Hydrothermal extraction from dates was done, and then 3 groups of the date extract liquid group (A), the date puree group (B) and the jujube skin flesh group (C) were prepared. And, the omija extract liquid was added to the 3 groups at respective rates of 3:7, 5:5 and 7:3. Then, total 9 medicated diet sources were prepared by adding cinnamon, the ginger extract liquid, honey and pectin. And their physicochenmical and sensory attributes were examined. Total sugar, free sugar and pH increased in the samples of the 3 groups as the addition amount of the jujube extract increased. In chromaticity of respective samples, the "L" value did not show any difference among the 3 groups, but the "a" value was the highest in the "A"group, and "b"value was the highest in the "B"group. In the date puree group and the jujube skin flesh group, their viscosity increased as the addition amount of the jujube extract increased, but in the jujube extract liquid group, no difference was found in its viscosity. As a result of conducting the discrimination test, it was identified that the as the addition amount of the jujube extract was increased, color and sweet taste were feltto be stronger and our taste was felt to be weaker in all 3 groups. In the jujube puree group and the jujube skin flesh group, as the addition amount of the jujube extract increased, the jujube fragrance was felt to be stronger, but in the jujube extract liquid group, no difference was found in its fragrance. And, no difference was found in brightness and viscosity between samples. As a result of conducting the palatability test, no difference was showed in the appearance, but as for the overall palatability including texture, taste and fragrance, preference increased as the addition amount of the jujube extract increased.
This study intended to compare the effects of the sugar-Omija extract tea (SO) and LuoHanGuo-Omija extract tea (LO) to blood glucose levels and metabolism among streptozotocin (STZ)-induced diabetic mice and to prove the positive effects of LuoHanGuo extract as a sugar substitute. According to the results, the rate of blood glucose increase and the blood glucose level was reduced, and the weight was increased. Aspartate aminotransferase (AST), alanine aminotransferase (ALT), triglyceride, low density lipoprotein cholesterol, and total cholesterol was decreased, and albumin, total protein, and high density lipoprotein cholesterol (HDL-C) was increased in the STZ/LO group as compared to the STZ group. Compared to the STZ/SO group, the rate of blood glucose increase and glucose tolerance was decreased. AST, ALT, and total cholesterol was decreased, and HDL-C level was high. In conclusion, the LO displayed hypoglycemic effect, prevented weight loss caused by diabetes, recovered liver function, and improved lipid metabolism. In addition, such positive effects were obviously shown during comparison with the experimental group treated with the SO. Therefore, the LO were considered as favorable food supplements that can be safely taken by persons with diabetes, obesity, hypertension, and heart disease; they have to control their sugar intake as well as their diet.
Omija seed oil was extracted from Omija seeds, a by-product of Omija, using an eco-friendly pressed extraction method. Physicochemical properties and antioxidant activities of 12 extracts using different combinations of seed moisture content (5, 7.5, and 10%) and extraction temperature (25, 49, 75, and 100℃) were then investigated. The highest extraction yield was 31.33% at moisture of 5% and temperature of 75℃. The lowest acid value was 4.18±0.25 at moisture of 5% and temperature of 49℃or moisture of 7.5% and temperature of 25℃. Peroxide value, iodine value, and saponification value were the lowest at 0.64±0.56 meq/kg, 159.38± 6.03, and 57.60±9.40, respectively, at moisture of 5% and temperature of 25℃. The content of total polyphenolics was the highest at 4,413±125 mg TAE/100 g when the moisture content was 10% and the extraction temperature was 25℃. DPPH radical scavenging activities of oil extracts at 20~100 mg/mL were 28.68±7.30~87.65±2.20%. The maximum antioxidant activity and IC50 were 87.65±2.20% and <20 mg/mL, respectively, for extract obtained at moisture of 10% and temperature at 100℃. As a result, the extraction yield, acid value, peroxide value, iodine value, saponification value, and so on were excellent at moisture content of 5% and extraction temperature of 25℃ or 49℃. However, the content of total polyphenolic compounds and antioxidant activity were the highest at moisture of 10% and extraction temperature of 100℃. In conclusion, extracting Omija seed oil from Omija seeds, a by-product of Omija, is effective with a pressed extraction method.
