Backbone 1H, 15N, and 13C resonance assignments and secondary structure prediction of SAV2228 (translation initiation factor-1) from Staphylococcus aureus

  • SAV2228 has an OB (Oligomer-Binding)-motif which is frequently used for nucleic acid recognition. To characterize the activity of translation initiation factor-1 (IF-1) from Staphylococcus aureus, SAV2228 was expressed and purified in Escherichia coli. We acquired 3D NMR spectra showing well dispersed and homogeneous signals which allow us to assign 94.4% of all $^1HN$, $^{15}N$, $^{13}C{\alpha}$, $^{13}C{\beta}$ and $^{13}CO$ resonances. We could predict a secondary structure of SAV2228 using TALOS and CSI from NMR data. SAV2228 was consisted of one ${\alpha}$-helix and five ${\beta}$-sheets. The predicted secondary structure, ${\beta}-{\beta}-{\beta}-{\alpha}-{\beta}-{\beta}$, was similar to other bacterial IF-1, but it was not completely same to the eukaryotic one. Assigned NMR peaks and secondary structre prediction can be used for the study on interaction with nucleic acid in the future.

공조용 압축기의 Cavity Resonance의 측정 및 저감에 관한 연구 (A Study on Measurement and Reduction of Cavity Resonance Based on the Internal Acoustic Modeling of Compressor)

  • Pressure pulsation Inside the discharge and suction cavity of rotary and scroll compressor are often a major source of objectionable noise and vibration. The key factor of these noise and vibration is due to the cavity resonance. It is not only necessary to understanding the characteristics of pulsation in order to reduce the excitation force of gas to the cavity but also to verifying the phenomena of cavity resonance. For the purpose of these understandings, measurement and simulation of cavity resonance can lead to a better understandings how they occur and be very important to identify the ways to reduce the noise efficiently. In this paper, modeling of the cavity(internal acoustics inside the shell) is discussed and simulated using FEM. Results from the simulation are compared with those measurement in experiments. In describing of cavity mode by experiments, it is very important to specify the exact conditions under which they are measured. Finally, this paper shows the one example of reduced cavity resonance in the compressor.

전자스핀공명을 이용한 자성체연구 소개 (ESR and its Application to Magnetic Research)

  • 전자스핀공명은 전자의 고유한 자기모멘트인 스핀과 마이크로파와의 상호작용을 통하여 물질 내의 상자성 이온에 관한 정보를 얻어내는 실험방법이다. 자성물성에 관한 연구를 포함하여 물리, 화학, 생물학, 의학 및 양자계산과 같은 넓은 분야에 응용되고 있다. 전자스핀공명실험으로 어떻게 상자성 이온의 g-텐서, 스핀 및 오비탈 상태에 대한 정보를 얻고 스핀간의 교환 상호작용력과 스핀-격자 및 스핀-스핀 완화시간을 측정할 수 있는가를 간단한 예들과 함께 소개한다.

Characterization of biotin-avidin recognition system constructed on the solid substrate

  • The biotin-avidin complex, as a model recognition system, has been constructed through N-hydroxysuccinimide(NHS) reaction on a variety of substrates such as a smooth Au film, electrochemically roughened Au electrode and chemically modified mica. Stepwise self-assembled monolayers (SAMs) of biotin-avidin system were characterized by surface-enhanced resonance Raman scattering (SERRS) spectroscopy, atomic force microscopy (AFM) and surface plasmon resonance (SPR). A strong SERRS signal of rhodamine tags labeled in avidin from the SAMs on a roughened gold electrode indicated the successful complex formation of stepwise biotin-avidin recognition system. AFM images showed the circular shaped avidin aggregates (hexamer) with ca. $60{\AA}$ thick on the substrate, corresponding to one layer of avidin. The surface coverage and concentration of avidin molecules were estimated to be 90% and $7.5{\times}10^{-12}mol/cm^2$, respectively. SPR technique allowed one to monitor the surface reaction of the specific recognition with high sensitivity and precision.

Structural Analysis and Single-Crystal EPR Study of Dimeric Cu(I) Complex with TTF Derivative

  • A Cu(I) complex with an asymmetric TTF derivative (CET-EDTTTF) is prepared from the slow-diffusion method using CET-EDTTTF and Cu(I)Br solutions and characterized by X-ray crystallography and EPR spectroscopy. Structural analysis shows Cu(I) ions are tetrahedrally coordinated to two bridging bromides, one terminal bromide, and one S atom from CET-EDTTTF. Detailed geometrical and EPR analysis identified that the dimmer molecule contains [Cu$_2Br_4]^{2-}$ anion between two [CET-EDTTTF]$^+$ radical cations. Single-crystal EPR investigation of the complex reveals that the ganisotropy is unusually big, compared to those of the previously reported TTF+ cation radicals, implying that there is significant contribution of the Cu d-orbital to the HOMO of the complex.

