한국산 1미기록종, Acanthaphritis unoorum (농어목, 꼬리점눈퉁이과)의 어란 및 자치어의 분자동정 및 형태기재 (Molecular Identification and Morphological Descriptions of the Eggs, Larvae and Juvenile of the Previously Unrecorded Species Acanthaphritis unoorum (Perciformes, Percophidae) in Korean Waters)

  • We presented detailed morphological descriptions of the eggs, larvae and juvenile of Acanthaphritis unoorum based on specimens collected with bongo nets from Korean waters during the period May 2017-July 2018. We collected 18 individuals including eggs (n= 4, 0.77-0.85 mm in egg diameter), preflexion larvae (n= 6, 4.11-6.31 mm in standard length, SL), flexion larvae (n= 4, 6.60-7.82 mm SL), postflexion larvae (n= 3, 8.94-13.46 mm SL), and one juvenile (n= 1, 14.67 mm SL). The mitochondrial (mt) DNA 16S rRNA sequences of the eggs, and the cytochrome c oxidase subunit I (COI) sequences of the larvae were identical to those of A. unoorum adults (genetic distances <0.01). The A. unoorum larvae and the juvenile that we collected were morphologically similar to those of Dactylopsaron dimorphicum, but the A. unoorum specimens were readily distinguishable by the presence of lateral melanophores. This is the first record of A. unoorum in Korean waters. We propose a new Korean name for A. unoorum: "O-ri-bu-ri-nuntung-i".

A Rapid and Universal Direct PCR Method for Macrofungi

  • Macrofungi are valuable resources as novel drug candidates, new biomaterials, and edible materials. Recently, genetic approaches pertaining to macrofungi have been continuously growing for their identification, molecular breeding, and genetic engineering. However, purification and amplification of fungal DNA is challenging because of the rigid cell wall and presence of PCR inhibitory metabolites. Here, we established a direct PCR method to provide a rapid and efficient method for PCR-grade macrofungal DNA preparation applicable to both conventional PCR and real-time PCR. We first optimized the procedure of lysis and PCR using the mycelia of Lentinula edodes, one of the most widely consumed macrofungal species. Lysates prepared by neutralizing with (NH4)2SO4 after heating the mycelia in a mixture of TE buffer and KOH at 65℃ for 10 min showed successful amplification in both conventional and real-time PCR. Moreover, the addition of bovine serum albumin to the PCR mixture enhanced the amplification in conventional PCR. Using this method, we successfully amplified not only internal transcribed spacer fragments but also low-copy genes ranging in length from 500 to 3,000 bp. Next, we applied this method to 62 different species (54 genera) of macrofungi, including edible mushrooms, such as Pleurotus ostreatus, and medicinal mushrooms such as Cordyceps militaris. It was found that our method is widely applicable to both ascomycetes and basidiomycetes. We expect that our method will contribute to accelerating PCR-based approaches, such as molecular identification, DNA marker typing, gene cloning, and transformant screening, in macrofungal studies.

  • Objective: Men's sexual health plays an important role in male fertility and childbearing, as it is associated with factors such as sexual desire, healthy spermatogenesis, and erectile function. In various cultures, medicinal plants have been utilized to address male sexual issues, including infertility and erectile dysfunction. Despite recent advancements in medical science for treating male impotence, some men opt for herbal supplements as an alternative, given that numerous herbs have the potential to enhance male sexual performance. The Apiaceae family is one of the oldest plant families used for medicinal purposes. Ferula, a genus within this family, comprises approximately 170 different species worldwide. Members of this genus possess numerous therapeutic properties due to the presence of various compounds. This article aims to explore the potential impacts of Ferula plants on the male reproductive system. Methods: This review article was prepared by searching for terms including Ferula and "aphrodisiac," Ferula and "spermatogenesis," and Ferula and "male reproductive system." Relevant information was gathered through electronic databases, including ISI Web of Knowledge, PubMed, and Google Scholar. Results: The findings indicated that relatively comprehensive studies have been conducted in this area, revealing that certain Ferula species have been employed in folk medicine to boost fertility and libido. Recent research has corroborated these effects. Conclusion: It is hoped that new aphrodisiac compounds with fewer side effects can be isolated from Ferula plants in the future.

Alpha-Glucosidase Inhibitory Activity of Saponins Isolated from Vernonia gratiosa Hance

  • Species belonging to the Vernonia (Asteraceae), the largest genus in the tribe Vernonieae (consisting of about 1,000 species), are widely used in food and medicine. These plants are rich sources of bioactive sesquiterpene lactones and steroid saponins, likely including many as yet undiscovered chemical components. A phytochemical investigation resulted in the separation of three new stigmastane-type steroidal saponins (1 - 3), designated as vernogratiosides A-C, from whole plants of V. gratiosa. Their structures were elucidated based on infrared spectroscopy (IR), one-dimensional (1D) and two-dimensional nuclear magnetic resonance (2D NMR), high-resolution electrospray ionization mass spectrometry (HR-ESI-MS), and electronic circular dichroism analyses (ECD), as well as chemical reactivity. Molecular docking analysis of representative saponins with α-glucosidase inhibitory activity was performed. Additionally, the intended substances were tested for their ability to inhibit α-glucosidase activity in a laboratory setting. The results suggested that stigmastane-type steroidal saponins from V. gratiosa are promising candidate antidiabetic agents.

