\pm$9.7 g/L.

Ultrasound-assisted extraction (UAE) has attracted growing interest, as it is an effective method for the rapid extraction of bioactive compounds from plants with a high extraction efficiency comparable to the conventional extraction. In this study, UAE was used for the extraction of polyphenols from lipid extracted microalgae (Tetraselmis KCTC 12236BP) and the effects of five extraction variables on the total phenolic compounds (TPC) were studied. For the optimization of extraction parameters, particle size, solid-to-liquid (L/S) ratio, ethanol concentration, extraction temperature and extraction time have been examined as independent variables. All variables exhibited the significant effects on the extraction of TPC and extraction temperature showed the most significant effect among five variables. The optimal extraction conditions were the extraction using mixed particle, S/L ratio of 10%, ethanol concentration of 60%, extraction temperature of $100^{\circ}C$ and extraction time of 30 min, which gave the 8.7 mg GAE/g DW for TPC. Compared with conventional hot-water extraction, TPC extraction under UAE was increased by up to 1.8 fold with same extraction condition. This study showed that UAE under low temperature and short extraction time was proven to be an effective extraction process for TPC production from LEA compared to conventional hot-water extraction process.
In order to increase the content of germanium in ginseng adventitious roots, the effects of chelating agents on germanium content and root growth were investigated in the submerged cultures of ginseng adventitious roots. Chelating agents such as citric acid, oxalic acid, phosphoric acid, EDTA (Ethylenediamine tetraacetic acid) or EGTA (Ethylene glycol-bis $({\beta}-aminoethylether)-tetraacetic$ acid) were administrated in the submerged culture of ginseng root containing 50 ppm $GeO_2$. After 6 weeks of cultivation, fresh weight, germanium and saponin contents in the roots were analyzed. Among chelating agents, addition of 1.0mM phosphoric acid was found to be best for germanium accumulation. Under this condition, germanium content increased 1.4 times as compared to that of the control. The germanium content in the adventitious roots also increased with addition of EDTA or EGTA, while they inhibited the growth of ginseng adventitious root. Citric and oxalic acids were not effective for increasing germanium content in adventitious roots. As the results, it suggests that the phosphoric acid can be proved as the optimal agent for the enhancement of germanium accumulation in ginseng adventitious roots. These results can be served as a guideline for the mass production of ginseng adventitious roots containing germanium by large-scale production.
A vector harboring double cassettes; a heterologous gene expression cassette of pHCE-InaN-antigen and a ghost formation cassette of pAPR-cI-E lysis 37 SDM was constructed and introduced to E. coli DH5a. For the production of a bacterial ghost vaccine, bacterial ghosts from E. coli / Streptococcus iniae with four different types of antigens - enolase, GAPDH, sagA and piaA - were produced by the optimization of fermentation parameters such as a glucose concentration of 1 g/l, agitation of 300 rpm and aeration of 1 vvm. Efficiency of ghost bacteria formation was evaluated with cultures of OD$_{600}$=1.0, 2.0 and 3.0. The efficiency of the ghost bacteria formation was 99.54, 99.67, 99.99 and 99.99% with inductions at OD$_{600}$=3.0, 1.0, 2.0 and 1.0 for E. coli/S. iniae antigens enolase, piaA, GAPDH and sagA, respectively. Ghost bacteria as a vaccine was harvested by centrifugation. The antigen protein expressions were analyzed by SDS-PAGE and western blot analysis, and the molecular weights of the enolase, piaA, GAPDH and sagA were 78, 26, 67 and 26 kDa, respectively. The molecular weights of the expressed antigens were consistent with theoretical sizes obtained from the amino acid sequences.

Planting date shift is one of the means of adapting to climate change in Kimchi Cabbage growers in major production areas in Korea. This study suggests a method to estimate the potential yield of Kimchi Cabbage based on daily temperature accumulation during the growth period from planting to maturity which is determined by a plant phenology model tuned to Kimchi Cabbage. The phenology model converts any changes in the thermal condition caused by the planting date shift into the heat unit accumulation during the growth period, which can be calculated from daily temperatures. The physiological maturity is estimated by applying this model to a variable development rate function depending either on growth or heading stage. The cabbage yield prediction model (Ahn et al., 2014) calculates the potential yield of summer cabbage by accumulating daily heat units for the growth period. We combined these two models and applied to the 1km resolution climate scenario (2000-2100) based on RCP8.5 for South Korea. Potential yields in the current normal year (2001-2010) and the future normal year (2011-2040, 2041-2070, and 2071-2100) were estimated for each grid cell with the planting dates of July 1, August 1, September 1, and October 1. Based on the results, we divided the whole South Korea into 810 watersheds, and devised a three - dimensional evaluation chart of the time - space - yield that enables the user to easily find the optimal planting date for a given watershed. This method is expected to be useful not only for exploring future new cultivation sites but also for developing cropping systems capable of adaptation to climate change without changing varieties in existing production areas.

Collagenases are generally defined as enzymes that are capable of degrading the polypeptide backbone of native collagen under conditions that do not denature the protein. An extracellular collagenase-producing bacterial strain was isolated from kimchi and identified to be Bacillus subtilis JS-17 through morphological, cultural, biochemical characteristics and 16S rDNA sequence analysis. Optimum culture condition of Bacillus subtilis JS-17 for the production of collagenase was $1.5\%$ fructose, $1\%$ yeast extract, $0.5\%\;K_2HPO_4,\;0.4\%\;KH_2PO_4,\;0.01\%\;MgSO_4\cdot7H_2O,\;0.01\%\; MnSO_4\cdot4H_2O,\;,0.1\%$ citrate and $0.1\%\;CaCl_2$. The production of collagenase was optimal at $30^{\circ}C$ for 72 hr. A collagenase was isolated from the culture filtrate of Bacillus subtilis JS-17. The enzyme was purified using Amberlite IRA-900 column chromatography, Sephacryl S-300 HR column chromatography and DEAE-Sephadex A-50 column chromatography The purified collagenase has an specific activity 192.1 units/mg. The molecular weight of the purified enzyme was estimated to be 28 kDa by SDS-PACE. The purified collagenase has $100\%$ activity up to $55^{\circ}C$.