Operational Characteristics of Flux-lock Type SFCL using Series Resonance

  • We analyzed the fault current limiting characteristics of a flux-lock type $high-T_c$ super­conducting fault current limiter (HTSC-FCL) using series resonance between capacitor for series resonance and magnetic field coil which was installed in coil 3. The capacitor for the series resonance in the flux-lock type HTSC-FCL was inserted in series with the magnetic field coil to apply enough magnetic field into HTSC element, which resulted in higher resistance of HTSC element. However, the impedance of the flux lock type HTSC-FCL has started to decrease since the current of coil 3 exceeded one of coil 2 after a fault accident. The decrease in the impedance of the FCL causes the line current to increase and, if continues, the capacitor for the series resonance to be destructed. To avoid this operation, the flux-lock type HTSC-FCL requires an additional device such as fault current interrupter or control circuit for magnetic field. From the experimental results, we investigated the parameter range where the operation as mentioned above for the designed flux-lock type HTSC-FCL using series resonance occurred.

횡동요에 기인하는 전복에 대한 1-자유도계 모형의 이론해석 (Theoretical Analysis at One Degree-at-Freedom Model for Rolling at Ships with Focus on Capsize)

  • Recent studies have shown that the short time solution of the equation of motion for the rolling of ships is important in deciding the possibility of capsize of ships due to the excessive heel. Since most of known solutions for nonlinear equations of motion are long time or steady periodic solutions, here a simple way is described to get the short time solutions of the Duffing equation, which was chosen for deriving a criterion for the capsize of the ship. With the small external rolling moment, we first assume the state of the small damping and near resonance. Then, for cases when the frequency of the external moment is higher than the resonant one, an inequality was derived as a criterion for the capsize. This gives the range of the initial condition and the magnitude of the external moment which should be avoided for a ship to be safe from capsize. Furthermore, from the linearized equation, it is also shown that a simple and self-explanatory solution can be obtained consistent with that for the case of no damping, which yields the well-known linear growth with time.

  • Resonance fluorescence is the manifestation of the interaction between the physical system under consideration and the vacuum-field fluctuation. The fluorescence spectrum provides such physical informations as the energy-level structure of the system, instabilities and relative populations of the energy levels, etc.. One of the typical fluorescence spectra is the Mollow triplet appearing when two-level atoms are driven by a strong coherent field in free space$^{(1)}$ . In the weak field limit, the singlet instead of the triplet is obtained with a reduced linewidth due to the squeezing of one quadrature phase of the induced atomic dipole$^{(2)}$ . On the other hand, when the atoms are put inside a cavity rather than in free space, a doublet spectrum due to the vacuum Rabi-splitting is achieved, showing clearly the coupling of atoms and the cavity in the single-quantum limit$^{(3)}$ .   • This paper describes a type of piezoelectric transformer with improved design principles. It consists of a thin rectangular piezoelectric ceramic plate having two input electrodes and one output electrode. The length of each input and output electrode is nearly one third of that of piezoelectric transformer itself. In the driving section, the ceramic plate is poled in the thickness direction reversely with each other, and has electrodes on both main faces. The electric input near its fundamental resonance frequency is applied to the driving section in parallel and the output voltage from the generating section is connected to the resistor load. Its equivalent circuit is derived from the Mason\`s model. The frequency characteristics near the resonance frequency under no load have been investigated. Moreover, using this piezoelectric transformer we measured the LCD backlight characteristics.

풍력발전기 드라이브트레인 공진 해석 (Analysis of Resonance for Drive-train in Wind Turbine)

  • This study investigated the problems in current practice of drive-train resonance analysis procedure and suggested solutions. The first problem is the resonance occurrence at the un-identified resonance point by the current practice, as for a solution the force spectrum analysis for each critical force transmitting component was suggested. The second one is the inaccurate estimation of potential resonance point in eigenfrequency analysis because of the non-consideration about the eigenfrequency dependency on rotor-speed, the fine linearization at each rotor speed point all over operational range was proposed to account for the affection. Lastly the insufficient time for resonance activation under run-up simulation condition was recognized as a problem in resonance load increasing analysis, as an alternative, steady state condition was suggested to estimate the maximum load increasing level.
