무지개송어(Oncorhynchus mykiss)와 은연어(O. kisutch)간의 잡종 및 잡종 3배체 생산 (Production of Hybrid and Allotriploid between Rainbow Trout (Oncorhynchus mykiss) and Coho Salmon (O. kisutch))

  • 경제성이 높은 어종인 무지개송어와 은연어를 대상으로 두 종간의 장점을 갖춘 신품종을 개발하기 위하여 종간 잡종 및 잡종 3배체를 유도하였다. 그 결과, 잡종의 부화율은 $51.7\%$로 여타 대조군에 비해 낮았으나 잡종 3배체의 부화율은 $77.6\%$로 무지개송어 3배체와 유사한 부화율을 보였다. 또한 부화 2개월후의 생존율은 잡종인 경우 $1.7\%$로 매우 낮았으나 잡종 3배체는 $54.5\%$로 무지개송어 3배체의 $56.6\%$ 생존율과 유사하였다. 잡종의 세포 및 핵의 평균 크기는 양친으로 사용된 두 종의 중간을 보였으며 잡종 3배체의 세포 및 핵의 크기는 잡종에 비해 크게 나타났다. 잡종의 염색체수는 60개로 나타나 n=30의 염색체수를 가진 무지개송어 난자와 n= 30의 염색체수를 가진 은연어 정자와 교배된 개체만이 생존력을 갖는 것으로 나타났다. 잡종 3배체의 경우 그의 염색 체수는 $90\~93$개로 Robertsonian형의 전좌에 의한 염색체 다형현상을 보여주었다.

해수순치에 따른 무지개송어 (Oncorhynchus mykiss)의 프로락틴 및 성장호르몬 유전자의 발현 변화 (Changes in Prolactin and Growth Hormone Gene Expression of Rainbow Trout Oncorhynchus mykiss Adapted to Seawater)

  • Prolactin (PRL) plays an important role in freshwater (FW) osmoregulation by preventing the loss of ions and the uptake of water in fish. Growth hormone (GH) promotes acclimation to seawater (SW) in several teleosts. We acclimated rainbow trout Oncorhynchus mykiss weighting $68.2{\pm}16.6$, $138.3{\pm}24$, and $287.5{\pm}42.1$ g in separate experiments to SW under slow-acclimation (SSW) or acute-acclimation (ASW) conditions, and then examined the PRL and GH mRNA levels using the real-time quantitative polymerase chain reaction. The PRL mRNA levels in all three experimental groups decreased significantly with both the SSW and ASW treatments, as compared to a control group kept in FW for 30 days. The GH mRNA levels increased with ASW in the largest fish, whereas the levels in the other groups did not change significantly. The mortality rate of the largest fish was lower than for the other groups, whereas the growth rate among the three experimental groups did not differ significantly. The growth rate of the ASW group was highest for the smallest fish. These results suggest that SW acclimation is associated with the gene expression levels of PRL and GH in relatively large rainbow trout. In addition, the fish mortality and growth rate on FW-SW transfer seem to be related to body weight, and the SW acclimation method may be applied to the hatcheries industry.

Dietary inclusion of mealworm (Tenebrio molitor) meal as an alternative protein source in practical diets for rainbow trout (Oncorhynchus mykiss) fry

  • Background: An 8-week feeding trial was designed to evaluate the potential of yellow mealworm (MW; Tenebrio molitor) as a locally available nutrient-rich feedstuff for rainbow trout fry (Oncorhynchus mykiss). Methods: Triplicate groups of fish (mean ± SE; 1.11 ± 0.01 g) were assigned to each of the five isonitrogenous and isocaloric practical diets containing graded level of a full fat MW (0, 7, 14, 21, and 28%) at the expense of fish meal (designated as MW0, MW7, MW14, MW21, and MW28, respectively). Results: Fish growth performance in terms of weight gain and specific growth rate significantly increased with increasing dietary MW level up to 14% and then declined when dietary MW levels further increased to 28%. Significantly higher protein efficiency ratio and lower feed conversion ratio were found in fish fed with diets containing MW compared to fish fed the control MW0. Myeloperoxidase activity was significantly higher in fish fed MW7 diet compared to fish fed the MW0 diet. Fish fed the MW14 and MW28 diets had significantly higher lysozyme activities than those fed the MW0 diet. Conclusions: Overall, the efficacy of MW as promising alternative to fish meal in practical diets for rainbow trout fry has been proved not only in relation to growth rates and feed utilization, but also from the viewpoint of immunopotentiation effects.