Grape products have been known to exert greater antioxidant and anti-obesity than anti-hyperglycemic effects in animals and humans. Omija is used as an ingredient in traditional medicine, and it is known to have an anti-hyperglycemic effect. We investigated whether the combined extracts of grape pomace and omija fruit (GE+OE) could reduce fat accumulation in adipose and hepatic tissues and provide beneficial effects against hyperglycemia and insulin resistance in type 2 diabetic mice. C57BL/KsJ-db/db mice were fed either a normal control diet or GE+OE (0.5% grape pomace extract and 0.05% omija fruit extract, w/w) for 7 weeks. GE+OE decreased plasma leptin and resistin levels while increasing adiponectin levels and reducing the total white adipose tissue weight. Furthermore, GE+OE lowered plasma free fatty acid (FFA), triglyceride, and total-cholesterol levels as well as hepatic FFA and cholesterol levels. Hepatic fatty acid synthase and glucose 6-phosphate dehydrogenase activities were decreased in the GE+OE group, whereas hepatic ${\beta}$-oxidation activity was increased. Furthermore, GE+OE supplementation not only reduced hyperglycemia and pancreatic ${\beta}$-cell failure but also lowered blood glycosylated hemoglobin and plasma insulin levels. The homeostasis model assessment of insulin resistance levels was also decreased and the decrease seems to be mediated by the lowered activities of hepatic glucose-6-phosphatase and phosphoenolpyruvate carboxykinases. The present data suggest that GE+OE may have the potential to reduce hyperglycemia, insulin resistance, and obesity in patients with type 2 diabetes.
The principal objective of this study was to conduct a physiological evaluation of Korean Ginseng pickles with added Chija and Omija pigment. Prior to the processing of 3 different colors of pickles, SOD-liked activity, hydroxyl radical scavenging activity, total phenolic acid contents, and lecithin oxidation inhibitory effects of Korean Ginseng, Chija and Omija water extracts were assessed. After processing the 3 colors of pickles, sensory evaluation and color values were conducted. SOD-liked activity of Korean Ginseng(42.58%) and Chija(41.88%) water extracts were similar to those of tocopherol(54.62%), but were significantly higher than those of Omija(29.01%). The hydroxyl radical scavenging activity of Ginseng water extract(87.85%) was similar to that of BHT(83.13%) and tocopherol(71.57%), but were significantly higher than those of Chija(68.01%) and Omija(37.15%). The total phenolic acid contents of Korean Ginseng, Chija, and Omija water extracts were measured at $1.01{\sim}1.66\;mg/m{\ell}$, levels similar to those of tocopherol($1.26\;mg/m{\ell}$) but significantly lower than that of BHT($3.89\;mg/m{\ell}$)(p<0.05). The lecithin oxidation inhibitory effects of Ginseng water extract(98.86%) was similar to that of BHT(92.82%) and tocopherol(89.13%), but was significantly higher than that of Chija(64.28%) and Omija (53.34%). With regard to the results of sensory evaluation for the 3 colors of Ginseng pickles, the color and overall quality of P2 were significantly higher than those of P1 and P3(p<0.05). With regard to luminance, P1 scored significantly higher than P2 and P3(p<0.05). The a value of P3 was significantly higher than that of P1 and P2, and the b values of P2 were significantly higher than those of P1 and P3.
Onion vinegar, which has an undesirable flavor and taste formed through alcohol and acetic acid fermentation, possesses additives that can improve sensory quality. Thus, the objective of this study was to present an optimized blending ratio using response surface methods for an onion vinegar beverage by adding Omija extracts. This study was performed to formulate an Omija-onion vinegar beverage (OOVB) and investigate its antioxidant properties and antimicrobiological effects. The experimental design was conducted using an optimal mixture model of response surface methodology which generated eighteen experimental trials with overall acceptance as the responses. According to the statistical analyses, OOVB demonstrated a ratio containing onion vinegar, water, brown sugar, apple extracts and Omija extracts of 10, 72.3, 4.4, 12.2 and 1.1 (weight ratio), respectively. The OOVB revealed desirable nutrition values (phenolics compounds 19.3 mg/100 g, total flavonoids 3.1 mg/100 g, quercetin 1.9 mg/100). The OOVB displayed antibacterial effects in Gram negative Enterobacter aerogenes, Escherichia coli, Salmonella typhimurium and Gram positive Staphylococcus aureus. The findings revealed that OOVB was 18% in DPPH radical inhibitionand 11% in superoxide dismutase-like activity thus, OOVB has nutritional value and good quality as well as potential biological activities for functional beverages.
This study was carried out to comparison of the contents of general and mineral components between the original sample and water extract in each part of omija. The content of Carbohydrate was highest in endocarps, and that the crude protein and crude lipid in seeds. The contents of K and Mn in the fruits, endocarps, and seeds were all higher than those of the other cations. The content of K and Zn in endocarps were three to four times as much as those of seeds. and the content of Na, Ca, and Cu in endocarps were 1.5 to 1.75 times of seeds. Mineral contents by water extract in each part were ordered as K, Mg, Ca, and Fe. As the percentage of each ion in water extract on the basis of original sample, Fe was the highest ratio of behavior, and Mn was lowest.