납자루아과(Pisces: Acheilognathinae) 어류 3종의 숙주조개에 대한 산란양상 및 재첩(Corbicula fluminae) 내 산란 국내 최초 보고 (Spawning patterns of three bitterling fish species (Pisces: Acheilognathinae) in host mussels and the first report of their spawning in Asian clam(Corbicula fluminae) from Korea)

  • 본 연구는 낙동강 지류인 고령 회천 유역의 납자루아과어류 3종(줄납자루, 낙동납자루, 각시붕어)을 대상으로 숙주조개 종에 대한 산란양상을 조사하고 재첩 내 산란한 종을 식별하였다. 현장에서 채집된 총 200개 조개 중 납자루아과 어류 산란율은 말조개(45.2%)와 작은대칭이(45.5%)가 유사하였으나, 재첩은 12.1%로 낮게 나타났다. 재첩을 산란조개로 이용한 납자루아과 어류는 줄납자루와 낙동납자루로 확인되었다. 말조개에서는 납자루아과 3종 모두 산란이 확인되었으며 작은대칭이에선 줄납자루만 산란이 확인되었다. 기존 문헌에 의하면, 재첩은 말조개류보다 크기가 작고 아가미 구조가 달라 납자루아과 어류의 난 및 치어가 생존에 불리하다고 보고된 바 있다. 이러한 연구 결과들을 토대로 재첩과는 산란숙주조개로서 이용될 가능성이 없는 것으로 알려졌지만 재첩 내 납자루아과 어류 산란이 최근 중국에서 보고된 바 있고, 본 연구를 통해 재첩내 납자루아과 산란이 국내 최초로 확인되었다. 이는 재첩이 납자루아과의 산란숙주조개로 이용되었다는 것을 의미 하지만, 산란 후 발생과정을 거쳐 유영능력을 획득하여 조개 밖으로 이동해 독립된 생활사를 지속하는지에 대한 여부는 추후 확인이 필요하다. 재첩 내 산란이 산란초기에 나타나는 현상이거나 높은 경쟁관계와 같은 생물학적 환경요인에 의한 현상일수도 있는점을 고려해 추후 5, 6월 시기에 조사가 필요하다. 또한 낙동강수계의 다른 하천을 대상으로 추가적인 연구를 수행하여 납자루아과 어류의 재첩에 대한 산란양상 파악을 보다 면밀히 구명할 필요가 있다. 말조개 33개체 중 19개체(57.6%)에서 납자루아과 어류 2종 이상의 난이 관찰되었고, 이러한 결과는 산란장을 차지하기 위한 납자루아과 어류 종간경쟁의 강도가 매우 높다는 것을 의미한다. 본 연구는 낙동강에 서식하는 납자루아과 담수어류의 산란양상을 보다 정확히 규명하고 이들 자연개체군의 효과적인 유지, 관리 및 보전 방안 마련을 위한 생태, 유전적 기초 데이터를 제공할 것으로 기대한다.

명태(Gadus chalcogrammus) 수정란에서 신경괴사증바이러스(nervous necrosis virus) 모니터링 (Monitoring of nervous necrosis virus in fertilized eggs of walleye pollock (Gadus chalcogrammus))

  • 2017년 1월에서 4월까지 강원도 고성에서 사육중인 명태 친어로부터 수정란을 채집하여 신경괴사증바이러스(nervous necrosis virus, NNV)의 검출을 시도하였다. 매 회 50 mg씩 수정란을 채집하여 이를 1 set로 간주하였으며 총 37 set를 제작하였다. RNA를 추출하고 cDNA를 합성하여 NNV를 대상으로 reverse transcriptase PCR을 실시하였다. 그 결과, one-step PCR법으로는 37 set의 시료가 모두 음성이었으며, two-step PCR법으로는 5.4% (2/37)의 시료가 양성을 나타내었다. 그러나, band의 농도가 매우 낮아 시퀀싱은 불가능하였다. 본 연구의 결과 및 이전 연구의 결과로부터 현재 국내에서 양식하고 있는 명태에서 NNV 감염에 의한 폐사는 발생하지 않는 것으로 추정된다. 하지만, 지속적인 모니터링 및 양성 개체 출현 시 바이러스 분리의 시도 등은 필요할 것으로 생각된다.