해수산 무지개송어(Oncorhynchus mykiss) 가공부산물(프레임육)의 식품학적 품질 특성 조사 (Investigation of Food Quality Characterization of Processing By-product (Frame Muscle) from the Sea Rainbow Trout Oncorhynchus mykiss)

  • This study compared the meat quality of sea rainbow trout Oncorhynchus mykiss frame muscle (S-FR-TY) and fillet (S-FI-TY) with the frame muscle of freshwater rainbow trout (F-FR). There was a difference of < 1% in the proximate composition of S-FR-TY vs. S-FI-TY, and of S-FR-TY vs. F-FR. The Hunter a value of F-FR-TY was lower compared with that of F-FR cultured in Jecheon (F-FR-JC), but higher than that of F-FR cultured in Pyeongtaek (F-FR-PT). However, no difference in the Hunter a value of S-FR-TY compared with any other F-FR was observed. The odor intensity of S-FR-TY was lower compared with that of F-FR, while the taste of S-FR-TY was milder than that of S-FI-TY, but stronger than that of F-FR. Although its total amino acid content was lower, the essential amino acid content of S-FR-TY was higher compared with S-FI-TY; the potassium content of S-FR-TY was also significantly greater. S-FR-TY contained high levels of nutritional and functional components, such as anserine, lysine, threonine, and eicosapentaenoic and docosahexaenoic acid. These results suggest that S-FR-TY could be used as a resource of canned food or fish jerky for children.

무지개 송어(Oncorhynchus mykiss)의 영양 성분에 대한 astaxanthin 첨가사료의 영향 (Effect of an Astaxanthin-supplemented Diet on the Nutritional Composition of Rainbow Trout (Oncorhynchus mykiss))

  • This study investigated the effects of astaxanthin, which is used to improve the muscle color of fish, on the nutritional composition of rainbow trout (Oncorhynchus mykiss). The trout were fed extruded pellets containing astaxanthin (pigmented rainbow trout) or without astaxanthin (non-pigmented rainbow trout). No significant differences in the contents of crude protein and crude ash between the two muscles of the two groups were observed. However, the crude fat composition of the muscle of pigmented rainbow trout was about two times higher than that of control muscle. In the muscle of pigmented rainbow trout, the contents of docosahexaenoic acid (DHA, 22:6n-3) and eicosapentaenoic acid (EPA, 20:5n-3), which are functional polyunsaturated fatty acids, were 1.6 fold higher than in non-pigmented muscle. In addition, the contents of minerals such as Zn and Fe, vitamins (B group, C, and E), free amino acids, carotenoids and astaxanthin were increased in the muscle of rainbow trout fed pellets supplemented with astaxanthin. Specifically, the content of the bioactive compound astaxanthin, was increased six times in the pigmented muscle, as compared to the control muscle. The results suggest that a diet supplemented with astaxanthin improves the nutritional composition of rainbow trout muscle as well as improving the muscle color.

해수순치 무지개송어 (Oncorhynchus mykiss)의 Vibrio anguillarum 감염증 (Vibrio anguillarum infection in rainbow trout Oncorhynchus mykiss during seawater adaption)

  • 본 연구에서는 2013년 4월 제주 소재 육상수조에서 무지개송어 해수순치 과정중에 대량폐사가 발생하여 질병검사를 실시하였다. 병어의 대부분은 체표에 궤양이 형성되어 있었고, 비장 비대, 유문수 부위 및 복강 근육의 출혈 및 위속에 액성물질이 가득 차 있었다. 병원체 검사결과, 기생충, 진균, 바이러스는 검출되지 않았지만 검사 개체 5마리에서 간, 비장 및 신장조직에서 동일한 형태의 세균 colony가 시료 당 200개 이상 분리되었다. 분리세균의 16S rDNA 염기서열 분석 결과, Vibrio anguillarum과 100% (875/875 bp) 일치하였다. 이상의 결과, 무지개송어 해수 순치과정 중에 발생된 질병은 Vibrio anguillarum 감염증으로 사료되었다.

대두박 사료에 대한 무지개송어(Oncorhynchus mykiss)의 소화 특성 (Digestive Characteristics of Rainbow Trout Oncorhynchus mykiss on Soybean Meal Based Diets)

  • To evaluate the digestive characteristics and bioavailability of dietary soybean meal (SBM), the post-prandial changes in the gastric contents of rainbow trout Oncorhynchus mykiss were determined for 24 h after feeding of SBM diets. A curve estimation of regression diagnostics using a comparison of the adjusted $r^2$ and probability was performed to test the tendency of the post-prandial changes and gave a quadratic polynomial (exponential) regression for all experimental groups. The gastric evacuations rates (GER) for higher-SBM groups were slower than those for lower-SBM groups. The estimated GER (digestion time) for 75% gastric content for fish fed a 70% SBM diet was 1.63 times longer than that for fish fed the control diet. Despite the fact that the pH values in the gastric contents rose from 4.05 at 0 h to 5.12-5.38 at 1 h after feeding, then dropped to 4.57-4.83 at 9 h, with no significant differences among experimental groups, the gastric moisture contents increased significantly in the higher-SBM groups. This is most likely due to an increase in digestive juices in the higher-SBM groups, rather than water introduced externally. The percentage of soluble nitrogen in the gastric contents of fish fed the higher-SBM diets was higher than that in the fish fed the control diet, and the SBRs (stomach weight/body weight${\times}100$) in the higher-SBM groups (diets 4, 5, 6 and 7) were also significantly higher than the SBR of the control group (P<0.05). This may indicate that the protein in SBM can be digested slowly due to physiological digestive adaptation in rainbow trout.