본 연구는 식빵의 주재료인 밀가루의 일부를 오미자청으로 발효시킨 쌀가루 sourdough로 대체하여 식빵을 제조하여 품질 특성과 기호도를 살펴봄으로써 적정 수준의 쌀가루 sourdough 첨가량을 제시하고자 시도되었다. 쌀가루 sourdough는 오미자청을 이용해 전통적 sourdough 제법을 이용하여, 쌀가루 sourdough를 0% (대조군), 15, 30, 45, 60% 첨가하여 식빵을 제조한 후 pH, 색도, 부피, 질감과 기호도를 측정하였고 SEM을 이용해 식빵의 단면을 촬영하였다. pH는 쌀가루 sourdough 첨가량이 증가할수록 유의적으로 감소하였으며, 색도는 쌀가루 sourdough 첨가량이 증가할수록 백색도, 녹색도 및 황색도 모두 유의적으로 감소하는 경향을 보였다(p<0.05). 부피는 대조군과 15%군 간에는 유의적인 차이가 없었으나 쌀가루 sourdough 첨가량이 증가할수록 유의적으로 낮아졌다(p<0.05). 경도는 쌀가루 sourdough 첨가량과 저장시간이 증가할수록 유의적으로 증가하였으나(p<0.05), 대조군, 15, 30%군 간에는 유의적인 차이가 없는 것으로 보아 쌀가루 sourdough는 노화 억제 효과가 있는 것으로 생각된다. 탄력성은 쌀가루 sourdough 첨가량이 증가할수록 유의적으로 감소하는 경향을 보였고(p<0.05) 대조군과 15%군간에는 유의적인 차이가 없는 것으로 보아 쌀가루 sourdough를 15%까지는 첨가하여도 밀가루만으로 제조한 식빵이 가지는 탄력성을 유지하는 것으로 생각된다. SEM으로 촬영한 결과 대조군과 15%군의 식빵 단면은 유사한 모양을 보였다. 기호도는 모든 항목에서 대조군, 15, 30%가 유의적(p<0.05)으로 높게 평가되었다. 이상의 결과를 종합하면 오미자청으로 발효시킨 쌀가루 sourdough 15% 첨가한 식빵은 대조군과 비교시 손색이 없는 품질을 기대할 수 있는 것으로 사료된다. 다만 오미자 발효액에 내재된 미생물은 수확 시기나 지역, 배합율, 발효온도, 숙성기간에 따라 sourdough의 품질에 변화를 일으켜 오미자청에 존재하는 미생물에 관한 보완 연구가 필요할 것으로 생각된다.
오미자 5 g에 100 mL의 세 가지 용매(아세톤, 에탄올, 메탄올)를 각각 가하여 추출하여 농축한 다음, DMSO 용매에 녹여 항산화활성과 항유전독성을 조사하였다. 그 결과 총페놀 함량에서 메탄올 추출물이 1183.3 mg/100 g으로 가장 높았다. 항산화활성을 50, 100, 250, 500, $1000\;{\mu}g/mL$ 농도에서 측정하였을 때 DPPH radical 소거능과 SOD 유사활성은 각각 농도에 의존하여 결과 값이 향상되었으며, DPPH radical 소거능에서 높은 $IC_{50}$값을 나타낸 것은 에탄올 추출물로 $1411.1\;{\mu}g/mL$을 나타내었고, SOD 유사활성에서 $IC_{50}$값은 오미자 메탄올 추출물과 에탄올 추출물이 $905.7\;{\mu}g/mL$, $970.3\;{\mu}g/mL$ 순으로 비슷한 활성을 나타내었다. 또한 comet assay에서는 오미자 추출물을 1, 5, 10, $50\;{\mu}g/mL$의 농도로 백혈구에 처리한 후 $H_2O_2\;200\;{\mu}M$의 농도로 처리하여 DNA 손상을 유도한 결과 손상된 $50\;{\mu}g/mL$ 농도에서 DNA tail 부분의 DNA 함량을 측정한 % tail DNA inhibition이 각각 AE(73.1%), ME(63.7%), EE(62.8%)로 $H_2O_2$ 처리 양성 대조구에 비해 유의적으로 증가하였다. 이는 오미자에 함유되어 있는 페놀 화합물 외에 lignan 화합물을 포함한 여러 가지 다른 생리활성 물질들의 항산화력으로 인해 DPPH radical 소거능과 SOD 유사활성, 항유전독성 활성이 상호 의존적이 지 않음을 나타내고 있다고 사료된다. 따라서 오미자의 항산화 활성과 항유전독성의 메카니즘적 상호관계를 규명하는 연구가 향후 수행되어져야 할 것이다. 이상의 결과에서 오미자의 용매에 따른 추출물들은 각각 항산화 활성과 항 유전독성이 있음을 알 수 있었고, 따라서 본 연구에서는 오미자의 천연 생리활성 물질로서의 활용 가능성을 규명할 수 있었다.