Microbiome Study of Initial Gut Microbiota from Newborn Infants to Children Reveals that Diet Determines Its Compositional Development

  • To understand the formation of initial gut microbiota, three initial fecal samples were collected from two groups of two breast milk-fed (BM1) and seven formula milk-fed (FM1) infants, and the compositional changes in gut microbiota were determined using metagenomics. Compositional change analysis during week one showed that Bifidobacterium increased from the first to the third fecal samples in the BM1 group (1.3% to 35.1%), while Klebsiella and Serratia were detected in the third fecal sample of the FM1 group (4.4% and 34.2%, respectively), suggesting the beneficial effect of breast milk intake. To further understand the compositional changes during progression from infancy to childhood (i.e., from three weeks to five years of age), additional fecal samples were collected from four groups of two breast milk-fed infants (BM2), one formula milk-fed toddler (FM2), three weaning food-fed toddlers (WF), and three solid food-fed children (SF). Subsequent compositional change analysis and principal coordinates analysis (PCoA) revealed that the composition of the gut microbiota changed from an infant-like composition to an adult-like one in conjunction with dietary changes. Interestingly, overall gut microbiota composition analyses during the period of progression from infancy to childhood suggested increasing complexity of gut microbiota as well as emergence of a new species of bacteria capable of digesting complex carbohydrates in WF and SF groups, substantiating that diet type is a key factor in determining the composition of gut microbiota. Consequently, this study may be useful as a guide to understanding the development of initial gut microbiota based on diet.

Age and Growth of the Mudskipper, Scartelaos gigas(Perciformes, Gobiidae) from Korea

  • Age and growth of the mudskipper, Scartelaos gigas were investigated using the second actinost bone of the pectoral girdle, based on an analysis of 560 individuals collected from the mud flats in south west Korea. Specimens were collected semimonthly from March to September of 2003. Actinost and ring radii of each ring group showed a direct one-to-one relationship, with ring radius increasing in tandem with actinost radius. Monthly change in the marginal indices showed that ring formation occurred between May and July(mainly June), being supported by the monthly change in the gonadosomatic(highest in June) and hepatosomatic(lowest in July) indices. Because the species has not been found since November, when air temperature was less than about $10^{\circ}C$, it is thought to enter to hibernation. Therefore, it was suggested that the two new actinost rings may appear a year: one during the spawning season(May-July) and another during hibernation(since November). Although the growth of S. gigas must be limited to short periods each year from March to April and from August to October, its growth rate was considerably fast. The von Bertalanffy growth parameters of S. gigas were $TL_{\infty}$=179.36 mm, K=0.78 $year^{-1}$, $t_0$=-0.7762, and age-length key were $TL_1$=134.3 mm, $TL_2$=158.6 mm, $TL_3$=169.1 mm, $TL_4$=175.0 mm.

Characteristics of Potential Gamma-Aminobutyric Acid-Producing Bacteria Isolated from Korean and Vietnamese Fermented Fish Products

  • Gamma-aminobutyric acid (GABA) is a neurotransmitter that exerts several physiological functions and positive effects on human health. The aim of this study was to isolate and characterize the strains that had GABA-producing abilities from various fermented fish products. A total of 91 acid-producing strains were isolated from 41 samples of fermented fish products, and 27 strains showing GABA-producing abilities were identified by the 16S rDNA sequences. Among the strains, 31% strains tolerated at high-salt environment of 10-20% throughout the fermentation of fish sauces. The 27 isolates that produced GABA at various concentrations did so in the range of 5 to 454 mM. These GABA-producing isolates were identified as lactic acid bacteria of 14 strains, which included twelve Lactococcus lactis, one Enterococcus faecium, and one Lactococcus pentosus; eight Bacillus cereus group, which included seven B. thuringiensis and one B. cereus; and five Staphylococcus spp. Interestingly, with Vietnamese fish sauces, we mostly identified species of B. thuringiensis and Staphylococcus spp., while with Korean fermented fish products, the majority of the strains identified belonged to L. lactis. Among the strains, B. thuringiensis LH2134 produced the highest levels of GABA at 366 mM among the strains identified from Vietnamese fish sauces, whereas L. lactis LA43, a new strain isolated from Korean jeotgal (salted shrimp paste), produced the highest amount of GABA at 454 mM and the glutamate concentration in the medium was essential for GABA accumulation. Therefore, such the isolates might serve as good starters for development of more GABA-reinforced foods among fermented fish products.

두릅나무 순의 Saponin에 관한 연구 (II) - Saponin 의 동정 - (Studies on the Saponins in the Shoot of Aralia Elata (II) -Identification of the Saponins-)

  • 두릅나무 순의 saponin을 추출하여 그 구조를 밝혀 본 결과는 다음과 같다. 1. 자연산 날 것 식용적기의 crude saponin에서 aglycon으로 oleanolic acid와 hederagenin외에 1,3-methylenedioxy-3-dehydroxyoleanolic acid를 동정하였으며 당으로 ${\alpha},\;{\beta}-glucose$와 arabinose, rhamnose를 확인하였다. 2. 자연산 날 것 식용적기에 가장 많은 ‘b’ saponin(elatoside $Fh_2$)을 분리하여 동정한 결과 그 구조는 3-0-$[{\alpha}-L-arabinopyranosyl(1{\rightarrow}2)-{\beta}-D-glucopyranosyl]$-28-O-${\beta}-D-glucopyranosyl$ oleanolic acid라 추정된다.