무지개송어의 해수순치과정에 일어나는 삼투조절세포의 미세구조 (Ultrastructural Change of Osmoregulatory Cells during Seawater Adaptation in Rainbow Trout (Oncorhynchus mykiss))

  • 양어장에서 염분농도가 각각 $5^{\circ}/_{\circ\circ}$, $10^{\circ}/_{\circ\circ}$, $15^{\circ}/_{\circ\circ}$$1.5m^3$인 사육조에서 순치 적응된 평균 체중이 130g인 smolt 단계의 치어와 870g인 성어의 무지개송어 (Oncorhynchus mykiss) 총 150마리를 공시어로 이용하여 아가미 및 신장의 미세구조의 변화를 알기 위해서 광학 및 전자현미경으로 관찰하여 얻어진 결과는 다음과 같다. 해수에 순치 적응된 무지개송어에서 아가미엽의 비대와 아가미엽 상피세포의 분리 그리고 신사구체의 수축, 신장의 응혈 등의 조직학적 변화가 나타났다. 해수에 순치 적응된 개체의 삼투압은 당수에 서식하는 개체의 삼투압보다 높게 나타나서 평균 300 m Osmol 이상으로 나타났다. 삼투압의 증가는 염분과 무기질의 배출효율을 높이기 위해서 혹은 체내의 삼투압의 균형을 맞추기 위해서 삼투압이 증가하는 것으로 사료된다. 아가미엽의 염세포가 증가하는 발생정도는 염분농도의 증가와 더불어 높게 나타났고, 아가미엽 표면은 염분농도가 높아질수록 요철상태가 심한 구조로 나타났으며, 길게 변형된 미토콘드리아가 관찰되었다. 무지개송어는 조직병리학적 변화에도 불구하고 해수에 순치 적응하는 것으로 나타났다.

  • 무지개송어, Oncorhynchus mykiss 와 산천어, O. masou 간 잡종 및 잡종 3배체가 유도되었다. 잡종 3배체의 3배체 유도율은 92%로 유도된 잡종 3배체의 초기 생존율은 잡종에 비해 높게 나타났다. 잡종의 염색체수는 2n=63 이었으며 잡종 3배체의 염색체수는 3n=93으로 나타났다. 잡종은 적혈구 세포 및 핵 크기에서 잡종유도시 사용된 무지개송어, 산천어의 적혈구 세포 및 핵 크기의 중간치를 나타냈으며 잡종 3배체는 잡종에 비해 배수화에 기인되어 큰 적혈구 세포 및 핵 크기를 나타내었다. 이러한 배수화는 인형성부위 수에서도 나타나 잡종은 1개 혹은 2개의 인형성부위를 보인 반면 잡종 3배체는 1개, 2개 혹은 3개의 인형성부위를 나타내었다.

사료 과다공급으로 유도된 비만 무지개송어(Oncorhynchus mykiss)의 생리·조직학적 변화 (Physiological and Histological Changes of Overfeeding-induced Obese Rainbow Trout Oncorhynchus mykiss)

  • Obesity could cause immune-physiological disorders in fish. Yet, little is known about the impact of obesity on stress and histological responses. This study aimed to determine histological and physiological changes in and vulnerability of overfeeding-induced obese rainbow trout Oncorhynchus mykiss exposed to stress condition. Control, intermediate and overfed groups were fed at 1.5, 2.5 and 3.09% of their body weight per day, respectively, for eight weeks. Weight gain, body mass index, hepatosomatic index and serological parameters, and histology of liver were measured in five fish from each group at week 0, 2, 4, and 8. At week 8, 20 fish from each group were exposed to heat stress by increasing water temperature at a rate of 3℃ per day from 15 to 25℃ and maintaining the final temperature for 10 days. Overall, overfed fish showed significantly higher weight gain, body mass index, and serological parameters than those of fish in the other groups. Fish in the overfed and intermediate groups displayed multifocal infiltration of inflammatory cells in hepatic parenchyma. Mortality rate and serological parameters of fish in the overfed group exposed to heat stress were significantly higher than those of fish in the other groups, indicating increased vulnerability to environmental stress.
